Changes of Protein During Growth of Soybean Sprout (Gel filtration에 의한 콩나물 제조중(製造中) 단백질(蛋白質)의 변화조사(變化調査))

  • Gel filtration of water soluble protein of soybean and cotyledon of sprout at various growth stages by using Sephadex G-200 showed 5 fractions (A,B,C,D and E). According to gel filteration and disc gel electrophoresis, fraction B,C and D were identified as 11S,7S and 2S, respectively. Fraction A was turbid substance and fraction E showed 1 peak. During growth of sprout 7S decreased firstly, 2S secondly and 11S lastly in cotyledon. Fraction A increased until 6th day and decreased thereafter while E increased throughout the growth. In axis only two fractions (11S+7S and E) were showed, and 11S+7S fraction was little changed and fraction E increased slightly with the growth.

Evaluation of Glyceryl Monooleate(GMO) W/O Emulsion Stability by using Turbiscan®LAB (Turbiscan®을 이용한 Glyceryl Monooleate(GMO) 함유 W/O 유제의 안정성 평가)

  • The main object of this study was to prepare of w/o emulsion including glyceryl monooleate(GMO) and to evaluate its stability by using the recently developed $Turbiscan^{(R)}LAB$. GMO is the polar oily surfactant with the low HLB value, and it forms the gel phase of cubic structures after dissolves in aqueous media. Phosphate buffer solution (PBS) of pH 7.4 was prepared as the water phase and Marcol 52(mineral oil) was used as the oil phase in this study. GMO was used as the surfactant of W/O emulsion. W/O emulsion using GMO alone as a surfactant was very unstable. But the emulsion using both GMO and poloxamer 407 was more stable. The stability of W/O emulsions was evaluated after centrifuging the emulsions. But it was difficult with naked eye because an opaque and concentrated system like W/O emulsion was very turbid. So $Turbiscan^{(R)}LAB$ was used to detect the destabilization phenomena in non-diluted emulsion. As a result, the W/O emulsion using the proper amounts of GMO and poloxamer 407 was more stable among them using GMO of various amounts. But it seems that the other element for the stability of W/O emulsion including GMO was required. Furthermore, the $Turbiscan^{(R)}LAB$ was a very efficient analyzer for evaluating the physical stability of emulsion.

Phase Behavior and Spontaneous Vesicle Formation in Aqueous Solutions of Anionic Ammonium Dodecyl Sulfate and Cationic Octadecyl Trimethyl Ammonium Chloride Surfactants

  • Phase behavior for the mixed aqueous surfactant systems of cationic octadecyl trimethyl ammonium chloride (OTAC)/anionic ammonium dodecyl sulfate (ADS)/water was examined. Below the total surfactant concentrations of 1.5 m molal, mixed micelles were formed. At the total surfactant concentrations higher than 1.5 m molal, there appeared a region where mixed micelles and vesicles coexist. As the surfactant concentration increased, the systems looked very turbid and much more vesicles were observed. The vesicles were spontaneously formed in this system and their existence was observed by negative-staining transmission electron microscopy (TEM), small-angle neutron scattering (SANS) and encapsulation efficiency of dye. The vesicle region was where the molar fraction α of ADS to the total mixed surfactant was from 0.1 to 0.7 and the total surfactant concentration was above 5 × 10-4 molality. The size and structure of the vesicles were determined from the TEM microphotographs and the SANS data. Their diameter ranged from 450 nm to 120μm and decreased with increasing total surfactant concentration. The lamellar thickness also decreased from 15 nm to 5 nm with increasing surfactant concentration and this may be responsible for the decrease in vesicle size with the surfactant concentration. The stability of vesicles was examined by UV spectroscopy and zeta potentiometry. The vesicles displayed long-term stability, as UV absorbance spectra remained unchanged over two months. The zeta potentials of the vesicles were large in magnitude (40-70 mV) and the observed longterm stability of the vesicles may be attributed to such high ζ potentials.

  • The characteristic floc growth of Al-based coagulants was investigated in the aspect of mixing intensity and visualization of generated flocs during coagulation and flocculation processes. Zeta potential of turbid particles in the artificial water nearly approached to zero at pH 8-9, in which TDS and conductivity were minimized. The removal rate of turbidity and phosphate was maximized at the optimal mixing intensity of rapid and slow mixing stages. After the rapid mixing stage of coagulation process, small particles ($3-5{\mu}m$) were abruptly generated, and higher mixing intensity made more numbers of flocs. With the progress of slow mixing stage, the number of small particles were decreased with the simultaneous increase of intermediate particles ($7-21{\mu}m$). The number of large particles (>$23{\mu}m$) were maximized at the lowest rapid mixing intensity of $95.1sec^{-1}$, whereas small particles (<$5{\mu}m$) were maximized at the highest rapid mixing intensity of $760.7sec^{-1}$.

