Effects of Hyunggyeyungyotang and Kamihyunggyeyungyotang administration on the anti-inflammation, analgesia and anti-allergic reaction in mice (형개연교탕(荊芥連翹湯)과 가미형개연교탕(加味荊芥連翹湯)이 소염(消炎).진통(鎭通) 및 항(抗)알레르기에 미치는 영향(影響))

  • Hyunggyeyungyotang has been used for treatment of sinusitis and otitis media in oriental medicine since ancient times. It is reported that Hyunggyeyungyotang has good effects on inflammatory and allergic diseases of otorhinolaryngology in clinical medicine. Kamihyunggyeyungyotang was made by adding several herbs to Hyunggyeyungyotang which has such good effects. To investigate the effects of Hyunggyeyungyotang and Kamihyunggyeyungyotang on inflammatory, algesic and allergic diseases, the author examined the analgesic effect by acetic acid reaction, studied the anti-inflammatory effect through the experiments of the protein thermo-denaturation and circumscribed edema. Besides researched the anti-allergic effect through the vascular permeability response to Chemical Mediator and the delayed type hypersensitivity response to Picryl Chloride. The obtained results were as follows ; 1. In the analgesic effect of Hyunggyeyungyotang and Kamihyunggyeyungyotang extract by acetic acid method, both of the sample groups showed the analgesia, but didn't show useful effect. 2. In the anti-inflammatory effect on the protein thermo-denaturation, the sample groups revealed the inhibitory effect in proportion to concentration as compared with the control group. 3. In the inhibitory action on circumscribed edema induced by Caraggeenin, both of Hyunggyeyungyotang and Kamihyunggyeyungyotang administration showed the significant effect after 4 hours in comparison to the control group. 4. In the delayed type hypersensitivity response to Picryl Chloride, both of the sample groups revealed the significant effects. 5. Both of the sample groups decreased the vascular permeability induced by Histamine in comparison with the control group, but the significancy was admitted in only Hyunggyeyungyotang administration. According to above results, Hyunggyeyungyotang and Kamihyunggyeyungyotang are considered to be used for treament of the inflammatory diseases including sinusitis.

  • Sinichengpaeum has been widely used in the oriental medical treatment of nasal diseases. These studies were performed done to investigate the effect of Sinicheng- paeum on the anti-allergic action We studied the vascular permeability response induced by the histamine and serotonin injection(allergy Ⅰ type), homologous PCA provoked by the IgE-like antibody against EWA(allergy Ⅳ type), contact dermatitis induced by picryl chloride(allergy Ⅳ type), delayed type hypersensitivity response to SRBC (allergy Ⅳ type), and the amount of eosinophil and IgE. The results were as follows: 1. The effect of Sinichengpaeum on vascular permeability responses to intradermal histamine and serotonin were significant. 2. In the homologous PCA provoked by the IgE-like antibody against white egg albumin, Sinichengpaeum showed a significant effect. 3. In the delayed type hypersensitivity responses to picryl chloride, Sini-chengpaeum provoked a significant effect. 4. After reaction provoked by picryl chloride, Sinichengpaeum showed an insignficant effect on amount of eosinophil, but a significant effect in IgE amount. 5. In the delayed type hypersensitivity responses to SRBC, Sinichengpaeum provoked a significant effect. 6. After reaction provoked by SRBC, Sinichengpaeum showed a significant effect on amount of cosinophil, but an insignificant effect in IgE amount.

  • The object of thes research is to lucidate the clinical effects of Gami-Cheongshimyeonjayeum on the anti-allergic and gimmune response to rats and mice. The obtaind results of Gami-Cheongshimyeonjayeum administration are as follows : 1. The cytotoxic index increased slightly at over concentration $1{\times}10^{-2}/ml$, but it is thought that the cytotoxic effect show no significant value under the concentration $1{\times}10^{-3}/ml$. 2. Gami-Cheongshimyeonjayeum water extract seems to suppress the leakage volume of Evans blue to decrease the vascular permeability. 3. Gami-Cheongshimyeonjayeum show the inhibitory effect of footpad edema. 4. Protein leakage is inhibited by the administration of Gami-Cheongshimyeonjayeum. 5. The delayed type hypersensitivity to SRBC is suppressed at the 24hours after the administration of Gami-Cheongshimyeonjayeum on the experimental group 1, 2. 6. Hemolysin and hemagglutinin titer, and RFC increase to bulid up the immune function.

