Relative Hydrophobicity of Triphenylmethane Dyes as Revealed by Interaction with Tetraarylborate Anions

  • The ion aggregates formed between cationic triphenylmethane dyes and tetraphenylborate(TPB) or tetrakis(4-fluorophenyl) borate (TFB) anions have been investigated spectroscopically. The photometric sensitivities of the dyes are found to be increasing in the order pararosaniline < malachite green < methyl violet 2B < crystal violet < ethyl violet. Cetyltrimethylammonium bromide(CTAB) and Triton X-100(TX-100) destroy the ion aggregates. By comparing the concentration of surfactant beyond which dye-borate mixed solutions behave identically with the dye blank, the order of hydrophobicity appears to be parallel with that of photometric sensitivity.

Development of In Vitro Porcine Oocytes Following Intracytoplasmic Injection of Sperm-Mediated GFP Gene

  • Transgenic animals production tools have been valuable for research and purpose. The current methods of gene transfer, microinjection and nuclear transfer, which are widely used in transgenic animal production, but all most methods has only had limited success in production of larger species. Here, we report the possibility of a sperm-mediated gene transfer method in porcine embryos. Oocytes were collected from ovaries harvested at a local slaughterhouse were matured in 500${mu}ell$ drops of TCM-199 under mineral oil at 38.5$^{\circ}C$ in a humidified atmosphere of 5%CO2 in air. After 42-43h of in vitro maturation oocytes were denuded. for sperm injection into the cytoplasm of the porcine oocytes, sperm suspension in NIM medium are subjected extraction with TritonX-100 before mixing with a green fluorescent gene (GFP). Sperm with Tritonx-100 were prepared by adding TritonX-100 to a final volume of 0.05% in the sperm suspension and mixing by trituration for 60s before two wishes in NIM medium at 2$^{\circ}C$. A(ter wishing, sperm were mixed with TritonX-100 at $25^{\circ}C$ followed by washes at 2$^{\circ}C$. Sperm were resuspended in ice cold NIM to a final volume of 400${mu}ell$ and 2-20ng/${mu}ell$ DNA were triturated on ice for 60s. All microinjection was performed in HEPES-buffered CZB medium at room temperature within 2h. After culture in NCSU-23 for 72h, percent of porcine embryos transfected GFP gene are 20.7%(6/29) in 20ng/${mu}ell$ sperm-DNA mixed group and other groups were 3.7 %(2/54)and 4.7%(3/67). These data suggests that sperm-mediated gene transfer method should be used to the production tool of transgenic pig efficiently.

Effects of Pretreatments of Surfactants, Germicides, Sucrose, or Hormones on the Vase Life of Cut Rose 'Red Sandra' (계면활성제, 살균제, 자당 및 호르몬 전처리가 절화장미(cv. Red Sandra) 수명에 미치는 영향)

  • In order to develop a pretreatment solution for cut rose, the effects of surfactants [Tween 20, Triton X-100, polyoxyethylene 4 lauryl ether (PLE)], germicides (aluminum sulfate, $AgNO_3$, dichloroisocyanuric acid, STS, benzalkonium chloride, 8-hydroxyquinoline sulfate), sucrose, and hormones (ABA and kinetin) on the longevity and quality of 'Red Sandra' were investigated in environment-controlled room. Although 20 and 50 ppm Tween 20, and 500 ppm PLE appeared, in appearance, to be effective in retarding blueing and wilting, respectively, they didn't show statistical differences as compared to distilled water control. Among germicides, $AgNO_3$ was the most effective in delaying petal blueing, petal withering, and reduction of fresh weight, regardless of its concentration, while, in the case of STS, only 1mM treatment was effective in delaying of petal withering. Only 5% sucrose treatment delayed petal blueing, petal withering, and bent neck, but showed no significant difference as compared to 500 ppm aluminum sulfate. Finally, single or combination treatments of ABA and cytokinin were found to rather stimulate the senescence of cut rose.

