Archvillin C-Terminus-Binding Proteins in Human Skeletal Muscle

  • Archivillin, a muscle-specific isoform of supervillin, is a component of the costameric cytoskeleton of muscle cells. The purpose of this study was to determine which protein in the skeletal muscle collaborates with archvillin C-terminus. For this purpose, a yeast two-hybrid screening of human skeletal muscle cDNA library was performed using the C-terminal region of archvillin as bait. This study shows that seven human skeletal muscle proteins, namely, nebulin, xeplin, archvillin, GAPDH, TOX4, PITRM1, and YME1L1 interact with archvillin C-terminus. Especially, xeplin is a newly discovered protein interacts with archvillin C-terminus. These results indicate that archvillin C-terminus acts as a bridge between nebulin and xeplin at costameres. Archvillin C-terminal region interacts with nebulin C-terminal region at Z-discs and interacts with xeplin at the vicinity of sarcolemma. I propose that these interactions may contribute to formation of costameric structure and muscle contraction.

  • In the case of railway freight, there's a lot of process at railway freight terminal. In these days, for operation efficiency improvement, Load/Unload planning is being carried out research. Load/unload problem can be classified as track assignment, container assignment, yard truck route problem, unload equipment scheduling, container yard plan. In this paper, we focus on track assignment and container assignment among the load/unload problem. we analysis information architecture to solve this problem. And we compares two precesses for existing method and optimization method of track/container assignment processes. An contents different analysis of two methods can shows reformation of an information architecture for load/unload process in the railway freight terminal.

  • The purpose of this paper is to analyze transport ability of AGV(Automated Guided Vehicle) and SHC(SHuttle Carrier). The main difference between two types of transport vehicles is that AGV depends on container crane or transfer crane to do loading/unloading container, but SHC is very independent to it. Therefore, the transport ability of SHC is expected to be higher than AGV. So, in this paper, we established simulation model to evaluate two types of transport vehicles and analyzed the results. Simulation model was established to automated container terminal with perpendicular yard layout, and applied closed loop operation of transport vehicle between apron and stacking yard. In the result, SHC showed very superior than AGV aspect of container crane productivity and vehicle fleets.

Crystal Structure of the Pneumococcal Vancomycin-Resistance Response Regulator DNA-Binding Domain

  • Vancomycin response regulator (VncR) is a pneumococcal response regulator of the VncRS two-component signal transduction system (TCS) of Streptococcus pneumoniae. VncRS regulates bacterial autolysis and vancomycin resistance. VncR contains two different functional domains, the N-terminal receiver domain and C-terminal effector domain. Here, we investigated VncR C-terminal DNA binding domain (VncRc) structure using a crystallization approach. Crystallization was performed using the micro-batch method. The crystals diffracted to a 1.964 Å resolution and belonged to space group P212121. The crystal unit-cell parameters were a = 25.71 Å, b = 52.97 Å, and c = 60.61 Å. The structure of VncRc had a helix-turn-helix motif highly similar to the response regulator PhoB of Escherichia coli. In isothermal titration calorimetry and size exclusion chromatography results, VncR formed a complex with VncS, a sensor histidine kinase of pneumococcal TCS. Determination of VncR structure will provide insight into the mechanism by how VncR binds to target genes.

Investigation of the Relationships Between Death Recognition, Terminal Care Stress, and Terminal Care Performance in Terminal Hospital Nurses (요양병원간호사의 죽음인식, 임종간호스트레스와 임종간호수행간의 관계 조사)

  • The purpose of this study is to investigate the relationships between death recognition, terminal care stress, terminal care performance, and other influencing factors of terminal care performance in terminal care hospital nurses. Two hundred forty nurses working in 11 hospitals for the elderly located in G, N, C city were surveyed. They were asked to complete three questionnaires: one on death recognition, one on terminal care stress, and a final one on terminal care performance. The data collected were analyzed using descriptive statistics, Pearson's correlation, and stepwise regression using SAS 9.1. The findings suggest that the nurses' own mental and occupational stress was the cause of many difficulties in the practice of end-of-life care, where the nurse will often experience the death patients, which results in increased dysentery stress. There was a negative correlation between species stress and terminal care performance. Death recognition and terminal care stress were correlated with terminal care performance. The results suggest that the longer the clinical experience, the higher the quality of terminal care performance. Therefore, it is suggested that terminal care should be performed considering the longevity of the careers career and their death tendency. Further to this study, it is necessary to find out how to reduce stress and to evaluate other variables affecting the care of the patient.

