Cytotoxicities of Tumor-specific T Lymphocytes Primed by Glioma Apoptotic Body - or Glioma Cell Lysate-pulsed Dendritic Cells

  • Objective : The choice of tumor antigen for dendritic cell[DC]-loading has still been an unresolved problem in the DC-based vaccine strategies against malignant gliomas that has not been found well-characterized tumor specific antigens. In this study, we compare tumor-specific T cell response induced by glioma apoptotic body[GAB]-pulsed DCs to response induced by glioma cell lysate-pulsed ones quantitatively. Methods : DCs generated in the presence of granulocyte macrophage-colony stimulating factor and interleukin[IL]-4 from peripheral blood mononuclear cells[PBMCs] of HLA-A2 positive healthy donors were cultured. Each GABs and glioma cell lysate generated from HLA-A2 positive T98G glioblastoma cells were co-incubated with DCs. $CD8^+$ T lymphocytes isolated from PBMCs of same donors were cultured in media containing IL-2 and either stimulated by GAB- or lysate-pulsed DCs three times at a weekly interval. The interferon[IFN]-${\gamma}$ concentrations of each cell culture supernate were measured by enzyme immunoassay technique. Cytolytic activity of the generated cytotoxic $CD8^+$ T cells either stimulated with GAB- or lysate-pulsed DCs was determined by a standard 4-h $^{51}Cr$-release assay. Results : IFN-${\gamma}$ production and cytolytic activity of effector T cells stimulated by GAB-pulsed DCs were significantly higher than those of T cells stimulated by lysate-pulsed ones. Conclusion : These results indicate the choice of antigen is a critical determinant in the induction of antitumor immunity against malignant glioma. Antigen preparations from GABs represent a promising alternative to glioma cell lysate in DC-based glioma vaccine strategies.

  • The chromatographic separation of the MeOH extract of the aerial parts from Aster hispidus (Compositae) led to the isolation of eight compounds. Their structures were established by spectroscopic methods to be ${\beta}-amyrin$ (1), oleanolic acid (2), (2R)-1, 2-O-(9Z, 12Z, $15Z-dioctadecatrienoyl)-3-O-{\beta}-D-galactopyranosyl\;glycerol$ (3), trans-phytol (4), 9, 12, 15-octadecatrienoic acid (5), kaempferol (6), 3,5-dicaffeoyl quinic acid (7), 3,4-dicaffeoyl quinic acid (8) and kaempferol-3-O-rutinoside (9). Compounds 1, $3{\sim}6$ and 9 showed non-specific moderate cytotoxicity against five human tumor cell lines $(5.44{\sim}23.51\;{\mu}g/ml)$. The other compounds were of marginal activity against tested five human cancer cell lines $(9.05{\sim}>30.0\;{\mu}g/ml)$.

Effect of Lactobacillus Fermentation on the Anti-Inflammatory Potential of Turmeric

  • Curcumin, the major bioactive constituent of turmeric, has been reported to have a wide range of pharmacological benefits; however, the low solubility in water has restricted its systemic bioavailability and therapeutic potential. Therefore, in the current study, we aimed to investigate the effect of turmeric fermentation on its curcumin content and anti-inflammatory activity by using several lactic acid bacteria. Fermentation with Lactobacillus fermentum significantly increased the curcumin content by 9.76% while showing no cytotoxicity in RAW 246.7 cells, as compared to the unfermented turmeric, regardless of the concentration of L. fermentum-fermented turmeric. The L. fermentum-fermented turmeric also promoted cell survival; a significantly higher number of viable cells in lipopolysaccharide (LPS)-induced RAW 264.7 cells were observed as compared to those treated with unfermented turmeric. It also displayed promising DPPH scavenging ($7.88{\pm}3.36%$) and anti-inflammatory activities by significantly reducing the nitrite level and suppressing the expression of the pro-apoptotic tumor necrosis factor-alpha and Toll-like receptor-4 in LPS-induced RAW 264.7 cells. Western blot analysis further revealed that the anti-inflammatory activity of the fermented turmeric was exerted through suppression of the c-Jun N-terminal kinase signal pathway, but not in unfermented turmeric. Taken together, the results suggested that fermentation with lactic acid bacteria increases the curcumin content of turmeric without increasing its cytotoxicity, while strengthening the specific pharmacological activity, thus, highlighting its potential application as a functional food ingredient.

