Reemergence of porcine epidemic diarrhea virus on Jeju Island

  • Porcine epidemic diarrhea virus (PEDV) strains responsible for recent outbreaks in the United States have been occurring in Mainland Korea since late 2013. Over the past 10 years, PEDV outbreaks have not been reported on Jeju Island. However, in late March of 2014, PEDV re-emerged on Jeju Island and was found to be genetically identical to PEDV strains currently circulating in Mainland Korea. The present study was conducted to provide a better understanding of the epidemiology of PEDV and more effective preventive measures against PED.

Characterization of the genomes of Aujeszky's disease virus isolated in Korea (국내분리 오제스키병 바이러스의 게놈 유전자 특성 분석)

  • The molecular genetic characterization of Aujeszky's disease virus (ADV) Yangsan strain (ADVYS), a Korean isolate, was investigated by analyzing the electrophoresis patterns and the physical maps of the viral DNA digested with various endonucleases. To establish DNA library for ADV-YS, twelve major BamHI restricted segments were cloned. Each location of the segments in the ADV genome was determined by sequence comparison with the sequences reported in Genbank and those sequences of the both termini of the segments. Physical maps were constructed based on the electrophoresis patterns of the digested viral DNA by restriction endonuclease and the results of Southern blot analyses with various DIG labeled probes originated from those of enzyme restricted segments of virulent (Shope) and avirulent (Bartha) strain. Comparing ADV-YS with a standard strain of Kaplan in the maps of restriction enzymes, following major respects were identified: (i) disappearance of BamHI restriction site between the first and second BamHI segments, (ii) creation of the BamHI restriction site in the fifth segment, and (iii) generation of the BglII site in the unique short (US) region. The genome of ADV-YS also contains a type 2 herpesvirus DNA molecule (in which the US region only inverts itself relative to the unique longregion) like all other ADV strains except Norden strain(type3), analyzed up to date. The size of the ADV genome estimated from the sizes of the restriction enzyme fragments, was approximately 145.3 kb (BamHI) or 145.4 kb (BglII). BamHI enzyme cleavage patterns were compared among the five Korean ADV isolates: Yangsan, Yongin, Dangjin, Jincheon and Iksan strains. Difference either in the number or in the size of the DNA fragments, suspected regions of termini of IR and TR, could be detected among all five strains.

Genotypic Variations among Human Caliciviruses in Korea: 1987-1994 (한국에 산재하는 사람 Caliciviruses의 다양한 유전자군: 1987-1994년)

  • Sequence comparison of the RNA-dependent RNA polymerase of human caliciviruses (HuCVs) from Korean children with gastroenteritis revealed significant genetic variation among them. cDNA clones were produced from the HuCVs collected from pediatric population during a period of 1987-1994. The application of reverse transcription-polymerase chain reaction (RT-PCR) using primers directed to the RNA-dependent RNA polymerase region within ORF1 of Norwalk virus (NV) showed that 13.7% of HuCVs yielded PCR products of similar size to the NV prototype, NV8FIIa/68/US, with exceptions of HuCV 185/87/Korea and HuCV 1115/90/Korea. Computer analyses showed that the PCR products had a continuous protein encoding frame on the positive strand, and contained GLPSG and YGDD amino acid motifs at the predicted distance from primers. Alignment of the amino acid sequences of HuCVs with previously published sequences for Snow Mountain agent (SMA), NV, and Sapporo/82/Japan indicated that these strains can be divided into four major genogroups. There were 10 (45%) SMA-like CVs, one (4.5%) NV-like HuCVs, two (9%) Sapporo-like HuCVs, and nine (41%) unidentified HuCVs. This fourth genogroup should be investigated further. HuCV 185/87/Korea and HuCV 1115/90/Korea, Sapporo-like CVs, were genetically distinct from previously characterized HuCVs and more closely related to known animal CVs. One of the animal CV-like strain, HuCV 185/87/Korea, showed nucleotide and amino acid homology of only 67% and 73% with the prototype Sapporo/82/Japan. Further characterization of animal and human CV genomes and studies of possible cross-transmission of CVs from animals to humans are likely to be beneficial in understanding the epidemiology of HuCVs.

