The disinfection effect of UV-C and calcium hypochlorite to shrimp farm instruments contaminated with EHP (Enterocytozoon hepatopenaei) (EHP (Enterocytozoon hepatopenaei)에 오염된 사육기구에 대한 UV-C와 차아염소산칼슘의 소독 효과)

  • In this study, nylon mash and silicone tube mainly used as shrimp farm equipment were contaminated with Enterocytozoon hepatopenaei (EHP) which is the cause of Hepatopancreatic microsporidiosis (HPM), and were treated with calcium hypochlorite or UV-C disinfection methods for EHP eradication. As a result, similar with the control group (not disinfected), EHP was detected on the nested PCR until the 10 days in the UV-C single treated group. On the other hand, EHP was not detected from 7 days in calcium hypochlorite single treated group (total concentration 200 ppm as available chlorine), and combination of calcium hypochlorite and UV-C treated group revealed no detection of EHP from 3 days. It is appropriate that treated with UV-C and calcium hypochlorite for 3 days or single treated with calcium hypochlorite for 7 days to eradicate EHP on contaminated instrument used in shrimp farms. In contrast, disinfection effect of only using UV-C is very low.

Efficiency of Silver Ion-Silica Solid Phase Extraction for Elimination of Sulfur Compounds during Pesticide Multiresidue Analysis in Allium Species (파속류의 잔류농약분석과정에서 silver ion-silica solid phase extraction에 의한 황화합물의 제거 효율성)

  • Since organic sulfur compounds respond to GC/ECD sensitively, they interfere with quantitative separation of pesticides during residual pesticide analysis of Allium species. In this study, it was intended to develop a rapid and simple method for pesticide multi-residues analysis through clean-up and interferences by a solid-phase extraction (SPE). An SPE method employing silver nitrate impregnated Florosil cartridge was developed and evaluated for the elimination of sulfur compounds from the test solution of Allium species during pesticide residues analysis. The silver nitrate impregnated Florosil cartridge was prepared by efflux of 3 ml of 20% silver nitrate solution through Florosil cartridge (1 g packing, 6 ml tube). The extracts equivalent to 2, 4 6, and 10 g of each sample were loaded onto the cartridge and allowed to exude, and then the exudations were analyzed by GC/ECD. More than 95% of sulfur compounds were removed from the loaded extracts equivalent, up to 6 g in onion, 4 g in spring onion and 4 g in shallot, respectively. 40 pesticides were spiked in the Allium species and loaded onto the cartridge to determine the recoveries; from this, the recoveries of 34 pesticides were within 70~120%.

Acaricidal and Repellent Activities of Cnidium officinale-derived Compounds Against Haemaphysalis longicornis (Acari: Ixodidae) (작은소피참진드기에 대한 다양한 식물체 및 천궁유래물질의 살비 및 기피 효과)

  • This study investigated the acaricidal and repellent activities of various plant extracts and Cnidium officinale-derived materials against Haemaphysalis longicornis. Among the 24 methanolic plant extracts, C. officinale showed the highest acaricidal activity (93.3%) against H. longicornis. Platycodon grandiflorus, Phytolacca americana, Magnolia denudata, Houttuynia cordata, C. officinale, and Pueraria montana showed significant differences in their repellent activity. Consequently, the activity of solvent-fractionated materials and the derived material of the C. officinale, was investigated and a 90% acaricidal effect was observed three days after hexane fraction treatment, indicating that the repellent response was statistically significant. Butylidenephthalide, which is a C. officinale-derived compound, showed 90% acaricidal activity seven days after treatment. Additionally, all H. longicornis moved to the untreated area in the t-tube olfactometer experiment, confirming the repellent activity of butylidenephthalide. Therefore, the results of this study suggest the use of C. officinale crude extract and butylidenephthalide as potent acaricidals and repellents against H. longicornis, respectively.

PCR Analysis for Halal Authentication of Cosmetics Containing Carrot, Oyster Mushroom, and Pine Needle Extracts (당근, 느타리버섯 및 솔잎 추출물이 첨가된 화장품의 할랄인증을 위한 PCR 분석)

  • In recent years, interest in halal authentication from the domestic food and cosmetics field has been growing for advances into the overseas halal market. For halal authentication, the product must not contain haram ingredients derived from pig, dog, human, GMO, etc. In this study, the presence of haram ingredients in plant extracts (carrot, oyster mushroom, and pine needle) treated with papain and bromelain and cosmetics (mask pack and cream) containing these extracts were analyzed by PCR to confirm whether these cosmetics were suitable for halal authentication. Detection limits of the PCR method that specifically detected template DNA of human, pig, dog, and GMO were $1.29{\times}10^3$, $1.14{\times}10^3$, $1.24{\times}10^2$ and $2.02{\times}10^3copies/tube$, respectively. PCR was not inhibited by the plant extracts or cosmetic ingredients. Results of PCR for the plant extracts or cosmetics containing these extracts were all negative. This PCR method could be used to rapidly identify the presence of haram ingredients in raw materials or final products during the manufacturing process of food and cosmetics.