맥문동 물 추출물의 선천면역 활성과 염증억제 효과 (The Effects of Liriopis Tuber Water Extract on Innate Immune Activation and Anti-Inflammation)

  • Objectives: This study was designed to examine the anti-cancer activity by innate immunomodulating and anti-inflammatory effects of liriopis tuber water extract (LPE). Methods: Cell cytotoxicity was tested with 4T1 mouse mammary carcinoma cells, spleen cells, macrophage, and RAW264.7 cells. To investigate innate immunomodulating effects of LPE on macrophage, we measured tumor necrosis factor-alpha (TNF-α), interleukin-12 (IL-12), and interleukin-10 (IL-10). To investigate innate immunomodulating effects of LPE on RAW264.7 cell, we measured TNF-α, interleukin-6 (IL-6). In addition, TNF-α and nitric oxide (NO) induced by lipopolysaccharide (LPS) were measured after treating with LPE to observe innate immunomodulating effect of LPE on RAW264.7 cell. Also, mitogen-activated protein kinase (MAPK) and nuclear factor κB (NF-κB) were examined by western blot analysis. Results: In an in vitro cytotoxicity analysis, LPE affected tumor cell growth above specific concentration. As compared with the control group, the production of TNF-α, IL-12, and IL-10 were increased in macrophage. As compared with the control group, the production of TNF-α and IL-6 were increased in RAW 264.7 cell. The expression of TNF-α and NO induced by LPS after treating LPE was decreased. In addition, treatment of RAW 264.7 cell with LPE increased the phosphorylation levels of p-extracellular signal-regulated kinase (p-ERK), p-Jun N-terminal kinase (p-JNK), and p-p38. Conclusions: LPE might have impact on the anti-cancer effect by activation of innate immune system and inflammation control.

Essential Oil of Tridax procumbens L Induces Apoptosis and Suppresses Angiogenesis and Lung Metastasis of the B16F-10 Cell Line in C57BL/6 Mice

  • Background: To determine the effect of essential oil obtained from a traditionally used medicinal plant Tridax procumbens L, on lung metastasis developed by B16F-10 melanoma cells in C57BL/6 mice. Materials and Methods: Parameters studied were toxicity, lung tumor nodule count, histopathological features, tumor directed capillary vessel formation, apoptosis and expression levels of $P^{53}$ and caspase-3 proteins. Results: In vitro the MTT assay showed cytotoxicity was found to be high as 70.2% of cancer cell death within 24hrs for $50{\mu}g$. In vivo oil treatment significantly inhibited tumor nodule formation by 71.7% when compared with untreated mice. Formation of tumor directed new blood vessels was also found to be inhibited to about 39.5%. TUNEL assays also demonstrated a significant increase in the number of apoptotic positive cells after the treatment. $P^{53}$ and caspase-3 expression was also found to be greater in the essential oil treated group than the normal and cancer group. Conclusions: The present investigation showed significant effects of the essential oil of Tridax procumbens L in preventing lung metastasis by B16F-10 cell line in C57BL/6 mice. Its specific preventive effect on tumor directed angiogenesis and inducing effect on apoptosis warrant further studies at the molecular level to validate the significance of Tridax procumbens L for anticancer therapy.

길경(桔梗)이 면역활성(免疫活性) 및 항암(抗癌)에 미치는 영향 (Antitumor and Immunomodulatory Activities of Platycodon grandiflorum)

  • Methods: Intravenous administration of Platycodon grandiflorum was performed 2 days before tumor inoculation, then mice were killed 14 days after tumor inoculation, then number of tumor colonies were counted. Methanol extracts of Platycodon grandiflorum was added to colon26-M3.1 carcinoma cells, L5178Y-R lymphoma cells and Hela cells, and then cell growth was counted. To observe the immunomodulating effects of Platycodon grandiflorum, production of IL-6, IL-10, IL-12 and TNF-$\alpha$ were measured with ELISA assay and the cell growth of macrophage were also counted. Furthermore, antimetastatic experiment after depletion NK cells by injection of anti-asialo GM1 serum was also administered. Results: Intravenous administration of Platycodon grandiflorum significantly inhibited metastasis of colon26-M3.1 carcinoma cells. In an in vitro cytotoxicity analysis, Platycodon grandiflorum affected tumor cell growth above specific concentration. As compared with control, the production of IL-6, IL-10, IL-12 and TNF-$\alpha$ were incresed. And depletion NK cell completly abolished the inhibitory effect of metastasis. Conclusion: Platycodon grandiflorum appears to have considerable activity on immunomodulating effects and inhibit the metastasis of tumor. Further evaluation is needed for settling this.

