작약 레드참 꽃잎의 이온화원-푸리에 변환 질량분석과 기능성 연구 (Fourier Transform Ion Cyclotron Resonance (FT-ICR) MASS Spectrophotometric Analysis of Flower Petal from Paeonia lactiflora cv. ‘Red Charm’ and Evaluation of its Functional Activity)

  • Little attention has been paid to the functional aspect of the flower petal of Paeonia lactiflora, compared to that of its root. To determine the components of flower petal of Paeonia lactiflora, we conducted the Fourier transform ion cyclotron resonance (FT-ICR) MASS spectrophotometric analysis. We detected the 24 different types of ingredients from the 70% ethanol extracts of flower petal of peonia lactiflora cv. ‘Red Charm’. The main compounds were quercetin glucopyranosides, methyl gallate, paonioflolol and kaemperol glucopyranosides. We further tested its functional activity. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity of the extracts was 87.9-90.4% at 0.1mg/ml. This result showed that these flower extracts have approximately 5-fold stronger antioxidant potential than a previous report with root extracts (Bang et al. 1999). The result of tyrosinase inhibition assay of Paeonia lactflora extract was almost similar to that of arbutin except significantly higher effect in the coral sunset extract at 0.1% concentration. Hyaluronidase inhibition assay showed 76.5% inhibition at 5% concentration of this flower extract, indicating that Peaonia lactiflora flower extracts have the major anti-inflammatory, anti-oxidant and brightening effects. Taken together, these results suggest these three Paeonia lactiflora species extracts might provide the basis to develop a new natural brightening agent.

Tyrosinase Inhibitors Isolated from the Edible Brown Alga Ecklonia stolonifera

  • Extracts from seventeen seaweeds were determined for tyrosinase inhibitory activity using mushroom tyrosinase with L-tyrosine as a substrate. Only one of them, Ecklonia stolonifera OKAMURA (Laminariaceae) belonging to brown algae, showed high tyrosinase inhibitory activity. Bioassay-guided fractionation of the active ethyl acetate (EtOAc) soluble fraction from the methanolic extract of E. stolonifera, led us to the isolation of phloroglucinol derivatives [phloroglucinol (1), eckstolonol (2), eckol (3), phlorofucofuroeckol A (4), and dieckol (5)]. Compounds 1~5 were found to inhibit the oxidation of L-tyrosine catalyzed by mushroom tyrosinase with $IC_{50}$ values of 92.8, 126, 33.2, 177, and 2.16 ${\mu}g$ /mL, respectively. It was compared with those of kojic acid and arbutin, well-known tyrosinase inhibitors, with $IC_{50}$ values of 6.32 and 112 ${\mu}g$ / mL, respectively. The inhibitory kinetics analyzed from Lineweaver-Burk plots, showed compounds 1 and 2 to be competitive inhibitors with $K_i$ of $2.3{\times}10^{-4}\;and\;3.1{times}10^{-4}$ M, and compounds 3~5 to be noncompetitive inhibitors with $K_i$ of $1.9{\times}10^{-5},\;1.4{\times}10^{-3}\;and\;1.5{\times}10^{-5}$ M, respectively. This work showed that phloroglucinol derivatives, natural compounds found in brown algae, could be involved in the control of pigmentation in plants and other organisms through inhibition of tyrosinase activity using L-tyrosine as a substrate.

한방 생약재 발효액의 항산화 활성 및 tyrosinase 저해 활성 (Tyrosinase Inhibition Activity and Antioxidant Capacity by Fermented Products of Some Medicinal Plants)

  • 국내에서 자생하고 있는 한방 생약재의 생리활성 물질을 이용한 기능성 미백 화장품 소재 개발을 위한 기초연구의 일환으로 40종의 한방 생약재 열수 추출물을 상황버섯 균사체로 발효시켜 얻은 발효 한방액의 phenolics, flavonoids, minerals, decursin 및 decursinol angelate 함량과 항산화 및 tyrosinase 저해 활성을 측정 하였다. 페놀성 화합물 함량은 추출물에서 Corydalis turtschaminovill 0.24%~Syringa velutina 11.42% (건물기준) 범위 였으며, 발효 한방액에서는 Poria cocos 0.18%~S. velutina 12.27% 범위였다. 한방 생약재 추출물의 코직산 함량은 Poria cocos Sclerotium 0.02~S. velutina 9.67 mM 범위였으며, 발효액에서는 P. cocos 0.33~S. velutina 10.32 mM 범위였다. 특히 Syringa velutina은 높은 농도의 페놀성 화합물과 코직산을 함유하고 있었는데 추출물 보다는 발효액에서 더 높았다. 높은 항산화 활성(>60%)은 A. sessiliflorum, Citrus nobillis 및 Angelica gigas의 추출물과 A. sessiliflorum의 발효액에서 다른 한방 생약재 보다 높게 나타났다. Tyrosinase의 높은 저해 활성(>50%)은 Morus alba, Glycyrrhiza glabra 및 Rubus coreanus 추출물과 G. glabra, Cnidium officinale 및 S. velutina 발효액에서 높았다. 이상의 실험 결과 40종의 한방 생약재 중에서 G. glabra, C. officinale 및 S. velutina 3종은 발효에 의해 tyrosinase 저해 활성과 코직산이 증가함으로서 이를 활용한 기능성 미백 화장품 개발에 적용할 수 있는 기초 자료를 제공해주는 데 유용하게 사용될 것이다.

