Identification of differentially expressed cDNAs in Acanthamoeba culbertsoni after mouse brain passage

  • Free-living amoebae of the genus Acanthamoeba are causative agents of granulomatous amebic encephalitis and amebic keratitis. Because the virulence of Acanthamoeba culbertsoni cultured in the laboratory is restored by consecutive brain passages, we examined the genes induced in mouse brain-passaged A. culbertsoni by differential display reverse transcriptase polymerase chain reaction (DDRT-PCR). Enhanced A. culbertsoni virulence was observed during the second mouse brain passage, i.e., infected mouse mortality increased from $5\%\;to\;70\%.$ Ten cDNAs induced during mouse brain passage were identified by DDRT-PCR and this was confirmed by northern blot analysis. BlastX searches of these cDNAs indicated the upregulations of genes encoding predictive NADH-dehydrogenase, proteasomal ATPase, and GDP-mannose pyrophosphorylase B, which have previously been reported to be associated with A. culbertsoni virulence factors.

  • Tropical forests constitute almost half of the global forest cover, account for 35% of the global net primary productivity and thereby have potential to contribute substantially to sequester atmospheric $CO_2$ and offset climate change impact. However, deforestation and degradation lead by unsustainable management of tropical forests contribute to the unprecedented species losses and limit ecosystem services including carbon sequestration. Sustainable forest management (SFM) in the tropics may tackle and rectify such deleterious impacts of anthropogenic disturbances and climatic changes. However, the existing dilemma on the definition of SFM and lack of understanding of how tropical forest sustainability can be achieved lead to increasing debate on whether climate change mitigation initiatives would be successful. We reviewed the available literature with a view to clarify the concept of sustainability and provide with a framework towards the sustainability of tropical forests for enhanced carbon stock and climate change mitigation. We argue that along with securing forest tenure and thereby reducing deforestation, application of reduced impact logging (RIL) and appropriate silvicultural system can enhance tropical forest carbon stock and help mitigate climate change.

Effect of chitosan coating combined with hypotaurine on the quality of shrimp (Litopenaeus vannamei) during storage

  • This study aimed to investigate the effect of different coating materials on the quality of shrimp (Litopenaeus vannamei) during chilled storage for 10 days. Fresh shrimp were randomly divided into five groups: the control group, the hypotaurine treatment group (2%), the chitosan group (1%), the hypotaurine + chitosan group (2% hypotaurine solution with 1% of chitosan), and the sodium metabisulfite treatment group (1.25%). Compared with other treatments, the lower accumulation of total visible counts (TVC, 5.25 Log10 CFU/g), total volatile basic nitrogen (TVB-N, 22.5 mg/100 g) and thiobarbituric acid values (TBA, 0.58 mg MDA/kg) suggested that coating of chitosan-hypotaurine could retard the microbial activity, protein degradation and lipid oxidation of shrimp. Meanwhile, results demonstrated that the chitosan coating combined with hypotaurine showed an excellent performance in inhibiting quality deterioration (pH 7.5, ∆E 7.0, hardness 393 g, and elasticity 0.69). Furthermore, the melanosis degree of shrimp was alleviated, and the sensory parameters, including appearance, odor and texture, were maintained to the acceptable level by chitosan based hypotaurine treatment during the chilled storage.

Morphological studies of fly puparia (Diptera: Calliphoridae) in the soil from a Joseon Dynasty grave in Korea

  • Archaeoentomological investigations were conducted on soil contents from a grave belonging to the Joseon Dynasty as part of the Urban Environment Maintenance Project (UEMP) in Cheongjin 12-16 dong (districts), Jongno-gu, Seoul, Korea, from December 01, 2008 to February 19, 2011. A total of 28 insect puparia with hard shells of the common green bottle fly Lucilia sericata were identified in the soil. Evidence suggested that the corpse was placed outside for some days instead of being buried immediately after death. This is the first report of fly puparia in soil samples from a tomb of the Joseon Dynasty during 16-17 AD in Korea. Our findings may help determine the timeframe of burial and offer archaeological insights into the funerary customs of the period.

Molecular and Biochemical Properties of a Cysteine Protease of Acanthamoeba castellanii

  • Acanthamoeba spp. are free-living protozoa that are opportunistic pathogens for humans. Cysteine proteases of Acanthamoeba have been partially characterized, but their biochemical and functional properties are not clearly understood yet. In this study, we isolated a gene encoding cysteine protease of A. castellanii (AcCP) and its biochemical and functional properties were analyzed. Sequence analysis of AcCP suggests that this enzyme is a typical cathepsin L family cysteine protease, which shares similar structural characteristics with other cathepsin L-like enzymes. The recombinant AcCP showed enzymatic activity in acidic conditions with an optimum at pH 4.0. The recombinant enzyme effectively hydrolyzed human proteins including hemoglobin, albumin, immunoglobuins A and G, and fibronectin at acidic pH. AcCP mainly localized in lysosomal compartment and its expression was observed in both trophozoites and cysts. AcCP was also identified in cultured medium of A. castellanii. Considering to lysosomal localization, secretion or release by trophozoites and continuous expression in trophozoites and cysts, the enzyme could be a multifunctional enzyme that plays important biological functions for nutrition, development and pathogenicity of A. castellanii. These results also imply that AcCP can be a promising target for development of chemotherapeutic drug for Acanthamoeba infections.

