Potential to mitigate ammonia emission from slurry by increasing dietary fermentable fiber through inclusion of tropical byproducts in practical diets for growing pigs

  • Objective: Research was conducted to test the effect of including fiber-rich feedstuffs in practical pig diets on nutrient digestibility, nitrogen balance and ammonia emissions from slurry. Methods: Three Vietnamese fiber sources were screened, namely cassava leaf meal (CL), cassava root residue (CR), and tofu by-product (TF). Accordingly, a control diet (Con) with 10% of dietary non-starch polysaccharides (NSP) and three test diets including one of the three fiber-rich feedstuffs to reach 15% of NSP were formulated. All formulated diets had the same level of crude protein (CP), in vitro ileal protein digestible and metabolisable energy, whereas the in vitro hindgut volatile fatty acid (VFA) production of the test diets was 12% to 20% higher than the control diet. Forty growing barrows with initial body weight at $28.6{\pm}1.93kg$ ($mean{\pm}standard$ deviation) were allocated to the four treatments. When pigs reached about 50 kg of body weight, four pigs from each treatment were used for a nitrogen balance trial and ammonia emission assessment, the remaining six pigs continued the second period of the feeding trial. Results: The TF treatment increased fecal VFA by 33% as compared with the control treatment (p = 0.07), suggesting stimulation of the hindgut fermentation. However, urinary N was not significantly reduced or shifted to fecal N, nor was slurry pH decreased. Accordingly, ammonia emissions were not mitigated. CR and CL treatments failed to enhance in vivo hindgut fermentation, as assessed by fecal VFA and purine bases. On the contrary, the reduction of CP digestibility in the CL treatment enhanced ammonia emissions from slurry. Conclusion: Dietary inclusion of cassava and tofu byproducts through an increase of dietary NSP from 10% to 15% might stimulate fecal VFA excretion but this does not guarantee a reduction in ammonia emissions from slurry, while its interaction with protein digestibility even might enhance enhanced ammonia emission.

Bleomycin Inhibits Proliferation via Schlafen-Mediated Cell Cycle Arrest in Mouse Alveolar Epithelial Cells

  • Background: Idiopathic pulmonary fibrosis involves irreversible alveolar destruction. Although alveolar epithelial type II cells are key functional participants within the lung parenchyma, how epithelial cells are affected upon bleomycin (BLM) exposure remains unknown. In this study, we determined whether BLM could induce cell cycle arrest via regulation of Schlafen (SLFN) family genes, a group of cell cycle regulators known to mediate growth-inhibitory responses and apoptosis in alveolar epithelial type II cells. Methods: Mouse AE II cell line MLE-12 were exposed to $1-10{\mu}g/mL$ BLM and $0.01-100{\mu}M$ baicalein (Bai), a G1/G2 cell cycle inhibitor, for 24 hours. Cell viability and levels of pro-inflammatory cytokines were analyzed by MTT and enzyme-linked immunosorbent assay, respectively. Apoptosis-related gene expression was evaluated by quantitative real-time reverse transcription-polymerase chain reaction (qRT-PCR). Cellular morphology was determined after DAPI and Hoechst 33258 staining. To verify cell cycle arrest, propidium iodide (PI) staining was performed for MLE-12 after exposure to BLM. Results: BLM decreased the proliferation of MLE-12 cells. However, it significantly increased expression levels of interleukin 6, tumor necrosis factor ${\alpha}$, and transforming growth factor ${\beta}1$. Based on Hoechst 33258 staining, BLM induced condensation of nuclear and fragmentation. Based on DAPI and PI staining, BLM significantly increased the size of nuclei and induced G2/M phase cell cycle arrest. Results of qRT-PCR analysis revealed that BLM increased mRNA levels of BAX but decreased those of Bcl2. In addition, BLM/Bai increased mRNA levels of p53, p21, SLFN1, 2, 4 of Schlafen family. Conclusion: BLM exposure affects pulmonary epithelial type II cells, resulting in decreased proliferation possibly through apoptotic and cell cycle arrest associated signaling.

