생균활성 보존을 위한 유산균 미세캡슐 개발 (Development of Probiotic Microcapsules for the Preservation of Cell Viability)

  • 본 연구에서는 생균제로서의 L. fermentum YL-3의 미세캡슐화를 시도하였고, 균체에 대한 캡슐 재료의 영향을 조사함으로써 캡슐제조시 균에 대한 손상을 최소화하고자 하였다. 그리고 제조된 캡슐의 내산성과 저장성을 조사하여 유산균의 내산성과 저장능의 증진에 적합한 캡슐의 제조 및 보존 방법을 연구하였다. 먼저, 인체에 무독성인 sodium alginate를 캡슐의 원료로 선택하여 microcapsule을 제조한 결과 생균제가 alginate 캡슐에 포집되어 존재하는 것을 확인할 수 있었고, pH 2에서 120-126 ${\mu}m$ 크기의 견고한 캡슐을 제조하였으며, pH가 증가할수록 크기가 커지지만 pH 3 이상에서는 견고성이 너무 떨어져 캡슐의 형성이 어려워 오히려 pH 3에서의 캡슐보다 작은 형태를 나타내었다. 반면, 캡슐 후의 생균제의 생존율은 pH가 증가할수록 높게 나타났다. 그리고 캡슐제조시 사용되는 재료가 균체에 미치는 영향을 조사한 결과 캡슐의 주재료인 sodium alginate와 $CaCl_{2}$는 균의 활성을 떨어뜨리는 것으로 조사되었고, 그 외의 재료인 sodium citrate, Tween 80, Span 80, soybean oil은 균의 활성에 별다른 영향을 주지 않는 것으로 나타났다. 또한 microcapsule과 macrocapsule을 제조하여 각각의 캡슐의 내산성과 저장성을 비교한 결과, 내산성은 microcapsule이 가장 우수한 것으로 나타났고, 저장성은 $4^{\circ}C$에서는 microcapsule, $25^{\circ}C$에서는 macrocapsule에서의 생균제 활성이 좋은 것으로 확인되었다.

Purification and Characterization of a Novel Extracellular Thermostable Alkaline Protease from Streptomyces sp. M30

  • A novel alkaline protease from Streptomyces sp. M30, SapHM, was purified by ammonium sulfate precipitation, hydrophobic interaction chromatography, and DEAE-Sepharose chromatography, with a yield of 15.5% and a specific activity of 29,070 U/mg. Tryptic fragments of the purified SapHM were obtained by electrospray ionization quadrupole time-of-flight mass spectrometry. Nucleotide sequence analysis revealed that the gene sapHM contained 1,179 bp, corresponding to 392 amino acids with conserved Asp156, His187, and Ser339 residues of alkaline protease. The first 24 amino acid residues were predicted to be a signal peptide, and the molecular mass of the mature peptide was 37.1 kDa based on amino acid sequences and mass spectrometry. Pure SapHM was optimally active at 80℃ in 50 mM glycine-NaOH buffer (pH 9.0), and was broadly stable at 0-50℃ and pH 4.0-9.0. The protease relative activity was increased in the presence of Ni2+, Mn2+, and Cu2+ to 112%, 113%, and 147% of control, respectively. Pure SapHM was also activated by dimethylformamide, dimethyl sulfoxide, Tween 80, and urea. The activity of the purified enzyme was completely inhibited by phenylmethylsulfonyl fluoride, indicating that it is a serine-type protease. The Km and Vmax values were estimated to be 35.7 mg/ml, and 5 × 104 U/mg for casein. Substrate specificity analysis showed that SapH was active on casein, bovine serum albumin, and bovine serum fibrin.

퍼지앤트랩-기체크로마토그래피(PT-GC)를 이용한 식품첨가물 중 1,4-디옥산 및 클로로히드린류 분석 (Analysis of 1,4-Dioxane and Chlorohydrins in Food Additives by Purge & Trap GC)