  • Objectives This research was carried out to establish the clinical practice guideline(CPG) for Yin-Deficit Diurnal-Heat (Eumheo-oyeol) symptomatology of Soyangin disease. Methods Dongeuisusebowon(sinchuk edition) and several kinds of literatures including journal articles concerning this symptomatology of Soyangin disease were collected and classified. Sasang constitutional specialists' conference was held to make an agreement on the conflicting issues as well. Consensus was drawn as a result of the conference. Results & Conclusions 3 papers were selected as an inclusion and exclusion criteria for the relevant articles to Yin-Deficit Diurnal-Heat (Eumheo-oyeol) symptomatology of Soyangin disease. Yin-Deficit Diurnal-Heat (Eumheo-oyeol) symptomatology consists of two aspects : Yin-Deficit Diurnal-Heat (Eumheo-oyeol) severe pattern and Yin-Deficit Diurnal-Heat (Eumheo-oyeol) critical pattern. In Yin-Deficit Diurnal-Heat (Eumheo-oyeol) severe pattern contains 1 disease, namely, Clear Yang Depletion of Large Intestine (Daejang-cheongyang Moson) pattern (Lower wasting-thirst (Haso) pattern). In Yin-Deficit Diurnal-Heat (Eumheo-oyeol) critical pattern contains 2 diseases, Yin-Deficit Diurnal-Heat (Eumheo-oyeol) pattern and Yin-Deficit Diurnal-Heat (Eumheo-oyeol) advanced pattern. Yin-Deficit Diurnal-Heat (Eumheo-oyeol) symptomatology has several kinds of symptoms like dry mouth, disliking to drink much water, diurnal body fever, coldness on the back and nausea as well as body fever, chest discomfort, constipation or dry stool as a common symptoms of Interior Heat disease. Clear Yang Depletion of Large Intestine (Daejang-cheongyang Moson) pattern (Lower wasting-thirst (Haso) pattern) has above mentioned symptoms and much urine/turbid urine, thin thigh and knee joints and twinge of joint pain over the body. Yin-Deficit Diurnal-Heat (Eumheo-oyeol) pattern has symptoms like indigestion and epigastric discomfort, abdominal pain and vomiting in addition. Yin-Deficit Diurnal-Heat (Eumheo-oyeol) advanced pattern has symptoms like hematemesis as well.

Blood Pressure Lowering Effect of Korean turbid rice wine (Takju) Lees Extracts in Spontaneously Hypertensive Rat (SHR) (막걸리박 열수추출물 첨가 식이가 선천성 고혈압쥐(SHR)의 혈압 저하에 미치는 영향)

  • This study was conducted to investigate the effects of Takju lees hot water extracts on the blood pressure in spontaneously hypertension rats (SHR). Twenty eight male SHR were grouped by their blood pressure and fed a control diet or experimental diets containing 1% (G1), 2% (G2), or 4% (G4) Takju lees extracts for 4 weeks. Food intake was not significantly different among the groups. However, body weight gain was significantly lower in groups fed the Takju lees extract than the control group. The systolic blood pressure was significantly lower in the Takju lees extract containing groups (especially in G4 group) than the control groups. In addition, mean blood pressure {(systolic+dyastolic)/2} decreased with an increase in the amount of Takju lees extract in the diet and feeding period. Takju lees extract decreased angiotensin I converting enzyme (ACE) activity in a dose-dependent manner. These results suggest that the Takju lees extract exert an antihypertensive effect by decreasing ACE activity.

Comparison of Larval Fish Survival of Pale chub (Zacco platypus) Exposed to Different Levels Turbidity (탁수조건에 따른 피라미 치자어의 생존률 비교)

