Malaria Prevalence in a Low Transmission Area, Jazan District of Southwestern Saudi Arabia

  • Detailed description of malaria in low transmission areas is crucial for elimination. The current study aimed to provide a comprehensive description for malaria transmission in Jazan, a low transmission district, southwestern Saudi Arabia. Patients at a tertiary care hospital were recruited in our study between August 2016 and September 2018. Malaria diagnosis was performed through a species-specific nested polymerase chain reaction (nested PCR), microscopy and Paramax-$3^{TM}$ rapid detection test (RDT). Malaria was detected in 30 patients by the PCR, with point prevalence of 10.9%. Of these malaria infections, 80% was imported, 26.6% was asymptomatic and 23.3% was sub-microscopic. Malaria was reported throughout the year, with February/March and September/October peaks. Infection was significantly more in males than in females (P=0.01). Likewise, infections were detected more in febrile than in non-febrile patients (P=0.01). Adult aged 15-24 years, fever and travel were identified as high-risk factors. Malaria was primarily attributed to Plasmodium falciparum mono-infections, followed by P. vivax mono-infections and lastly to falciparum/vivax mixed infections accounting 76.6%, 16.6%, and 6.6% of PCR-confirmed malaria cases, respectively. The nested PCR was superior to the smear microscopy (sensitivity 76.6%; specificity 100%) and the RDT (sensitivity 83.3%, specificity 94.2%). The overall percent agreement between microscopy and the RDT was 92.7% (kappa=0.63). High proportion of imported malaria including sub-microscopic and sub-patent cases were described. We suggest that incorporation of molecular tool into the conventional malaria diagnosis is beneficial in Jazan district.

Molecular Surveillance of Pfkelch13 and Pfmdr1 Mutations in Plasmodium falciparum Isolates from Southern Thailand

  • Artemisinin-based combination therapy (ACT) resistance is widespread throughout the Greater Mekong Subregion. This raises concern over the antimalarial treatment in Thailand since it shares borders with Cambodia, Laos, and Myanmar where high ACT failure rates were reported. It is crucial to have information about the spread of ACT resistance for efficient planning and treatment. This study was to identify the molecular markers for antimalarial drug resistance: Pfkelch13 and Pfmdr1 mutations from 5 provinces of southern Thailand, from 2012 to 2017, of which 2 provinces on the Thai- Myanmar border (Chumphon and Ranong), one on Thai-Malaysia border (Yala) and 2 from non-border provinces (Phang Nga and Surat Thani). The results showed that C580Y mutation of Pfkelch13 was found mainly in the province on the Thai-Myanmar border. No mutations in the PfKelch13 gene were found in Surat Thani and Yala. The Pfmdr1 gene isolated from the Thai-Malaysia border was a different pattern from those found in other areas (100% N86Y) whereas wild type strain was present in Phang Nga. Our study indicated that the molecular markers of artemisinin resistance were spread in the provinces bordering along the Thai-Myanmar, and the pattern of Pfmdr1 mutations from the areas along the international border of Thailand differed from those of the non-border provinces. The information of the molecular markers from this study highlighted the recent spread of artemisinin resistant parasites from the endemic area, and the data will be useful for optimizing antimalarial treatment based on regional differences.

  • This study aimed to investigate the tick species and give background for tick-borne investigations in Korea. Ticks were collected from the area within 2 km radius of the 4 domestic animal farms, where they were located in mountainous areas and raising animals on pasture, and from animal bodies in 2014 and 2015. In total, 7,973 nymphal and adult ticks were collected from the farms - 7,758 Haemaphysalis longicornis, 198 Haemaphysalis flava, and 17 Ixodes nipponensis, and 1,763 were collected from animals - 729 H. longicornis from cattle; 569 H. longicornis from goats; and 297 H. longicornis, 118 H. flava, 1 I. nipponensis, and 49 Amblyomma testudinarium from wild boars. As more species of ticks were collected from wild boars than domesticated animals and their habitats, various animal hosts should be considered while investigating tick species.