The Effect of Surfactant on Controlled Release of Amino acids Through Poly(2-Hydroxyethyl Methacrylate) Membrane (Poly(2-Hydroxyethyl Methacrylate)막을 통한 아미노산의 방출 조절에 대한 계면활성제의 효과)

  • The transport phenomena of the free amino acids through poly(hydroxyethyl methacrylate)[P(HEMA)] have been investigated with and without various kinds of surfactants solution and in the mixed surfactants solution. Glutamine has the highest diffusivity among 4 amino acids at 1CMC of cetyldimethylethylammonium bromide(CTABr) surfactant. Glutamic acid is not affected by the concentration of CTABr. Methionine and Lysine shows slight decreased diffusivity at 0.5 CMC, but increase its diffusivity at 1CMC and 2CMC due to the structure change of membrane and the viscosity change of surfactant solution. Glutamic acid has the highest diffusivity among four amino acids at sodium dodecyl sulfate(SDS) and Triton X-100 surfactant. In mixed surfactant solution, each amino acids shows high diffusivity through 45% water content membrane at the 0.5 mole fraction of SDS in the SDS/TX-100 surfactant mixtures. It has been found that not only the property of membrane but also the effects of solute-solvent interactions and solvent effect are very important as the permeation of amino acids occurs through P(HEMA) membrane. The diffusivities of free amino acids through membrane depend upon their molecular shape, size and charge.

  • Spectrophotometric determination of rare earth elements with MTB and the composition ratio were investigated in the presence of surfactants of cetylpyridinium chloride (CPC), Triton X-100, dodecyltrimethylammonium bromide (DTMAB) and cetyltrimethylammonium bromide (CTMAB) at pH 6.5. The colour development between MTB and rare earths in the presence of cationic surfactants was very stable and more sensitive than that in the absence of surfactants. The largest absorbance increase was provided by CPC, which was therefore chosen for determination of rare earth elements. REE-MTB-CPC complex has absorption maxima at 650 nm and obeys the Beer's law in the range of 0∼100 ng/ml. Molar absorptivity is $6.6{\sim}9.4{\times}10^4\;mol^{-1}l\;cm^{-1}$.

  • Trace cadmium was determined in urine by flame and nameless AAS. Inflame method, cadmium was extracted into MIBK from Cd-DDTC complex. In flameless method, samples were pretreated with 1%, Triton X-100. 1% $HNO_3$ and matrix modifier $Pd(NO_3)_2$. Analysis of cadmium was, performed at $450-750^{\circ}C$ ashing temperature by use of pyrocoated tubes. Results of analysis, flameless method superior in sample treatment and reproducibility to flame method and highest absorbance was obtained at $550^{\circ}C$ ashing temperature. 100 mg/L $Pd(NO_3)_2$ concentration. Proposed method were applied to the determination of cadmium in standard urine Lononorm-Metalle 3.

Isolation and Identification of Proteins Increasingly Expressed in Beef Loin on Maturation (성장기 소의 등심에 발현되는 단백질들의 분리 및 동정)

  • Protein profiles of beef loin were constructed by comparing two-dimensional gel electrophoresis patterns of the proteins from different growth stages of Hanwoo, Korean cattle. Proteins from the lean muscle of 0, 6, 12 and 24 months old Hanwoo were separated by isoelectric focusing (IEF) on 16 cm tube gels, and further processed second dimensionally by 12% SDS-polyacrylamide gel electrophoresis (PAGE) using $18{\times}20$ cm gel. Proteins with pI values ranging 3.0 to 9.0 and molecular weight of 15 to 100 kDa could be clearly detected on gel by silver staining. Interestingly, many of the proteins significantly increased and decreased during growth happened to be low molecular ones. To isolate the increased proteins, the soluble proteins were obtained from the tissue by 1% Triton X-100 extraction, then, fractionated by 30% and 50% ammonium sulfate. The isolation condition of each particular protein was determined. According to the conditions, two of the increased proteins were isolated, and transferred to PVDF membrane and microsequenced.

Factors Affecting Electrofusion of Plant Protoplasts (식물 Protoplast의 전기자극 융합에 관여하는 인자)