정공피 추출물의 항산화 활성 및 미백효과에 관한 연구 (Antioxidant and Whitening Effects of Sorbus commixta HEDL Cortex Extract)

  • 본 연구에서는 정공피 물 추출물의 미백 효과를 확인하기 위하여 항산화 효능을 검증하였고, 멜라닌 생성의 첫 단계인 tyrosinase 활성, mouse melanoma B16BL6 세포 생존율 및 TRP-1, TRP-2 mRNA 발현 정도를 측정하였다. 정공피 물추출물은 DPPH 및 hydroxy 라디칼 소거 작용을 통하여 높은 항산화 효능을 나타내었고, tyrosinase, TRP-1, TRP-2의 활성을 효과적으로 억제하는 것을 확인하였다. 이는 정공피 물 추출물이 직접적으로 B16BL6 melanoma 세포의 tyrosinase 단백질의 활성을 저해시킴으로써 멜라닌 생성을 억제하는데 효과적임을 의미한다. 결론적으로 정공피 추출물은 항산화 활성과 미백 효과가 우수하여 기능성 화장품의 천연 소재로서 활용 가능성이 매우 높은 것으로 판단된다.

인간 표피 멜라닌 세포에서의 단삼 추출물의 미백효과에 관한 연구 (Whitening Effect of Salvia miltorrhiza Bunge Water Extract in Human Epidermal Melanocyte)

  • 본 연구에서는 단삼 열수 추출물의 human epidermal melanocye (HEM) 세포에서의 미백활성을 측정하였다. 버섯유래의 tyrosinase 저해능 측정결과 $1,000{\mu}g/mL$의 농도에서 42%의 저해율을 나타내었으며, HEM 세포내의 tyrosinase, 멜라닌 생합성 저해능은 $5{\mu}g/mL$에서 26, 25%의 저해능을 나타내었다. 단삼 추출물의 미백과 관련된 tyrosinase, tyrosinase related protein 1 (TRP-1), tyrosinase related protein 2 (TRP-2), microphthalmia associated transcription factor (MITF)의 단백질 및 유전자 양이 감소함을 나타내었다. 또한 그 상위 단계인 cAMP 발현이 $5{\mu}g/mL$ 농도에서 41% 감소하는 것을 확인하였다. 본 실험을 통하여 단삼 추출물이 cAMP를 효과적으로 억제함으로써, 이를 통해 tyrosinase 활성뿐만 아니라 TRP-1, TRP-2 그리고 MITF 발현을 효과적으로 저해 한다는 사실을 확인하여 기능성 미백 화장품 소재로서의 활용 가능성을 제시하고자 한다.

홍화자, 향부자, 형개 추출물의 미백효과에 관한 연구 (A Study on the Depigmenting Effect of Carthamus tinctorius Seed, Cyperus rotundus and Schizonepeta tenuifolia Extracts)

  • 본 연구에서는 3가지 홍화자, 향부자, 형개 추출물의 미백효과를 연구하였다. 멜라닌 생성 첫 단계인 tyrosinase 억제활성과 멜라닌 생성 억제효과를 측정한 결과, 홍화자 에탄올 추출물이 tyrosinase 활성과 B16F10 melanoma 세포의 멜라닌 생성 억제하였다. 그 결과, 홍화자 추출물은 B16F10 melanoma 세포에서 melanogenesis 따른 tyrosinase 형성 억제에 따른 멜라닌 합성 관련 인자 MITF, tyrosinase, TRP-1, TRP-2 의 발현을 억제함에 따라 홍화자 추출물의 미백효과를 확인하였다. 따라서 홍화자는 미백효과를 가진 천연 기능성 재료로서 가능성이 매우 높은 것으로 판단된다.

Fermented Unpolished Black Rice (Oryza sativa L.) Inhibits Melanogenesis via ERK, p38, and AKT Phosphorylation in B16F10 Melanoma Cells

