A Study on the Application of New Cosmetic Materials of Whitening Effect and the Physiological Activities of Chestnut Inner Shell (율피의 생리활성 몇 미백효과를 이용한 화장품신소재에 관한 연구)

  • Objectives : This is the study of the application as the ingredients of cosmetics through the examination of the function for physiological activity of Chestnut inner shell. Methods : Chestnut inner shell, which had been extracted, concentrated, and freeze drying with water and ethanol, have been used for the experiment. The effects on electronic donating ability, SOD-like activity, xanthine oxidase inhibition, whitening effect, nitric oxide inhibition have been investigated in the physiological activity measurement of function experiment. Results : We used BHA and kogic acid for the comparative. As a result of testing electron donating ability, at over 100ppm of water extract and ethanol extract, BHA showed relatively high donating ability by more than 90%. And as a result of measuring SOD like activity, 1000ppm of water extract showed an effect of 30% and ethanol extract showed an effect of 40%, BHA showed an effect of 30%. In the xanthine oxidase inhibition test, 1000ppm of water extract showed an effect of 70% and ethanol extract showed an effect of 63%, BHA showed an effect of 100%. In the tyrosinase inhibition test, 1000ppm of water extract showed an effect of 55% and ethanol extract showed an effect of 87%, Kogic acid showed an effect of 98%. In the anti-inflammatory test, the water extract and ethanol extract inhibited the generation of nitric oxide. Conclusions : The results indicated that extract of Chestnut inner shell can be used as a natural ingredients with biological function in cosmetics ingredients.

  • In this study, we synthesized Oligo Peptide (Lys-Val-Ala-Arg-Pro: KVARP) and peptide derivatives using Solid Phase Peptide Synthesis(SPPS). KVARP was commonly known to improve whitening of skin. We measured bio-activity of the synthesized compounds. The whitening effect was measured in tyrosinase inhibition and the result showed to be highly effective with 93% inhibition rate at $5000{\mu}g/m{\ell}$ of Geranic-KVARP, on the other hand the IC50 value was $68{\mu}g/m{\ell}$. The wrinkle-reducing effect was measured by elastase inhibition at a concentration of 63% at $400{\mu}g/m{\ell}$ of Salicylic-KVARP, and the IC50 value was $253{\mu}g/m{\ell}$. In the DPPH assay, Caffeic-KVARP showed more than 95% antioxidant activity at $400{\mu}g/m{\ell}$ with high concentration and IC50 value was $31{\mu}g/m{\ell}$. The anti-inflammatory effect of Nitric Oxide inhibition was 67% at $400{\mu}g/m{\ell}$ of Lipoic-KVARP. Therefore, the four types of KVARP derivative that were synthesized from various experiments has shown that it could have potential to be used to develop new medicines, cosmetics as well as in various industries.

  • Objective : In this study, anti-oxidant and cosmeceutical activities of Isatis tinctoria L. extracted from water, ethanol and supercritical fluid condition were confirmed to investigate cosmeceutical activities for utilization as cosmetic ingredient. Methods: Anti-oxidant and cosmeceutical activities were investigated by using electron donating ability, xanthine oxidase, tyrosinase, astringent effect. Result : Isatis tinctoria L. extracts by supercritical fluid, water and ethanol showed good electron donating ability which were 82.7%, 62.6% and 44.8% at the concentration at 1,000ppm, respectively. Xanthine oxidase activity related with purine metabolism was inhibited by ethanol extract about 52.3% at the concentration at 1,000ppm. Tyrosinase inhibition effects, by supercritical fluid extract, ethanol extract and water extract, were 83.3%, 52.9% and 41.2% respectively at 1,000ppm. In the measurement of astringent effect, supercritical fluid extract at the concentration to 5,000ppm showed 85.7% in related activity. The water extract showed 95.9% nitrite scavenging activity at 5,000ppm. Conclusion : According to these results, it is possible that the extract of Isatis tinctoria L. can be used as a new natural material of cosmetic industry.

Anti-oxidative and Whitening Efects of 4'-O-methylalpinumisoflavone Isolated from Fruit of Maclura Tricuspidata Carrière (꾸지뽕나무 열매에서 추출한 4'-O-methylalpinumisoflavone의 항산화 및 미백 효과)

  • The anti-inflammatory effects of 4'-O-methylalpinumisoflavone (OMAI) has been reported in recent years. To develop effective and safe skin whitening agents, we investigated the anti-oxidative and melanogenic effects of OMAI isolated from fruit of Maclura tricuspidata Carrière (Cudrania tricuspidata) in macrophage and melanoma cell lines. In our results, OMAI showed effective superoxide scavenging activity and suppressed production of lipopolysaccharide (LPS)-induced intracellular reactive oxygen species (ROS) in RAW264.7 cells. In addition, α-melanocyte stimulation hormone (MSH)-induced production of melanin was also reduced by OMAI in B16F10 cells. Finally, OMAI significantly inhibited tyrosinase activity in B16F10 cells. These results suggest that OMAI suppressed melanin production via scavenging reactive oxygen species and inhibition of tyrosinase activity.

