  • A tyrosinase inhibitor, Lonjapoin, was isolated from the ethanolic extracts of Flos Lonicera by activity-guided fractionation. Lonjapoin ($IC_{50}$/; 2.07 $\mu\textrm{g}$/mL) was found to be potentially as effective inhibitor of production of melamin. Flos Lonicera (Jinyinhua) is the dry flower buds of Lonicera japonica. Thunb. It is used as an antibacterial and antiphlogistic agent in the treatment of abscess, laryngeal catarrh, erysipelas, dysentery, cold and fever. In our continuing search for natural source as cosmetic ingredients from Chinese medicinal herbs and found the extracts of Flos Lonicera showed the significant tyrosinase inhibition activity. In this study the active constituent of Lonicera japonica. obtained with the process of tyrosinase activity assay and column chromatography.

Antioxidant Activities and Tyrosinase Inhibitory Effects of Different Extracts from Pleurotus ostreatus Fruiting Bodies

  • We evaluated the antioxidant activity and tyrosinase inhibitory effects of Pleurotus ostreatus fruiting bodies extracted with acetone, methanol, and hot water. The antioxidant activities were tested against $\beta$-carotene-linoleic acid, reducing power, 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity, and ferrous chelating ability. Furthermore, phenolic acid and flavonoid contents were also analyzed. The methanol extract showed the strongest $\beta$-carotene-linoleic acid inhibition as compared to the other exracts. The acetone extract (8 mg/mL) showed a significantly high reducing power of 1.54 than the other extracts. The acetone extract was more effective than other extracts for scavenging on 1,1-diphenyl-2-picrylhydrazyl radicals. The strongest chelating effect (85.66%) was obtained from the acetone extract at 1.0 mg/mL. The antioxidant activities of the extracts from the P. ostreatus fruiting bodies increased with increasing concentration. A high performance liquid chromatography analysis detected seven phenolic compounds, including gallic acid, protocatechuic acid, chlorogenic acid, naringenin, hesperetin, formononetin, and biochanin-A in an acetonitrile and 0.1 N hydrochloric acid (5 : 1) solvent extract. The total phenolic compound concentration was $188{\mu}g$/g. Tyrosinase inhibition of the acetone, methanol, and hot water P. ostreatus extracts increased with increasing concentration. The results revealed that the methanol extract had good tyrosinase inhibitory ability, whereas the acetone and hot water extracts showed moderate activity at the concentrations tested. The results suggested that P. ostreatus may have potential as a natural antioxidant.

역미셀계를 이용한 지용성 식물체 추출물의 tyrosinase 저해효과 분석 (Ability of Lipophilic Extract Obtained from Plants to Inhibit Tyrosinase Activity in Reverse Micelles)

  • 체내에서 멜라닌 생합성을 촉매하는 tyrosinase에 대한 천연의 지용성 저해제 발굴을 위하여, isooctane/AOT(100 mM)/인산완충용액(20 mM, pH 8.0)으로 구성되고 tyrosinase(105.3 units/mL)와 3,4-dihydroxyphenyl-alanine(0.18 mM)을 함유한 역미셀계에서 식물체 75종의 petroleum ether 추출물이 tyrosinase 반응을 억제하는 효과를 분석하였다. 조사대상 식물체중에서 마늘이 in vitro 멜라닌 합성 반응을 완전히 저해하였고(100%), 모과, 고구마, 양파, 무순, 사과의 저해활성이 60% 이상이었다. 한약재와 향신료 중에서는 로즈마리, 고수, 계피, 산사자, 측백엽, 오매, 두충, 박하, 정향이 60% 이상의 저해능을 보였다. 그리고 각 식물체의 tyrosinase 저해효과와 더불어 추출수율을 함께 고려했을 때, 단위 원재료로부터 tyrosianse 저해제 생산을 많이 할 수 있는 소재를 선정할 수 있었다. 본 연구에서는 역미셀계를 이용함으로서 기존에는 분석이 어려웠던 지용성 물질의 tyrosinase 저해효과를 측정할 수 있었다.

영지버섯, 여주, 쓴메밀 및 혼합 시료의 메탄올 추출물의 생리활성 (Biological Activity of Methanolic Extract from Ganoderma lucidum, Momordica charantia, Fagopyrum tataricum, and Their Mixtures)

  • 영지버섯, 여주, 쓴메밀 및 이들 혼합 시료의 70% 메탄올 추출물을 이용하여 생리활성물질(폴리페놀 화합물 및 플라보노이드) 농도와 항산화(DPPH radical 소거 활성, 생체막 지질과산화, Fe/Cu 환원력), ${\alpha}$-glucosidase 저해, tyrosinase 저해 및 항혈전 활성에 대하여 검토하였다. 추출 수율과 폴리페놀 화합물 및 플라보노이드의 함량은 혼합 추출물에서 가장 높았다. 또한 혼합 추출물에서 ${\alpha}$-glucosidase 저해, tyrosinase 저해, 항산화, 환원력 및 항혈전 활성은 혼합 추출물에서 가장 높았으며, 이러한 효과는 처리 농도 의존적 이였다. 이상의 실험 결과에서 영지버섯, 여주, 쓴메밀의 혼합 메탄올 추출물은 폴리페놀 화합물과 플라보노이드를 많이 함유함으로서 높은 ${\alpha}$-glucosidase 저해 및 항산화 활성과 관련성이 높아 보여 항당뇨 건강 기능성식품의 소재 개발에 활용될 가능성이 높아 보인다.

