Two endemic foci of heterophyids and other intestinal fluke infections in southern and western coastal areas in Korea (전북 부안군과 경남 사천군 해안지역에서 발견한 이형흡충류 및 기타 장흡충류의 유행지)

  • Two endemic foci of heterophyid infections were discovered in coastal villages of Puan-gun. Chollabuk-do, and Sachon-gun. Kyongsangnam-do, Korea. Fecal examina- tions were performed on 153 inhabitants of Puan-gun and 138 of Sachon-gun, using cellophane thick smear and formalin-ether sedimentation technique. The helminth egg and/or protozoan cyst positive rate was 21.5% (33/153) in Puan-gun and 39.1% (54/138) in Sachon-gun. In Puan-fun, the egg Positive rate of heterophyids was the highest, 17.6%, and that of other parasites was 0.7-2.6% by parasite species. In Sachon-gun, that of heterophyids wa's 18.8%, followed by Clonorchis sinensis (12.3%). and other parasites (0.7-5.OBEI . Twenth-two (Puan-gun) and six (Sachon-gun) heterophyid egg Positive cases were treated loth praziquantel, and adult flukes were collected from their diarrheic stools. A total of 3,284 adult flukes of Heterophyes nocens was collected from all of the 22 patients treated in Puan-gun (3-778 individually) , and other trematodes were also collected from 2- 15 patients; Ftgidiopsis sumac, Stellcntchcsmus JcEcctus, Metogoninus Wokogaupci, M. miyntci, Stictodora Juscnta, Heterophwopsis continua, Acnnthopcrwphium kurognmo, and GWmnophaELoines seoi. In Sachon-gun, M. yokognwci (3,007 specimens), H. nocens (120), aud S. falcatus (46) were collected from 5 of 6 treated patients, and H. continuo and 5. tan each from one patient. The present study revealed that heterophyid flukes, especially If. nocens and M. Wokofowai, are prevalent in the southern and western coastal areas of Korea where fresh and/or brackish water fishes are popularly eaten raw.

Changes in enzyme activity and expression of DHFR of Toxoplumc gondii by antifolates (Antifolate 약제에 대한 톡소포자충의 DHFR 효소활성 및 유전자 발현의 변화)

  • The responses to antifolales of ToxopLasmc Bondii were investigated by measuring the dihydrorolate redLlctase (DHFR) activity. quantity of DHFR mRNA, and single-strand conformational polymorphism (SSCP) pattern. Pyrimethamine (PYM) and methotrexate (MTX) were tested ds anlifolates. When T. gondii was treated wish PYM, the viability was decreased by the increasing concentration of PYM. DHFR activity tended to increase as the passage proceeded. and the quantity of mRNA expressed was also increased according to passages. The viability of 7. gonnii was decreased by the increasing concentration of MTX, but it was maintained over 40% up to $100{\;}{\mu\textrm{m}}$ MTX. DHFR activity was 77.4% in the 1st passage ($1{\;}{\mu\textrm{m}}$). 82.2% in the 4th passage ($10{\;}{\mu\textrm{m}}$), and 141.3% in the 7th passage ($100{\;}{\mu\textrm{m}}$) But no changes were detected in SSCP pattern of T gondii rxposvd to FYM and MTX. both. These results suggested that the response of T gondii to FYN was rofulalcd by transrriptional level and that, in MTX. the viability of T. gonnii was derived from increasing DHFK activity.

