Characterization of Trueperella pyogenes isolated from caseous lymphadenitis lesions in Korean native goats

  • The study characterized Trueperella pyogenes (T. pyogenes) from caseous lymphadenitis (CLA) in Korean native goats (KNG). CLA samples (n=191) were obtained from abattoir-slaughtered KNG. Pus were obtained and Corynebacterium pseudotuberculosis was detected by PCR using PLD gene. Three PLD-negative samples (1.57%) from supramammary lymph node were subjected to bacterial culture. Isolates were identified using 16S rRNA and biochemically characterized by APIZYM. Phylogenetic and biochemical analyses identified the isolates as T. pyogenes, and have shown separate clusters and varied enzyme reaction, suggesting genotypic and phenotypic diversity. This first report of T. pyogenes-induced CLA in KNG provided information for further investigations.

도축 젖소에서 Trueperella pyogenes 감염에 의한 턱 밑 화농육아종 증례보고 (A case of submandibular pyogranuloma caused by Trueperella pyogenes in the slaughtered dairy cows)

  • Multiple enlarged submandibular nodules were discovered during a routine antimortem examination at slaughter in 3 dairy cows of 20-month-old. Grossly, submandibular nodules were firm, reddish, and oval in shape. On cut surface, multiple yellowish abscesses in the parenchyma were observed. Histopathologically, the nodules were characterized by severe diffuse pyogranulomatous inflammation and chronic fibrosis with multiple eosinophilic clubs of Splendore-Hoeppli material containing central bacterial colonies. Trueperella pyogenes was isolated from the lesions. This case was diagnosed as submandibular pyogranuloma caused by Trueperella pyogenes in slaughtered dairy cows in Korea.

An Advanced Understanding of Uterine Microbial Ecology Associated with Metritis in Dairy Cows

  • Metritis, the inflammation of the uterus caused by polymicrobial infections, is a prevalent and costly disease to the dairy industry as it decreases milk yield, survival, and the welfare of dairy cows. Although affected cows are treated with broad-spectrum antibiotics such as ceftiofur, endometrial and ovarian function are not fully recovered, which results in subfertility and infertility. According to culture-dependent studies, uterine pathogens include Escherichia coli, Trueperella pyogenes, Fusobacterium necrophorum, and Prevotella melaninogenica. Recent studies using high-throughput sequencing observed very low relative abundance of Escherichia coli, Trueperella pyogenes, and Prevotella melaninogenica in cows with metritis. Herein, we propose that metritis is associated with a dysbiosis of the uterine microbiota, which is characterized by high abundance of Bacteroides, Porphyromonas, and Fusobacterium.
