Objectives: The aim of this study was to compare the push-out bond strength and dentinal tubule penetration of root canal sealers used with coated core materials and conventional gutta-percha. Materials and Methods: A total of 72 single-rooted human mandibular incisors were instrumented with NiTi rotary files with irrigation of 2.5% NaOCl. The smear layer was removed with 17% ethylenediaminetetraacetic acid (EDTA). Specimens were assigned into four groups according to the obturation system: Group 1, EndoRez (Ultradent Product Inc.); Group 2, Activ GP (Brasseler); Group 3, SmartSeal (DFRP Ltd. Villa Farm); Group 4, AH 26 (Dentsply de Trey)/gutta-percha (GP). For push-out bond strength measurement, two horizontal slices were obtained from each specimen (n = 20). To compare dentinal tubule penetration, remaining 32 roots assigned to 4 groups as above were obturated with 0.1% Rhodamine B labeled sealers. One horizontal slice was obtained from the middle third of each specimen (n = 8) and scanned under confocal laser scanning electron microscope. Tubule penetration area, depth, and percentage were measured. Kruskall-Wallis test was used for statistical analysis. Results: EndoRez showed significantly lower push-out bond strength than the others (p < 0.05). No significant difference was found amongst the groups in terms of percentage of sealer penetration. SmartSeal showed the least penetration than the others (p < 0.05). Conclusions: The bond strength and sealer penetration of resin-and glass ionomer-based sealers used with coated core was not superior to resin-based sealer used with conventional GP. Dentinal tubule penetration has limited effect on bond strength. The use of conventional GP with sealer seems to be sufficient in terms of push-out bond strength.
The sealing effect of a few cavity liners upon the dentinal tubule were studied in vitro. The materials employed in this study were Silcot (SP$\'{E}$CIALIT$\'{E}$S SEPTODONT, M.-T. GENDRAULT, Pharmacien), Hypo-Cal(Ellman Dental Mfg.Co.Inc.), Cavity Lining(De Trey), and Copaute(Harry J. Bosworth Co.). Freshly extracted human teeth were devided into 5 groups by age-under twenty, twenties, thirties, forties, and over fifty. Class V cavities were prepared routinely. The cavity walls of eight teeth of each group were lined by Silcot, Copalite, Cavity Lining, and Hypo-Cal. Remaining eight were not lined as a control. These specimens were immersed in dye solution (2 gm eosin Y to 800 cc distilled water) for 48 hours to allow maximum dye penetration into dentinal tubules. Each specimen was sectioned longitudinally including Class V cavity floor under water spray. Dye penetration into dentinal tubules were examined and following results were obtained. 1. Liners used on this study showed more or less dye penetration into dentinal tubules. But compared with the teeth without lining, the dye penetration of lined specimens were decreased. 2. Of these liners tested, Silcot was the most effective sealer upon the dentinal tubules. Copalite was the moderate sealer and Cavity Lining showed a tendency similar to Copalite. Hypo-Cal revealed the greatest dye penetration. 3. As the age was increased, the more the dye penetration into the dentinal tubules was decreased.

The purpose of this study is to examine the sealing effect and efficiency of root canal filling MTA (Endoseal, Endoseal MTA). A total of 106 extracted single rooted teeth were used and classified with group AH (AH-26), group PR (ProRoot MTA), group ES (Endoseal) and group EM (Endoseal MTA) depending on filled sealers. Time was measured in each group when sealers were filled. The groups were divided into subgroup A and subgroup B. The sealing of root canal walls and penetration of sealer in the dentinal tubule were evaluated, respectively. According to the results, the sealing of root canal walls and dentinal tubule penetration of root canal filling MTA were inferior to AH-26 (p < 0.05). When compared with ProRoot MTA, however, there was no significant difference in sealing of root canal walls (p > 0.05), but dentinal tubule penetration was high (p < 0.05). Working time was shorter in root canal filling MTA than ProRoot MTA and AH-26 (p < 0.05). In conclusion, root canal filling MTA has lower root canal sealing effect than resin-based sealer, however, when in MTA needed root canal filling, it could be an effective alternative.