  • 폴리소르베이트류, 염화콜린, 주석산수소콜린, 변성전분 및 향신료 중 1,4-디옥산 및 클로로히드린류의 분석법을 개발하고 확립된 방법으로 잔류량을 조사한 결과는 다음과 같다. 분석에 사용된 트랩으로는 Vocarb 3000이 적합하였고 이때의 검출한계는 DOX 1.38$\mu\textrm{g}$, EPC 0.23$\mu\textrm{g}$, PCH 3.30$\mu\textrm{g}$, ECH 3.97$\mu\textrm{g}$, DCP 20.43$\mu\textrm{g}$이었다. 검체 온도증가와 무수황산나트륨 농도 증가에 따라 분석감도는 증가하였으나 6$0^{\circ}C$ 이상에서는 상대표준편차(RSD, %)가 6%를 초과하여 재현성이 떨어져 분석온도를 5$0^{\circ}C$로 설정하였다. 폴리소르베이트류에서의 회수율은 염을 첨가하지 않고 측정하였을 때 EPC(66%)를 제외하고는 90%이상을 나타내었다. 염화콜린 및 주석산수소콜린의 경우에는 대부분 회수율이 100% 이상이었으나 주석산수소콜린의 EPC는 20%로 낮게 나타났다. 히드록시프로필인산이전분, 히드록시프로필전분 및 찰옥수수전분에서는 검체를 가수분해한 후 회수율을 측정한 결과 검체종류별 큰 차이 없이 회수율이 90% 이상을 나타내었다. 향신료의 분석에서는 무수황산나트륨을 첨가하여 분석한 결과 회수율이 90%이상이었다. 구매한 폴리소르베이트류, 염화콜린, 주석산수소콜린, 향신료 중 1,4-디옥산 및 클로로히드린류를 분석한 결과 트윈 80에서 DOX가 2.5ppm 검출되었으나 이외의 검체에서는 모두 불검출되었고 직접 제조한 히드록시프로필전분에서도 모두 불검출되었다.

상추 펠렛종자의 피복물질 탐색과 영양물질 첨가가 발아력에 미치는 영향 (Identification of Pelleting Materials and Effect of Nutrient Addition on the Germination of Pelleted Lettuce Seeds)

  • 상추종자는 크기가 불균일하여 기계화 파종이 어렵다. 펠렛의 목적은 종자크기를 증가시켜 기계화 파종을 가능하게 하여 파종과 솎음노력을 절감하고 종자를 절약하는데 있다. 펠렛 접착제 종류 및 농도에 따라 발아율과 발아일수에는 큰 차이가 있었다. 사용된 접착제 가운데 polyvinyl alcohol(PVA)에서 전반적으로 발아가 원활하였고 다음이 polyvinyl pyrrolidone(PVP), hydroxyethyl cellulose(HEC), methyl cellulose(MC), tween 80 순으로 나타났다. 펠렛 파복물질의 종류에 따라 발아율이 달랐는데 전반적으로 diatomaceous earth 및 talc + calcium carbonate가 다른 펠렛 피복물질에 비해 발아율도 높고 발아일수도 다눅되어 상추의 종자 펠렛에 적합한 피복물질이었다. 반면 Limestone, calcium oxide, bentonite 등은 발아율이 저조하였다. 상추종자는 펠렛 후 종자의 크기가 33배 증가되었다. 펠렛종자에 영양물질 첨가는 발아율이 약간 감소되었고, 발아일수도 지연되는 경향이었다. 첨가되는 영양물질의 급원에 따라서도 펠렛종자의 발아력에도 차이가 있었는데, 대체적으로 MS medium이 monosodium phosphate 보다 발아억제 정도가 낮았다. Priming 후 펠렛된 종자는 무처리 종자를 펠렛한 경우에 비해 발아율이 높았고 조기발아 하였다.

바이오 계면활성제에 의한 토양/지하수내 유기성 오염물질 제거 (Use of Biosurfactant for the Removal of Organic Pollutants in Soil/Groundwater)

  • 토양/지하수내에 존재하는 소수성 유기오염물을 제거하기 위한 방법의 적용성 검토를 위하여 바이오 계면활성제인 HPCD에 유기오염물이 흡수되는 현상에 대한 실험을 실시하였다. 유기오염물의 HPCD와의 반응은 아주 빠르게 진행되어 95% 정도의 반응이 10분 이내에 일어났다. 얻어진 흡수계수는 용액의 pH. 이온강도 및 유기오염물의 농도 변화에 대하여 약간의 영향이 있는 것으로 나타났다. 두 가지 방법에 의하여 구한 토양표면에 흡착되는 HPCD의 양은 무시할 정도로 나타났으며 HPCD가 고체상에 대한 친화성이 없다는 사실은 토양/지하수내 존재하는 유기오염물을 제거시 큰 장점으로 작용할 수 있다. 결론적으로 HPCD는 지표 밑의 유기오염물을 제거하는데 효과적으로 사용될 수 있다. HPCD에 의한 유기오염물의 흡수는 오염물의 크기, 소수성 등에 의하여 큰 영향을 받으며 합성된 계면활성제와의 사용 타당성 비교시 오염되어 있는 유기물 종류뿐 아니라 재료비, 2차 오염 유발 가능성 등을 종합적으로 고려해야 한다.

