The Roles of Hydroxyl Substituents in Tyrosinase Inhibitory Activation of Flavone Analogues (Flavone 유도체들의 Tyrosinase 저해활성화 반응에서 Hydroxyl 치환기들의 역할)

  • Molecular docking of polyhydroxy substituted flavone analogues (1-25) as substrate molecules to the active site of tyrosinase (PDB ID: Deoxy-form (2ZMX) & Oxy-form (1WX2)) and Free-Wilson analysis were studied to understand the roles of hydroxyl substituents ($R_1-R_9$) in substrate molecules for the tyrosinase inhibitory activation. It is founded from Free-Wilson analysis that the $R_1$=hydroxyl among $R_1-R_9$ substituents had the strongest influence on the tyrosinase inhibitory activity. H-bonds between the hydroxyl substituents of substrate molecules and amino acid residues in the active site of tyrosinase were contributed to make a stable substrate-receptor complex compound. Particularly, it is proposed from the findings that the noncompetitive inhibitory activation would take place via H-bonding between peroxide oxygen (Per404) atom in the active site of tyrosinase and the hydroxyl substituents in substrate molecule.

A Study on the Tyrosinase Inhibitory and Antioxidant Effect of Microalgae Extracts

  • Reactive oxygen species (ROS) disrupt the cellular redox balance, exert cytotoxic effects, and consequently promote the development of various diseases in humans. Previous studies have reported that antioxidants counteract the adverse effects of ROS. Several studies examine the whitening effects of various agents based on their ability to inhibit tyrosinase activity. Tyrosinase is a critical enzyme involved in the synthesis of melanin, which protects the skin against radiation. Various agents exhibiting antioxidant and tyrosinase inhibitory activities have been synthesized. However, these synthetic drugs are associated with toxicity, decreased safety, and poor skin penetration in vivo, which has limited the clinical application of synthetic drugs. This study examined the antioxidant and tyrosinase inhibitory activities of some microalgae. The methanol, dichloromethane, and ethyl acetate extracts of four microalgal species (Tetraselmis tetrathele, Dunaliella tertiolecta, Platymonas sp., and Chaetoceros simplex) were prepared. The physiological and whitening effects of microalgal extracts were investigated by measuring the antioxidant and tyrosinase inhibitory activities. The ethyl acetate extract of D. tertiolecta exhibited the highest antioxidant and tyrosinase inhibitory activities. Future studies must focus on examining the whitening effects of microalgae on cell lines to facilitate the development of microalga-based therapeutics for skin diseases, functional health foods, and whitening agents. Thus, microalgae have potential applications in the pharmaceutical, food, and cosmetic industries.

Inhibition Effect of the Extracts of Trichloma matsutake Mycelia on Tyrosinase Activity (송이[Tricholoma matsutake]에서 분리된 균사 배양액의 Tyrosinase 억제효과)

  • In this study, the optimum conditions for mycelium culture of the mushroom Tricholoma matsutake and the inhibitory effect of the mycelium extracts no tyrosinase activity have been examined. When the extracts of the Tricholoma matsutake mycelia were tested for inhibitory activity on tyrosinase, it was found that the components extracted with ethyl acetate and water showed the highest inhibitory activity. The effect of antioxidants on the growth of mycelium and tyrosinase-inhibiting activity was also investigated. The results showed that tocopherol inhibited the growth in a concentration-dependent manner. In terms of tyrosinase-inhibiting activity, however, tocopherol was found to enhance the inhibitory activity.

