The Effect of Pretreatment with Various Mutagens on Glycoconjugates of Plasma Membrane in HeLa Cells (HeLa 세포에서 원형질 막의 glycoconjugate에 대한 수종 돌연변이원의 전처리 효과)

  • The present investigation has been performed to elucidate the effect of pretreatment with low dose of ultraviolet radiation (UV), ethyl methansulfonate (EMS), and bleomycin (BLM) on cell survival and lectin-binding glycoconjugates of plasma membrane in HeLa cells treated with mutagen. The percentage of survival of cells pretreated with 1 mM EMS following treatment with 10 mM EMS was higher than that of cells treated with 10 mM EMS alone. Wheat germ agglutinin (WGA) staining intensity of cells pretreated with 1 mM EMS and subsequently treated with 10 mM EMS was stronger than that of cells treated with 10 mM EMS alone. But, succinylated wheat germ agglutinin (sWGA) staining intensity of cells pretreated with 1 mM EMS and subsequently treated with 10 mM EMS was similar to that of cells treated with 10 mM EMS alone. These results suggest that the acquired resistance to EMS is related to the glycoconjugates containing sialic acid of plasma membrane involved in multidrug resistance or adaptive response in HeLa cells.

Effect of β-Carotene and Vitamin C on Chlorophyll-Induced Photooxidation (클로로필의 광산화에 미치는 β-카로텐과 비타민 C의 영향)

  • Skin is continously exposed to ultraviolet (UV) radiation, the major cause of skin disorders including skin aging. Chlorophylls were well known as photosensitizer initiating subsequent chemical reactions such as photooxidative deterioration of cellular structures. This experiment was designed to elucidate the effects of $\beta$-carotene and ascorbic acid with chlorophylls on UVB-induced photooxidation in linoleic acid emulsion model system and skin homogenate of ICR mouse. In linoleic acid emulsion model system, the addition of chlorophyll and $\beta$-carotene accelerated the photooxidation, while high concentration of ascorbic acid prevented. The combination of chlorophylls, $\beta$-carotene and ascorbic acid, which concentrations are simplified from mustard leaf kimchi, prevented UVB-induced photooxidation. Although single treatment of $\beta$-caretene accelerated photooxidaiton, $\beta$-caretene acted as antioxidant in the combination with ascorbic acid. Similarly the addition of individual chlorophylls and $\beta$-carotene accelerated the UVB-induced photooxidation in skin homogenate of ICR mouse. 50 ppm of ascorbic acid did not show the any preventive effect, however 500 ppm of ascorbic acid effectively prevented the oxidation. Photooxidation was prevented in the combination of chlorophylls and $\beta$-carotene with 500 ppm of ascorbic acid and concentration rate of ascorbic acid plays an important role in the prevention of UVB-induced photooxidation.

  • The mutant strain M6 derived from Acetobacter sp. A9, which produces high levels of the bacterial cellulose derived by random mutagenesis with N-methyl-N'-nitro-N-nitrosoguanidine or UV treatment, was selected by a Hestrin and Schramm medium (HSB) plate assay. The characterization of the cellulose production was studied in flask culture to improve the productivity of bacterial cellulose by $Acetobacter$ sp. A9 and mutant strain M6. The yield of cellulose production was superior to mutant M6 than $Acetobacter$ sp. A9. Cellulose was produced 0.12 g $L^{-1}$ by $Acetobacter$ sp. A9 at HS medium and the mutant M6 produced the cellulose 6.95 g $L^{-1}$at HS medium. Strain M6 produced less amount of gluconic acid than A9, thus showing that cellulose production is negatively relted with the gluconic acid production.

Effects of Chlorophyll a on UVB-induced Cellular Responses and Type I pN Collagen Synthesis in vitro (클로로필 a가 UVB 유도성 산화적 스트레스와 matrix metalloproteinases (MMPs) 활성화 및 콜라겐 합성에 미치는 영향)

  • Several studies have demonstrated that chlorophyll has many beneficial properties, including anti-inflammatory, anti-tumor, and antioxidant properties. Chlorophyll has been also shown to have an excellent chemopreventive potential against skin tumor. Its preventive mechanism against skin tumor, however, has not been examined in detail. Furthermore, the effects of chlorophyll on UVB-induced cellular responses have not been investigated. Solar UV radiation, in particular its UVB component, is the primary cause of many adverse biological effects, which is responsible for the photoaging and skin cancer. We investigated the preventive effects of chlorophyll a on UVB-mediated responses in human immortalized HaCaT kerationocytes and normal human fibroblast. We found that treatment of chlorophyll a markedly inhibited UVB-induced generation of reactive oxygen species and lipid peroxidation. Chlorophyll a also prevented UVB-induced MMP-1 expression and MMP-2 activation and increased Type I pN collagen synthesis. These results suggest that chlorophyll a is a potent candidate for the prevention and treatment of UVB-induced skin cancer and photoaging.

