GLC에 의한 버섯의 Amino Acid 정량(定量) (Quantitative Analysis of Protein Amino Acid in Agaricus Bisporus by GLC)

  • Quantitative analysis was achieved by gas-liquid chromatographic method (GLC) with a single column system of OV-17 for 16 of protein amino acids in mushroom (agaricus bisporus). The quantities of protein amino acids in mushroom were determined $48.32{\sim}255.94mg%$ alanine, $108.6F{\sim}364.82mg%$ glycine, $124.30{\sim}314.17mg%$ Valine, $32.99{\sim}418.79mg%$ leucine and isoleucine, $151.78{\sim}669.07mg%$ threonine, $88.12{\sim}4F6.3Fmg%$ Serine, $21.9F{\sim}114.94mg%$ Hydroxyproline, $20.F4{\sim}174.63mg%$ proline, $34.52{\sim}173.59mg%$ Methionine, $225.25{\sim}1417.61mg%$ Aspartic Acid, $10F.00{\sim}392.17mg%$ Phenylalanine, $12F.46{\sim}535.65mg%$ Glutamic Acid and Lysine, Tyrosine in trace amount.

버들명아주의 성분(成分)에 관한 연구(硏究) (Studies on the Constituents of Chenopodium acuminatum)

  • White needle crystalline compound, mp.\;78{\sim}79$, was isolated from the petroleum ether fraction of the aerial parts of Chenopodium acuminatum. This compound was supposed to be cycloalkane by chemical properties, spectral data and elemental analysis. After extraction with petroleum ether, the methanolic extract of this plant found to contain aspartic acid, threonine, serine, glutamic acid, glycine, cystine, valine, isoleucine, tyrosine, and phenylalanine. Inorganic elements contained in this plant are as following: Mg 4.84%, Ca 9.602%, Zn 1.11%, K 29.104%, Mn 5.124%, Fe 27.039%, Co 0.31%, Pb 0.995%, Na 0.039% and Cu, 0.0076%.

임신말 태아의 아미노산영양에 대한 연구 (A Study on the Fetal Amino Acids Nutrition at Term of Human Pregnancy)

  • This study was performed to investigate the placental transfer and the human fetal utilization of amino acids at term of pregnancy. The plasma levels of 23 free amino acids in both the umbilical circulation ( umbilical vein and artery ) and the uterine circulation ( uterine vein and iliac artery ) of 34 pregnant women were measured at delivery by the cesarean section. In the umbilical circulation, 9 amino acids ( alanine, lysine, valine, leucine, arginine, isoleucine, ornithine, cystine, ${\alpha}-aminobutyrate$ ) were significantly higher and 2 amino acids ( glutamate, aspartate ) were significantly lower in the umbilical vein than in the umbilical artery. In the uterine circulation, alanine, tyrosine and methionine were significantly lower in the uterine vein than in the iliac artery. Glutamate was significantly lower in the uterine vein than in the iliac artery. According to these results, the origin of fetal plasma amino acids was discussed in terms of the metabolic conversions which would occur in the Placenta and the fetal utilization of amino acids was estimated.

Recommendations for the Selective Labeling of [$^{15}N$]-Labeled Amino Acids without Using Auxotrophic Strains

  • The strategy to incorporate [$^{15}$ N]-labeled amino acids were discussed. Instead of using specific auxotrophic strains for selective labeling, the prototrophic strain, BL2l(DE3), was used with a plasmid, pLysS, and found to be very effective for several amino acids including alanine, lysine, leucine, and threonine. Isoleucine, valine, glutamine, and tyrosine were also found to be effective despite some diffusion into other amino acids. Interesting result was obtained when [$^{15}$ N]-labeled glycine was tried: only glycines were labeled when amino acid mixture was added in the growth medium, and serines were co-labeled when amino acids were omitted. These results can be used as a guideline when selective labeling strategy is considered, and when the resulting data are interpreted.