The purpose of this study was to observe the sealing ability and durability of All-Bond 2, Gluma Bonding System, Scotch bond 2 and Superbond D-liner which are dentin bonding agents used as desensitizing agents. The durability of the sealing ability of the materials were compared after 0, 140, 420, 840 tooth brushing strokes. 120 extracted teeth were divided into 5 groups and the agents were applied to the exposed dentin. No agents was applied on group I, the control group. Each specimen went through thermocycling from $5^{\circ}$ to $55^{\circ}C$, 200 times. Each group was devide into 4 subgroups and artificial tooth brushing strokes were done for 0, 140, 420, 840 times. Finally the specimens were stored in 0.5 % methylene blue solution for 24 hours in a incubator set at $37^{\circ}C$. The tooth were sectioned perpendicular to the long axis and the dye penetration ratio to the pulp was measured. The following results were obtained. 1. All four dentin bonding agents initially showed excellent sealing ability. 2. All-bond 2, Gluma Bonding System and Superbond D-liner showed durability of dentinal tubule sealing effect after 840 strokes(6-week) artificial tooth brushing. 3. Scotchbond 2 showed a significant decrease in sealing ability after 420 strokes(3-week) artificial tooth brushing. (P<0.05).

Objectives: This study evaluated the dentinal penetration depth of 2.5% sodium hypochlorite (NaOCl) in root canals with and without preparation and different irrigant activation protocols. Materials and Methods: Sixty-three bovine mandibular incisors were randomly allocated to 6 groups (n = 10): G1, preparation + conventional needle irrigation (CNI); G2, preparation + passive ultrasonic irrigation (PUI); G3, preparation + Odous Clean (OC); G4, no preparation + CNI; G5, no preparation + PUI; G6, no preparation + OC; and CG (negative control; n = 3). Samples were filled with crystal violet for 72 hours. Irrigant activation was performed. Samples were sectioned perpendicularly along the long axis, 3 mm and 7 mm from the apex. Images of the root thirds of each block were captured with a stereomicroscope and analyzed with an image analysis software. One-way analysis of variance, followed by the Tukey post hoc test, and the Student's t-test were used for data analysis, with a significance level of 5%. Results: The NaOCl penetration depth was similar when preparation was performed, regardless of the method of irrigation activation (p > 0.05). In the groups without preparation, G6 showed greater NaOCl penetration depth (p < 0.05). The groups without preparation had a greater NaOCl penetration depth than those with preparation (p = 0.0019). Conclusions: The NaOCl penetration depth was similar in groups with root canal preparation. Without root canal preparation, OC allowed deeper NaOCl penetration. The groups without preparation had greater NaOCl penetration than those undergoing root canal preparation.

Objectives: This study investigated the cytotoxicity, radiopacity, pH, and dentinal tubule penetration of a paste of 1.0% calcium-doped zinc oxide nanocrystals (ZnO:1.0Ca) combined with propylene glycol (PRG) or polyethylene glycol and propylene glycol (PEG-PRG). Materials and Methods: The pastes were prepared by mixing calcium hydroxide [Ca(OH)2] or ZnO:1.0Ca with PRG or a PEG-PRG mixture. The pH was evaluated after 24 and 96 hours of storage in deionized water. Digital radiographs were acquired for radiopacity analysis and bubble counting of each material. The materials were labeled with 0.1% fluorescein and applied to root canals, and images of their dentinal tubule penetration were obtained using confocal laser scanning microscopy. RAW264.7 macrophages were placed in different dilutions of culture media previously exposed to the materials for 24 and 96 hours and tested for cell viability using the MTT assay. Analysis of variance and the Tukey test (α = 0.05) were performed. Results: ZnO:1.0Ca materials showed lower viability at 1:1 and 1:2 dilutions than Ca(OH)2 materials (p < 0.0001). Ca(OH)2 had higher pH values than ZnO:1.0Ca at 24 and 96 hours, regardless of the vehicle (p < 0.05). ZnO:1.0Ca pastes showed higher radiopacity than Ca(OH)2 pastes (p < 0.01). No between-material differences were found in bubble counting (p = 0.0902). The ZnO:1.0Ca pastes had a greater penetration depth than Ca(OH)2 in the apical third (p < 0.0001). Conclusions: ZnO:1.0Ca medicaments presented higher penetrability, cell viability, and radiopacity than Ca(OH)2. Higher values of cell viability and pH were present in Ca(OH)2 than in ZnO:1.0Ca.