Risk of Recrudescence of Lymphatic Filariasis after Post-MDA Surveillance in Brugia malayi Endemic Belitung District, Indonesia

  • Belitung district in Bangka-Belitung Province, Indonesia with a population of 0.27 million is endemic for Brugia malayi and 5 rounds of mass drug administration (MDA) were completed by 2010. Based on the results of 3 transmission assessment surveys (TAS), the district is declared as achieving elimination of lymphatic filariasis (LF) in 2017. The findings of an independent survey conducted by the National Institute of Health Research and Development (NIHRD) in the same year showed microfilaria (Mf) prevalence of 1.3% in this district. In 2019, NIHRD conducted microfilaria survey in 2 villages in Belitung district. Screening of 311 and 360 individuals in Lasar and Suak Gual villages showed Mf prevalence of 5.1% and 2.2% with mean Mf density of 120 and 354 mf/ml in the respective villages. Mf prevalence was significantly higher among farmers and fishermen compared to others and the gender specific difference was not significant. The results of a questionnaire based interview showed that 62.4% of the respondents reported to have participated in MDA in Lasar while it was 57.7% in Suak Gual village. About 42% of the Mf positive cases did not participate in MDA. Environmental surveys identified many swampy areas supporting the breeding of Mansonia vector species. Persistence of infection is evident and in the event of successful TAS3 it is necessary to monitor the situation and plan for focal MDA. Appropriate surveillance strategies including xenomonitoring in post-MDA situations need to be developed to prevent resurgence of infection. Possible role of animal reservoirs is discussed.

Morphological and Molecular Identification of Spirometra Tapeworms (Cestoda: Diphyllobothriidae) from Carnivorous Mammals in the Serengeti and Selous Ecosystems of Tanzania

  • Spirometra tapeworms (Cestoda: Diphyllobothriidae) collected from carnivorous mammals in Tanzania were identified by the DNA sequence analysis of the mitochondrial cytochrome c oxidase subunit 1 (cox1) and internal transcribed spacer 1 (ITS1), and by morphological characteristics. A total of 15 adult worms were collected from stool samples and carcasses of Panthera leo, Panthera pardus, and Crocuta crocuta in the Serengeti and Selous ecosystems of Tanzania. Three Spirometra species: S. theileri, S. ranarum and S. erinaceieuropaei were identified based on morphological features. Partial cox1 sequences (400 bp) of 10 specimens were revealed. Eight specimens showed 99.5% similarity with Spirometra theileri (MK955901), 1 specimen showed 99.5% similarity with the Korean S. erinaceieuropaei and 1 specimen had 99.5% similarity with Myanmar S. ranarum. Sequence homology estimates for the ITS1 region of S. theileri were 89.8% with S. erinaceieuropaei, 82.5% with S. decipiens, and 78.3% with S. ranarum; and 94.4% homology was observed between S. decipiens and S. ranarum. Phylogenetic analyses were performed with 4 species of Spirometra and 2 species of Dibothriocephalus (=Diphyllobothrium). By both ML and BI methods, cox1 and ITS1 gave well supported, congruent trees topology of S. erinaceieuropaei and S. theileri with S. decipiens and S. ranarum forming a clade. The Dibothriocephalus species were sisters of each other and collectively forming successive outgroups. Our findings confirmed that 3 Spirometra species (S. theileri, S. ranarum, and S. erinaceieuropaei) are distributed in the Serengeti and Selous ecosystems of Tanzania.

Growth performance of planted population of Pinus roxburghii in central Nepal

  • Background: Climate change has altered the various ecosystem processes including forest ecosystem in Himalayan region. Although the high mountain natural forests including treelines in the Himalayan region are mainly reported to be temperature sensitive, the temperature-related water stress in an important growth-limiting factor for middle elevation mountains. And there are very few evidences on growth performance of planted forest in changing climate in the Himalayan region. A dendrochronological study was carried out to verify and record the impact of warming temperature tree growth by using the tree cores of Pinus roxburghii from Batase village of Dhulikhel in Central Nepal with sub-tropical climatic zone. For this total, 29 tree cores from 25 trees of P. roxburghii were measured and analyzed. Result: A 44-year long tree ring width chronology was constructed from the cores. The result showed that the radial growth of P. roxburghii was positively correlated with pre-monsoon (April) rainfall, although the correlation was not significant and negatively correlated with summer rainfall. The strongest negative correlation was found between radial growth and rainfall of June followed by the rainfall of January. Also, the radial growth showed significant positive correlation with that previous year August mean temperature and maximum temperature, and significant negative correlation between radial growth and maximum temperature (Tmax) of May and of spring season (March-May), indicating moisture as the key factor for radial growth. Despite the overall positive trend in the basal area increment (BAI), we have found the abrupt decline between 1995 and 2005 AD. Conclusion: The results indicated that chir pine planted population was moisture sensitive, and the negative impact of higher temperature during early growth season (March-May) was clearly seen on the radial growth. We emphasize that the forest would experience further moisture stress if the trend of warming temperatures continues. The unusual decreasing BAI trend might be associated with forest management processes including resin collection and other disturbances. Our results showed that the planted pine forest stand is sub-healthy due to major human intervention at times. Further exploration of growth climate response from different climatic zones and management regimes is important to improve our understanding on the growth performance of mid-hill pine forests in Nepal.