  • To quantitatively assess the effects of turbidity on egg development and larval fish survival, a laboratory fish rearing experiment was applied to different life stages (newly hatched larval stage, juvenile stage and pre-adult stage) of the Zacco platypus, one of the most universal and tolerant species in Korea. According to the stress index of turbidity in water with exposure time, three different treatments, including a reference condition (1~7 NTU) as well as intermediate (20~150 NTU) and high turbidity conditions (400~1,000 NTU) were applied, and egg hatching and larval fish mortality rates were observed. The mortality rates of newly hatched larval fish were significantly different among treatments (ANOVA, $F_{2,3}$=17.79, p<0.05). Average rates of survival to hatching were 20.9% (${\pm}0.1%$) for reference condition, 11% (${\pm}6.9%$) for intermediate level and 3.2% (${\pm}3.7%$) for high level conditions, respectively. A sudden change of mortality at the high level was observed within 5 days of the experiment. About 84% of juvenile fish survived until 20 experimental periods under conditions of reference turbidity, while survival under conditions of intermediate turbidity was over 80% of larval fish until day 13 of the experiment, but dropped to less than 10% after day 14 and 15 of two experiments. Fish mortality appeared from day 6 of the high turbidity experiment, and 50% mortality was achieved at day 9 to 10 of experiment. Full mortality occurred at day 14 of the experiment (RM-ANOVA, $F_{2,38}$, p<0.005). In the pre-adult stage experiment, no mortality was observed during the experiment at reference level treatment (20 days), while only slight mortality rates were observed for both intermediate and high levels until day 5 of the experiment, however, no further fish died in either experiment. It was significantly different compared to reference condition (RM-ANOVA, $F_{2,20}$=8.28, p<0.01), but no difference was observed between intermediate and high level conditions. Consequently, this tolerant species has been determined to be well adapted to high levels of turbidity in its adult stage, but more vulnerable throughout earlier life stages.

Seasonal Variation of Transparency in the Southeastern Yellow Sea (황해 남동해역 투명도의 계절 변화)

  • The data of Secchi disc observation collected during $1966\~1990$ were analyzed to investigate the seasonal variation of transparency in the southeastern Yellow Sea. The bimonthly distribution of mean transparency showed that the isolines of transparency were roughly parallel to the isobaths. The transparency was low (3 m in february and 8 m in August) if the shallow water less than 20 m depth in comparition to the higher values (10 m in february and 17 m in August) in the deeper water. The lowest transparency was found in winter. The transparency increased in spring and the highest transparency occurred in summer. The water becomes turbid in autumn. Suspended solid concentrations in winter are ranged from 28 to 130 $mg/{\ell}$, and from 8 to 60 $mg/{\ell}$ in summer. The seasonal variation of transparency seems to be mainly affected by resuspension of solid from the bottom. The amounts of suspended solid are large in winter due to the vertical convection by cooling effect and tubulence by the strong wind, and small in summer due to the strong stratification and weak wind.

Characteristics of turbid water on watershed in rainy season (하천유역의 강우기 탁수발생 특성)

  • 강우기에 하천 유역에서의 탁수 발생은 대상 지역의 토양, 토지 이용형태와 식생 분포, 강우의 강도, 토지의 경사 등에 따라 큰 영향을 받게 된다. 본 연구에는 토지의 이용형태에 따른 강우 시 탁수 발생량을 평가하고자 용담댐 유역의 탁수 발생특성 조사를 실시하였다. 2005년의 조사지점은 유역면적이 가장 작은 주자천 유역과 구량천, 금강본류를 대상으로 하였다. 구량천과 금강본류는 용담댐 유역을 구성하는 가장 큰 두 소유역이며 다를 지역에 비해 오염원이 많아 용담댐수질에 많은 영향을 주는 하천이다. 현장조사는 6월 27일부터 7월 3일 까지 실시하였다. 2007년 조사 대상 지역은 주자천 유역의 주천교 지점과 금강본류 유역의 월곡교 지점을 선정하였다. 현장조사는 6월21${\sim}$22일, 9월14${\sim}$15일 2회에 걸쳐 실시하였으며 시간별 현장 탁도 측정 및 시료 채취를 통한 분석을 병행하였다. 2005년에 실시한 조사결과 오염원이 많고 유역이 넓은 천천은 7월 1일에 유입량이 최대로 348.16CMS였으며, 그날 탁도는 오후 2시 10분조사시에 최대 2060NTU를 나타냈다. 구량천은 같은날 조사를 실시하여 유량은 최대 200.33CMS였고, 최대 탁도는 763NTU를 나타냈다. 반면 주자천에서는 최대 탁도가 98.4NTU를 나타내 가장 탁도가 높은 금강본류와는 약 13배의 차이를 나타냈다. 2007년 1차 조사는 6월 21${\sim}$22일에 걸쳐 이루어 졌다. 주천교 지점은 시간최대 강우 17mm가 발생한 시점에 최대 92NTU의 탁도를 나타내었고 그 이후 감소하여 20NTU이하의 탁도를 유지하였다. 반면, 월곡교 지점은 시간최대 23mm의 강우가 발생하였고, 최대 탁도는 3,610NTU에 도달하였으며 1,000NTU이상의 고탁도가 6시간 이상 유지된 후, 시간이 지남에 따라 점차 감소하였다. 이러한 결과는 미개발지역인 주천교 지점의 경우 강우에 의한 탁수 발생이 상대적으로 적음을 보여주고 있으며 월곡교 지점과 같이 개발로 인해 노출된 토양의 경우 강우에 의해 쉽게 침식이 일어나 고탁도의 탁수를 발생시킴을 보여준다. 그러나 주천교지점에서 2005년에 조사한 자료와 비교해 보면 2007년이 적은 강우임에도 불구하고 탁도는 높게 나타난 결과를 나타내고 있으며 이는 토지이용의 고도화에 따른 영향으로 판단되었다. 연구 결과 유역내에서 탁수 발생은 같은 강도의 강우에 있어서 큰 차이를 보이고 있음을 확인할 수 있었다. 따라서 강우기에 저수지에서의 탁수 문제의 해결을 위해서는 상류 유역의 탁수 발생원 관리가 중요함을 보여주고 있으며, 토지 이용 형태에 따른 토양 침식 방지 대책이 강우기의 댐 내 탁도 문제를 개선할 수 있는 중요한 방안으로 판단된다.

