Tissue Distribution of Tropane Alkaloids in Rats and its Determination by GC/MS After the Oral Administration of Scopolia Rhizome (GC/MS에 의한 tropane alkaloids의 분석 및 흰쥐의 생체내 분포)

  • Scopolia rhizome is mistaken as an atractylodes rhizome because of their similarities in shape. That is why atractylodes rhizome imported from China sometimes contain scopolia rhizome, which is very toxic. 8 persons were intoxicated atractylodes after taking imported atractylodes rhizome which is tainted. In kampo medicine prepared with such imported atractylodes rhizome, the level of tropane alkaloids ranged from 1.12∼4.34 mg/dose. In this study, we tried to investigate the tissue distribution of scopolia rhizome in rats. The extracts of scopolia was administered orally to rats (a single dose of 10mg/kg, 20mg/kg and 7 days repeated dose of 10mg/kg). Their blood was collected at 0.5, 1, 2, 4, 6 hrs, and liver, kidney, lung and spleen were collected after 6 hrs. The tissue homogenate was applied to solid phase extraction column for the determination of tropane alkaloids. After the oral administration of 20mg/kg scopolia extracts, l-hyoscyamine was detected in rat blood to 2 hrs after dosing. The concentration of tropane alkaloids was the highest in liver followed by lung, kidney and spleen. However, lung, kidney and spleen were similar in amount.

  • The effects of temperature on Vitali test for tropane alkaloids were investigated. At low temperatures, after nitration occurred at para-position, the electron force of the nitro group caused hydrolysis of ester and oxidation, p-nitrobenzoic acid being produced. At $140^{\circ}C(high{\;}temperature)$, pyrolytic deesterification first took place and then nitration followed, yielding 2,4-dinitrobenzoic and 3,5-dinitrobenzoic acids.

Selection of Tropane Alkaloids High-Producing Lines by Single Cell Cloning of Hyoscyamus niger L. Root Cultures

  • Hyoscyamus species is sources of the hypnotic and sedative drugs hyoscyamine and scopolamine. Single cells of Hyoscyamus niger were dissociated from suspension cultures and adventitious roots obtained from single-cell clones which were cultured on B5 medium containing 3% (w/v) sucrose, 0.1 mg/L IBA and 0.4% (w/v) gelrite. H. niger adventitious root lines showed wide variation in tropane alkaloids production and growth. An effective selection of 200 root lines was made possible by the application of the 'Dragendorff's reagent' for qualitative detection of the alkaloids from root. A high correlation coefficient (r=0.9390) was observed between the values obtained with the two methods based on HPLC and Dragendorff's reagent analysis. Among the selected roots, the highest scopolamine content was 16.64 mg/g DW (Hn-59), which was 8.82-fold more productive than the lowest alkaloid producing line (Hn-25). Here, we established an efficient selection method on tropane alkaloids production and suggest that the Dragendorff's reagent is of great practical value in selection of invisible compounds.

Studies on the Determination of Hyoscine-N-butylbromide by ORD and CD(I) (ORD 및 CD에 의한 Tropane Alkaloid의 정량에 관한연구(I) - Hyoscine-N-butylbromide의 정량)

  • The ORD and CD for hyoscine-N-butylbromide present curves of negative Cotton effect in 190-250nm region. The values of .alpha./sub 232/ .alpha./sub 208/, vertical bar .alpha./sub 232/ vertical bar + vertical bar .alpha./sub 208/ and .theta./sub 221.5/ have linearities in the range of lower concentration (hyoscine-N-butylbromide 0.01-0.1w/v%). It is possible to determine the optical purity of hyoseine-N-butylbromide by using these linearities. This experiment is extremely simple and less time-consuming than other methods for estimating optical purity.

Gas-liquid Chromatographic Analysis of Some Tropane Alkaloids

  • The aim of this investigation was to develop a quantitative gas-liquid chromatographic method of analysis forhyosyamine and scopolamine, and to apply this method to the analysis of preparations. The trimethylsily(TMS) derivatives of the alkaloids were found to be far superior to the nonsilylating compounds in charomatagrams. Bis(trimethylsilyl) acetamide(BSA) was evaluated and found to be a good reagent for silyation of the alkaloids. The optimum derivatization conditions were heating the alkaloids in a closed tube at $70^{\circ)$ for 30 min with a 150 molar excess of BSA to the alkaloids were found to be alkaloids. Calibration curves for the two alkaloids were alkaloid. The standard deviations were 1.1% for hyoscyamine and 1.5% for scopolamine. The minimum detectable amount using the hydrogen flame ionization edtector was determined to be 2$\times$10$^{-11}$ moles of each alkaloid injected.