High Prevalence of Enterobius vermicularis Infection among Schoolchildren in Three Townships around Yangon, Myanmar

  • In order to determine the status of Enterobius vermicularis infection among schoolchildren in suburban areas of Myanmar, 761 primary schoolchildren in 3 different townships around Yangon City were subjected to a survey using cello-tape anal swabs. The subjected schoolchildren were 383 boys and 378 girls who were 5-7 years of age. Only 1 anal swab was obtained from each child. The overall egg positive rate of E. vermicularis was 47.2% (359 positives), and sex difference was not remarkable (48.6% in boys and 45.8% in girls). However, the positive rate was the highest in South Dagon (54.6%) followed by Hlaing Thayar (43.8%) and North Dagon (34.8%). This difference was highly correlated with the living standards of the people in each township. Nucleotide sequence of the 5S rDNA from the eggs on the cello-tape (2 children) revealed 99.7% identity with that of E. vermicularis reported in GenBank. The results indicated that E. vermicularis infection is highly prevalent among primary schoolchildren around Yangon, Myanmar.

Evaluation and Intercomparisons of the Estimated TOVS Precipitable Waters for the Tropical Plume (Tropical Plume 에 대한 TOVS 추정 가강수량의 평가와 상호비교)

  • Precipitable Water(PW) are retrieved over the tropical and subtropical Pacific Ocean from TOVS infrared and microwave channel brightness temperature and OLR observations by means of stepwise linear regression. The retrieved TOVS PW fields generated by PW$_{sfc}$(71.1 % of the variance and 0.62 g cm$^{-2}$ standard error over the surface) and PW$_{700500}$(71.7 % and 0.17 g cm$^{-2}$ over the 700 - 500 hPa layer) revealed more evolving synoptic signals over the tropical and subtropical Pacific Ocean. The PW$_{sfc}$ dose not show significantly the TP feature because of the representation of the lower PW for high-level clouds not associated with deep convection. There exists some elusion to trace the TP on the PW$_{sfc}$ field if any supplementary information does not provide. But ECMWF analysis has a general tendency of drying the subtropics and moistening the ITCZ (InterTropical Convergence Zone) and SPCZ(South Pacific Convergence Zone). However, although ECMWF analysis is fairly successful in capturing mean patterms, it is unsuccessful in following active synoptic signal like a tropical plume. Similarly, SMMR-PW does not represent the TP well which consists of the highand middle-level clouds, but PW$_{sfc}$ shows underestimated moistness of TP and does not depict significant signal of TP. In the PW field derived from microwave observations, the TP can not be recognized well. Furthermore, the signature of PW$_{sfc}$ was different from OLR for the TP, which implies the presence of high- and middle-layer thin clouds, but in a closer agreement for deep and active convection areas which contain thick middle- and lower-layer clouds; though OLR represented the cloudiness in the tropics well. In synoptically active regions, it differed from OLR analysis, primarily bacause of actual differences in water vapor and cloud features. The signature of PW$_{sfc}$ was different from OLR for the TP.

Seroprevalence of Toxoplasma gondii among School Children in Pyin Oo Lwin and Naung Cho, Upper Myanmar

  • Toxoplasma gondii is an apicomplexan parasite that can cause toxoplasmosis in a wide range of warm-blooded animals including humans. In this study, we analyzed seroprevalence of T. gondii among 467 school children living in the rural areas of Pyin Oo Lwin and Naung Cho, Myanmar. The overall seroprevalence of T. gondii among school children was 23.5%; 22.5% of children were positive for T. gondii IgG, 0.4% of children were positive for T. gondii IgM, and 0.6% of children were positive for both T. gondii IgG and IgM. Geographical factors did not significantly affect the seroprevalence frequency between Pyin Oo Lwin and Naung Cho, Myanmar. No significant difference was found between males (22.2%) and females (25.0%). The overall seroprevalence among school children differed by ages (10 years old [13.6%], 11-12 years old [19.8%], 13-14 years old [24.6%], and 15-16 years old [28.0%]), however, the result was not significant. Polymerase chain reaction analysis for T. gondii B1 gene for IgG-positive and IgM-positive blood samples were negative, indicating no direct evidence of active infection. These results collectively suggest that T. gondii infection among school children in Myanmar was relatively high. Integrated and improved strategies including reinforced education on toxoplasmosis should be implemented to prevent and control T. gondii infection among school children in Myanmar.

Temporal and Spatial Variability of Precipitation and Evaporation over the Tropical Ocean

  • Temporal and spatial variability of precipitation (P), evaporation (E), and moisture balance (P-E; precipitation minus evaporation) has been investigated over the tropical ocean during the period from January 1998 to July 2001. Our data were analyzed by the EOF method using the satellite P and E observations made by the Tropical Rainfall Measuring Mission (TRMM) Precipitation Radar (PR) and the Special Sensor Microwave/Imager (SSM/I). This analysis has been performed for two three-year periods as follow; The first period which includes the El Ni${\tilde{n}}$o in early 1998 ranges from January 1998 to December 2000, and the second period which includes the La Ni${\tilde{n}}$o events in the early 1999 and 2000 (without El Ni${\tilde{n}}$o) ranges from August 1998 to July 2001. The areas of maxima and high variability in the precipitation and in the P-E were displaced from the tropical western Pacific and the ITCZ during the La Ni${\tilde{n}}$o to the tropical middle Pacific during the El Ni${\tilde{n}}$o, consistent with those in previous P studies. Their variations near the Korean Peninsula seem to exhibit a weakly positive correlation with that in the tropical Pacific during the El Ni${\tilde{n}}$o. The evaporation, out of phase with the precipitation, was reduced in the tropical western Pacific due to humid condition in boreal summer, but intensified in the Kuroshio and Gulf currents due to windy condition in winter. The P-E variability was determined mainly by the precipitation of which the variability was more localized but higher by 2-3 times than that of evaporation. Except for the ITCZ (0-10$^{\circ}$N), evaporation was found to dominate precipitation by ${\sim}$2 mm/day over the tropical Pacific. Annual and seasonal variations of P, E, and P-E were discussed.