새삼, 실새삼 및 갯실새삼 추출물이 Clone M-3 세포주의 Melanin 생합성 및 Tyrosinase 활성에 미치는 영향과 세포독성 및 항산화효과 (Inhibitory Effects on Melanin Biosynthesis and Tyrosinase Activity; Cytotoxicity in Clone M-3 and Antioxidant Activity by Cuscuta japonica, C. australis, and C, chinensis Extracts)

  • Water extracts, ethanol extracts, and juice of Cusuta japonica, C, australis, and C. chinensis were prepared, and their cytotoxicity, antioxidant activity and inhibitory effects on tyrosinase activitiy and melanin biosynthesis were estimated by using melanoma Clone M-3. From this study; the following conclusions were attained. Extracts of Cuscuta japonica, C. australis, and C. chinensis showed noticeable cytotoxicity except ethanol extracts from the stem of C. australis. A maximual cytotoxicity was observed with tile ethanol extract from the seed of C, australis (87.39%). While the ethanol extract from the seed of C. japonica (91.88%) showed the most pronounced inhibitory effect on melanin biosynthesis, the water extract from the stem of C. japonica (1.05%) possessed very little inhibitory effect. The most inhibitory effect on tyrosinase activity was observed with the water extract from the stem of C. australis (76.67%). Howeverr the water extract from the stem of C. japonica showed a very poor effect on the inhibition of tyrosinase activity All the preparations, except extracts from the seed of C. australis were able to remove reactive oxygen species (ROS) in a dose-dependent manner. The juice of C. japonica demonstrated the strongest activity (59.02%).

국화류 추출물 및 분획물의 생리활성 비교연구 (Comparison of Biological Activities on Extracts and Fractions in Compositae Plants)

  • 본 연구는 우리나라 전역에 자생하는 국화류3종 학슬, 희렴, 한련초를 대상으로 MeOH 80% 추출물 및 헥산, 클로로포름, 에틸아세테이트, 부탄올 및 물분획을 이용한 항균활성, 항산화 및 tyrosinase 저해활성효과에 대한 연구를 통해 기능성 화장품소재로서의 이용가능성을 확인하고자 하였다. 연구결과 항균활성의 경우 MeOH 80% 추출물에서는 한련초와 용매분획 중에서는 학슬의 헥산분획, 희렴의 클로로포름분획, 에틸아세테이트 분획, 한련초의 클로로포름분획을 중심으로 항균력을 나타내었다. 항산화 활성의 경우 3종 모두 용매분획 중 에틸아세테이트, 부탄올 분획에서 90%이상의 효과를 나타내었다. Tyrosinase 저해활성의 경우 희렴과 학슬의 에틸아세테이트 분획물에서 우수한 활성을 나타내었다. 이상의 국화류3종 MeOH 80%추출물 및 분획물에서 항산화 및 미백작용이 우수하므로 기능성화장품소재 개발 활용이 가능할 것으로 생각된다.

건칠의 항주름 효과와 미백효과 연구 (The effect of wrinkle care and whitening by Rhus verniciflua Stokes)

  • Objectives : Recently, the demands for the effective and safe depigmentating and anti-aging agents of the skin have increased due to the medical, pharmaceutical and cosmetic reasons. The aim of this study is to search new materials from the traditional herbal medicines which inhibit the aging process of skin in vitro.Methods : Human dermis cell (HS68) was used to test the effect of Rhus verniciflua Stokes (RVS). 80% ethanol or water extracts were screened for their inhibitory activities against elastase. Elastase inhibition effect was tested by microplate reader instrument. And MMP-1 suppression effect of RVS was tested by western blot. These cells were investigated the viability by MTS assay. And also the inhibition effect of tyrosinase by RVS was tested.Results : RVS (final concentrstion 1 ㎎/㎖ appeared over 30% of inhibition of elastase activity. So we are investigated anti wrinkle effects of Rhus verniciflua Stokes look through MMP-1 inhibition activity, also Extracts of RVS showed higher anti-tyrosinase activity than arbutin as final concentration 1 mg/ml. These results suggest that herbal medicines could be strong potential sources of inhibition of anti-aging and whitening effects for the skin.Colclusions : RVS was the best suppressor candidate of elastase activity among other control oriental drugs. It was found that RVS did not have toxicity to cells. We found proper concentration of RVS to treat in HS68 culture and investigated not only wrinkle care effect but also whitening effect of RVS.

