Radioisotope Labelling Method of Pine Gall Midge (Thecodiplosis japonensis UCHIDA et INOUYE) (솔잎혹파리에 대한 방사선동위원소 표식방법에 관한 연구)

  • This experiment was carried out to develope a technique for labelling pine gall midge with ·radioactive phosphorus and calcium. This technique evolved would help to obtain basic information on the appropriate concentration of $^{32}P\; and\; ^{45}Ca$ in tagging and determination of adult flight ranges which considered as one of the factors in dissemination. 1. Labelling at the larval stage of pine gall midge by injection of $^{45}Ca$ into pine tree trunk appeared not to be effective for tagging the adults because radioactivity was gradually decreased to background by emergence. 2. The radioactivities of larvae in the galls gradually increased in September but from October 1 the activity was gradually decreased for 10 days then equilibrium was maintained thereafter. The results seem to indicate that larval diet termination occured early October. 3. A maximum level of radiation was detected at 3 weeks after $^{32}P$ injection and thereafter it was gradually reduced. This result implies that $^{32}P$ is not appropriate radioisotope in labelling pine gall midge through trunk injection method. 4. After washing in running water for 10 minutes, the optimum$^{32}P$ concentration for treated overwintered larvae and pupae was found to be 0.5 micro curie per ml. for 30 minutes dipping.

  • Field experiment was conducted to develop a model of controlled release of oxyfluorfen by using various split applications. Rice and some weed species was included in this study. The models of split applications were 10-10-0. 10-10-10. 20-20-0, 20-10-10. 20-20-20, 0-40-0, and 0-0-0g/Ha at 3days before transplanting, and 11 days after transplanting, respectively. Rice injury appeared dispeared at 20 g/Ha of oxyfluorfen on low leaf sheath but disappeared at few days. The injury was reduced by split application even at the same rate of application. However, no injury was siginificant at 30 days after transplanting. Barnyardgrass, Monochoria, and arrowhead were most susceptible to oxyfluorfen, but bulruch and most perennial weeds recovered after temporary growth inhibition. Therefore, to develop oxyflourfen for use in rice transplanting of adult rice seedling, split application with reduced rates, and development of expected to be tank-mixed or premixed with other perennial herbicides to obtain droad spectrum of weeds.

  • Five parents which showed differential response to thiobencarb and their $F_1$ hybrids were treated with thiobencarb (S-[(4-chlorophenyl) methyl]diethylcarbamothioate) at 2 and 4 kg ai/ha in order to investigate the inheritance of rice (Oryza sativa L.) to the herbicide. In the analysis of diallel crosses for tolerance of rice cultivars to the herbicide, highly significant GCA (general combining ability) and SCA (specific combining ability) effects were observed at both herbicide rates using the parameters of plant height and dry weight ; the GCA effect being greater than the SCA effect. The additive effects appeared to be more prevalent than dominance effects, and partial dominance was recognized in relation to herbicide tolerance in the different rice cultivars. Higher heritabilities in the narrow and broad senses were observed at 4 kg ai/ha. The heritabilities in the narrow sense for plant height and dry weight were 0.726 and 0. 743, respectively. IR9660-50-3-1 which showed the highest GCA effects seemed to possess more dominant genes related to herbicide tolerance than the other rice cultivars tested.

Effect of Xylanase on Performance and Apparent Metabolisable Energy in Starter Broilers Fed Diets Containing One Maize Variety Harvested in Different Regions of China

  • The objective of this study was to investigate the variability in broiler performance, apparent metabolisable energy (AME) and ileal digestible energy (IDE) between five different maize samples fed with and without xylanase at 16,000 U/kg. Various in vitro characterisations were conducted to determine if any could predict performance or AME. Samples of the maize were harvested in five diverse regions and fed individually in a mash diet as follows (g/kg): test maize 608.3; soya bean meal (SBM) 324.1; poultry fat 25.2; salt 4.6; met 2.6; lys 1.6; thr 0.5; limestone 9.7, dical 18.4; vit/min 5.0; CP 210 and ME (kcal/kg) 3,085. The diets were fed to 720 broilers with 6 replicates, each containing 12 birds per treatment, from 0 to 18 d of age. Maize samples were analysed for starch, protein, crude fibre, fat, protein solubility index (PSI) and vitreousness using near infra red reflectance spectroscopy (NIR). They were also assayed using an in vitro starch digestibility method. The results showed that there was no effect of harvest region on the feed intake (FI), body weight gain (BWG) or feed conversion ratio (FCR) of the broilers over the 18 d period (p = 0.959, 0.926, 0.819 respectively). There was an improvement in all parameters with the addition of xylanase (FI p = 0.011; BWG and FCR p<0.001). There was a significant positive effect of xylanase on IDE, AME, IDE Intake (IDEI) and AME intake (AMEI) (p<0.0001 in all cases). Although there was no significant effect of maize source, there was a strong trend towards variability in IDE (p = 0.066) and AME (p = 0.058). There were no significant correlations (p<0.05) between any proximate or physiochemical values and any performance or AME values. This may suggest that none of those selected were suitable predictors for performance or AME. The broilers performed well according to the breed guidelines, with slightly increased FI, increased BWG and similar FCR prior to the addition of xylanase. When FCR and BWG were analysed with FI as a covariate, xylanase addition remained significant suggesting that the improvement in BWG and FCR was driven by an increase in digestibility and nutrient availability.