Antioxidative Activity and Component Analysis of Broussonetia kazinoki SIEB Extracts (닥나무 추출물의 항산화 활성 및 성분 분석)

  • In this study, the antioxidative activities and component analysis of Broussonetia kazinoki SIEB (B. kazinoki). extracts were investigated. B. kazinoki extract showed the effective free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}=8.53{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) of the ethyl acetate fraction of B. kazinoki. extracts in the luminol-dependent $Fe^{3+}-EDTA/H_2O_2$ system was $1.69{\mu}g/mL$. The ethyl acetate fraction of B. kazinoki. extracts also exhibited more prominent cellular protective effects (${\tau}_{50}$, 183.3 min at $10{\mu}g/mL$) than that of typical antioxidant $\alpha$-tocopherol (${\tau}_{50}$ = 38.00 min) in the $^1O_2$-induced photohemolysis of human erythrocytes. Components of the ethyl acetate fraction obtained from B. kazinoki extracts were analyzed by TLC, HPLC chromatogram, LC/ESI-MS/MS and $^1H$-NMR. Consequently, Components, components were identified as the kazinol J of kazinol series and luteolin (2-(3,4-dihydroxyphenyl)-5,7-dihydroxy-4-chromenone) of flavonoid series having antioxidant activities. These results indicate that extract/ fraction of B. kazinoki can be used as antioxidants in biological systems, particularly skins exposed to UV radiation by quenching and/or scavenging $^1O_2$ and other ROS, and protecting cellular membranes against ROS. Thus, the extract/fraction of B. kazinoki could be applicable to new cosmeceuticals.

Anti-Wrinkle Effects of Korean Rice Wine Cake on Human Fibroblast (양조 부산물인 주박의 주름개선 효과)

  • Skin aging is related to genetic and environmental factors (e.g., gene mutation and UV radiation respectively). To develop a new anti-wrinkle cosmetic or functional food by using Korean rice wine cake, we examined the effects of Korean rice wine cake, a brewery byproduct, on antioxidant effect, collagen synthesis and expression of MMP-1. Interestingly, we found that Korean rice wine cake has the ability to promote scavenging activity of DPPH radical. We also found that the cell proliferation and synthesis of collagen in HS27 cells was increased by Korean rice wine cake in a concentration-dependent manner. However, elastase inhibitory activity was not changed. In addition, the expression of MMP-1 was inhibited by Korean rice wine cake in a concentration-dependent manner. All these results suggest that Korean rice wine cake can be effectively used for the prevention of wrinkles in human skin.

Cellular Protective Effect and Component Analysis of Euphorbia humifusa Extracts (땅빈대 추출물의 세포 보호 효과 및 성분 분석에 관한 연구)

  • In this study, the cellular protective effect, antioxidative property and component analysis of Euphorbia humifusa extracts were investigated. The ethyl acetate fraction ($3.68\;{\mu}g/mL$) and aglycone fraction ($3.15\;{\mu}g/mL$) of Euphorbia humifusa extract showed prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity ($FSC_{50}$). Reactive oxygen species (ROS) scavenging activity ($OSC_{50}$) of Euphorbia humifusa extract on ROS generated in $Fe^{3+}$-EDTA/$H_2O_2$ system was investigated using the luminol-dependent chemiluminescence assay. The ethyl acetate fraction ($0.43\;{\mu}g/mL$) and aglycone fraction ($0.35\;{\mu}g/mL$) of extract showed higher ROS scavenging activity than L-ascorbic acid ($1.50\;{\mu}g/mL$). The cellular protective effects of fractions of Euphorbia humifusa extract on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The ethyl acetate fraction and aglycone fraction of extract protected cellular membranes against ROS in a concentration dependent manner ($5{\sim}25\;{\mu}g/mL$), and was more effective than (+)-${\alpha}$-tocopherol, lipid peroxidation chain blocker. Aglycone fraction from Euphorbia humifusa extract showed 2 bands in TLC and 2 peaks in HPLC. In HPLC chromatogram of aglycone fraction, peak 1 and peak 2 were identified as quercetin and kaempferol, respectively. And these components are very effective as antioxidant. Thus, these results indicate that fractions of Euphorbia humifusa extracts can function as antioxidant in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. Fractions of Euphorbia humifusa extracts can be applicable to new functional cosmetics for antioxidant.