황색종 잎담배의 숙성에 따른 아미노산의 조성 (Amino Acid Composition of Flue-cured Leaf Tobacco before and after the Aging)

  • The amino acid content and constitution of domestic flue-cured tobacco before and after the aging were investigated. The major amino acids of domestic flue -cured tobacco were histidine, serine, aspartic acid and proline and leucine, isoleucine and glycine were minority in them. Phenylalanine was the most changeable components through the aging. While the contents of tryptophan, sering and Proline which have herbaceous, harsh and bitter taste and aroma in smoke were reduced, tyrosine which adds smoothing and enhances smoke aroma was increased after the aging. The leaf tobacco aged at 4$0^{\circ}C$ for 30 days had a amino acid composition as good leaf tobacco storaged in natural condition for 2 years.

  • The earthworm(Annelid) is a herbicine which has traditionally been used in the treatment of infectious fever, jaundice, infection of middle ear, laryngitis, pharyngitis, nephritis, headache, toothache and certain urinary tract infections from the olden times. Before the isolation and purification of biologically active components we analyzed the basic constituents(proteins, amino acids, mineral, etc.) with lyophilized powder of Lamnodrilus gotai Hatai. The results were as follows: Minerals detected and quantitatively analyzed were $Ca^#$, $Mg^#$, Fe, Zn, Mn, Cu, Co, Cr, Ni, Ge and Se. Amino acids detected were alanine, phenylalanine, leucine, glutamic acid, tyrosine, threonine, arginine, aspartic acid, methionine, lysine, serine, histidine, isoleucine, glycine, proline and etc. The constituents of proteins, fat, fiber, ash and phosphorous were measured. These constituents were compared and discussed with those of other investigations.

Influence of the Structural Characteristics of Amino Acids on Direct Methylation Behaviors by TMAH in Pyrolysis

  • Direct methylation behaviors of 20 amino acids with tetramethylammonium hydroxide (TMAH) were studied under diluted conditions with silica. Amino acid concentration was controlled by dilution with silica ($SiO_2$) and the molar ratios of amino acid/silica were 0.20, 0.50, and 2.0. The molar ratios of amino acid/TMAH (0.51 - 4.64) also varied. It was found that arginine, asparagine, aspartic acid, cysteine, glutamic acid, and glutamine did not generate any directly methylated pyrolysis products, whereas alanine, glycine, isoleucine, leucine, methionine, phenylanaline, valine, and proline generated all the directly methylated pyrolysis products. Tri- and tetra methylated products of lysine consisted of two types. Histidine and threonine hardly generated the partly methylated products. Mono- and dimethylated products of serine, tryptophan, and tyrosine were not observed. Relative intensities of the methylated products varied with the amino acid concentration, TMAH concentration, and pyrolysis temperature. Direct methylation behaviors of amino acids were explained by the structural characteristics of amino acids.

대구알젓의 맛성분 제I보 : 대구알젓의 유리아미노산 (The Taste Conpounds of the Fermented Cod-roe, Gadus Macrocephlus - Changes of Free Amino Acids during the Fermentation of Cod-roe -)

  • Changes of free amino acids as taste compunds during the fermentation of cod-roe, Gadus marcrocephlus, were analyzed by a amino acid autoanalyzer. Leucine, lysine, valine, isoleucine and arginine were dominent amino acids in fresh cod-roe extracts, and the amounts of leucine (797.3mg%, on moisture and salt free base), lysine (577.7mg%) and valine(487.4mg%0 were 54.9% of total free amino acids. The contents of methionine, histidine, tyrosine and phenylanine were lower, and aspartic acid, glycine, threonine, glutamic acid and serine were detected trace amount. The free amino acids analyzed in this experiment were not changed in composition but changed in the amounts during 80 days of the fermentation. Total free amino acids were increased until 59 days of fermentation and than decreased gradually.