Turbid Water Simulation Model for Downstream of the Imha and Andong Dam (임하 및 안동댐 하류하천 탁도모의 시스템 구축)

  • 우리나라는 몬순기후대에 속하여 매년 홍수가 발생하고 이시기에 저수지로 유입된 탁수로 말미암아 저수지 인근 및 하류하천에 거주하는 주민들의 민원의 대상이 되고 있다. 이에 따라 유역, 저수지, 및 하천을 포함한 포괄적인 대책을 수립하기 위하여 유역과 저수지 그리고 저수지와 하천을 연계한 통합시스템을 구축함으로써 탁수의 거동을 고려한 저수지 운영에 대한 의사결정을 지원하고자 한다. 본 연구에서는 안동 및 임하댐의 연계운영을 통한 하류하천 탁도관리 방안을 수립하고자 동적수질모형인 Koriv1-win 모형을 구축하여 저수지 수질모의 시스템과 연계 가능하도록 하였다. 모형은 안동댐하류로부터 구미수위표 지점까지 134 km에 달하는 구간에 대하여 구축하였으며 대상구간 내에는 내성천을 비롯 5개의 주요지천이 존재하게 된다. 그림 1에는 수리모듈 및 수질모듈의 검증결과를 나타내고 있으며 탁도모의결과를 검증하기 위하여 임하댐 관리단에서 측정한 자료를 이용하였다. 수리해석결과 구미지점의 유량변동을 잘 반영하는 것으로 모의되었고 수질해석결과도 관측치와 상당히 비슷하게 나타나는 것을 알 수 있었다. 한편 안동댐방류수와 임하댐 방류수가 혼합되는 영역에서 2차원적 흐름양상과 각 댐으로부터 방류되는 방류수의 수온과 탁도의 혼합양상을 모의하였으며 실측을 통하여 모의결과와 비교해 보았다. 저수지 하류하천에 대하여 1차원 및 2차원적 수리해석과 수질해석결과를 통하여 하천에서의 탁수전파 거리 및 고탁도 탁수의 도달 시간을 산정할 수 있도록 함으로써 저수지 연계운영을 통한 하류단 탁도저감에 대한 계획수립이 가능하다.을 것으로 판단된다. 철거 선정 체계는 보 철거를 위해서 보 주변 지역 거주민을 설득하며, 하천의 환경 개선을 위한 합리적인 대안 제시를 목적으로 하고 있다. 선정 체계를 바탕으로 주민 협의 및 대안 제시를 통해 결정된 대상 보는 선정 체계 안에서 보 철거 영향 판단 절차에 따라서 보 철거로 인한 수문, 수리, 지형, 수질, 생태 영향을 판단하게 되며, 이와 더불어 사회 경제적인 영향을 평가하게 된다. 평가결과에 따라서 보를 완전히 철거하거나 다른 대안을 고려하여 보를 부분적으로 철거하거나 개량하게 된다.곳으로 1/3의 자기 생산을 담당하고 있었다. $\ulcorner$경상도지리지$\lrcorner$(慶尙道地理志)에는 $\ulcorner$세종실록$\lrcorner$(世宗實錄) $\ulcorner$지리지$\lrcorner$(地理志)와 동년대에 동일한 목적으로 찬술되었음을 알 수 있다. $\ulcorner$경상도실록지리지$\lrcorner$(慶尙道實錄地理志)에는 $\ulcorner$세종실록$\lrcorner$(世宗實錄) $\ulcorner$지리지$\lrcorner$(地理志)와의 비교를 해보면 상 중 하품의 통합 9개소가 삭제되어 있고, $\ulcorner$동국여지승람$\lrcorner$(東國與地勝覽) 에서는 자기소와 도기소의 위