Antioxidant Activity and Biological Activity of Specialty Barley (Hordeum vulgare L.) Extracts

  • Specialty barley extracts were prepared and investigated for its antioxidant activity and biological activity. Hunter $L^*$ values of the Iksan 86 extracts had higher than that of the Iksan 87 and Zasoojeongchal extracts. The extraction yields of Iksan 86, Iksan 87, and Zasoojeongchal was 8.08, 6.62, and 7.30%, respectively. The contents of total phenolic compounds of the Iksan 86, Iksan 87, and Zasoojeongchal extracts were 16.24, 15.51, and 13.95 GAE mg/g of sample, respectively. The DPPH radical scavenging activity of Iksan 86, Iksan 87, and Zasoojeongchal extracts were 50.00, 33.27, and 7.56% at a 500 ppm, respectively. The samples showed an inhibition of xanthin oxidase. ACE inhibition effect of specialty barley extracts, Iksan 86, Iksan 87, and Zasoojeongchal, was 39.81, 41.06, and 27.78%, respectively. Tyrosinase inhibition rates (%) of Iksan 86, Iksan 87, and Zasoojeongchal extracts were 26.21, 24.57, and 20.00%, respectively. Results indicated that specialty barley extracts possesses various biological activities including antioxidative capacity, xanthin oxidase inhibition activity, angiotensin converting enzyme inhibition activity, and tyrosinase inhibition activity.

효모에서 분리한 멜라닌 생성 억제 물질의 작용 기전 (Mechanism of Melanogenesis Inhibition by Melanoston Isolated from Yeast)

  • 본 연구에서는 효모에서 분리한 melanoston이라고 명명된 멜라닌 생성을 억제하는 물질의 작용 기전을 밝히기 위한 것이다. $\alpha$-MSH를 처리한 B16 melanoma 세포에서 melanoston은 tyrosinase mRNA 발현양을 10% 정도 저해되는데 그쳤으며 western blotting을 이용한 단백질 측정에서도 이와 비슷한 정도의 단백질 생성 억제를 보였다. 그러나 B16 세포 배양액에 melanoston을 첨가할 경우 세포내 tyrosinase 활성이 33% 까지 감소되는 것으로 나타나 metanoston이 tyrosinase inhibitor는 아니지만 세포내 tyrosinase 활성화 (activation) 과정을 억제하는 것으로 추측할 수 있었다. 또한 광학 현미경을 이용한 morphology 관찰에서 $\alpha$-MSH를 처리한 세포에서는 많은 dentrite가 형성되면서 세포분화가 일어나는 반면 melanoston를 처리한 경우에는 dendrite가 감소하면서 세포형태가 대조군과 비슷하게 회복 되는 것을 알 수 있었다. 또 FITC-anti-tyrosinase-Ab를 이용한 형광 염색을 통해서는 $\alpha$-MSH만 처리한 세포에서는 tyrosinase의 분포가 dendrite를 포함한 세포 전체로 퍼져나가는 것을 관찰할 수 있었고 $\alpha$-MSH와 melanoston을 동시에 처리한 세포에서는 대조군과 비슷하게 tyrosinase가 핵 주변에서만 관찰되어 melanoston이 B16 melanoma 세포의 분화과정에서 이를 억제하는 효과를 주고 있음을 알 수 있었다. 이상의 결과들을 종합해 볼 때 melanoston은 $\alpha$-MSH에 진행되는 B16 세포의 분화를 억제하고 이 과정에서 멜라닌 생성의 주된 효소인 tyrosinase의 활성화를 억제하며 결과적으로는 멜라닌 생성을 저해하는 것으로 사료된다.

수산폐기물 재활용을 위한 미더덕껍질 용매 추출물의 생리활성 탐색 (Screening of Biological Activity of Solvent Extract from Styela clava Tunic for Fishery Waste Recycling)

  • Styela clava tunic is generated in large amounts as a waste from S. clava processing plants and causes environmental problem. Although biological activities of S. clava were reported by many investigators, study on S. clava tunic was little. In this study, therefore, tyrosinase inhibition and antioxidative activities of extracts from S. clava tunic using different solvent were investigated for recycling of the fishery waste. Among extraction methods tested, autoclaved extraction (25.7%) and hot water extraction (18.2%) appeared to be effective for extraction. The highest total phenolic content was 46.6 mg/g in autoclaved extract while the highest flavonoid content was 23.0 mg/g in chloroform extract. All extracts possessed tyrosinase inhibition activity and the inhibition activity was concentration-dependent. Inhibition concentration ($IC_{50}$) against tyrosinase activity was $0.36{\times}10^4$ mg/ml in ethanol extract, $0.11{\times}10^3$ mg/ml in acetone extract and 0.27 mg/ml in n-butanol extract. Among extracts tested, hot water and autoclaved extracts displayed higher antioxidative activity than organic solvent extracts. Therefore, our data suggest that extract from S. clava tunic may potential candidate for cosmetic product with whitening effect and medicine for diseases caused by various oxidative stresses.