Characterization of Cellulolytic and Xylanolytic Enzymes of Bacillus licheniformis JK7 Isolated from the Rumen of a Native Korean Goat

  • A facultative bacterium producing cellulolytic and hemicellulolytic enzymes was isolated from the rumen of a native Korean goat. The bacterium was identified as a Bacillus licheniformis on the basis of biochemical and morphological characteristics and 16S rDNA sequences, and has been designated Bacillus licheniformis JK7. Endoglucanase activities were higher than those of ${\beta}$-glucosidase and xylanase at all temperatures. Xylanase had the lowest activity among the three enzymes examined. The optimum temperature for the enzymes of Bacillus licheniformis JK7 was $70^{\circ}C$ for endoglucanase (0.75 U/ml) and $50^{\circ}C$ for ${\beta}$-glucosidase and xylanase (0.63 U/ml, 0.44 U/ml, respectively). All three enzymes were stable at a temperature range of 20 to $50^{\circ}C$. At $50^{\circ}C$, endoglucanse, ${\beta}$-glucosidase, and xylanase had 90.29, 94.80, and 88.69% residual activity, respectively. The optimal pH for the three enzymes was 5.0, at which their activity was 1.46, 1.10, and 1.08 U/ml, respectively. The activity of all three enzymes was stable in the pH range of 3.0 to 6.0. Endoglucanase activity was increased 113% by $K^+$, while $K^+$, $Zn^+$, and tween 20 enhanced ${\beta}$-glucosidase activity. Xylanase showed considerable activity even in presence of selected chemical additives, with the exception of $Mn^{2+}$ and $Cu^{2+}$. The broad range of optimum temperatures (20 to $40^{\circ}C$) and the stability under acidic pH (4 to 6) suggest that the cellulolytic enzymes of Bacillus licheniformis JK7 may be good candidates for use in the biofuel industry.

Determination of Optimized Growth Medium and Cryoprotective Additives to Enhance the Growth and Survival of Lactobacillus salivarius

  • The beneficial effects of lactic acid bacteria (LAB) have been intensively investigated in recent decades with special focus on modulation of the host intestinal microbiota. Numerous discoveries of effective probiotics are driven by a significantly increasing demand for dietary supplements. Consequently, technological advances in the large-scale production and lyophilization are needed by probiotic-related industries for producing probiotic LAB for commercial use. Our study had a dual objective, to determine the optimum growth medium composition and to investigate appropriate cryoprotective additives (CPAs) for Lactobacillus salivarius, and compare its responses with other Lactobacillus species. The one-factor-at-a-time method and central composite design were applied to determine the optimal medium composition for L. salivarius cultivation. The following composition of the medium was established (per liter): 21.64 g maltose, 85 g yeast extract, 1.21 ml Tween 80, 6 g sodium acetate, $0.2g\;MgSO_4{\cdot}7H_2O$, $0.02g\;MnSO_4{\cdot}H_2O$, $1g\;K_2HPO_4$, $1.5g\;KH_2PO_4$, $0.01g\;FeSO_4{\cdot}7H_2O$, and 1 g sodium citrate. A cryoprotective additive combination comprising 10% (w/v) skim milk and 10% (w/v) sucrose supplemented with 2.5% (w/v) sodium glutamate was selected for L. salivarius, and its effectiveness was confirmed using culture-independent methods in the freeze-dried cells of the Lactobacillus strains. In conclusion, the optimized medium enhanced the species-specific cultivation of L. salivarius. On the other hand, the cryoprotective effects of the selected CPA mixture may also be dependent on the bacterial strain. This study highlights the necessity for precise and advanced processing techniques for large-scale production of probiotics in the food and feed industries.

