Matrix solid phase dispersion isolation and high performance liquid chromatographic determination of five benzimidazole anthelmintics in bovine muscle, liver and omasum (시료고체상분산처리와 액체크로마토그라피를 이용한 소의 근육, 간 및 천엽에서의 벤지미다졸계 구충제 잔류분석)

  • Simultaneous multiresidue analysis using liquid chromatography determination for five benzimidazole anthelmintics(thiabendazole, oxibendazole, albendazole, mebendazole and fenbendazole) in bovine muscle, liver and omasum has been described. Blank or benzimidazole-fortified samples(0.5g) were blended with bulk $C_{18}$($40{\mu}m$, 18% load, endcapped, 2g). A column made from the resultant $C_{18}$/animal tissue matrix was first washed with hexane($8m{\ell}$), following which the benzimidazoles were eluted with acetonitrile($8m{\ell}$). Analytes of extracted sample were determined by liquid chromatography with UV detector at 290nm. Correlation coefficients of standard curves for individual benzimidazole isolated from fortified samples, using internal standardization, were linear($0.991{\pm}0.007$ to $0.996{\pm}0.005$) with average relative percentage recoveries from $62.1{\pm}3.8(%)$ to $92.3{\pm}7.5(%)$ for the concentration range($0.2{\sim}6.4{\mu}g/g$), respectively. Recoveries rates of TBZ, MBZ in liver, OBZ, MBZ in muscle and TBZ, MBZ in omasium from fortified benzimidazole were 92.%, 87.3%, 74.5%, 82.7%, 75.2% and 83.5% at condition II, respectively. Condition II showed higher recoveries rates than condition I. These results indicated that the matrix solid phase dispersion(MSPD) methodology is acceptable for the determination of 5 benzimidazole anthelmintics and may also suitable for other matrixes of food animal origin.

Characteristics of Ti-SPAC as Fluidizing Phase Photocatalyst (Ti-구형활성탄의 유동상 광촉매 특성 평가)

  • In this sturdy, spherical activated carbon(SPAC) contained $TiO_2$ was made by ion-exchanged treatment and heat treatment for applying fluidizing bed system. The ion-exchange resin was treated by $TiCl_3$ aqueous solution. The treated resin and raw resin were heat-treated under nitrogen condition to convert into Ti-SPAC. During the heat-treatment, burn-off weight amounts and the element were measured by means of TGA and TGA/MS, individually. The physicochemical properties of Ti-SPAC was characterized by means of XRD, SEM, EDS, BET, EPMA, ESR, intensity and titanium content. The Ti-SPAC had spherical shape with diameter size about $350{\mu}m{\sim}400{\mu}m$ and $617m^2/g$ specific surface area. Structure of $TiO_2$ in Ti-SPAC was anatase and rutile form. Also, $TiO_2$ on SPAC were found that the $TiO_2$ were uniformly distributed through EPMA analysis. Moreover, the Ti-SPAC showed indirect photocatalyst activity estimation through ESR analysis, characteristics of photocatalyst potentially. Over all results, Ti-SPAC was used in fluidizing bed UV/photocatalyst system to remove HA(Humic Acid). That results were HA removal efficiency was about 70% and Ti-SPAC intensity was preserved during reaction. Ti-SPAC showed practical possibility as photocatalyst in fluidizing bed system.

Beneficial Effect of Curcumin on Epidermal Permeability Barrier Function in Hairless Rat (무모쥐에서 자외선에 의한 피부 장벽 손상에 미치는 커큐민의 보호 효과)

  • Recent research has shown that curcumin has beneficial effects in a variety of skin diseases, including scleroderma, psoriasis, and skin cancer. In this study, we assessed the effects of curcumin on epidermal permeability barrier function in vivo and in vitro. In order to evaluate the effects of curcumin on epidermal permeability barrier function in vivo, hairless rats were exposed to UVB irradiation, and curcumin was administered orally at a dosage of 150 mg/kg per day for 8 weeks. Transepidermal water loss (TEWL) and epidermal thickness were measured at the end of the experiment. The expression of filaggrin, a marker of keratinocyte differentiation, and serine palmitoyltransferase (SPT), a marker of the formation of the stratum corneum lipid barrier, in human HaCat keratinocytes were analyzed. The in vivo results showed that an 8 week administration of curcumin markedly prevented the UVB-induced increase in TEWL. The UV-induced increase in epidermal thickness was also reduced significantly by curcumin treatment. The in vitro results demonstrated the concentration-dependent effects of curcumin on the expression of both filaggrin and SPT in HaCat cells, reflecting the notion that curcumin can induce epidermal keratinocyte differentiation and can improve the recovery of skin barrier functions. These results show that curcumin is a promising candidate for the improvement of epidermal permeability barrier function.

