Experimental study on the effects of SIMNETONG, GANGJIYOUNSOO-DAN and CHUNGSIMJOKITANG(Xinnao-Tong, Jiangzhiyanshou-Dan and Qingxintiaoqi-Tang) on hyperlipidemia (심뇌통(心腦通),강지연수단(降脂延壽丹) 및 청심조기탕(淸心調氣湯)의 고지혈증(高脂血症)에 대(對)한 실험적(實驗的) 연구(硏究))

  • In order to evaluate the effects of SIMNETONG, GANGJIYOUNSOODAN and CHUNGSIMJOKITANG(Xinnao-Tong, Jiangzhiyanshou-Dan and Qingxintiaoqi-Tang) on hyperlipidemia, the experimental study was performed in rats without or with hyperlipidemia induced by 2% cholesterol fed-diet or Triton WR-1339. And the levels of serum total cholesterol, triglyceride, HDL-cholesterol, LDL-cholesterol and phospholipid were measured. The results are summarized as follows : In the model of hyperlipidemia induced by 2% cholesterol Fed-diet 1. Both low & highly concentrated GANGJIYOUNSOODAN showed significantly decreasing effects on the level of serum total cholesterol while SIMNETONG & CHUNGSIMJOKlTANG didn't. 2. GANGJIYOUNSOODAN, SIMINETONG and CHUNGSIMJOKITANG showed decreasing effects on the level of serum triglyceride with no statistical significance. 3. Only highly concentrated GANGJIYOUNSOODAN showed increasing effects on the level of serum HDL-cholesterol while the others had no effects. 4. Only highly concentrated GANGJIYOUNSOODAN had decreasing effects on the level of serum LDL-cholesterol while the others had no effects. 5. Highly concentrated GANGJIYOUNSOODAN and CHUNGSIMJOKITANG showed significantly decreasing effects on the level of serum phospholipid while the other had no effects. In the model of hyperlipidemia induced by Triton WR-1339 6. SIMNETONG, GANGJIYOUNSOODAN and CHUNGSIMJOKITANG showed slightly decreasing effects on the levels of serum total cholesterol and triglyceride with no statistical significance. 7. None of SIMNETONG, GANGJIYOUNSOODAN and CHUNGSIMJOKITANG showed any effects on the level of serum HDL-cholesterol. 8. Highly concentrated GANGJIYOUNSOODAN and CHUNGSIMJOKITANG showed decreasing effects on the level of serum LDL-cholesterol but the other had no effects. 9. All of highly concentrated SIMNETONG, GANGJIYOUNSOODAN and CHUNGSIMJOKITANG showed decreasing effects on the level of serum phospholipid with statistical significance. These results show that GANGJIYOUNSOODAN and CHUNGSIMJOKITANG, in orders, have antihyperlipidemic effects. Further study on the mechanism and on the herb screening of these prescription should be followed.

Studies on the Development of Antihyperlipidemic Drugs from Oriental Herbal Medicines(II) - Antihyperlipidemic Effects of Oriental Herbal Medicines - (한방약물로부터 항고지혈증 치료약물개발(2) - 수종 한약재의 항고지혈증 효과 -)

  • In the previous reports, we selected 80% MeOH extract of 7 herbs including Scutellariae Radix(SR), Paeoniae Radix Rubra(PRR), Moutan Cortex(MC), Angelicae Gigantis Radix(AGR), Crataegi Fructus(CF), Bambusae Caulis in Taeniam(BCT) and Cinnamomi Ramulus(CR), which exhibited the inhibitory effect on HMG-CoA reductase and DPPH free radical scavenging effect in vitro, and antihyperlipidemic effects on antihyperlipidemic rats induced by Triton WR 1339 in vivo. Among them, SR, MC, AGR and BCT showed significant suppression of elevated serum LDL-cholesterol level, and AGR and CF showed significant liver weight increase on high cholesterol diet induced hyperlipidemic mice. And, SR, PRR, AGR, BCT and CR significantly suppressed the elevated serum total cholesterol and triglyceride levels on corn oil induced hyperlipidemic rats. Then, in order to research new antihyperlipidemic agents from the oriental medicinal herbs, we chose SR, AGR, CR and BCT which have the antihyperlipidemic effect in vitro and in vivo, and those herbs were systematically fractionated with organic solvent. EtOAc fraction of SR, hexane fraction of BCT, AGR and chloroform fraction of CR exhibited remarkably inhibitory effect on HMG-CoA reductase activity.