  • One of the words that describe unique features of human beings is "Homo Ludens (Playing Man)". As humans enjoy abundant and prosperous lives and the quality of life is further improved, play is increasingly becoming a central value of human life. As part of play, ocean leisure is getting popular these days, and in turn demands for ocean leisure-related products in Korea, most of which are dependent on foreign markets, are growing. Therefore, it is required that local companies are quick to turn their eyes on the fast developing market. This research aims to help develop a design concept for bodyboard, more suitable for the domestic market, by analyzing products and reflecting customers' needs. The findings of this study are as follows: first, it is desirable that the target of bodyboard in domestic market is young people aged between 14 and 29 and its price ranges from 40,000 to 100,000 won; second, a tube-type bodyboard is appropriate for beaches in Korea mostly with small and gentle waves, as also shown in a survey that over 65% of those surveyed prefer a tube-type bodyboard, and lastly, the CMFF (Color, Material, Form, and Function) strategy was newly developed in this study in order to create new concept bodyboards.

Benefits of Palliative Office-Based Hyaluronic Acid Injection Laryngoplasty in Cancer-Related Unilateral Vocal Cord Paralysis Patients (악성 종양 관련 일측성 성대 마비 환자에서 고식적 Hyaluronic Acid 성대 주입 성형술의 유용성)

  • Background and Objectives : The effect of palliative injection laryngoplasty in cancer-related unilateral vocal cord paralysis patients on voice and swallowing function is uncertain and there are few previous studies of its suitability, benefits as a palliative treatment option. The purpose of this study is to confirm the objective results of voice and swallowing function after palliative office-based hyaluronic acid injection laryngoplasty in cancer-related unilateral vocal cord paralysis patients. Materials and Method : 36 patients who had unilateral vocal cord paralysis from non-thyroidal, extralaryngeal neoplasms were included in this study. To evaluate the clinical outcome, we analyzed perceptual GRBAS grading, acoustic analysis, aerodynamic study, Electroglottography (EGG), Voice Handicap Index (VHI-30) about voice function and disability rating scale (DRS), gastric tube dependency, aspiration pneumonia about swallowing function and 36-Item Short Form Survey version 2 (SF-36v2) about quality of life. Results : In GRBAS scale, G (p<0.001), R (p=0.004), B (p=0.001), A (p=0.011), and S (p=0.007) showed significant improvement. Jitter, shimmer, speaking fundamental frequency, maximal phonation time, VHI-30, DRS score, gastric tube dependency, aspiration pneumonia, and SF-36v2 were significantly improved after injection (p=0.016, p=0.011, p=0.045, p=0.005, p<0.001, p<0.001 p=0.003, p<0.001, and p<0.001 respectively). Conclusion : From this study we concluded office-based hyaluronic acid injection can be used as a useful palliative treatment option in cancer-related ill patients with unilateral vocal cord paralysis. Palliative hyaluronic acid injection laryngoplasty avoids the need for tube feeding, thus reducing the risk of aspiration pneumonia. These outcomes are accompanied by significant improvement in voice quality.

Fumigant Toxicity and the Repellent Effect of Essential Oils against Sitophilus zeamais Motschulsky (어리쌀바구미에 대한 식물정유의 훈증독성과 기피효과)

  • Stored grain insects appear frequently as grains are harvested, stored, and distributed on the market. Producers mostly depend on chemical fumigants to control and repel insect pests, but this application of chemicals may cause side effects for humans and the environment. Safer control approaches, including the use of natural products are required, and in this study, we assessed the control effect of essential oils on stored grain insects. The essential oils of 8 plant species from 6 families were tested for their fumigant toxicity and repellent effects using a Y-tube Olfactometer against Sitophilus zeamais adults. Pennyroyal and tea tree oil respectively exhibited 97% and 63% mortality at $10{\mu}{\ell}L^{-1}$ air concentration in tested fumigant toxicity. Of particular note, pennyroyal oil ($LT_{50}$, hour=3.87) showed higher adulticidal activities than tea tree oil ($LT_{50}$, hour=4.09). When assessing repellent effects, using a Y-tube Olfactometer at $10{\mu}{\ell}$/filter paper concentration, the cedarwood, cinnamon, cypress, patchouli and pennyroyal essential oils showed potent repellent activities against Sitophilus zeamais adults. Cinnamon, cypress and pennyroyal essential oils showed notably high repellent activities of 82.4%, 80.0% and 86.7%, respectively, at $1{\mu}{\ell}$/filter paper concentration.

