A small-scale survey on the status of intestinal parasite infections in rural villages in Nepal

  • The status of intestinal parasite infections was investigated in two rural villages (Chitrasar, Jerona) in Chitwan District, Nepal in 1999. Stool examination was performed with a total of 300 specimens from schoolchildren by formalin-ether sedinentation technique. The prevalence rate of intestinal parasite infections in the surveyed areas was 44.0%. The prevalence rate in Jerona was slightly higher than that in Chitrasar. The prevalence rate of intestinal parasite infections in female was slighly higher than that in male without statistically significant difference. Entamoeba coli was the most commonly found protozoan parasite (21.0%) followed by Giardia lamblia (13.7%) and others (5.3%). Hookworm was the most prevalent intestinal helminth (13.0%) followed by Trichuris trichiura (3.0%) and others (5.0%). forty-three specimens (14.3%) Showed mixed infections. It is necessary to implement large-scale treatment with anthelminthics, health education and sanitary improvement for intestinal parasite control in the surveyed areas.

  • An enzyme analysis of the liver fluke, Clonorchis sinensis from Kimhae, Korea and from Shenyang, China was conducted using a horizontal. starch gel electrophoresis in order to elucidate their genetic relationships. A total of eight enzymes was employed from two different kinds of buffer systems. Two loci from each enzyme of aconitase and esterase (${\alpha}-Na{\;}and{\;}{\beta}-Na$) : and only one locus each from six enzymes, gluucose-6-phosphate dehydrogenase (G6PD), ${\alpha}-glycerophosphate$ dehydrogenase (GPD), 3-hydroxybutyrate dehydrogenase (HBDH), malate dehydrogenase (MDH), phosphoglucose isomerase (PGI), and phosphoglucomutase (PGM) were detected. Most of loci in two populations of C. sinensis showed homozygous monomorphic banding patterns and one of them, GPD was specific as genetic markers between two different populations. However, esterase (${\alpha}-Na$), GPD, HBDH and PGI loci showed polymorphic banding patterns. Two populations of C. sinensis were more closely clustered within the range of genetic identity value of 0.998-1.0. In summarizing the above results, two populations of C. sinensis employed in this study showed mostly monomorphic enzyme protein banding patterns, and genetic differences specific between two populations.

Identification of Protein Arginine Methyltransferase 5 as a Regulator for Encystation of Acanthamoeba

  • Encystation is an essential process for Acanthamoeba survival under nutrient-limiting conditions and exposure to drugs. The expression of several genes has been observed to increase or decrease during encystation. Epigenetic processes involved in regulation of gene expression have been shown to play a role in several pathogenic parasites. In the present study, we identified the protein arginine methyltransferase 5 (PRMT5), a known epigenetic regulator, in Acanthamoeba castellanii. PRMT5 of A. castellanii (AcPRMT5) contained domains found in S-adenosylmethionine-dependent methyltransferases and in PRMT5 arginine-N-methyltransferase. Expression levels of AcPRMT5 were increased during encystation of A. castellanii. The EGFP-PRMT5 fusion protein was mainly localized in the nucleus of trophozoites. A. castellanii transfected with siRNA designed against AcPRMT5 failed to form mature cysts. The findings of this study lead to a better understanding of epigenetic mechanisms behind the regulation of encystation in cyst-forming pathogenic protozoa.

Metagonimus yokogawai: a 100-kDa Somatic Antigen Commonly Reacting with Other Trematodes

  • This study was undertaken to characterize the properties of a 100 kDa somatic antigen from Metagonimus yokogawai. Monoclonal antibodies (mAbs) were produced against this 100 kDa antigen, and their immunoreactivity was assessed by western blot analysis with patients' sera. The mAbs against the 100 kDa antigen commonly reacted with various kinds of trematode antigens, including intestinal (Gymnophalloides seoi), lung (Paragonimus westermani), and liver flukes (Clonorchis sinensis and Fasciola hepatica). However, this mAb showed no cross-reactions with other helminth parasites, including nematodes and cestodes. To determine the topographic distribution of the 100 kDa antigen in worm sections, indirect immunoperoxidase staining was performed. A strong positive reaction was observed in the tegumental and subtegumental layers of adult M. yokogawai and C. sinensis. The results showed that the 100 kDa somatic protein of M. yokogawai is a common antigen which recognizes a target epitope present over the tegumental layer of different trematode species.

Monitoring of Deforestation and Fragmentation in Sarawak, Malaysia between 1990 and 2009 Using Landsat and SPOT Images

  • Sarawak is the largest state in Malaysia that covers 37.5% of the total land area. Multitemporal satellite images of Landsat and SPOT were used to examine deforestation and forest fragmentation in Sarawak between 1990 and 2009. Supervised classification with maximum likelihood classifier was used to classify the land cover types in Sarawak. The overall accuracies of all classifications were more than 80%. Our results showed that forests were reduced at 0.62% annually during the two decades. The peat swamp forest suffered a tremendous loss of almost 50% between 1990 and 2009 especially at coastal divisions due to intensified oil palm plantation development. Fragmentation analysis revealed the loss of about 65% of the core area of intact forest during the change period. The core area of peat swamp forest had almost completely disappeared during the two decades.