  • The present study intended to evaluate the influences of MetagonimMi yokogawai on the activities of brush border membrane bound enzymes of the small intestine. Mice were infected with 500 metacercariae respectively, and the worm recovery, morphological changes and enzyme activities were observed chronologically. A part of them were followed after the treatment. Recovered worms decreased in number continuously after the infection, and they were less than 10% after 2 weeks and almost Bero after 28 weeks. Villous atrophy and stromal inflammation were found at two locations of the proximal jejunum from 2 weeks to 4 weeks after the infection. The enzymes, alkaline phosphatase, leucine aminopeptidase and disaccharidases (sucrase, lactase, maltase, and trehalase), showed lowered activities in the duodenum and proximal jejunum of the infected mice but they increased in the distal jejunum for the first two weeks. From three weeks after the infection, the activities were gradually recovered. In one week treated mice, they recovered the activities at 2 weeks from the treatment, but there found no differences of the activities between the 3 week treated group and infected controls. The present data reveal that M. yckogawai infection induces degenerative changes of the host's intestinal mucosa not only morphologically but functionally during the initial phase of infection. The lowered enzyme activities in acute metagonimiasis should be associated with malabsorption and diarrhea.

Epidemiological Studies of Digenetic Trematodes in Yongyang County, Kyungpook Province (경북 영양퇴역 흡충류 역학조사)

  • The present study was undertaken to determine the endemicity of trematode infections in Yongyang County, Kyungpook Province, Korea. One hundred and seventy-seven out of 955 residents examined were found to be infected with one or more kinds of helminthes. The prevalence rate was 18.5%. Among them, 70 were found to be infected with Clenorchis sinensis, 74 with Meta-gonimus sp. and 2:l with both flukes. The eggs of Fasciolidae were demonstrated from two specimens. The prevalence rate of clonorchiasis in males was 12.6%, while that in females was 3.6% (p<0.05). The prevalence of metagonimiasis in males was 12.0% and was also significantly higher than 6.1% in females (p<0.05). The intensity of both infections was significantly heavier in males than in females. The prevalence of both nukes was higher in residents of oyer 30 years of age than in those below that age. No correlation was found between the intensity and the age group. Among eight species of the fresh-water fish collected at the Panbyon River, Zacco temmincki was found to be the most frequently and heavily infested with metacercariae of Metagonimus sp., and Gnathopogon asromaculatus alone was found to be infested with metacercariae of Clonorchis sinensis. The present results suggest that Yongyang area still remains endemic with Clonorchis sinensis and Metagonimus sp. infection.

Toxoplasma Antibodies by Indirect Latex Agglutination Tests in Zoo Animals (동물원 동물에서의 Latex응집반응에 의한 Toxoplasmu항체가)

  • Total of 216 animals conserved in Seoul Grand Park were examined on the antibody titers of Toxoplasma by the indirect latex agglutination test, 20 out of 131 cases (15.3%) in mammals, 2 out of 75 cases (2.7%) in birds, and none in reptiles, according to species, 15 out of 68 species (22.1%) in mammals, 2 out of 36 species (5.6 %) in birds showed positive antibody titers when the titers of 1 : 32 or higher were regarded as positive. In mammals, it appeared as positive in 2 out of 6 cases (1 out of 3 species) in marsupials, 1 out of 15 cases (1 out of 11 species) in primates, lout of 1 case in bats, 6 out of 13 cases (5 out of 10 species) in carnivores, 1 in 12 cases (1 species out of 3) in odd-toed ungulates, 9 out of 80 cases (6 species out of 38) in even-toed ungulates, and none in rodents and in whales. In birds, 1 out of 21 cases (1 out of 7 species) in gallinaceous birds and lout of 6(5 species) in parrots appeared to have the positive antibody titers of Toxoplasma. And, none of reptiles showed positive. Frequencies of positive antibody titers were high in 1 : 64, 9 cases in mammals, followed by 1 : 32, 6 cases, 1 : 128, 3 cases, and 1 : 256, 2 cases, respectively. Two positive cases in birds appeared to be 1 : 64.