The Inhibitory Effects of Nelumbo nucifera Gaertner Extract on Melanogenesis (연자육 추출물의 멜라닌 합성 저해효과)

  • In order to develop new skin whitening agents, we prepared the $CH_2Cl_2$ layer (NGC) and BuOH layer (NGB) of 75% EtOH extract of the Nelumbinis nucifera Gaertner. We measured their tyrosinase inhibitory activity in vitro and melanin synthesis inhibitory activity in B16-F1 melanoma cells. They did not show inhibitory activity against mushroom tyrosinase but showed melanin synthesis inhibitory activity in a dose-dependent manner. In a melanin synthesis inhibition assay, NGC and NGB suppressed melanin production up to 52% and 46% at a concentration of $100{\mu}g/mL$, respectively. To elucidate the mechanism of the inhibitory effects of NGC and NGB on melanogenesis, we measured the expression of melanogenesis-related proteins by western blot assay. As a result, NGC suppressed the expression of tyrosinase, tyrosinase related protein 1 (TRP-1), tyrosinase related protein 2 (TRP-2), phosphorylated cAMP responsive element binding (p-CREB) protein, and microphthalmia associated transcription factor (MITF). And NGB inhibited the protein expression of tyrosinase and MITF, but had no significant effect on TRP-1, TRP-2, and p-CREB expression. Moreover, NGB increased the expression of phosphorylated extracellular signal-regulated kinase (p-ERK). In addition, we examined the inhibitory effect on the glycosylation of tyrosinase. As a result, NGC and NGB inhibited the activity of ${\alpha}$-glucosidase in vitro and the glycosylation of tyrosinase in B16-F1 melanoma cells. From these results, we concluded that NGC and NGB could be used as active ingredients for skin whitening.

Effect of Rubus coreanum Extracts on Tyrosinase Promoter (복분자 추출물이 티로시나아제 프로모터에 미치는 효과)

  • To estimate the inhibitory effect of Rubus coreanum extract on melanin biosynthesis, we tested its inhibitory effects on tyrosinase promoter in B16 mouse melanoma cells. Rubus coreanum extract by methanol had inhibitory effect approximately 10% on tyrosinase promoter at $100{\mu}g/mL$. In the MTT assay, the same extract exhibited very low cytotoxicity under $100{\mu}g/mL$. The fractions of methylene chloride, butyl alcohol and water did not showed the inhibitory effect on tyrosinase promoter, but the fraction of ethyl acetate exhibited inhibitory effect approximately 11% at $100{\mu}g/mL$.

Phenolic Compounds Content and Tyrosinase Inhibitory Effect of Unripe Apple Extracts

  • This study consisted of a quantitative analysis of five phenolic ingredients in differently sized unripe apple extracts, and their tyrosinase inhibitory effects were examined. In the HPLC analysis of phenolic ingredients, small ($4{\pm}1\;g$ per one) unripe apple extracts were observed to have significantly higher quercetin content than larger ($8{\pm}1\;g$ per one) unripe apple and ripe apple extracts. The amount of catechin, epicatechin, p-coumaric acid and chlorogenic acid contents were similar in both the small and large unripe apple extracts. For the results of the tyrosinase assay, small unripe apple extracts provided a potent tyrosinase inhibitory effect, showing 89.2% at 1000 ppm. The tyrosinase inhibitory effects of large unripe apple and ripe apple extracts were weaker than those of the small unripe apple extract. These results suggest that the small unripe apple extract could be useful for de-pigmenting material, while quercetin could be responsible for the potent tyrosinase inhibitory properties of small unripe apple extracts.

The Isolation of the Inhibitory Constitutents on Melanin Polymer Formation from the Leaves of Cercis chinensis (박태기나무의 잎으로부터 피부멜라닌 색소생성 억제성분의 분리)

  • Tyrosinase plays an important role in the process of melanin polymer biosynthesis. Therefore, the enzyme inhibitors have been of great concern as cosmetics to have skin-whitening effects on the local hyperpigmentation. During the search for new inhibitory compounds on melanin polymer biosynthesis from natural sources, MeOH extracts of 589 higher plants were tested for the inhibitory effect on tyrosinase activity by the muschroom tyrosinase assay in vitro. Among plants tested, the leaves of Cercis chinensis exhibited potent inhibitory effect on mushroom tyrosinase activity. Subsequently seven active compounds were isolated from the ethyl acetate soluble part of acetone extract of the leaves of C. chinensis by the activity guided fractionation monitoring the inhibitory effect on tyrosinase activity. Their chemical structures were identified as $kaempferol-3-0-{\alpha}-L-rhamnoside$, quercitrin, $myricetin-3-0-{\alpha}-L-rhamnoside$, myricetin-3-0-(2'-O-galloyl)- ${\alpha}$ -L-rhamopyranoside (desmanthin), (-)-epicatechin-3-0-gallate, (-)-epigallocatechin-3-0-gallate, and methyl gallate on the basis of the speculation of spectral data and chemical reaction. Among the flavonol rhamnosides, myricetin-3-0-(2'-O-galloyl)- -L-rhamnoside(desmanthin) showed most potent inhibitory effect on tyrosinase activity and the structure of B-ring in flavonol moiety was related to the activity. (-)-Epigallocatechin-3-O-gallate having pyrogallol group in flavan-3-ol moiety exhibited more potent inhibitory effect than (-)-epicatechin-3-0-gallate having catechol group in flavan-3-ol moiety on mushroom tyrosinase activity.