Residue Patterns of Insecticide Flubendiamide by Varieties of Peaches (살충제 Flubendiamide의 복숭아 품종에 따른 잔류양상)

  • BACKGROUND: This research has investigated the residue patterns of insecticide flubendiamide on three species of peaches with different surface forms, and the residue amounts of them when mixed with a spreader. METHODS AND RESULTS: Pesticide used for field application on peaches was 20% flubendiamide of suspension concentrate(SC) and was sprayed at a recommended rate. The residue amounts of flubendiamide in peach were analyzed by HPLC equipped with UV detector. After the observation with a microscope, the rank of fuzz amount on peach's surface was Kurakatawase, Wolmi in descending order and Cheonhong did not have any fuzz. The residue amounts of flubendiamide were 0.54 mg/kg for Kurakatawase, 0.43 mg/kg for Wolmi and 0.10 mg/kg for Cheonhong, respectively. When flubendiamide was used with a spreader, polyoxy ethylene methylpoly siloxane, the residue amount for Kurakatawase barely changed at 0.55 mg/kg regardless of mixing with a spreader, and at 0.53 mg/kg for Wolmi. In Cheonhong, the residue amount was 0.48 mg/kg, which increased by 4.8 times due to the use of a spreader. CONCLUSION: This result indicates that the residue amounts of flubendiamde were affected by the surface forms of peaches, and in the presence of a spreader the residue amount did not increase in fuzzy species, but was affected greatly for species without fuzz.

Development of a Flavor-Enriched Yeast Extract with a High Glutathione Content (글루타치온 고함유 정미성 효모추출물 개발)

  • Yeast strains with good sensory properties were selected, and those yeasts were subjected to mutation to develop high glutathione producing yeasts. In addition, the antiradical activity and flavoring effect of the yeast extract were evaluated. A total of 68 strains were screened, and three strains of Saccharomyces utilis, four strains of Candida utilis, and one strain of Zygosaccharomyces rouxii were selected based on the flavoring effect. Among them, a random mutation was elicited against SEM-Y8, resulting in a high flavoring effect and growth rate. The glutathione production by SEM-Y8 increased 2.0-fold following the mutation, and the DPPH radical quenching effect of the SY8-M2-1-derived extract increased 3.2-fold compared to that of the wild type. The sensory properties of the SY8-M2-1-derived extract were better than those of garlic or onion extract in umami and mouthfulness. Thus, the SY8-M2-1 extract could be used as a functional flavoring material with improved antiradical activity.

Characterization of Mutant Strain, Lactobacillus paracasei ML-7 Isolated from Kimchi, and Its Effect on the Growth of Broiler (김치 유래의 유산균 변이주 Lactobacillus paracasei ML-7의 특성 및 육계 성장에 대한 영향)

  • A mutant strain of Lactobacillus paracasei (ML-7) was derived by ultraviolet irradiation of the parent L. paracasei LS-2 strain, followed by colony selection on pH 3.5 MRS agar plates, on which L. paracasei LS-2 cannot grow. The L. paracasei ML-7 mutant strain produced an increased amount of organic acids compared to the L. paracasei LS-2 parent strain. Broiler chickens were randomly fed with one of three experimental diets: a basal diet as a control, and basal diets supplemented with 0.3% of the L. paracasei LS-2 and L. paracasei ML-7 strains. When concluding the study, the final body weights of the broilers fed with the supplemented basal diets were found to be significantly higher (1.9% for LS-2 and 6.2% for ML-7) than the control broiler group. Broilers fed with basal diet supplemented with the mutant L. paracasei ML-7 strain showed increased body weight and food intake, compared to broilers fed with the basal diet with or without the parent L. paracasei LS-2 strain.

Preparation and Characterizatino of Nano-sized Liposome Containing Proteins Derived from Coptidis rhizoma (황련유래 단백질이 함유된 나노리포좀의 제조 및 특성)

  • Coptidis Rhizoma, an antimicrobial agent from natural source, is known to have the antiviral effect on the Candida albicans that causes the infectious dermatitis. The valuable protein was extracted from the Coptidis Rhizoma, To prevent denaturalization from external stimulus and improve adsorption onto the skin, the nano-sized liposomes were prepared as a carrier. The CPR-containing liposomes showed an average diameter of 187 nm, surface charge of 3.337 mV and 33% encapsulation efficiency. The release behavior of CRP from the liposome was investigated with various temperature and releasing time. The PVA solution was coated on the surface of liposome to improve the stability. The coated liposome showed slow release behavior in comparison with the non-coated liposome. The CRP in the liposome maintained the effect on the Candida albicans after treating it at 50 and with ultraviolet for 24 h.