한국산 해산어류의 Anisakid유충 감염률 및 형태학적 분류 (Morphological Classification and Infection Rate of Anisakid Larvae in Marine Fishes)

  • Anisakiasis occur after the ingestion of raw marine fish and squid containg anisakid larvae. In recent years about 40 cases of anisakiasis have been recorded in Korea. Considering, however, the Korean style of eating raw fish, many more cases would exist and prevention is necessary. We describe the infection rate of anisakid larvae in 13 species of marine fish and squid which were caught in the Korean sea. And each extracted larva is classified according to morphological characters. The results are following ; Scomber japonicus, Pseudosciaena manchurica, Trichiurus haumela showed high infection rate of anisakid larvae. Although Sepia esculenta showed low infection rate, most of anisakid larvae found in Sepia edulis are embedded in muscles. So relative high frequent rate of anisakiasis may developed by Sepia esculenta. Five type (Anisakis Type I, Terranova Type B, Raphidascaris sp., Contracaecum Type A, Contracaecum Type D) of anisakid larvae are classified according to their morphological characters.

자유생활 아메바의 냉동보관 (The maintenance of free-living amoebae by cryopreservation)

  • 자유생환 아메바를 효과적으로 유지할 목적으로 냉동보관을 시도하였다. Glycerol과 Dimethylsulfoxide를 각 각 최종농도 7.5%가 되도록 아메바현탁액에 첨가하였다. 아매바 영양형을 냉동하는 과정에서 온도 하강속도가 빠를때 (평균 $1.3^{\circ}C/min$)보다 느릴때 (평균 $0.7^{\circ}C/min$) 생존력이 더 높았다. 액체질소 속에서 60일동안 보관한 후의 생존율은 처음 넣어준 아메바 영양형 수의 2~39%에 달하였다. 냉동 보관했던 아메바를 해동했을 때 어떤 형태적 변화나 배양했을 때의 문제점도 관찰되지 않았다.

A Retrieved Sparganum of Spirometra erinaceieuropaei from a Korean Man during Mechanical Thrombectomy

  • Human sparganosis is a zoonotic disease caused by infection and migration of the plerocercoid of Spirometra spp. Although sparganosis were reported from most parts of the body, the sparganum parasitizing inside cerebral artery is remarkably uncommon. We report a case of cerebral intravascular sparganosis in an elderly patient with acute ischemic stroke who was diagnosed by retrieving sparganum during mechanical thrombectomy. Finally, the parasites were identified as Spirometra erinaceieuropaei using multiplex PCR and cox1 gene sequencing.

Indoor Occurrence of the Ghost Ant Tapinoma melanocephalum (Hymenoptera: Formicidae) in Urban Homes in Korea

  • The ghost ant Tapinoma melanocephalum is a common household pest worldwide. The present study examined the occurrence of the species in urban homes in Korea. During the period of September 2014 to January 2016, T. melanocephalum workers were collected from 58 homes at 29 different localities using bait traps with 10% sugar solution. The species was widely distributed throughout urban homes at 29 different localities, and the indoor occurrence of T. melanocephalum was highest in Seoul (32.7%) and metropolitan areas of Gyeonggi-do (Province) (29.3%). The indoor incidence rate of T. melanocephalum peaked in September (22.8%), remained moderate from October through April, and peaked again in May (15.7%). In contrast, a low incidence was observed from June to August (7.0%). The present study provides evidence that native ants, such as T. melanocephalum, are potential indoor pests of homes in Korea throughout the year.

Study on the Mechanical Properties of Tropical Hybrid Cross Laminated Timber Using Bamboo Laminated Board as Core Layer

  • This study was performed to analyze the mechanical properties of tropical hybrid cross-laminated timber (CLT) with bamboo laminated board as the core layer in order to evaluate the possibility of its use as a CLT material. Bamboo board was used as the core layer and the tropical species Acacia mangium willd., from Indonesia, was used as the lamination in the outer layer. The modulus of elasticity (MOE), modulus of rupture (MOR), and shear strength of the hybrid CLT were measured according to APA PRG 320-2018 Standard for Performance-Rated Cross-Laminated Timber. The results show that the bending MOE of the hybrid CLT was found to be 2.76 times higher than SPF (Spruce Pine Fir) CLT. The reason why the high MOE value was shown in bamboo board and hybrid CLT applied bamboo board is because of high elasticity of bamboo fiber. However, the shear strength of the hybrid CLT was 0.8 times lower than shear strength of SPF CLT.