  • The optimum conditions of electric stimulation for electrofusion of protoplasts of petunia, carrot and soybean, and the effects of calcium, magnesium, protease, trypsin, triton X-100, concanavalin A, dimethyl sulfoxide(DMSO), glycerol monooleate and spermine on fusion frequency and/or viability of petunia protoplast were investigated. The optimum frequencies(Hz)-amplitudes(V/cm) of AC Pulse for protoplast pearl-chain formation were 10 kHz-20 V/cm and 1 MHz-60 V/cm for petunia, 100 kHz-40 V/cm and $1\;MHz-40{\sim}60\;V/cm$ for carrot, and $1\;MHz-40{\sim}80\;V/cm$ for soybean, respectively. The optimum condition of DC pulse treatment at the 1 MHz-60 V/cm-15sec treatment of AC for electrofusion of petunia protoplasts was 2.5 kV/cm-40 sec, and under this condition the fusion frequency and viability of protoplasts were 45 % and 10 %, respectively, Both of the protoplasts of carrot and soybean were not fused under the AC and DC conditions tested in this experiment. The electrofusion of petunia protoplasts was stimulated by calcium, and the fusion frequency and the viability of the protoplasts were 43 % and 11 % , respectively at the calcium concentration of 140 mM. Although fusion frequency was not affected by magnesium only, magnesium stimulated fusion frequency in the presence of calcium, and the viability and fusion frequency of petunia protoplasts were 45 % and 13 %, respectively, at 140 mM of magnesium-140 mM of calcium. The relative fusion frequencies of petunia protoplasts to the controls were increased by 2.4, 2.1, 1.6, 1.4, 1.8, 1.5 and 2.2 folds, respectively, by the treatments of protease, trypsin, triton X-100, concanavalin A, DMSO, glycerol monooleate, and spermine. The viabilities of petunia protoplasts were decreased by these substances.

Selective Isolation and Phylogeny of the Yeast Species Associated with Aloe vera and Aloe saponaria (알로에 베라(A. vera)와 알로에 사포나리아(A. saponaria)로 부터 효모의 분리 및 계통분석)

  • BACKGROUND: Several yeast species have potential applications in biotechnology and the identification of such yeast species is of great interest. The first step in the identification of yeasts is the establishment of an effective isolation method. Thus, we compared the efficacy of different yeast media in the isolation of yeast associated with Aloe vera and Aloe saponaria. METHODS AND RESULTS: In this study, we spread homogenized A. vera and A. saponaria leaves onto 4 different yeast selective media containing chloramphenicol, streptomycin, Triton X-100 and L-sorbose. We observed high selectivity for yeast and many colonies on media. We isolated 67 yeast strains from A. vera and 42 yeast strains from A. saponaria. We used phylogenetic analysis to identify the yeast isolates based on ITS region sequencing and performed sequence analysis on representative isolates from each agar plate. Further, we compared the sequences obtained with reference sequences. The yeast species isolated from A. vera were as follows: 56 isolates of Meyerozyma, 9 isolates of Cryptococcus, and 1 isolate each of Rhodotorula and Sporobolomyces. Those isolated from A. saponaria were as follows: 41 isolates of Rhodosporidium and 1 isolate of Sporobolomyces. CONCLUSION(S): All the isolates obtained using large agar plate containing chloramphenicol, streptomycin, Triton X-100 and L-sorbose were identified as yeast. Therefore, we concluded that this method is useful for selective screening of yeast species.

Graphene Oxide/Polyimide Nanocomposites for Gas Barrier Applications (산화그래핀이 함유된 폴리이미드 나노복합막의 기체차단성 평가 및 활용)

  • Polymeric films for gas barrier applications such as food packaging and electronic devices have attracted great interest due to their cheap, light and easy processability among gas barrier materials. Especially in electronic devices, extremely low gas permeance is necessary for maintaining the device performance. However, current polymeric barrier films still suffer from relatively high gas permeance than other materials. Therefore, there have been strong needs to enhance the gas barrier performance of polymeric barrier films while keep their own advantages. Recently, graphene is highlighted as a 2D-layered material for gas barrier applications. However, owing to the poor workability and difficulty to produce in engineering scale, graphene oxide (GO) is on the rise. GO consists of oxygen-containing functional groups on surface with intrinsic 2D-layered structure and high aspect ratio, and it can be well-dispersed in aqueous polar solvents like water, resulting in scalable mass production. Here, we prepared GO incorporated polyimide (PI) nanocomposites. PI is widely used barrier polymer with high mechanical strength and thermal and chemical stability. We demonstrated that PI/GO nanocomposites could perform as a gas barrier. Furthermore, surfactants (Triton X-100 (TX) and Sodium deoxycholate (SDC)) are introduced to enhance the gas barrier performance by improving the degree of dispersion of GO in PI matrix. As a result, TX enhanced the gas barrier performance of PI/GO nanocomposites which is similar to predicted value. This finding will provide new insight to polymer nanocomposites for gas barrier applications.