북동태평양 해역의 용존 자유아미노산 분포 및 조성 연구 (Vertical Distribution and Composition of Dissolved Free Amino Acids in the Northeast Pacific Ocean)

  • 북동태평양($9^{\circ}54'{\sim}10^{\circ}27'N$, $131^{\circ}43'{\sim}131^{\circ}53'W$)에서의 용존 자유아미노산 농도는 15.9~1778.2 nM의 범위를 갖으며, 평균 407.2 nM을 보였다. 표층 혼합층에서의 용존 자유아미노산은 60.1~1411.9 nM의 범위로 평균. 535.2 nM을 보였으며, 계절 수온약층이 형성된 수심 50~150 m의 수층에서는 91.7~1778.2 nM의 범위와 평균 588.5 nM의 값으로 최대치를 포함하고 있었다. 또한 저층까지의 수층에서는 15.9~384.2 nM의 범위와 평균 175.1 nM을 나타내었다. 연구해역의 모든 정점에서 glycine, alanine, glutamic acid, serine 그리고 valine 등이 전체 농도의 약 60% 이상을 차지하며 우점하였고, isoleucine, tyrosine, methionine 그리고 phenylalanine 등은 낮은 비율을 보였다. 아미노산 작용기단별 몰%는 aliphatic neutral이 평균 59%로 가장 높았으며 aliphatic hydroxy 16%, acidic 12%의 순으로 나타났다. 총아미노산의 농도가 수층별로 유광층에서 높고 저층이 상대적으로 낮은 분포를 보이고 있으나, 각 아미노산의 조성 비율은 매우 유사하게 나타나고 있었으며 이러한 결과는 미생물작용 및 각 아미노산들의 수용성 등에 의하여 일정하게 유지되고 있는 것으로 사료된다.

  • 오종(五種)의 숙성(熟成) 내장(內臟)젓을 수집(蒐集)하여 분석(分析)하였던 바 다음과 같은 결과(結果)를 얻었다. 1) 오종(五種)의 창젓에 관(關)한 일반분석결과(一般分析結果)는 다음과 같다. 항목(項目) 수분(水分)(%) 조단백(粗蛋白)(%) 조회분(粗灰分)(%) 염분(鹽分)(%) 회분(灰分)(%) 시료(試料) 전복창젓 61.7, 7.13, 21.8, 20.5, 1.3 조기창젓 62.5, 11.19, 20.1, 18.5, 1.8 갈치창젓 59.8, 10.38, 24.8, 22.4, 2.2 명태창젓(속초) 64.8, 8.56, 23.5, 21.97, 1.54 명태창젓(제주) 72.8, 8.66, 16.1, 15.3, 0.8 2) 창젓의 염분(鹽分)을 보통 $18{\sim}22%$ 정도이다. 3) 오종(五種)의 창젓중(中)에서 paper chromatography 법(法)에 의(依)해서 다음 같은 amino acids가 검출(檢出)되었다. Amion acids Free amino acids 종류(種類) 전복창젓 Cys., Hyd ypro., Thr., Lys., Glu., Ser., His., Try., Gly., Thr., Arg., Ala., met., phe., Leu., Isoleu., Pro. 조기창젓 Asp., Cys., Iys., Ser., His., Try., Gly., Thr., Arg., Hydpro., Tyr., ala., Val., met., phe., Leu., Isoleu., Pro. 갈치창젓 Cys., Lys., Gly., Ser., His., Try., Glu., 쏙., Arg., Hyd pro., Tyr., Ala., Met., Phe. 명태창젓(속초) Asp., Cys., Lys., Ser., His., Try., Gly., Thr., Hyd pro., Tyr., Ala., Met., Phe., Leu., Isoleu., Pro. 명태창젓(제주) Asp., Cys., Lys., Glu., Ser., His., Try., Gly., Thr., Hyd pro., Tyr., Ala., Val., met., phe., Leu., Isoleu., Pro. 4) 창젓류(類)에는 valine을 제외(除外)한 7종(種)의 필수(必須) amino acids Ieucine, isoleucine, phenylalanine tryptophan, methionine, threonine, lysine, 이 모두 존재(存在)한다. 5) 7종(種)의 필수(必須) amino acids 중(中)에서 tryptophan 함량(含量)은 극(極)히 적었다. 6) 창젓중(中) 함량(含量)이 제일 많은 것은 tyrosine 이며, 그다음이 hydroxyproline이다.