Inhibitory Effects of Butyl Alcohol Extract from Caesalpinia sappan L. on Melanogenesis in Melan-a Cells (소목의 부탄올 추출물에 의한 Melan-a 세포의 멜라닌생성 억제효과)

  • Melanogenesis is a physiological process resulting in the synthesis of melanin pigments, which play a crucial protective role against skin photocarcinogenesis. The heart wood of Caesalpinia sappan L.(C. sappan) has long been commonly used in Oriental folk medicines to promote blood circulation, and as an emmenagogue, analgesic or anti-inflammatory agent as well as a remedy for thrombosis. From the heartwood, many constituents have been purified and among them, brazilin and hematoxylin are two of the most abundant. This present study was designed to investigate the inhibitory effect of butanol extract from C. sappan on proliferation and melanogenesis in Melan-a cells. After 48 h treatment of these cells with various concentrations of butanol extract, the cells showed a dose-dependent inhibition in their proliferation without apoptotic cell death. Therefore, the growth retardation by the extract may be due to the cell arrest or cell differentiation. We also estimated total melanin content as a final product and activity of tyrosinase, a key enzyme, of melanogenesis in Melan-a cells. The melanin content and tyrosinase activity were deσeased in extract-treated cells in a dose dependent manner compared to control group. The butanol extract also resulted in a decrease of melanin content in ${\alpha}-melanocyte-stimulating$ hormone (MSH)-induced melanogenesis, indicating that butanol extract of C. sappan could be developed as skin whitening components of cosmetics.

Sageretia thea fruit extracts rich in methyl linoleate and methyl linolenate downregulate melanogenesis via the Akt/GSK3β signaling pathway

  • BACKGROUND/OBJECTIVES: Sageretia thea is traditionally used as a medicinal herb to treat various diseases, including skin disorders, in China and Korea. This study evaluated the inhibitory effect of Sageretia thea fruit on melanogenesis and its underlying mechanisms in B16F10 mouse melanoma cells. The active chemical compounds in anti-melanogenesis were determined in Sageretia thea. MATERIALS/METHODS: Solvent fractions from the crude extract were investigated for anti-melanogenic activities. These activities and the mechanism of anti-melanogenesis in B16F10 cells were examined by determining melanin content and tyrosinase activity, and by performing western blotting. RESULTS: The n-hexane fraction of Sageretia thea fruit (HFSF) exhibited significant anti-melanogenic activity among the various solvent fractions without reducing viability of B16F10 cells. The HFSF suppressed the expression of tyrosinase and tyrosinase-related protein 1 (TRP1). The reduction of microphthalmia-associated transcription factor (MITF) expression by the HFSF was mediated by the Akt/glycogen synthase kinase 3 beta ($GSK3{\beta}$) signaling pathway, which promotes the reduction of ${\beta}-catenin$. Treatment with the $GSK3{\beta}$ inhibitor 6-bromoindirubin-3'-oxime (BIO) restored HFSF-induced inhibition of MITF expression. The HFSF bioactive constituents responsible for anti-melanogenic activity were identified by bioassay-guided fractionation and gas chromatography-mass spectrometry analysis as methyl linoleate and methyl linolenate. CONCLUSIONS: These results indicate that HFSF and its constituents, methyl linoleate and methyl linolenate, could be used as whitening agents in cosmetics and have potential for treating hyperpigmentation disorders in the clinic.

Study of Fermentation Extract Made from Rice Bran and Dendropanax on the Whitening Effects in B16F10 Cell Line (미강·황칠 발효추출물이 B16F10 세포의 미백에 미치는 효과)

  • The purpose of this study is to research the whitening effects of fermentation extract made from the Rice bran and Dendropanax(FRD) Fermentation conditions were as follows; 1) Dendropanaxand and Rice bran were blended in a ratio of 1 to 1, 2) a weight of sugar was 10% of the total weight, 3) an amount of enzyme was 0.1%, and 4) a temperature was 20℃. It has been fermented for 90 days. In order to observe the whitening effects of FRD, the author measured the cell viability and the inhibition rate of the melanin biosynthesis, the activity of tyrosinase and SOD (superoxide dismutase) in malignant melanoma, B16F10 cells. As a result, FRD significantly inhibited the cell viability of B16F10 in more than 500 ㎍/㎖. FRD significantly suppressed the generation of melanin, and that induced by α-melanocyte stimulating hormone (α-MSH) in more than 1,000 ㎍/㎖. FRD significantly decreased the activity of tyrosinase and that induced by α-melanocyte stimulating hormone (α-MSH) in more than 500 ㎍/㎖. FRD did not changed the activity of SOD in dose dependent manner. Therefore, the author considered that the fermentation extract made from a Rice bran and Dendropanax will be able to produce high value-added products, if used as a commercial. Study on the Physiological Activities of Cleyera japonica Extract (비쭈기 나무(Cleyera japonica) 추출물의 생리활성에 대한 연구)