Discrimination and Authentication of Eclipta prostrata and E. alba Based on the Complete Chloroplast Genomes

  • Eclipta prostrata and E. alba are annual herbal medicinal plants and have been used as Chinese medicinal tonics. Both species are widely distributed in tropical and subtropical regions as well as in Korea. Both species have similar morphological features but E. alba has smoother leaf blade margins compared with E. prostrata. Although both species are utilized as oriental medicines, E. prostrata is more widely used than E. alba. Morphological semblances have confounded identification of either species. Here, we report the complete chloroplast genomes of both species to provide an authentication system between the two species and understand their diversity. Both chloroplast genomes were 151,733-151,757 bp long and composed of a large single copy (83,285-83,300 bp), a small single copy (18,283-18,346 bp), and a pair of inverted repeats (25,075-25,063 bp). Gene annotation revealed 80 protein coding genes, 30 tRNA genes and four rRNA genes. A phylogenetic analysis revealed that the genus Eclipta is grouped with Heliantheae tribe species in the Asteraceae family. A comparative analysis verified 29 InDels and 58 SNPs between chloroplast genomes of E. prostrata and E. alba. The low chloroplast genome sequence diversity indicates that both species are really close to each other and are not completely diverged yet. We developed six DNA markers that distinguish E. prostrata and E. alba based on the polymorphisms of chloroplast genomes between E. prostrata and E. alba. The chloroplast genome sequences and the molecular markers generated in this study will be useful for further research of Eclipta species and accurate classification of medicinal herbs.

Prevalence of Toxoplasma gondii Antibodies in Stray Dogs from Various Locations in West and East Malaysia

  • Toxoplasmosis is caused by an obligate intracellular protozoan parasite; Toxoplasma gondii, which is one of the most important zoonotic parasite worldwide. In dogs, the sexual reproductive cycle of T. gondii is lacking, and the animals are not widely consumed as food, but they are vital in the mechanical transmission of the parasite. However, there is no present data on the exposure of stray dogs to T. gondii in Malaysia. The objective of this serological survey was to determine the prevalence of T. gondii antibodies (IgG) and associated factors in stray dogs in East and West Malaysia. Antibodies to T. gondii were determined in serum samples from 222 stray dogs from 6 different states in East and West Malaysia (Peninsular Malaysia) using an Indirect ELISA. The seroprevalence for T. gondii was 23.4% (Confidence interval: CI 17.8-29.2%). Stray dogs from Selangor and Kuala Lumpur had the highest seroprevalence (32.4%; CI 13.2-45.5%) and lowest in those from Penang and Kedah (12.5%; CI 1.3-23.5%). Gender and breed were not associated with T. gondii seropositivity. However, adult dogs were more likely to be seropositive for T. gondii (OR=2.89; CI 1.1-7.7) compared with younger dogs. These results revealed that T. gondii is prevalent in stray dogs in the studied areas in Malaysia, and indicative of the level of environmental contamination of this parasite especially in urban areas.

저온에서 벼의 발아율 및 발아속도 관련 양적형질 유전자좌(QTL) 분석 (QTL Analysis of Germination Rate and Germination Coefficient of Velocity under Low Temperature in Rice)

  • 자포니카 벼의 저온 스트레스 내성 증진을 위하여, RIL 계통을 이용하여 저온 스트레스 내성 QTL을 탐색하였다. 이를 통하여 (1) 5, 9번 염색체에서 저온발아에 관련한 '기호벼' 유래 QTL, qLTG5와 qLTG9를 확인하였으며, 7, 9번 염색체에서 저온 발아속도에 관련한 '밀양23호' 및 '기호벼' 유래 QTL, qOGCV7, qOGCV9를 확인하였다. (2) Duncan 검정결과, 그룹VII [qLTG5+qLTG9 (qOGCV9)], 그룹VIII [qLTG5+qOGCV7+qLTG9 (qOGCV9)]의 계통들이 저온 스트레스에 내성이 있는 것으로 확인 되었다. (3) 최근 발표된 RIL 집단 담수내성 계통과 비교한 결과, 저온 스트레스에도 내성이 있으면서 담수발아에도 내성이 있는 것으로 확인된 총 2개의 유망 유전자원을 선발하였다. 본 연구의 결과를 통해 저온 및 혐기 관련 QTL의 집적은 벼의 저온에서의 발아 및 초기 입모율을 높여 저온스트레스 내성 개선에 도움이 되는 것으로 판단 되었으며, 선발된 유망 계통은 향후 직파재배 품종 육성에 유용한 유전자원으로 활용되어 직파재배의 안정성 증대에 기여할 것으로 기대된다.