Salinity Tolerance of Blackgram and Mungbean: II. Mineral Ions Accumulation in Different Plant Parts

  • Blackgram (Vigna mungo) is more salt tolerant than mungbean (Vigna radiata). This study was initiated to know whether the accumulation pattern of mineral ions in different plant parts plays a significant role in the differences in salt tolerance between the two Vigna species. Different mineral ions, viz. N, Cl, Na, K, Mg and Ca in different organs of two varieties of each of blackgram- Barimash-l (susceptible one) and Barimash-2 (tolerant one), and mungbean-Barimung-3 (tolerant one) and Barimung-4(susceptible one), were analyzed after growing with 0, 50, 75 and 100 mM NaCl solutions. The two crops showed a decreased but similar pattern of total N accumulation under saline conditions. The tolerant variety of both the crops showed a less reduction in total N than the susceptible one. Leaves showed the maximum while stem the minimum N, irrespective of levels of salinity. C $l^{[-10]}$ and N $a^{+}$ accumulation increased with the increasing salinity levels. Interestingly, similar to a halophyte, the salt tolerant blackgram exhibited conspicuously higher amount of N $a^{+}$ in the shoot than the salt-susceptible mungbean. However, the tolerant varieties showed less amount of N $a^{+}$ than the susceptible one, especially in blackgram. Seeds of both Vigna spp. accumulated the minimum amount of N $a^{+}$ than other plant parts. $K^{+}$ accumulation decreased by salinity in most of the plant parts, except seeds. Blackgram showed larger reduction in K than mungbean. The $Mg^{++}$ increased in leaves, petioles and stem by salinity while decreased in the roots, podshells and seeds in both the crops. Salinity increased $Ca^{++}$ accumulation in all plant-parts except roots of both Vigna spp. Apparently, the leaves of mungbean accumulated higher concentration of $Ca^{++}$ than blackgram. Varietal differences in the accumulation pattern of $K^{+}$, $Mg^{++}$ and $Ca^{++}$ were not clear. It was concluded that blackgram, presumably, possesses a similar salt tolerance mechanism to halophyte, and the pattern of accumulation of mineral ions in blackgram and mungbean was not fully ascribed to the differences in salinity tolerance between the two Vigna species.gna species.ies.s.ies.

Lactation Performance of German Fawn Goat in Relation to Feeding Level and Dietary Protein Protection

  • Effects of high and low levels of feeding with or without protected protein on the performance of lactating goats were studied. Twenty four German Fawn goats either from 1st ($43.37{\pm}3.937$ kg and 2 year old) or 3rd $62.64{\pm}6.783$ kg and 4-5 year old) parity were used for the trial. Feeding levels were 7.2 (I) and 5.2 (II) MJ ME/litre of milk of 3.5% fat in addition to that of the maintenance allowance. At each feeding level, diet had either unprotected (U) or formaldehyde protected (P) soya-meal. Thus, four diets were IU, IP, IIU and IIP, having six animals in each. The diets were composed of hay and pellet (10:4:1 of beet pulp : barley : soya-meal). Effect of feeding level, protein protection, parity, health status and kid number on intake, milk yield, milk composition, growth rate of goats were recorded across the 21 weeks of study. High feeding level resulted increase (p<0.001) in estimated metabolizable energy (ME) and metabolizable protein (MP) availability. Dietary inclusion of protected soya-meal increased (p<0.001) the estimated MP but not the ME availability. Animals in 1st parity ate more (p<0.001) DM (111 vs. 102 g/kg $W^{0.75}$/d) than those in 3rd parity. Animals with twin kids (110 g/kg $W^{0.75}$/d) had higher (p<0.001) DM intake than those with single kid (102 g/kg $W^{0.75}$/d). Fat (4%) corrected milk (FCM) yield was not effected by high (1,924 g/d) or low (1,927 g/d) feeding level but increased (p<0.001) with protected (2,166 g/d) compared with unprotected (1,703 g/d) soya-meal. FCM yield for four dietary combinations were 1,806, 2,078, 1,600 and 2,254 g/d for diets IU, IP, IIU and IIP, respectively. For unit increase (g) in estimated MP availability relative to ME (MJ) intake, FCM yield increased ($1,418{\pm}275.6$) g daily ($r^2$=0.58; p<0.001). Milk fat (3.14 vs. 3.54%; p<0.001) and protein (2.94 vs. 3.04% p<0.05) contents were lower at high than the low feeding level. Protected protein increased (p<0.001) the fat, lactose and net energy (NE) content of milk. Milk urea concentration of 175, 183, 192 and 204 mg/l for diets IU, IP, IIU and IIP, respectively indicated lower RDP content of these diets. The RDP contents were 6.97, 6.70, 7.30 and 6.83 g/MJ of ME for diets IU, IP, IIU and IIP, respectively. Live weight change over the experimental period were 41, 6, 17 and 19 g/d. Absence of any positive response of high feeding was probably due to inefficient rumen fermentation resulting from inadequate RDP supply. Protected protein improved production performance apparently by increasing MP:ME ratio in the absorbed nutrient.