Vitamin D Status of Breastfed Filipino Infants Aged Less Than 6 Months in an Urban Community

  • Purpose: This study aimed to determine the serum 25-hydroxy-vitamin D (25(OH)D) status of breastfed infants less than six months old and their mothers, and factors affecting the status. Methods: This cross-sectional study was done on breastfed, term, Filipino infants less than six months old who were seen at local health centers and clinics in an urban area. The serum 25(OH)D levels of these infants and their mothers were determined, and their demographic data, nutritional status, sun exposure behavior, and maternal vitamin D intake were analyzed for correlation using regression models. Results: Among the 131 infants, 101 (77%) had vitamin D deficiency (VDD), which was defined as having 25(OH)D levels <37.5 nmol/L, and 13 (10%) had vitamin D insufficiency (VDI), with levels >37.5-50 nmol/L. Conversely, maternal VDD with levels <50 nmol/L was seen in 31 (24%) mothers and maternal VDI with levels 50-75 nmol/L, in 63 (48%) mothers. Infant age and maternal 25(OH)D status were independent predictors of infant VDD. Infants less than three months old were found to have a six-time increased risk of infant VDD (p=0.004). Infants who had mothers with VDD had a six-time increased risk, whereas those with maternal VDI had a four-time increased risk of infant VDD (p=0.049 and p=0.020, respectively). Conclusion: Both infant and maternal VDD and VDI were seen to be highly prevalent in this tropical, urban community. Young infants and maternal VDD/VDI independently increased the risk of infant VDD, whereas lack of sun exposure of the mothers increased the risk for maternal VDI.

  • Global warming has affected the distribution of organisms for decades and has displayed rapid ascent recently. Research into the effects on tropical organisms are vital. Padina boryana is a resourceful marine microalgae in the Maldives Sea in the Laccadive region. A 70% ethanol extraction (PBE) of this seaweed was used to investigate its antioxidant potential. Both in vitro and in vivo models were implemented. PBE exhibited protective potential against H2O2 induced apoptosis. ROS levels were suppressed due to PBE. PBE expressed a cytoprotective nature. In vivo experiments involving the zebra fish model conformed its validity. The antioxidant efficacy of PBE was dose dependent. Study outcomes suggest PBE has potential as a novel and valuable marine resource to aid the functional food and cosmeceutical industries.

Microencapsulated basil oil (Ocimum basilicum Linn.) enhances growth performance, intestinal morphology, and antioxidant capacity of broiler chickens in the tropics

  • Objective: Microencapsulation is a technique to improve stability, bioavailability, and controlled release of active ingredients at a target site. This experiment aimed to investigate the effects of microencapsulated basil oil (MBO) on growth performance, apparent ileal digestibility (AID), jejunal histomorphology, bacterial population as well as antioxidant capacity of broiler chickens in a tropical climate. Methods: A total of 288 one-day-old female broilers (Ross 308) were randomly allocated into 4 groups (6 replicates of 12 birds), based on a completely randomized design. Dietary treatments were as follows: i) basal diet (NC), ii) basal diet with avilamycin at 10 ppm (PC), iii) basal diet with free basil oil (FBO) at 500 ppm, and iv) basal diet with MBO at 500 ppm, respectively. Results: Dietary supplementation of MBO improved average daily gain, and feed conversion ratio of broilers throughout the 42-d trial period (p<0.05), whereas MBO did not affect average daily feed intake compared with NC group. The broilers fed MBO diet exhibited a greater AID of crude protein and gross energy compared with those in other groups (p<0.05). Lactobacillus spp. and Escherichia coli populations were not affected by feeding dietary treatments. Both FBO and MBO had positive effects on jejunal villus height (VH), villus height to crypt depth ratio (VH:CD) and villus surface area of broilers compared to NC and PC groups (p<0.05). Superoxide dismutase level in the duodenal mucosa of MBO group was significantly increased (p<0.01), whereas malondialdehyde level was significantly decreased (p<0.01). Conclusion: Microencapsulation could be considered as a promising driver of the basil oil efficiency, consequently MBO at 500 ppm could be potentially used as a feed additive for improvement of intestinal integrity and nutrient utilization, leading to better performance of broiler chickens.

Status of Helminthic Infections in Residents around River Basins in the Republic of Korea for 10 Years (2011-2020)

  • The positive rate of Clonorchis sinensis is the highest among intestinal parasites in the Republic of Korea (Korea). More than 1.2 million people were at risk of C. sinensis infection in Korea in 2012. An intensive control program is being implemented for residents of the 5 major river basins to reduce helminthic infections, including C. sinensis infection. This study evaluated the continuous intensive control program for parasitic diseases including clonorchiasis in areas near the 5 major river basins in Korea over the past 10 years (2011-2020). A total of 335,020 fecal samples (one sample per resident) prepared by the modified sedimentation technic were microscopically examined. Those who expelled helminth eggs were treated with anthelmintics through local health centers and re-examined 3 months later. The overall positive rate of helminths egg was 7.1%. The annual positive rates were dramatically decreased from 14.4% (2011) to 5.9% (2020). The egg positive rate was highest in C. sinensis (5.3%), followed by heterophyid flukes (1.5%) and Trichuris trichiura (0.2%). The prevalence of C. sinensis was significantly higher in males (7.6%) than in females (3.7%), and the highest in the 50-59 years (7.0%) age group. Our results are beneficial to establish prevention and control policies against helminthiases including clonorchiasis in endemic areas in this country.