  • For the purpose of the development of a skin-whitening agent, Sophora flavescens was evaluated for tyrosinase inhibitory activity and its active principles were identified followed activity-guided isolation. The ethanol extract and dichloromethane fraction from S. flavescens showed significant inhibition of mushroom tyrosinase. From the dichloromethane fraction, three known prenylated flavonoids, sophoraflavanone G, kuraridin, and kurarinone, were isolated. Compared with kojic acid ($IC_50$=20.5 $\mu$M), these compounds possessed more potent tyrosinase inhibitory activity. (omitted)

Anti-melanogenesis Effect of Phenolic Compounds Isolated from Gastrodia elata (천마(Gastrodia elate) 추출물로부터 분리된 페놀성 물질의 멜라닌 생성 억제작용)

  • Melanin pigmentation in human skin is a major defense mechanism against ultraviolet light of the sun, but abnormal pigmentation such as freckles, liver spot could be a serious aesthetic problem. Nearly all studies are mainly concentrated on searching for the materials that have inhibitory activities on tyrosinase. In this work, to isolate phenolic compounds from Gastrodia elata, we purified the extract through solvent fractionation, column chromatography, and recrystallization. They were identified as 4-hydroxybenzyl alcohol 1, bis(4-hydroxyphenyl)methane 2, gastrodin (4-${\beta}$-D-glucopyranosyloxybenzyl alcohol) 3 on the base of spectroscopic evidences. In order to investigate their depigmentation effect, inhibitory activity against mushroom tyrosinase and inhibitory activity of melanin synthesis in B16 melanoma cells were evaluated in vitro. We have found that 4-hydroxybenzyl alcohol 1 and gastrodin (4- ${\beta}$-D-glucopyranosyloxybenzyl alcohol) 3 have no tyrosinase inhibitory activity, but inhibit the melanin synthesis in B16 melanoma cells. Tyrosinase inhibitory activities of bis(4-hydroxyphenyl)methane 2 (IC$\_$50/ = 400 $\mu\textrm{g}$/mL) and butanol fraction (IC$\_$50/ = 46 $\mu\textrm{g}$/mL) were lower/higher than that of arbutin (IC$\_$50/ = 114 $\mu\textrm{g}$/mL), but inhibitory activities of melanin synthesis in B16 melanoma cells were much higher than that of arbutin. Especially, tyrosinase inhibitory activities of isolated phenolic fraction (IC$\_$50/ = 2.37 $\mu\textrm{g}$/mL) from butanol fraction was very higher than that of arbutin (IC$\_$50/ = 114 $\mu\textrm{g}$/mL). Therefore, these results suggest that isolated phenolic compounds from Gastrodia elata have inhibitory activity against mushroom tyrosinase and inhibitory activity of melanin synthesis in 816 melanoma cells in vitro.

  • Inhibitory effect on tyrosinase activity and melanogenesis of the mycelia and mycelial culture broth of Macrolepiota procera were investigated. The methanol fraction of culture broth showed 56.6% of tyrosinase inhibitory activity and its IC50 was 8.78 mg/ml. Using Streptococcus bikiniensis for melanogenesis in vitro, the methanol extract of mycelia showed 94.0% inhibition of melanogenesis.