Scanning Electron Microscopical Findings of Echinochasmus japonicus Tegument (Echinochusmus juponicus 표피 미세구조의 주사전자현미경적 관찰)

  • Tegumental ultrastructures of Echinochasmus japonicus were observed by scanning electron microscopy. The worms were recovered from albino rats which were experimentally infected with the metacercariae obtained from Pseudorasbora parva. Followings are summarized findings. 1. The worms were minute and plumpy gourdshaped with attenuated anterior and round posterior end. The tegument of whole body was wrinkled transversely and covered with cobblestone-like cytoplasmic processes. 2. Head crown was armed with 24 collar spines which were embedded in cytoplasmic pockets. The spines were arranged in a row with an interruption at dorsomedian line, however, the 2nd and the 4th spines were outstretched more than others. Oral and ventral suckers were muscular with numerous type II sensory papillae, and genital pore opened between the two suckers. 3. Tegumental spines were spade-shaped with broad base and pointed tip. They were compact in ventra-lateral tegument or dorsal surface of anterior body. They were not found between the two suckers and dorsal surface of posterior body. 4. Two types of sensory papillae, uni-ciliated (Type I) and roundly swollen sensory papillae (Type II), were observed. The type I papillae were chiefly distributed on ventral surface of tegument and type II were on the lips of suckers. Arrangement of collar spines, shape and distribution of tegumental spines or sensory papillae are regarded as characteristic features of E. japonicus.

Ultrastructural studies on the surface of Puragonimus westermani metacercaria (폐흡충 피낭유충 표피의 형태학적 관찰)

  • The present study was undertaken to demonstrate the surface structure of Paragonimus westermani metacercaria in Korea with special reference to the distribution of sensory papillae. Metacercariae were isolated from crayfish, one of the second intermediate tost of P. westermani in Bogil island, Chollanam-do (Province), Korea, where has been known as an endemic area of human paragonimiasis. Isolated metacercariae were encysted and examined with light. scanning and transmission electron microscopes for morphological features. On the surface of iBetacercariae, three types of sensory FaFillae were identified. Large domed papillae ($3~5{\mu\textrm{m}}$), which were covered with wrinkled plasma n!embrane of the worm, were distributed on the oral and ventral suckers only. On the oral sucker, these large domed papillae were 12~13 in number. On the other hand large domed papillae on the ventral sucker were constantly 6 in number and hexagonal in distribution. Small domed papillae ($2~3{\mu\textrm{m}}$), of which surface was more smooth than those of large ones, were distributed symmetrically on the ventral (30~32 pairs) and dorsal surfaces (40~42 Pairs). Ciliated Papillae ($0.8~1.5{\mu\textrm{m}}$) were observed about 5~6 in number around the oral sucker and 3~5 pairs each on the ventral and dorsal surface of the body. Single Fcinted spines covered the entire surface of the body except around the excretory pore. Spines on the anterior fart of the body were 0.9~2.0${\mu\textrm{m}}$ in length and $45~55/100{\mu\textrm{m}}$2 in number, and were gradually reduced in length ($0.4~1.4{\mu\textrm{m}}$) and in nuns.her ($12~27/100{\mu\textrm{m}}$2) toward the posterior part. The body wall of p. westermoni metacercariae was consisted with anucleated syncytium layer, fibrous interstitial layer and musclar layer. In the anucleated syncytium, biconcave ($0.15~0.55{\mu\textrm{m}}$) and spherical ($0.08~0.16{\mu\textrm{m}}$) secretory granules, which were transferred from epidermal cells via protoplasmic tubules, mitochondria and rihoEorses, T-ere observed. Spines originated around the basement membrane protruded externally. Epidermal cells were consisted with a nucleus and a cytoplasm, and connected to syncytium with protoplasmic tubules. In the cytoplasm many secretory granules, mitochondria, Golgi complex, endoplasmic reticula, ribosomes and lipid droplets were observed.