Positive rates for Enterobius vermicularis eggs among preschool children in Yeosu-si, Jeollanam-do, Korea (2017-2021)

  • This study aimed to evaluate the positive rates for Enterobius vermicularis eggs among preschool children in Yeosu-si, Jeollanam-do, the Republic of Korea (Korea) over a period of 5 years (2017-2021). Perianal swab samples, obtained using cellotape, from 10,392 preschool children in 26 districts were examined microscopically for E. vermicularis eggs. The test results were notified through the local health center, and the families of children who tested positive were advised to provide them anthelmintics treatment. The annual positive rates were 5.0%, 5.2%, 4.4%, 2.2%, and 1.0% in 2017, 2018, 2019, 2020, and 2021, respectively. The overall positive rate was higher in boys than in girls (P< 0.05), and children aged 5-7 years were at a higher risk of being infected than those aged 0-4 years (P< 0.05). Although the rates of infection by E. vermicularis in the survey area, Yeosu-si, were still in the 1% range , the results of this study suggest that they can be significantly reduced through continuous intervention centered around the test-treatment strategy.

한국산 "붉평치"의 Lampris megalopsis(붉평치과, 이악어목)로의 학명 변경 (Change of the Scientific Name for the Opah in Korea to Lampris megalopsis (Lampridae, Lampriformes))

  • 우리나라에서 붉평치는 2014년 11월 삼척에서 정치망으로 채집되어 Jeong et al.(2015)에 의해 Lampris guttatus로 처음 보고되었다. 이후, Underkoffler et al. (2018)에 의해 1종(L. lauta)의 학명 부활 및 3종(L. australensis, L. incognitus, L. megalopsis)이 새롭게 보고되어, 국내 붉평치의 학명 재검토가 필요한 실정이다. 국내 미기록종 표본은 현재 서대문자연사박물관에 박제 표본으로 보존되어 있으며, 박제 표본은 다음의 형태적 특징을 가진다: 안경이 가랑이체장의 5%보다 크다; 머리, 흉부, 새개부를 포함한 몸 전체에 둥근 반점이 산재한다; 둥근 반점의 크기는 불규칙적이다; 지느러미가 모두 붉다. 또한, L. guttatus는 북대서양에 분포하는 반면 L. megalopsis는 전 세계의 온대 및 열대 해역에 분포한다. 따라서, 본 연구 결과는 형태 및 분포 특징에 의거 한국산 붉평치의 학명이 L. guttatus에서 L. megalopsis로 변경되어야 함을 시사한다.

Prospecting endophytic colonization in Waltheria indica for biosynthesis of silver nanoparticles and its antimicrobial activity

  • Endophytes ascertain a symbiotic relationship with plants as promoters of growth, defense mechanism etc. This study is a first report to screen the endophytic population in Waltheria indica, a tropical medicinal plant. 5 bacterial and 3 fungal strains in leaves, 3 bacterial and 1 yeast species in stems were differentiated morphologically and identified by biochemical and molecular methods. The phylogenetic tree of the isolated endophytes was constructed using MEGA X. Silver nanoparticles were biosynthesized from a rare endophytic bacterium Cupriavidus metallidurans isolated from the leaf of W. indica. The formation of silver nanoparticles was confirmed by UV-Visible spectrophotometer that evidenced a strong absorption band at 408.5 nm of UV-Visible range with crystalline nature and average particle size of 16.4 nm by Particle size analyzer. The Fourier Transform Infra-Red spectrum displayed the presence of various functional groups that stabilized the nanoparticles. X-ray diffraction peaks were conferred to face centered cubic structure. Transmission Electron Microscope and Scanning Electron Microscope revealed the spherical-shaped, polycrystalline nature with the presence of elemental silver analyzed by Energy Dispersive of X-Ray spectrum. Selected area electron diffraction also confirmed the orientation of AgNPs at 111, 200, 220, 311 planes similar to X-ray diffraction analysis. The synthesized nanoparticles are evaluated for antimicrobial activity against 7 bacterial and 3 fungal pathogens. A good zone of inhibition was observed against pathogenic bacteria than fungal pathogens. Thus the study could hold a key aspect in drug discovery research and other pharmacological conducts of human clinical conditions.