Inhibitory Effects on the Type 1 Hypersensitivity and Inflammatory Reaction of Herba Patriniae Aqua-acupuncture (패장약침(敗醬藥鍼)이 면역반응(免疫反應)에 미치는 영향)

  • Objective : Experimental studies were done to research the clinical effects of Herba Patriniae(HP) aqua-acupuncture ($BL_{13},\;BL_{17},\;BL_{13}{\cdot}BL_{17}$ and free points) on the anti-allergic inflammatory response. Methods : We measured active systemic anaphylatic shock induced by compound 48/80 and microvascular permeability increased by acetic acid. And we measured total IgE and plasma WBC level, serum total protein, albumin, immunoglobulin and NO levels induced by egg albumin. Results : HP aqua-acupuncture pretreatments at all acupoints inhibited active systemic anaphylatic shock induced by compound 48/80 and microvascular permeability increased by acetic acid. Total IgE and plasma WBC level inhibited by HP aqua-acupuncture pretreatment at $BL_{13}\;BL_{17}$ and free points. However, HP aqua-acupuncture didn't effect serum total protein, albumin, immunoglobulin and NO levels. Conclusion : These results suggest that HP aqua-acupuncture may be beneficial in the regulation of type Ⅰ allergic reaction, but is further required immunological studies on the allergic reaction.

Inhibitory Effects of Immediate-Type Allergic Reaction of Okbyungpoongsan-Gami by Anal Therapy (항장효법에 의한 옥병풍산가미의 즉각형 알레르기 반응 억제 효과)

  • Okbyungpoongsan-Gami (OG) has been used for the treatment of excessive sweating with general weakness and allergic rhinitis recently. Anal therapy is another way of taking Oriental medicine. It is an important pathway but not available in common clinical situation. This experiment was performed in order to study the inhibitory effect of immediate-type allergic reaction of OG by anal therapy. In addition, the experiment was practised by 1 H-NMR spectroscopy to examine molecular structure of OG. The results were obtained as follows : OG concentration-dependently inhibited compound 48/80- induced immediate type systemic allergic reaction with concentrations of 0.01-1.0g/kg by anal administration 1 h before injection of compound 48/80. OG also concentration-dependently inhibited compound 48/80- induced ear swelling response with concentrations of 0.01-1g/kg by anal administration 1h before injection of compound 48/80. OG also inhibited the passive cutaneous anaphylaxis activated by anti-dinitrophenyl (DNP) IgE antibody concentration- dependently at concentrations ranging from 0.01 to 1g/kg. When OG was pretreated at concentrations ranging from 0.01 to 1g/ℓ, the histamine release from the rat peritoneal mast cells induced by compound 48/80 was reduced in a concentration-dependent manner. OG (0.1-1g/ℓ) had a significant inhibitory effect on histamine release from IgE-induced activated mast cells. OG is seen to be a biochemical compound certainly by 1 H-NMR spectroscopy According to above results, anal therapy of OG may be beneficial in the treatment of systemic and local immediate-type allergic reactions by inhibition of histamine release from mast cells.

  • Experimental studies were done to research the effects of the Ursi fel and the UDCA on the anti-allergic effects. The results were obtained as follows: 1) In the experimental effects of the Ursi fel and the UDCA on the vascular permeability responses to intradermal Serotonin, though both of the Ursi fel and UDCA revealed the significant effect, the Ursi fel had stronger effect than the UDCA. 2) In the provocative effects of the Ursi fel and the UDCA on the vascular permeability responses to intradermal Histamin, though both of the Ursi powfel and UDCA showed the significant effect, the Ursi fel had moreerful effect than the UDCA. 3) In the 48 hours homologous passive cutaneous anaphylaxis provoked by the IgE-like antibody against white egg albumin, though both of the Ursi fel and UDCA revealed significant effect, the Ursi fel had stronger effect than the UDCA. 4) In the delayed type hypersensitivity response to Picryl Chlorede, though both of the Ursi fel and UDCA were proved to be effective significantly the Ursi fel showed stronger effect. 5) In the delayed type hypersensitivity responses to Sheep red blood cell, the Ursi fel revealed the significant effect, though the UDCA has no significant effect. According to above results, the Ursi fel was approved it could be used widely as antiallergic drug for immediate and delayed type allergic diseases. Although the UDCA revealed efficacy in immediated type allergic diseases, it had less powerful effects than the Ursi fel and it showed no effects in some experiment of delayed type allergy, so it would be difficult to be used clinically.