Detection of circulating antigens in rats experimentally infected with Paragonimus westermani by ELISA (ELISA법을 이용한 폐흡충 감염 백서의 혈청내 항원 검출)

  • Circulating antigens in rats experimentally infected with Paragonimus westermani were examined by ELISA. From a total of 22 albino rats, each fed with 25 metacercariae, blood samples were collected until 12 weeks after infection. The specific antibodies against P. westermani in the serum of an infected cat were purified by ammonium sulfate precipitation, DEAE anion-exchange chromatography and affinity chromatography serially. So-called double antibody sandwich ELISA method was used for the detection of circulating antigens. The results were as follows: Mean value of O.D. in control sera was O. 04 (S.D.=0. 04). After infection, mean O.D.(S.D.) values were changed serially: 0.03(0.01) at 0.5 week(3 days), 0.55(0.50) at 1 week, 0.69(0.45) at 1.5 week, O.20 (0.19) at 2 weeks and O.13(0.10) at 2.5 weeks of infection. They returned, thereafter, to the level before infection. When O. 16 (mean+3 S.D.) were considered as cut-off value, those higher than O. 16 were observed only in the sera collected between 1 and 2.5 weeks after infection. Average 8. 4 immature worms (2.2 from the lungs and pleural cavities; 6.2 from muscles) were recovered in a rat at 12 weeks after infection. The fact that circulating antigens were not detected after 3 weeks of infection was considered to the caused by the formation of antigen-antibady complexs.

Isolation and identification of Cyuptosporidium from various animals in Korea (한국산 동물로부터 크립토스포리디움의 분리 및 동정 III. 닭으로부터 Cryptosporidium baileyi의 분리)

  • Each of SPF chicken(Hi-Line strain, 2-day-old males) was inoculated with 2.5 or $5\times10^4$ oocysts by stomach tube. The oocyst was the medium type of Cryptosporidium previously isolated from Korean chicken origin, and passed in 2-day-old SPF chicken. The patterns of oocyst discharge were monitored daily, and in order to observe the ultrastructure of the developmental stages, the bursa of Fabricius of the chicken was examined by transmission electron microscopy (TEM) on the 12th day postinoculation. The prepatent period for 8 chicken was 5.9 days postinoculation on the average, and the patent period was 12.9 days. The number of oocysts discharged per day for the chicken was reached peak on day 12 postinoculation on the average. A large number of oocysts was found in fecal samples obtained from inoculated chicken on days 8~14 postinoculation. The ultrastructural feature of almost every developmental stage of the medium type from chicken was very similar to that of Cryptosporidium previously isolated from mammalia including human and birds except for the attachment site of C. tsuris to the mucus cell from mammalia, but dimension of the oocysts from fecal samples of the medium type was different from those of C. meleagridis and mammalia origin. The above results reveal that the medium type of Cryptosporidium of Korean chicken origin is identified as Cryptosporidium baileyi.

Epidemio-entomological survey of Japanese encephalitis in Korea (한국에 있어서 일본뇌염의 역학적, 매개동물학적 조사)

  • In order to determine the seasonal prevalence and population dynamics of Culex tritaeniorhynchus in relation to the epidemics of Japanese encephalitis, and ecology of these vector mosquito in Kyungpook Province, Korea, studies were con- ducted during the Period of 7 years from 1984 to 1990. Cx. tritaeniorhynchus first collected in June between 4th and 28th, and trapped in large numbers during the period from mid-August to early September, showed a simple sharply pointed one-peaked curve. There was a gradual decrease from mid-September, with a very small number of them collected until early October in every year. The average number of Cx. tritaeniorhynchus rapidly decreased after 1985, and the number became particularly low in 1989. The highest population density, which was observed in August during the initial three years, was found to be delayed in the following years, accompanied by a decrease in the number of mosquitoes. In the trend of nocturnal activity of Cx. tritaeniorhynchus, with oncoming darkness they become very active, gradually decreasing in activity toward mid night, but slightly increasing toward dawn. The immature stages of Cx. tritaeniorhynchus were first found in rice fields contributing to peak adult densities in mid-July. The highest average densities of Cx. trisaeniorhynchus was 14,900 per m2 on mid-August 19th. The larval Cx. tritaeniorhynchus showed high resistance levels and resistance ratios against 5 organophosphorus compounds. In the adult horisontal life table characteristics of Kyungsan colonies of Cx. tritaeniorhynchus under insectary condi- tions, life expectancy was 28.3 days for males and 59.8 days for females. The net reproductive rate was 7.8 and generation time was 25.6 days.