Selection of Herbicide - Tolerant Rice(Oryza sativa L.) Callus by Tissue Culture (조직배양(組織培養)을 통한 수도내성(水稻耐性) 카루스 선발(選拔))

  • The response of callus growth of rice (Oryza sativa L.) cultivars which showed different responses to herbicides as seedlings was investigated to select resistant or tolerant calli. Callus growth of IR28 which was susceptible to thiobencarb (S-[(4-chlorophenyl)methyl]diethylcarbamothioate) during callus induction was not inhibited by $10^{-5}$ M and $10^{-6}$ M thiobencarb, indicating that there was a difference in tolerance among callus induction and growth, and the intact plant level. A similar result was obtained with IR31917-45-3-2-2 to butachlor [N - (buthoxymethyl) -2-chloro-N- (2, 6-diethylphenyl) acetamide]. The fresh weight of IR28 callus transferred into $10^{-5}$ M thiobencarb after treatment at $10^{-6}$ M for 30 days was not affected by the herbicide, indicating that transferring callus into gradually higher herbicide concentrations can be a useful method for selection of herbicide-tolerant cell lines.

  • The response of rice (Oryza sativa L.) cultivars to mixtures of 2, 4-D(2, 4-dichlorophenoxyacetic acid) and other herbicides was investigated to determine if there was an interaction between them. When 2, 4-D was applied, shoot growth of Taipei 309 was more affected than that of IR28 at all concentrations used. In contrast, when thiobencarb (S-[(4-chlorophenyl)methyl]diethylcarbamothioate), butachlor [N-(buthoxymethyl) -2-chloro-N-(2, 6-diethylphenyl) acetamide], and glyphosate [N-(phosphonomethyl)glycine] were applied, the shoot growth of Taipei 309 was less affected than that of IR28 at all herbicide concentrations. Combination of 2, 4-D and the lowest thiobencarb concentration was antagonistic for shoot length for both cultivars, but at higher concentrations, it was synergistic. Synergism for shoot fresh weight between 2, 4-D and thiobencarb was observed with IR28 at all concentrations but, for Taipei 309, synergism was observed only at lower 2, 4-D concentrations. Mixing 2. 4-D with butachlor resulted in greater inhibition in shoot length and fresh weight of IR28 than Taipei 309 at all concentrations indicating a synergistic interaction. With combinations of 2, 4-D and glyphosate, an antagonistic interaction for shoot length was observed for both cultivars. A synergistic interaction for shoot fresh weight was observed with IR28 when combinations of the highest concentration of glyphosate and 2, 4-D were applied but there was an antagonistic interaction with Taipei 309.

  • About 136 weed species belonging to 44 families occurring in summer observed in Cheju island. The Compositae was the most widely occurring family covering 29 weed species, followed by 16 species in Graminae, 14 in Leguminosae, 13 in Rosaceae, 10 in Polygonaceae etc., The dominant weed species detected in range areas of Cheju island were Trifolium repens, Botrychium virginianum, Rosa multiflora, Miscanthus sinensis, Imperata cylindrica. The range areas of Cheju island consisted of the communities of Artemisia princeps-Botrychium virginianum, Artemisia princeps-Erigeron borariensis and Dianthus sinensis, and Artemisia-Lespedeza bicolor and Miscanthus sinensis and then the differential species of their communities were Botrychium virginianum, Erigeron boriensis, and Lespedeza bicolor and Miscanthus sinensis, respectively. The species of the high frequencies occurring in investigated sites were Artemisia princeps, Cirsium rhinoceros, Hydrocotyle ramijlora, Rosa multiflora, Sanguisorba officinalis, Rubus crataegifolius, Potentilla fragariodes, Erigeron annus, Plantago asiatica, Oxalis corniculata, Mosla dianthera in order.

Sensitivity Analysis of Nozzle Geometry Variables for Estimating Residual Stress in RPV CRDM Penetration Nozzle (원자로 상부헤드 관통노즐의 잔류응력 예측을 위한 노즐 형상 변수 민감도 연구)

  • Recently, several circumferential cracks were found in the control rod drive mechanism (CRDM) nozzles of U.S. nuclear power plants. According to the accident analyses, coolant leaks were caused by primary water stress corrosion cracking (PWSCC). The tensile residual stresses caused by welding, corrosion sensitive materials, and boric acid solution cause PWSCC. Therefore, an exact estimation of the residual stress is important for reliable operation. In this study, finite element simulations were conducted to investigate the effects of the tube geometry (thickness and radius) on the residual stresses in a J-groove weld for different CRDM tube locations. Two different tube locations were considered (center-hole and steepest side hill tube), and the tube radius and thickness variables ($r_o/t$=2, 3, 4) included two different reference values ($r_o$=51.6, t=16.9mm).