  • Objectives : We studied on anti-allergic effects of Semen Perillae Herbal Acupuncture(SPHA) and Semen Perillae Herbal Acupuncture Solution(SP). Methods : In vivo, Animals were herbal-acupunctured SPHA at both ST36 three times for 5 days. Then, we investigated compound 48/80-induced active systemic anaphylaxis (ASA) using ICR mice and anti-DNP IgE-induced passive cutaneous anaphylaxis (PCA) using Sprague Dawley rat. In vitro, we measured cell viability, b-hexosaminidase release, IL-4 and TNF-a from RBL-2H3 cells after treatment of SP of various concentrations. Results : In vivo, SPHA pretreatments at both ST36 inhibited compound 48/80-induced ASA. PCA was inhibited by SPHA pretreatments at both ST36 and optional points. In vitro, SP treatments were not affect on cell viability and inhibited b-hexosaminidase release, IL-4 and TNF-a. Conclusions : These results suggest that SPHA and SP may be beneficial in the inhibition of allergic inflammatory response.

  • Seungmagalgeuntang-gamibang has long been known to have anti-allergic effect. However, the mechanism of action of Seungmagalgeuntang-gamibang is not well investigated. The author analysed the effects of Seungmagalgeuntang-gamibang on the vascular permeability, delayed-type and contact hypersensitivities, and phagocytic function, the results obtained are as follows: 1. Administration of Seungmagalgeungtang-gamibang decreased the vascular permeability induced by serotonin in the mouse. 2. Administration of Seungmagalgeuntang-gamibang decreased the vascular permeability induced by histamine without statistical significant. 3. Administration of Seungmagalgeuntang-gamibang decreased the delayed-type hypersensitivity induced by sheep red blood cells. 4. Administration of Seungmagalgeungtang-gamibang decreased the contact hypersensitivity induced by dinitrochlorobenzene. 5. Seungmagalgeungtang-gamibang increased the phagocytic-activities of macrophages in vitro and in vivo. 6. Seungmagalgeungtang-gamibang enhanced the formation of reactive oxygen intermediates in vitro and in vivo. The above results demonstrate that Seungmagalgeuntang-gamibang suppresses the hypersensitivity reactions with increasing the phagocytic functions and formations of reactive oxygen intermediates from macrophages.

The Experimental Study on the Anti-allergic Effects of Sipjeondaebo-tang (십전대보탕이 알레르기반응에 미치는 영향)

  • This experimental research has been done to study the effects of Sipjeondaebo-tang(SOT) on the anti-allergic reaction. We found the several important results from the research which has been performed by two experiments toward immediately type and delayed type in order to study the effects of SDT on hypersensitivity response to mice. The results obtained from our research are as following. The survival rate of one group to which we injected only the compound 48/80 is almost 0% according to its density and timing test. In the other hand, the survival rates of the other group to which we injected both of the compound 48/80 and SDT are 20%, 10%, 30%, 10%, 40% and 70% according to 0.025, 0.05, 0.1, 0.25 0.5 and 1 (mg/g) of compound 48/80. Time dependency test also shows the 30% and 20% survival rates in 5 and 10 minutes. SDT revealed the significantly inhibitory effect on Compound 48/80 induced Mast cell degranulation. SDT showed the significantly inhibitory effect in the delayed type hypersensitivity response to picry1 chloride. SDT showed the significantly inhibitory effect in the delayed type hypersensitivity response to sheep red blood cell. Our research provides the important evidence that SDT is benificial to the prevention and treatment of allergy related diseases.