Biotin-Conjugated Block Copolymeric Nanoparticles as Tumor-Targeted Drug Delivery Systems

  • To achieve targeted drug delivery for chemotherapy, a ligand-mediated nanoparticulate drug carrier was designed, which could identity a specific receptor on the surfaces of tumor cells. Biodegradable poly(ethylene oxide)/poly$({\varepsilon}-caprolactone)$ (PEG/PCL) amphiphilic block copolymers coupled to biotin ligands were synthesized with a variety of PEG/PCL compositions. Block copolymeric nanoparticles harboring the anticancer drug paclitaxel were prepared via micelle formation in aqueous solution. The size of the biotin-conjugated PEG/PCL nanoparticles was determined by light scattering measurements to be 88-118 nm, depending on the molecular weight of the block copolymer, and remained less than 120 nm even after paclitaxel loading. From an in vitro release study, biotin-conjugated PEG/PCL nanoparticles containing paclitaxel evidenced sustained release profiles of the drug with no initial burst effect. The biotin-conjugated PEG/PCL block copolymer itself evidenced no significant adverse effects on cell viability at $0.005-1.0{\mu}g/mL$ of nanoparticle suspension regardless of cell type (normal human fibroblasts and HeLa cells). However, biotin-conjugated PEG/PCL harboring paclitaxel evidenced a much higher cytotoxicity for cancer cells than was observed in the PEG/PCL nanoparticles without the biotin group. These results showed that the biotin-conjugated nanoparticles could improve the selective delivery of paclitaxel into cancer cells via interactions with over-expressed biotin receptors on the surfaces of cancer cells.

L5187Y 세포에 대한 불화피리미딘 대사억제제 독성에 관한 Benzylacyclouridine의 영향 (The Effects of 5-benzylacyclouridine on the Cytotoxicities of Fluorinated Pyrimidine Antimetabolic Agents in L5178Y Cells)

  • Benzylacyclouridines (BAU and BBAU)는 uridine phosphorylase (UrdPase)의 선택적이고 강력한 상경억제제이다. 보고된 바에 의하면 (Cancer Res., 44: 1852, 1984) Benzylacyclouridines가 5-fluoro-2'-deoxyuridine (FdUrd)의 인체 암세포에 대한 독성을 증가시켜 준다고 하였지만, L5187Y 세포를 사용한 본 실험에서는 Benzylacyclouridines가 FdUrd를 포함하여 5-fluorouridine (FUra) 모두에 대해 조금도 세포 독성을 증가시키지 못하였을 뿐만아니라, 오히려 세포를 그들 독성으로부터 투여량에 비례하여 보호하였다. 복강내 주사에 의한 생체실험에서도 Benzylacyclouridines는 5-fluorinated pyrimidine에 의한 L5187Y를 지닌 쥐(mouse)의 life-span을 연장시켜 주지 못하였다. 본 실험에서 Benzylacyclouridines가 기대했던 fluorinated pyrimidine 항암제의 효과를 증진시키지 못한 이유는 nucleosides의 anabolism이 UrdPase와 orotate phosphoribosyltransferase이 의한 sequential 작용에 의하던가 또는 Benzylacyclouridines에 의한 nucleosides의 수송억제에 의하던가, 아니면 두가지 다 복합적으로 작용한 결과로 생각된다.

Effects of Inonotus obliquus Extracts on Immunomodulating Activity

  • Aim : This study aimed at elucidating the effects of Inonotus obliquus on anti-tumor effects in vivo and immune-based characterization of the mushroom as a potential candidate for cancer remedy. Methods : To investigate the immunomodulatory effects of Inonotus obliquus, we investigated macrophage functions and NK cell activities through the measurement of NO production of macrophage, NK cell cytotoxicity and expressions of cytokines and genes regulating immune responses, in addition to pulmonary metastasis model in vivo. Results : Inonotus obliquus showed general cytotoxicity at high concentrations over the 100 ${\mu}g/m{\ell}$ on the both of normal and cancer cell lines. Inonotus obliquus showed both inhibitory and promotive effects on pulmonary colonization of CT-26 cell depending on period or route of administration in vivo. Conclusion : From these results, it cannot be concluded that Inonotus obliquus has cancer-specific activity. Furthermore, Inonotus obliquus has the provability to show adverse effects differently according to the concentration and the method of administration.

Antimicrobial and Antitumor Photodynamic Effects of Phleichrome from the Phytopathogenic Fungus Cladosporium Phlei

  • Fungal perylenequinones have photodynamic activity and are promising photosensitizers for photodynamic therapy (PDT). Here, we investigated the bactericidal and antitumor activities of phleichrome from the fungal perylenequinone family in vitro. Photodynamic bactericidal activity of phleichrome was analyzed by agar-well diffusion method under dark and illuminated conditions. The photodynamic antitumor activity of phleichrome was analyzed in MCF-7, HeLa, SW480, and HepG2 human cancer cell lines using in vitro cytotoxicity assays. Photodynamic bactericidal activities against Gram-negative and Gram-positive bacteria were species-specific. Antitumor activity against all tumor cell lines increased under the illuminated condition. Depending on the results of the analyses, Phleichrome has potential for further drug development related to its antibacterial and antitumor activities.