Overexpression of sprA and sprB Genes is Tightly Regulated in Streptomyces griseus

  • The sprA and sprB genes, encoding the chymotrypsin-like proteases Streptomyces griseus protease A (SGPA) and Streptomyces griseus protease B (SGPB), and the sprT gene that encodes Streptomyces griseus trypsin (SGT) were cloned from S. griseus and were overexpressed in various strains of S. griseus. When the sprT gene was introduced into S. griseus, trypsin activity increased 2-fold in the A-factor deficient mutant strain, S. griseus HH1, and increased 4-fold in the wild strain, S. grise us IFO 13350. However, there was no detectable increase of chymotrypsin activity in the transformants of S. griseus with either sprA or sprB, in contrast to the results obtained from S. lividans as a heterologous host. To solve the negative gene dosage effects in S. griseus, either the sprA or the sprB genes with their own ribosome binding sites were linked to the downstream of the entire sprT gene, and the coexpression efficiency was examined in S. lividans and S. griseus. The transformants of S. lividans with either pWHM3-TA (sprT+sprA) or pWHM3­TB (sprT+sprB) showed 3-fold increase of trypsin activity over that of the control, however, only the transformant of pWHM3-TB demonstrated 7-fold increase in chymotrypsin activity, indicating that the pWHM3-TB has a successful construction for the overexpression of chymotrypsin in Streptomyces. When the coexpression vectors were introduced into S. griseus IFO 13350, the trypsin level sharply increased by more than 4-fold, however, the chymotrypsin level did not increase. These results strongly suggest that the overexpression of the sprA and sprB genes is tightly regulated in S. griseus.

Semiweekly Variation of Spring Population of a Mixotrophic Ciliate Myrionecta rubra (=Mesodinium rubrum) in Keum River Estuary, Korea (춘계 금강 하구에서 혼합영양 섬모류인 Myrionecta rubra (=Mesodinium rubrum) 개체군의 단주기 변동)

  • Myrionecta rubra, a mixotrophic ciliate, is a cosmopolitan red tide species which is commonly found in neritic and estuarine waters. M. rubra had long been listed as an “nculturable protist”until 2 different laboratory strains were finally established in 2 research groups at the beginning of this century, enabling us to perform initiative investigation into various aspect of the live M. rubra strains (Gustafson et al. 2000; Yih et al. 2004b; Johnson and Stoecker 2005). Field sampling was carried out on high tide at 2 fixed stations around Kunsan Inner Harbor (St.1 near the Estuarine Weir and St.2 off Kunsan Ferry Station) every other day for 4 months from mid-February 2004 to understand detailed figure of the recurrent spring blooms of M. rubra following the onset of the water gates operation of the Keum River Estuarine Weir on August 1994. With its maximum abundance of 272 cells mL$^{-1}$ in St.1, fluctuation pattern of the M. rubra population at the 2 stations was strikingly similar. Notable growth of M. rubra population started on late April, to cause M. rubra red tides during one month from mid-May in which “xceptionally low salinity days”without its red tide were intermittently inserted. High abundance of M. rubra over 50 cells mL$^{-1}$ was recorded at samples with their water temperature and salinity higher than 15${^{\circ}C}$ and 4.0 psu, respectively. During pre-bloom period when salinity fluctuation is moderate and the water temperature is cooler than 15°C, Skeletonema costatum, a chain-forming centric diatom, was most dominant. Cyanobacterial species such as Aphanizomenon flos-aquae and Phormidium sp. replaced other dominant phytoplankters on the days with “xceptionally low salinity”even during the main blooming period of M. rubra. To summarize, M. rubra could form spring blooms in Keum River Estuary when the level of salinity fluctuation was more severe than that for the dominant diatom Skeletonema costatum and milder than that for the predominance by freshwater cyanobacteria. Therefore, optimal control of the scale and frequency of freshwater discharges might lead us to partially modify the fluctuation pattern of M. rubra populations as well as the period of spring blooms by M. rubra in Keum River Estuary. Sampling time interval of 2 days for the present study or daily sampling was concluded to be minimally required for the detailed exploration into the spring blooms by M. rubra populations in estuaries with weirs like Keum River Estuary.

Thermal and Mechanical Properties of Poly(lactic acid) Specimens Fabricated by Various Equal-channel Angular Extrusion Processes (다양한 방식의 등통로각압축공정으로 가공된 Poly(lactic acid) 시편들의 열 및 기계적 물성)

  • We fabricated rod-like poly(lactic acid)(PLA) specimens through applying various methods of equal-channel angular extrusion(ECAE) process and investigated the change of thermal and mechanical properties of specimens before and after each ECAE process. Combining three re-injection routes(A, BC, and C) and three pass counts(1, 2 and 4) allowed us to fabricate 7 different PLA specimens. Thermal properties of each specimen were measured by both differential scanning calorimeter and thermo-gravimetric analyzer. Shear strains of each specimen with respect to applied loads were measured by indentation hardness tester. Field emmision scanning electron microscopy was used to observe internal microstructure of cross-section of each specimen. The observed microstructures qualitatively supported the explanation of hardness test results. Among 7 specimens, PLA-P2A showed the biggest shear strain probably due to its dense microstructure.