  • In this study, we investigated the applicability of functional materials by examining a variety of physiological activities with the extract of Cleyera japonica leaf. Cleyera japonica extract showed a low cytotoxicity against murine melanoma B16F10 cells. In little or no cytotoxicity at concentrations, we showed that the treatment with Cleyera japonica extract resulted in a significant increase in the DPPH radical scavenging activity (IC50, 22.90 ㎎/L), similar to ascorbic acid (IC50, 18.65 ㎎/L) and anti-microbial activities against Bacillus subtilis, Escherichia coli, and Candida albicans. In particular, anti-microbial activities against Gram-positive bacteria was high. These results suggest that Cleyera japonica extract could be used as a natural preservative. Additionally, Cleyera japonica extract showed the inhibition of tyrosinase activity (IC50, 178.90 ㎎/L), similar to kojic acid (IC50, 89.13 ㎎/L) and decreased melanin content (IC50, 101.90 ㎎/L) higher than the control arbutin level (IC50, 100.65 ㎎/L), especially. Therefore, these results indicate that Cleyera japonica extract may be an effective material for functional cosmetics such as skin whitening materials.

Studies on Skin Anti-aging Efficacy of Hydrolysate from Phellinus igniarius (상황버섯 가수분해물의 피부 항노화 효능에 대한 연구)

  • Objective : The object of this study was to assess the efficacy of hydrolysate from Phellinus igniarius(HPI) on anti-aging activities in vitro measurement and mini clinical study performed on 5 subjects. Methods : To evaluate skin anti-aging efficacy of HPI, 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical scavenging activity, type I collagen synthesis, inhibition of nitric oxide(NO) production, inhibiton of tyrosinase, hyaluronan synthase(HAS)2, 3 mRNA expression were measured in vitro. Also, mini clinical study of skin hydration was performed on 5 subjects using HPI in distilled water(DW) and 1,3-butylene glycol diluted solution(30% in DW). Results : 1. DPPH radical scavenging activity of HPI was increased in a dose-dependant. 2. Type I collagen synthesis was increased in 50, 100 and 500㎍/㎖ of HPI. 3. NO production was not inhibited in all concentrations of HPI. 4. Tyrosinase was inhibited in 500, 1000, 2500 and 5000㎍/㎖ of HPI. 5. HAS2 mRNA expression was increased in 50, 100, 150 and 200㎍/㎖ of HPI, HAS3 mRNA expression was increased in 100, 150, and 200㎍/㎖ of HPI. 6. In the mini clinical study of 5 subjects, there was a difference in skin hydration over time for each solutions, but it was not statistically siginificant. Conclusions : HPI increased DPPH radical scavenging activity, type I collagen synthesis, and HAS2, HAS3 mRNA expression. HPI also suppressed tyrosinase. The findings of this study suggest that HPI can be used as an skin anti-aging material.

Application of Isolated Tyrosinase Inhibitory Compounds from Persimmon Leaves (감나무 잎으로 부터 분리한 tyrosinase 억제물질의 응용)

  • Total phenolic content was the highest in 60% ethanol extracts at 21.91 mg/g, and inhibitory activity against tyrosinase of 60% ethanol extracts was higher than ethanol extracts of other concentration. The inhibitory compounds against tyrosinase from Persimmon leaves were purified using Sephadex LH-20, MCI-gel CHP-20 column chromatography with gradient elution. Two purified compounds were isolated as a result. The chemical structures of each compound were determined and identified using $^1H$-NMR and $^{13}C$-NMR, FAB-Mass. The compounds were confirmed as (+)-gallocatechin and prodelphinidin B-3. The tyrosinase inhibitory activities of purified (+)-gallocatechin and prodelphinidin B-3 were 29.5 and 40.2%, respectively. The inhibitory activities of (+)-gallocatechin and prodelphinidin B-3 against melanin biosynthesis in melanoma cell were 32.5 and 46.7%. The safety of essence with tyrosinase inhibitory compounds from persimmon leaves was also assessed by various safety profiles. First, changes in pH (4.90~4.95) and viscosity (23,000~26,000 cP) was not detected for 60 days. Essence also showed stability against temperature and light for 60 days. All these findings suggest that extracts from persimmon leaves have a great potential as a cosmetical ingredient with a potent whitening effect.