미세유화약물송달시스템을 이용한 로바스타틴의 생체이용률 향상 (Improvement of Bioavailability for Lovastatin using Self-microemulsifying Drug Delivery System)

  • A self-microemulsifying drug delivery system (SMEDDS) was developed to increase the dissolution rate, solubility, and ultimately bioavailability of a poorly water soluble drug, lovastatin. SMEDDS was thε mixtures of oils, surfactants, and cosurfactants, which emulsify under conditions of gentle agitation, similar to those which would be encountered in the gastro-intestinal (GI) tract. Various types of self-emulsifying formulations were prepared using four types of oil (Capryol 90, Lauroglycol 90, Labrafil M 1944 CS and Labrafil M 2125), two surfactants (Cremophor EL and Tween 80), and three cosurfactants (Carbitol, PEG 400 and propylene glycol). Thε efficiency of emulsification was studied using a laser diffraction size analyzer to determine particle size distributions of the resultant emulsions. Optimized formulations selected for bioavailability assessment were Carpryol 90 (40%), Cremophor EL (30%) and Carbitol (30%). SMEDDS containing lovastatin (20 mg and 5 mg) were compared to a conventional lovastatin tablet $(Mevacor^{\circledR},\;20\;mg/tab)$ by the oral administration as prefilled hard gelatin capsules to fasted beagle dogs for in vivo study. The arεa under the serum concentration-time curve from time zero to the last measured time in serum, $AUC_{0{\rightarrow}24h}$, was significantly greater in SMEDDS, suggesting that bioavailability increase 130% and 192% by the SMEDDS, respectively. The self-emulsifying formulations of lovastatin afforded the improvement in absolute oral bioavailability relative to previous data of lovastatin tablet formulation. These data indicate the utility of dispersed self-emulsifying formulations for the oral delivery of lovastatin and potentially other poorly absorbed drugs.

Development of Self-microemulsifying Drug Delivery System for Enhancing the Bioavailability of Atorvastatin

  • The objective of the study was to prepare self-microeulsifying drug delivery system (SMEDDS) incorporating atorvastatin calcium and evaluate its properties and oral bioavailability. Solubility of atorvastatin in various vehicles was determined. Pseudo-ternary phase diagrams were constructed to identify the good self-emulsification region. The droplet size distributions of the resultant emulsions were determined by dynamic light scattering measurement. The mean droplet size of chosen formulation (20% ethyl oleate, 40% tween-80, 40% Carbitol$^{(R)}$) was $23.4{\pm}1.3$ nm. The SMEDDS incorporating atorvastatin calcium appeared to be associated with better performance in dissolution and pharmacokinetic studies, compared with raw atorvastatin calcium. In dissolution test, the release percentage of atorvastatin from SMEDDS mixture could rapidly reach more than 95% within 3 min. Oral $AUC_{0{\rightarrow}8hr}$ values in SD rats was $1994{\pm}335\;ng{\cdot}hr/mL$, which significantly increased (P<0.05) compared with raw atorvastatin calcium. The SMEDDS formulation was relatively stable when stored at $4^{\circ}C$ during 3 months. Our studies illustrated the potential use of SMEDDS for the delivery of hydrophobic compounds, such as atorvastatin, by the oral route.

Effects of Non-ionic Surfactant Supplementation on Ruminal Fermentation, Nutrient Digestibility and Performance of Beef Steers Fed High-roughage Diets

  • Three experiments were conducted to determine the effects of non-ionic surfactant (NIS) supplementation on ruminal fermentation, nutrient digestibility and performance of beef steers fed high-roughage diets. The objective of experiment 1 was to investigate the effects of NIS supplementation on in vitro ruminal fermentation of cultures administered with corn and barley as grain substrate and rice straw and timothy hay as roughage substrate. The in vivo ruminal fermentation, nitrogen balance and digestibility of nutrients were also examined with steers fed a high-roughage diet in experiment 2. The aim of experiment 3 was to determine the responses to NIS of growing steers fed a high-roughage diet. In experiment 1, ammonia nitrogen concentration for NIS supplementation was higher (p<0.05) than for the control with all substrates. However, concentrations of total volatile fatty acid (VFA), acetate, butyrate and valerate of the incubated roughage substrates, rice straw and timothy hay, were higher (p<0.05) for NIS supplementation than for the control whereas VFA concentrations in the cultures of corn and barley were unaffected. These results indicated that effects of NIS on ruminal fermentation are diet dependent, specifically on roughage sources. In experiment 2, ruminal pH of steers supplemented with NIS was lower (p<0.05) than the control. Ruminal concentrations of ammonia nitrogen, acetate, total VFA and urinary concentrations of purine derivatives were increased (p<0.05) by NIS supplementation. In experiment 3, supplementation of NIS increased (p<0.05) intakes of total feed and corn silage, average daily gain, and feed efficiency of growing steers although they varied depending on supplementation level. Due to the roughage-specific feature of NIS effects, NIS appears to enhance ruminal fermentation of fibrous parts of feeds and, consequently, performance of steers fed a high-roughage diet.