The effect of moistening and air-drying of acid-conditioned dentin before priming on the formation of resin-dentin hybrid zone was investigated, Freshly extracted human molars were used and divided at random into 5 groups, Groups 1 - 3 consisted of specimens conditioned with 10 % phosphoric acid for 20 seconds; Group 1 served as a control in which the conditioned dentin was simply blot-dried with a damp facial tissue; Group 2 was air dried for 30 seconds ; Group 3 was air dried for 30 seconds and immediately remoistened for 10 seconds with air-water syringe. and then the specimen was blot-dried with a damp facial tissue. Groups 4-5 were not acid conditioned ; In group 4, the smear layer on the dentin was blot dried before primer placement; Group 5 was air dried only for 30 seconds, The acetone-based primer and bonding agent of All Bond 2 (Bisco. Inc., USA) and composite resin (Z-100, 3M Dental products, USA) were applied for acid conditioned dentin and non-conditioned dentin. The morphologic ultrastructure of resin-dentin hybrid zone was examined by the use of SEM and TEM. and the existence of inorganic material and analysis of Ca/P weight-percent ratio in the resin-dentin hybrid zone were revealed by the EDAX, The results were as follows : 1. In the moistened specimens from acid-conditioned groups, the resin penetrated about 3-$4{\mu}m$ into dentin and the denatured collagen smear layer was not present at the surface. The resin tag was formed to a thickeness of 3-$4{\mu}m$ at the upper part of dentinal tubule and compactively connected to each other by means of many lateral branching. 2. In the air-dried specimens from acid-conditioned groups, the resin penetrated about 2.0-$2.5\;{\mu}m$ into dentin and an upper thin black layer to a thickness of 30-35nm was identified between adhesive resin and demineralized collagen layer. The resin tag to have a diameter of $2.5{\mu}m$ was formed at the upper part of dentinal tubule. However the funnel shape of the tag was not notable compared to the moistened specimens. 3. In the remoistened specimens from acid conditioned groups, the resin penetrated about 2.0-$2.5{\mu}m$ into dentin and an upper black layer was not present. The resin tag at the upper part of dentinal tubule was formed less than $2{\mu}m$ and was weakly connected to each other by means of few lateral branching. 4. In the non-conditioned groups, the smear layer was formed to a thickness of $0.5{\mu}m$ at dentin surface. However, the resin-dentin hybrid zone was not identified by TEM. The evidence of resin penetration into intertubular and intratubular dentin did not show. 5. All the acid-conditioned groups showed that the detected calcium and phosphorus weight percent ratios at the $2{\mu}m$ upper portion from the resin-dentin interface into the resin were much higher than that at the $2{\mu}m$ lower portion from the resin-dentin interface to dentin. (P<0.01).