  • TThis study explored the requirement of fabric sensor that can measure the motion of the joint effectively by measuring and analyzing the variation in electric resistance of a sensor in accordance with bending and stretching motion of the arm by the implementation of a motion sensor utilizing conductive fabric. For this purpose, on both sides of two kinds of knitted fabric, namely 'L' fabric and 'W' fabric Single Wall Carbon Nano-Tube(SWCNT) was coated, fabric sensor was developed by finishing them in a variety of ways, and the sensor was attached to the arm band. The fabric sensor consisted of total 48 cases, namely background fabric for coating, the method of sensor attachment, the number of layer of sensors, the length of sensor, and the width of sensor. The performance of fabric motion sensors in terms of a dummy arm, that is, a Con-Trex MJ with 48 arm bands around it was evaluated. For each arm band, a total of 48, fastened around the dummy arm, it was adjusted to repeat the bending and stretching at the frequency : 0.5Hz, ROM : $20^{\circ}{\sim}120^{\circ}$, the voltage was recorded for each case after conducting three sets of repeat measurement for a total of 48 cases. As a result of the experiment, and as a consequences of the evaluation and analysis of the voltage based on the uniformity of the base line of the peak-to-peak voltage(Vp-p), the uniformity of Vp-p within the same set, and the uniformity of the Vp-p among three sets, the fabric sensors that have been configured in SWCNT coated 'L' fabric / welding / two layers / $50{\times}5mm$, $50{\times}10mm$, $100{\times}10mm$, and SWCNT coated 'W' fabric / welding / two layers / $50{\times}10mm$ exhibited the most uniform and stable signal value within 5% of the total variation rate. Through all these results of the experiment, it was confirmed that SWCNT coated fabric was suitable for a sensor that can measure the human limb operation when it was implemented as a fabric sensor in a variety of forms, and the optimal sensor types were identified.

Characterization of human cardiac mesenchymal stromal cells and their extracellular vesicles comparing with human bone marrow derived mesenchymal stem cells

  • Cardiac regeneration with adult stem-cell (ASC) therapy is a promising field to address advanced cardiovascular diseases. In addition, extracellular vesicles (EVs) from ASCs have been implicated in acting as paracrine factors to improve cardiac functions in ASC therapy. In our work, we isolated human cardiac mesenchymal stromal cells (h-CMSCs) by means of three-dimensional organ culture (3D culture) during ex vivo expansion of cardiac tissue, to compare the functional efficacy with human bone-marrow derived mesenchymal stem cells (h-BM-MSCs), one of the actively studied ASCs. We characterized the h-CMSCs as CD90low, c-kitnegative, CD105positive phenotype and these cells express NANOG, SOX2, and GATA4. To identify the more effective type of EVs for angiogenesis among the different sources of ASCs, we isolated EVs which were derived from CMSCs with either normoxic or hypoxic condition and BM-MSCs. Our in vitro tube-formation results demonstrated that the angiogenic effects of EVs from hypoxia-treated CMSCs (CMSC-Hpx EVs) were greater than the well-known effects of EVs from BM-MSCs (BM-MSC EVs), and these were even comparable to human vascular endothelial growth factor (hVEGF), a potent angiogenic factor. Therefore, we present here that CD90lowc-kitnegativeCD105positive CMSCs under hypoxic conditions secrete functionally superior EVs for in vitro angiogenesis. Our findings will allow more insights on understanding myocardial repair.

Metabolomics comparison of serum and urine in dairy cattle using proton nuclear magnetic resonance spectroscopy

  • Objective: The aim of the study was to conduct metabolic profiling of dairy cattle serum and urine using proton nuclear magnetic resonance (1H-NMR) spectroscopy and to compare the results obtained with those of other dairy cattle herds worldwide so as to provide a basic dataset to facilitate research on metabolites in serum and urine. Methods: Six dairy cattle were used in this study; all animals were fed the same diet, which was composed of total mixed ration; the fed amounts were based on voluntary intake. Blood from the jugular neck vein of each steer was collected at the same time using a separate serum tube. Urine samples were collected by hand sweeping the perineum. The metabolites were determined by 1H-NMR spectroscopy, and the obtained data were statistically analyzed by performing principal component analysis, partial least squares-discriminant analysis, variable importance in projection scores, and metabolic pathway data using Metaboanalyst 4.0. Results: The total number of metabolites in the serum and urine was measured to be 115 and 193, respectively, of which 47 and 81, respectively were quantified. Lactate (classified as an organic acid) and urea (classified as an aliphatic acylic compound) exhibited the highest concentrations in serum and urine, respectively. Some metabolites that have been associated with diseases such as ketosis, bovine respiratory disease, and metritis, and metabolites associated with heat stress were also found in the serum and urine samples. Conclusion: The metabolites measured in the serum and urine could potentially be used to detect diseases and heat stress in dairy cattle. The results could also be useful for metabolomic research on the serum and urine of ruminants in Korea.
