  • Six monoclonal antibodies to human cardiac troponin I (hcTnI) were produced to eventually develop an immunosensor for acute myocardial infarction (AMI). For the characterization of these antibodies, a set of 11 different peptides covering selected ranges of the complete amino acid sequence of hcTnI was prepared and used for epitope mapping. Such analysis allowed to select an appropriate pair of antibodies that can form a sandwich type of immune complexes and was consequently used for an immunoassay.

허혈성 뇌졸중 환자의 운동기능회복에 따른 중요 혈액인자들의 변화 (The Change of the Important Blood Factors According to the Recovery of Motor Function with Ischemic Stroke Patients)

  • Background: This study had been carried out with 18 ischemic stroke patients as its object for about eight months from October, 2006 to May, 2007 in order to observe the recovery of motor function and the change of important blood factors according to the different quantitative exercises. Methods: Subjects were assigned randomly either experimental group (n=19) or the control group (n=19), when the study began the halfway on this study dropout 20 patients, and final subjects remained experimental group's 9 patients and control group's 9 patients. Both groups received thermotherapy and functional electrical stimulation (FES), also taken different quantitative exercise therapy (experimental group 180 minutes, control group 80 minutes). Subjects were assessed for upper and lower extremities motor function Fugl-Meyer Scale; FMS), blood test (white blood count; WBC, low density lipoprotein -cholesterol; LDL-C, high density lipoprotein-cholesterol; HDL-C, Troponin) during pretest, after 2 months, after 3 months. Results: The results of this study were as follows; 1. FMS has no statistically significant difference with intergroup(p>.05). But there was a statistically significant difference with each groups (p<.05). 2. WBC has no statistically significant difference with intergroup (p>.05). But there was a statistically significant difference in control group (p<.05), without experimental group (p>.05). 3. LDL-C has no statistically significant difference with intergroup (p>.05). But there was a statistically significant difference in control group (p<.05), without experimental group (p>.05). 4. HDL-C has no statistically significant difference with intergroup (p<.05). But there was a statistically significant difference with each groups (p>.05). 5. Troponin Ⅰ has no statistically significant difference with intergroup (p>.05). Also there was no statistically significant difference with each groups (p>.05). Conclusion: These findings suggest that different quantitative exercises has no effect on FMS, LDL-C, HDL-C, WBC, Troponin Ⅰ with ischemic stroke patients. But the treatment period that there's less correlation between the recovery of motor function and the different quantitative exercise, also less correlation between the change of important blood factors and the different quantitative exercises with ischemic stroke patients.

Solution Structure of the D/E Helix Linker of Skeletal Troponin-C: As Studied by Circular Dichroism and Two-Dimensional NMR Spectroscopy

  • We have synthesized a 17-residue peptide with the amino acid sequence RQMKEDAKGKSEEELAD corresponding to residues 84-100 of chicken skeletal troponin C. This stretch of the protein sequence is in the middle one-third of the 32-residue 9-turn α-helix that connects the two globular domains of the dumbell-shaped molecule and includes the D/E linker helix. We describe here the solution conformation of the helix linker as studied by circular dichroism (CD) and two-dimensional nuclear magnetic resonance (2-D NMR) spectroscopy. The NOE connectivities together with the vicinal $^3J_{N{\alpha}}$ coupling constants suggest that the peptide exists in a fast conformational equilibrium among several secondary structure: a nascent helix near the N-terminus, a helix, and a substational population of extended and random coil forms. In addition, two interresidue α-α NOEs are observed suggesting a bent structure with a bend that includes the single glycine in position 92. These results are consistent with the ideas that in neutral solution the D/E linker region of the central helix in troponin C can adopt a helical conformation and the central helix may have a segmental flexibility around Gly 92.

  • The calcium-induced structural changes in the skeletal muscle regulatory protein troponin C (NTnC) involve a transition from a ‘closed’to an ‘open’structure with the concomitant exposure of a large hydrophobic interaction site for target proteins. Structural studies have served to define this conformational change and elucidate the mechanism of the linkage between calcium binding and the induced structural changes. There are now several structures of NTnC available from both NMR and X-ray crystallography. Comparison of the calcium bound structures reveals differences in the level of opening. We have considered the concept of a flexible open state of NTnC as a possible explanation for this apparent discrepancy. We also present simulations of the closed-to-open transition which are in agreement with the flexibility concept and with experimental energetics data.

Balloon valvuloplasty for severe subaortic stenosis in a Pomeranian dog

  • A nine-month-old Pomeranian dog with exercise intolerance and syncope was presented. The dog was depressed with grade 4 systolic murmur on cardiac auscultation. Based on cardiac examination, the dog was diagnosed with severe subaortic stenosis with involvement of the anterior mitral valve. ${\beta}-blocker$ administration was initiated and clinical signs were improved, but not fully resolved. Balloon valvuloplasty was performed and the dog survived for nearly one year without clinical sign and the cardiac troponin I level was normalized. This case describes successful management of severe subaortic stenosis in a small breed dog through balloon valvuloplasty.

  • 細胞의 分化에 관한 硏究의 一環으로 鷄胚의 筋細胞를 培養하면서 미오신, 악틴, 트로포미오신, 트로포닌 等 筋特異 蛋白質의 合成과 培養細胞에서 培養液內로 放出되는 蛋白質을 形態分化와 竝行하여 分析하였다. 筋特異 蛋白質중 미오신과 악틴은 細胞融合前에 활발히 合成되며 融合後에는 相對的으로 떨어지고, 트로포닌은 融合直後부터 활발히 合成되기 시작하며, 트로포미오신은 分化의 全期間에 걸쳐 合成율이 거의 一定하였다. 培養筋細胞는 細胞融合이 일어나기 前에 分子量 18,000 달톤과 20,000달톤 그리고 그 이상의 몇 가지 蛋白質을 培養液內에 放出시틴다는 것이 거의 確實하다. 이들 蛋白質은 培養筋細胞의 膜蛋白質일 것으로 推定되며 筋細胞의 同時 融合을 誘導하는 機能을 가진 것으로 생각된다.

  • 급성 심근경색 진단용 DNA 컴퓨팅 시스템 모듈로서, 트로포닌 I (troponin I, Tnl). 트로포닌 T (troponin T, TnT). 미오글로빈 (myoglobin), C-반응 단백질 (C-reactive protein, CRP) 과 각각 결합할 수 있는 네 가지 종류의 앱타머틀 선정하고, 이의 개발을 시도하여, 그 중 첫 번째로 C-반응 단백질-결합 앱타머를 SELEX 기법을 이용하여 선별해내었다. 또한, 선별된 앱타머 염기서열에 기초하여 각각 10-mer 길이의 FDNA 와 QDNA 를 제작하고, 표적 단백질 (CRP) 과 혼합시켜 형광발현 변화의 추이를 살펴보았다. 앱타머 및 FDNA. QDNA 가 결합할 경우에는 형광감쇄효과가 발생하므로, 형광감쇄효과가 일어나지 않은 경우에 비하여 현저하게 형광측정값이 저조하게 나타나는 현상을 확인할 수 있었다. 향후 연구로, 나머지 세 가지 종류의 앱타머를 SELEX기법을 이용하여 선별해내고. 기확보된 C-반응 단백질-결합 앱타머 모듈과 함께 논리회로를 구성하는 DNA 컴퓨팅 칩을 제작할 예정이다.