Echinostoma miyagawai Ishii, 1932 (Echinostomatidae) from Ducks in Aceh Province, Indonesia with Special Reference to Its Synonymy with Echinostoma robustum Yamaguti, 1935

  • Adult echinostomes having 37 collar spines collected from the intestine of Pitalah ducks in Aceh Province, Indonesia in 2018 were morphologically and molecularly determined to be Echinostoma miyagawai Ishii, 1932 (Digenea: Echinostomatidae). Among 20 ducks examined, 7 (35.0%) were found to be infected with this echinostome, and the number of flukes collected was 48 in total with average 6.9 (1-17) worms per duck. The adult flukes were 7.2 (6.1-8.5) mm in length and 1.2 (1.0-1.4) mm in width (pre-ovarian or testicular level) and characterized by having a head collar armed with 37 collar spines (dorsal spines arranged in 2 alternating rows), including 5 end group spines, and variable morphology of the testes, irregularly or deeply lobed (3-5 lobes) at times with horizontal extension. The eggs within the worm uterus were 93 (79-105) ㎛ long and 62 (56-70) ㎛ wide. These morphological features were consistent with both E. miyagawai and Echinostoma robustum, for which synonymy to each other has been raised. Sequencing of 2 mitochondrial genes, cox1 and nad1, revealed high homology with E. miyagawai (98.6-100% for cox1 and 99.0-99.8% for nad1) and also with E. robustum (99.3-99.8% for nad1) deposited in GenBank. We accepted the synonymy between the 2 species and diagnosed our flukes as E. miyagawai (syn. E. robustum) with redescription of its morphology. Further studies are required to determine the biological characteristics of E. miyagawai in Aceh Province, Indonesia, including the intermediate host and larval stage information.

피부 노화 치료로서 저분자콜라겐펩타이드의 피부 항당화와 콜라겐 합성 효과 (The Effect of Low Molecule Collagen Peptide on Skin Anti-glycation and Collagen Synthesis as a Skin Aging Therapy)

  • 콜라겐 가수 분해물(collagen hydrolysate, CH)은 피부 진피 섬유아세포를 자극하여 콜라겐과 엘라스틴 같은 세포 외 기질 합성을 촉진함으로써 피부 노화 방지에 도움을 주는 것으로 알려져 있다. 최근 피부 노화를 일으키는 다양한 요인 중 최종당화산물(advanced glycation end products, AGEs)이 주목을 받고 있음에도 불구하고, 아직까지 CH가 AGE축적에 미치는 영향은 연구된 바 없다. CH는 피부 구조단백질의 생성을 촉진하여 AGE 축적에 영향을 줄 수 있으므로, 이를 확인하기 위해 트리펩타이드25%, Gly-Pro-Hyp 4%를 함유한 CH인 저분자콜라겐펩타이드(low molecule weight collagen peptide, LMCP)를 이용하여 임상시험을 수행하였다. 피부 조직 내 AGE 축적량을 평가하기 위해 AGE reader를 사용하여 피부자가형광(skin autofluorescence, SAF) 값을 측정하였다. 0.5%와 1.0% LMCP 용액을 8 주 동안 피험자의 전박에 도포한 결과, 시험부위의 SAF값이 대조부위에 비해 유의하게 감소하였다. 추가적으로, LMCP에 의한 피부섬유아세포의 콜라겐 합성 촉진을 평가하기 위해 CCD-986sk를 이용하여 in vitro test를 수행하였다. 그 결과, 800 ㎍/mL의 LMCP는 CCD-986sk의 human pro-collagen Iα1(COL1A1) 합성을 증가시켰다. 본 연구를 통해 LMCP도포가 콜라겐 합성을 촉진하여 항당화 효과에 도움을 줄 수 있다는 것을 확인하였으며, 이는 LMCP가 노화 방지 화장품 원료로써 잠재력이 있음을 시사한다.