  • Melanin is a major factor that darkens skin color as one of the defense systems to prevent the harmful effects of UV light. However, darkened skin from the localized or systemic accumulation of melanin is viewed in many cultures as an esthetic problem. Consequentially, searching for anti-melanogenic agents from natural sources is very popular worldwide. Previous screening of fermented rice products, obtained from various rice cultivars fermented with different sources of loog-pang (Thai traditional fermentation starter), revealed that the highest ability to reduce the melanin content in B16F10 melanoma cells was from unpolished black rice fermented with a defined starter mixture of microbes isolated from loog-pang E11. The aim of this study was to investigate the mechanism of the fermented unpolished black rice (FUBR) on the inhibition of melanogenesis in B16F10 melanoma cells. The strongest reduction of cellular melanin content was found in the FUBR sap (FUBRS). The melanin reduction activity was consistent with the significant decrease in the intracellular tyrosinase activity. The FUBRS showed no cytotoxic effect to B16F10 melanoma or Hs68 human fibroblast cell lines. It also significantly reduced the transcript and protein expression levels of tyrosinase, tyrosinase-related protein 1 (TYRP-1), TYRP-2, and microphthalmia-associated transcription factor. Furthermore, it induced a significantly increased level of phosphorylated ERK, p38 and Akt signaling pathways, which likely contributed to the negative regulation of melanogenesis. From these results, a model for the mechanism of FUBRS on melanogenesis inhibition was proposed. Moreover, these results strongly suggested that FUBRS possesses anti-melanogenesis activity with high potential for cosmeceutical application as a skin depigmenting agent.

도화(Prunus persica Flos) 분획물의 미백 및 주름개선 효과 (Whitening and Anti-wrinkling Effects of Fractions from Prunus persica Flos)

  • 본 연구는 도화를 70% 아세톤 추출하여 sephadex LH-20으로 분리한 후 8개의 분획물을 얻을 수 있었다. 미백효과 측정으로 tyrosinase 저해활성을 측정한 결과 분획물 8의 경우 1,000 ppm에서 92.2%를 나타내었으며, 멜라노마 세포에 의한 tyrosinase 저해활성은 100 ppm에서 63.4%, 멜라닌 생합성은 저해능은 100 ppm에서 71.7%의 효과를 나타내어 분획물 8의 효능이 가장 우수하였다. 주름개선 효과 측정으로 elastase 저해활성을 측정한 결과 분획물 5, 7이 1,000 ppm에서 각각 71.4, 74.5%의 저해활성을 나타내었으며, collagenase 저해활성과 collagen 생합성량을 측정한 결과 분획물 8이 100 ppm에서 80.0% 이상의 저해활성을 나타내었다. 이상의 결과로 미루어 보아 도화 분획물의 미백 및 주름개선 효과가 있음을 확인할 수 있었고, 기능성 화장품 소재로서의 가능성을 확인할 수 있었다.

사간(射干) 에탄올추출물이 피부생리활성에 미치는 영향 (Effect of Ethanol Extract of Belamcandae Rhizoma on Dermal Bioactive Properties)

  • Objective : This experimental study was preformed to investigate the effect of ethanol extract of Belamcandae Rhizoma (EBR) on dermal bioactive properties related to cosmeceuticals such as radical scavenging activity and depigmentation. Methods : The free radical scavenging activity of EBR was assayed in cell free systems using 1,1-diphenyl-2-picrylhydrazyl (DPPH). Antibacterial potency was measured by the size of inhibition zone with change of volume. Cell viability was measured by MTT assay and tyrosinase activity was assessed using L-DOPA. Results : EBR showed slightly radical scavenging activity and protected against UVB-induced cell cytotoxicity. Futhermore, EBR showed antibacterial activities on S. epidermidis and S. aureus. Although the proliferation of B16/F10 cells was decreased by EBR, necrosis was not revealed. EBR augmented tyrosinase activity and dendrite outgrowth in B16/F10 cells. Conclusions : These results suggest that EBR has antioxidative & antibacterial activities, and stimulate melanogenesis.

Antimelanogenic effect of ginsenoside Rg3 through extracellular signal-regulated kinase-mediated inhibition of microphthalmia-associated transcription factor

  • Background: Panax ginseng has been used to prolong longevity and is believed to be useful for improving skin complexion. Ginsenosides are the most active components isolated from ginseng, and ginsenoside Rg3 (G-Rg3) in particular has been demonstrated to possess antioxidative, antitumorigenic, and anti-inflammatory properties. The aim of this study was to examine the ability of G-Rg3 to inhibit melanogenesis. Methods: The effects of G-Rg3 on melanin contents and the protein levels of tyrosinase, microphthalmia-associated transcription factor (MITF), and tyrosinase-related protein 1 (TRP1) were evaluated. Melanogenesis-regulating signaling molecules such as Akt and extracellular signal-regulated kinase (ERK) were also examined to explore G-Rg3-induced antimelanogenic mechanisms. Results: G-Rg3 was found to significantly inhibit the synthesis of melanin in normal human epidermal melanocytes and B16F10 cells in a dose-dependent manner. The activity of cellular tyrosinase and the expression of MITF, tyrosinase, and TRP1 were all reduced, whereas ERK was strongly activated. PD98059 (a specific inhibitor of ERK) attenuated the G-Rg3-induced inhibition of melanin synthesis and tyrosinase activity. Conclusion: Taken together, these results showed that G-Rg3 induces the activation of ERK, which accounts for its antimelanogenic effects. G-Rg3 may be a promising safe skin-whitening agent, adding to the long list of uses of P. ginseng for the enhancement of skin beauty.