The purpose of this study was to evaluate on the interfacial morphology between dentin and restorative materials. In this in vitro study, the cavity wall restorated with 3 different kinds of tooth colored restorative materials [resin-modified Glass Ionomer cement (Fuji II LC), composite resin (Z-100), compomer (Dyract)]. The thirty extracted human molar teeth without caries and/or restorations are used. The experimental teeth were randomly divided into three groups of ten teeth each. In each group, Wedge shaped cavities (width: 3mm, length: 2mm, depth: 1.5mm) were prepared at the cementoenamel junction on buccal and lingual surfaces. The adhesive of composite resin were mixed with rhodamine B. Primer of composite resin, Prime & Bond 2.1 of Dyract and liquid of Fuji II LC were mixed with fluorescein. In group 1, the cavity wall was treatment with dentin conditioner, and then restorated with Fuji II LC. In group 2, the cavity wall was treatment with Prime & Bond 2.1 and then restorated with Dyract. In group 3, the cavity wall was etching with 10% maleic acid, applied with primer and bonding agent and then restorated with Z-100. The interface between dentin and restorative materials was observed by fluoresence imaging with a confocal laser scanning microscope. The results were as follows : 1. In Glass ionomer group, adaptation of resin modified Glass-ionomer restoration against cavity wall is tight, but the crack formed inside of restoration were observed. 2. In Dyract group, the penetration of resin tag is shorter and the width of hybrid layer is narrower than composite resin group. 3. In Z-100 group, primer penetrated deeply through dentinal tubule. Also bonding agent was penetrated along the primer, but the penetration length is shorter than primer part, and in 3-D image, the resin tag is conical shape and lateral branch is observed.
If root and resection is done during surgical endodontic treatment, newly exposed dentinal tubules form pathways between the canal and the peripheral tissue. Nd : YAG laser was used to block this phenomenon, and its effect was studied with dye penetration and SEM techniques. 40 intact single rooted teeth were divided into 4 groups(10 each) : control group and test groups, in which retrograde cavity surface, cutting surface, retrograde cavity surface & cutting surface were treated with laser(1 watt 15pps) and finally retrograde filling with IRM was conducted. After that, they were stained with 2 % methylene blue, sectioned and evaluated by the maximum infiltration depth. And to observe surface change, they were prepared for SEM. The results were as follows ; 1. All experimental groups showed microleakage with variation in amount. 2. The 2nd group which treated both the retrograde cavity and cutting surface showed significantly less microleakage than the other groups(p<0.05). There was no significant difference between groups treated on one side only. 3. As a result of SEM observation of dentin surface, obstruction of dentinal tubules with marble shaped granules, which were different from normal dentin could be seen. Cracks could be seen also. 4. In summary of this experiment, it is thought that effort to obstruct the exposed dentinal tubules as well as retrograde cavity after root end resection is needed.
The purpose of this study is to measure the bonding strength at the enamel and dentin of extracted bovine incisors when they are irradiated by W Nd: YAG, EoQ Nd: YAG, and CW $CO_2$ laser, etched and then attached by composite resin. The laser-irradiated surface, the etched surface after the laser-irradiation and the interface of the treated surface and resin are observed by using scanning electron microscope. The NP Nd:YAG laser is used at the peam power of 207 KW. The EOQ Nd: YAG laser is used at the peak power of 1.15Mk. The CW $CO_2$ laser is used at the energy output of 5W and 10W in enamel; in dentin, at the energy output of 2W. The obtained results were as follows: 1. The shear bond strength of enamel decreased in the following order: EOQ Nd:YAG laser group, NP Nd:YAG laser group, CW $CO_2$ laser groups, unlased group. The unlased group showed a significant difference from EOQ Nd:YAG laser group (P<0.01), NP Nd:YAG laser group (P<0.05), but no significant difference from CW $CO_2$ laser group (P>0.05). 2. The shear bond strength of dentin decreased in the following order: CW $CO_2$ laser group, NP Nd:YAG laser group, unlased group, EOQ Nd:YAG laser group. The unlased group showed a significant difference from CW $CO_2$ laser group (P<0.01), but no significant difference from Nd:YAG laser groups (P>0.05). 3. The scanning electron microscope of enamel revealed irregular microcrack and pore at the surface in the NP Nd:YAG laser group and CW $CO_2$ laser group, but the crackless flat surface in the EOQ Nd:YAG laser group. 4. The scanning electron microscope of dentin revealed obstruction of in most of dentinal tubule and decrease of diameter. 5. The scanning electron microscope of the interface of resin and enamel revealed prominent penetration of resin tag in the EOQ Nd:YAG laser group.