Sensitivity Analysis of Satellite BUV Ozone Profile Retrievals on Meteorological Parameter Errors (기상 입력장 오차에 대한 자외선 오존 프로파일 산출 알고리즘 민감도 분석)

  • The accurate radiative transfer model simulation is essential for an accurate ozone profile retrieval using optimal estimation from backscattered ultraviolet (BUV) measurement. The input parameters of the radiative transfer model are the main factors that determine the model accuracy. In particular, meteorological parameters such as temperature and surface pressure have a direct effect on simulating radiation spectrum as a component for calculating ozone absorption cross section and Rayleigh scattering. Hence, a sensitivity of UV ozone profile retrievals to these parameters has been investigated using radiative transfer model. The surface pressure shows an average error within 100 hPa in the daily / monthly climatological data based on the numerical weather prediction model, and the calculated ozone retrieval error is less than 0.2 DU for each layer. On the other hand, the temperature shows an error of 1-7K depending on the observation station and altitude for the same daily / monthly climatological data, and the calculated ozone retrieval error is about 4 DU for each layer. These results can help to understand the obtained vertical ozone information from satellite. In addition, they are expected to be used effectively in selecting the meteorological input data and establishing the system design direction in the process of applying the algorithm to satellite operation.

Simultaneous Identification/Determination System for Sildenafil, Homosildenafil, Vardenafil and Tadalafil as Adulterants in Foods (식품중 불법함유된 발기부전치료제 성분 검출을 위한 동시분석법 연구)

  • An easily available, simultaneous identification/determination procedure for sildenafil, homosildenafil, tadalafil, vardenafil in adulterated health related foods was established by using a combination of three different analytical methods; thin layer chromatography(TLC), liquied chromatography-mass spectrometry (LC/MS) and high-performance liquied chromatography (HPLC)/photo-diode-array detector. The sample solution for TLC was applied to silica gel 60 $F_{254}$ plates with ethylacetate/acetonitrile/25%ammonia (90:10:5) as a developing solvent. Spots were located under UV radiation at 254 nm and dragendolfs reagent. Mass spectra of the compounds by LC/MS were investigated with electrospray ionization (ESI) interface, under positive ion mode. The HPLC analysis was performed on a column of capcell pack $C_{18}$ (UG120, 4.6${\times}$250mm I.D. 5 ${\mu}$m)with 0.1% sodium 1-hexansulfonate (in 0.1% phosphoric acid)/acetnitrile (73:27) as a mobile phase, and effluent was minitored with a photo-diode-applied to commercial foods, Sildenafil content was inthe range of 0.4mg/g~360.9 mg/g from 7 out of 35 samples. Homosildenafil content was in the range of 2.2 mg/g~336.0 mg/g from 7 out of 35 samples. Tadalafil content was 429.3 mg/g, 9.6 mg/500 mg from 2 out of 35 samples. The procedure described here is available for the screening of sildenafil, homosildenafil, tadalafil, vardenafil.

Antioxidative and Antiaging Effects of Sorbus commixta Twig Extracts (마가목 가지 추출물의 항산화 및 항노화에 관한 연구)

  • In this study, the antioxidative effects and inhibitory effects on tyrosinase and elastase of Sorbus commixta (S. commixta) twig extracts were investigated. The aglycone fraction of S. commixta twig extract showed the prominent free radical (1,1-diphenyl-2-picrylhydrazyl, DPPH) scavenging activity($FSC_{50}$, $13{\mu}g/mL$). Reactive oxygen species (ROS) scavenging activities of S. commixta twig extracts on ROS generated in $Fe^{3+}-EDTA/H_2O_2$ system were investigated by the luminol-dependent chemiluminescence assay. The 50 % ethanol extract among extracts showed the most prominent ROS scavenging activity ($OSC_{50}$, $0.189{\mu}g/mL$). The cellular protective effects of extract/fractions of S. commixta twig on the rose-bengal sensitized photohemolysis of human erythrocytes were investigated. The 50 % ethanol extract and ethyl acetate fraction showed the cellular protective effects against ROS in a concentration dependent manner ($5{\sim}50{\mu}g/mL$). The inhibitory effect of S. commixta twig extract on tyrosinase was investigated to assess the whitening efficacy. The ethyl acetate ($IC_{50}$, $113.2{\mu}g/mL$) and aglycone fraction($IC_{50}$, $105.3{\mu}g/mL$) on tyrosinase showed more remarkable inhibitory effect than arbutin($IC_{50}$, $226.88{\mu}g/mL$), known as the whitening agent. The inhibitory effect of aglycone fraction ($IC_{50}$, $6.9{\mu}g/mL$) on elastase was simillar to quercetin($IC_{50}$, $6.1{\mu}g/mL$), flavonoid known as reference compound. These results indicate that extract/fractions of S. commixta twig can function as antioxidants in biological systems, particularly skin exposed to UV radiation by scavenging $^1O_2$ and other ROS, and protect cellular membranes against ROS. S. commixta twig extracts can be applicable to new functional cosmetics for anti-aging products.