  • While demands for express service are rapidly increasing according to recent progress of electronic commerce, express service companies are struggling to take a larger delivery service market share through ongoing improvement in their service processes. Extension of cut-off time for express service centers can provide the express company with increase of total sales, but it may also cause to increase the possibility not to satisfy customer needs due to work delay in the consolidation terminal. Therefore, compromised decision for cut-off time of each service center should be made by taking operation characteristics of the consolidation terminal into account. This study suggests an approach for determining the cut-off time for express service centers according to operational characteristics of the consolidation terminal with the objective of maximizing expected incremental sales. The problem defined in this study can be represented as two successive models; one is an integer programing model in which the best cut-off time for each sales center are determined, and the other is a single machine scheduling model in which a working schedule in the consolidation terminal is obtained. A genetic algorithm is developed to solve the two models simultaneously. Finally, an example problem is carried out to verify applicability and performance of the algorithm with the data set collected from an express company.

Redescription of Gonostomum algicola and G. gonostomoida (Ciliophora: Spirotrichea: Sporadotrichida) Unknown from Korea

  • Two rare ciliates from soil under the oak tree and mosses in the top of mountain in Korea were identified as Gonostomum algicola Gellert, 1942 and G. gonostomoida (Hemberger, 1985), respectively. There is little information on their morphological features, therefore their detailed redescriptions are needed. The description was based on the observation of living and protargol impregnated specimens, and biometric analysis. Their diagnostic characteristics are as follows. Gonostomum algicola; $88-113\times30-40{\mu}m$ in vivo, colourless cortical granules, 20-31 adoral membranelles, two fronto-terminal cirri, five fronto-ventral cirri, no mid-ventral cirri, two transverse cirri, two to three micronuceli. Gonostomum gonostomoida; $60-121\times21-40{\mu}m$ in vivo, no cortical granules, 27-34 adoral membranelles, no fronto-terminal cirri, two fronto-ventral cirral rows with each row bearing three cirri, midventral cirral row with 11-14 cirri, two to three transverse cirri, one to six micronuceli. So far, total three species within the genus Gonostomum have been recorded from Korea by the present study.

Trajectory Optimization for Autonomous Berthing of a Twin-Propeller Twin-Rudder Ship

  • Autonomous berthing is a crucial technology for autonomous ships, requiring optimal trajectory planning to prevent collisions and minimize time and control efforts. This paper presents a two-phase, two-point boundary value problem (TPBVP) strategy for creating an optimal berthing trajectory for a twin-propeller, twin-rudder ship with autonomous berthing capabilities. The process is divided into two phases: the approach and the terminal. Tunnel thruster use is limited during the approach but fully employed during the terminal phase. This strategy permits concurrent optimization of the total trajectory duration, individual phase trajectories, and phase transition time. The efficacy of the proposed method is validated through two simulations. The first explores a scenario with phase transition, and the second generates a trajectory relying solely on the approach phase. The results affirm our algorithm's effectiveness in deciding transition necessity, identifying optimal transition timing, and optimizing the trajectory accordingly. The proposed two-phase TPBVP approach holds significant implications for advancements in autonomous ship navigation, enhancing safety and efficiency in berthing operations.

  • The most important thing in acontainer terminal is to handle the cargo effectively in the limited time available. To achieve this objective, many strategies have been introduced and applied. To create an automated container terminal, it is necessary for the cargo handling equipment to be equipped with more intelligent control systems. From the middle of the 1990's, automated rail-mounted gantry cranes (RMGC) and rubber-tired gantry cranes (RTG) have been widely used to handle containers in yards. Recently, many pieces of equipment, like CCD cameras and sensors, have beenmounted in these cranes to cope with the automated terminal environment. In this paper, we try to support the development of more intelligent automated cranes, which allow for more effective cargo handling in yards. For this purpose, the modeling, tracking control, anti-sway system design, skew motion suppressing, and complicated motion control and suppressing problems must be considered. Especially, in this paper, the system modeling and a new tracking control approach are discussed, and an experimental study is performed based on a two-degree-of-freedom (2DOF) servosystem design.