Desorption Kinetics and Structural Changes of Tetracycline Treated Barrier Membranes for Guided Tissue Regeneration (Tetracycline 처리된 조직유도재생술용 차폐막의 약제유리양상 및 구조적 변화)

  • Tetracycline is known to be effective in eliminating periodontopathogens and have collagenolytic activity. This study was performed to observe the desorption kinetics and structural changes of tetracycline-treated barrier membranes for guided tissue regeneration. Four kinds of barrier membranes were tested : $Tefgen^{(R)}$(American Custom Medical, USA) and $Gore-Tex^{(R)}$(W.L. Gore & Associates Inc., USA) as nonresorbable membranes ; Resolut(polyglycolide & polylactide copolymer, W.L. Gore & Associates Inc., USA) and $Biomend^{(R)}$(collagen, Collatec Co., USA) as resorbable membranes. The membranes were cut into discs(diameter : 4mm) and were immersed in 5% tridodecylmethylammonium chloride(TIMAC) ethanol and air-dried. The membrane discs were absorbed with $100{\mu}g/ml tetracycline solution(pH8) for one minute and dried. For desorption kinetics, TC treated discs were immersed in phosphate buffered saline solution (PBS, pH 7.4). PBS was exchanged daily and TC concentration was measured by absorbance at 276nm on UV spectrophotometer. To measure remaining antibacterial activity, discs of 1 day to 4 weeks after desorption were placed on Mueller Hinton agar containing Bacillus cereus and incubated aerobically in $37^{\circ}C$ for twelve hours and the inhibition diameters were measured. To observe the structural change of membranes after TIMAC treatment or immersion in PBS, the membrane discs were examined under SEM. The results were as follows : 1. Total amounts of TC absorbed into membrane discs($0.7536mm^2$) were $2000{\mu}g$, $1800{\mu}g$, $2625{\mu}g$ and $2499{\mu}g$ for $Tefgen^{(R)}$, $Gore-Tex^{(R)}$, $Biomend^{(R)}$ and $Resolut^{(R)}$. 2. The concentration of TC released from barrier membrane discs was maintained over $4{\mu}g/ml$ until the fifth day in nonresorbable membranes and $Resolut^{(R)}$, but until the fourth day in $Biomend^{(R)}$, Until the ninth day in nonresorbable membranes and until the seventh day in resorbable membranes, the TC concentration was maintained over $1{mu}g/ml$. 3. The four membrane discs in the first day showed similar size of inhibition zone. One to four weeks later, the inhibition zone was much smaller in resorbable membrane discs than nonresorbable membrane discs. 4. Any structural change due to treatment of TIMAC was not observed on the nonresorbable membranes. $Resolut^{(R)}$ did not show any structural change except fibrillar loosening during immersion period, but Biomend showed destruction of membrane structure from the first week of immersion. This study indicates that tetracycline treated barrier membranes lead to the sustained release of tetracycline for over 7 days. This slow release pattern of tetracycline may contribute to the favorable clinical outcome of guided tissue regeneration.

[ $5{\alpha}-Reductase$ ] Inhibitors from Native Plants and their Sebosuppressive Effects in Cultured Human Sebaceous Gland Cells (천연물 유래의 $5{\alpha}-Reductase$ 저해제의 개발과 인체 유래 피지선 세포의 배양을 이용한 피지분비 억제 효과 측정)

  • This study was performed to develop new cosmeceutical agents with sebosuppressive activity from native plant extracts in Korea. Inhibitory efforts of the extracts on $5{\alpha}-reductase$ (5-AR) were evaluated by enzyme kinetics analysis using UV-spectrophotometric method. Two kinds of enzyme suspensions as 5-AR sources were prepared from rat liver tissue and cultured hSG cells. The sebosuppressive effects were determined by measuring the total lipid quantity produced in cultured hSG cells after incubation with the extracts. As a result, Pinus thunbergii extracts showed the most potent 5-AR inhibitory effects. Its $K_i$ values were 0.0002% and 0.0014% for rat liver 5-AR and human sebaceous gland 5-AR, respectively. Addition of Pinus thunberii extract to hSG cells showed 48% reduction in total lipid production at 0.005% concentration. In conclusion, Pinus thunbergii extracts can be used as a cosmeceutical agent to regulate sebum production and to alleviate the sebum-involved skin diseases, such as acne and seborrheic dermatitis.

Mechanistic Studies for Electrochemical Oxidation of ${\iota}$-Sparteine (${\iota}$-Sparteine의 전기화학적 산화반응에 대한 메카니즘의 연구)

  • The mechanism for electrochemical oxidation of natural alkaloid, ${\iota}$-sparteine (SP) was studied in acetonitrile solvent. The cyclic voltammogram of SP shows two irreversible anodic peaks at +0.75 V and +1.45 V vs. Ag/AgCl (0.1M AgNO$_2$ in acetonitrile) electrode. Coulometry reveals that the number of electrons involved in each oxidation peaks is in the range of 1.2∼1.3 respectively. Neutral imine radical was produced by fast deprotonation of SP radical cation formed by oxidation of one nitrogen atom in SP. Two pathways are possible for the reaction of the neutral radical: Due to the disproportionation of the radical, SP and enamine were mainly produced. Also, the 1,2-dehydrosparteinium cation was formed as minor product through the second one electron transfer oxidation of this radical. The (+)-lupanine was produced by treatment of sparteinium cation with potassium hydroxide. We have isolated and confirmed the electrolysis products using IR, GC-MS, UV-Vis, and thin-layer spectroelectrochemical method.