Antihyperlipidemic Effects of Red Ginseng, Crataegii Fructus and Their Main Constituents Ginsenoside Rg3 and Ursolic Acid in Mice

  • Anti-hyperlipidemic effect of red ginseng (RG; the steamed root of Panax ginseng C.A. Meyer) and Crataegii fructus (CF, the fruit of Crataegus pinnatifida BGE), which are used frequently in China and Korea as herbal medicines to treat arteriosclerosis, were investigated. Treatments of RG and CF significantly reduced blood triglyceride (TG) and total cholesterol (TC) levels in Triton WR-1339-induced hyperlipidemic mice and serum TG levels in corn oil-induced hypertriglyceridemic mice. Ginsenoside Rg3 and ursolic acid, the main constituents of RG and CF, respectively, also reduced TG and TC levels in hyperlipidemic mice. RG and CF significantly lowered the high blood TG and TC levels and body and epididymal mass weights induced by long-term feeding of a high-fat diet and increased the high-fat diet-induced decrease in blood HDL cholesterol levels. RG and Rg3 reduced the blood TC levels more than CF and ursolic acid. However, blood TG level were reduced by CF and ursolic acid more than RG and Rg3. RG, CF, and their constituents also inhibited pancreatic lipase and HMG-CoA reductase activities. The most potent inhibitor was Rg3. These findings suggest that RG and CF may be suitable for the therapies of hypercholesterolemia and triglyceridemia, respectively.

Lactic Acid Bacteria Increases Hypolipidemic Effect of Crocin Isolated from Fructus of Gardenia jasminoides

  • The fructus of Gardenia jasminoides Ellis (GF) has been widely used as a natural colorant in Asian countries, and also as a Chinese traditional medicine for its homeostatic, antiphlogistic, analgesic, and antipyretic effects. In the present study, its main component, crocin, was fermented with lactic acid bacteria (LAB) and their antihyperlipidemic activity was measured. The GF extract, fermented GF (F-GF), crocin, and fermented crocin (F-crocin) significantly inhibited the increase of serum triglyceride (TG) level in corn oil feeding-induced triglyceridemic mice, as well as that of serum TG and total and LDL cholesterol levels in Triton WR-1339-induced hyperlipidemic mice. These agents also showed hypolipidemic activity in hyperlipidemic mice induced by high fat diet for 5 weeks. F-GF and F-crocin exhibited more potent hyperlipidemic effects than GF and crocin. The results suggest that the hypolipidemic effect of GF and crocin can be synergistically activated by LAB, and that F-GF and F-crocin may improve hyperlipidemia in clinic, compared with GF and crocin.

An Experimental Studies on the Effects of Chungsimondamtang (청심온담탕(淸心溫膽湯)의 효능(效能)에 관(關)한 문헌적(文獻的) 고찰(考察))

  • In order to study oriental medical literature and clinical effects of Chungsimondamtang, the experiments of analgesic action, sedative action, anticonvulsive action, action on the isolated ileum, action on the circulatory system and hyperlipidaemia were observed. The results were as follows: 1. The analgesic effects were significantly noted. 2. The prolongaton of hypnotic time was noted. 3. The inhibitory effects on the convulsion induced by strychnine, picrotoxin and caffeine were significantly recognized. 4. The spontaneous momentum of isolated ileum was evidently inhibited, and recognized anti Ach. and anti Ba. effects. 5. The effects of descending blood pressure in normal rats and anesthetized rabbits were strongly recognized. 6. The effects of inhibition on the increase of TG. and TC. levels in hyperlipidaemia rats induced by Triton WR-1339 was significantly recognized. 7. The effects of inhibition on the increase of TG. and TC. levels in hyperlipidaemia rats induced by 75% fructose was significantly recognized. With the genelalization of the above-mentioned experimental results, literatual and clinical effects of Chungsimondamtang were approximate to actual experimental results.

Rhei Rhizoma and Chunghyuldan Inhibit Pancreatic Lipase

  • Pancreatic lipase-inhibitory activity of the rhizome of Rhei Rhizoma and its antihyperlipidemic activity were measured. Rhei Rhizoma inhibited pancreatic lipase with $IC_{50}$ value of 6.5 mg/ml (triolein as a substrate). Rhei Rhizoma significantly inhibited serum TG level in corn oil feeding-induced mice, and serum TG and cholesterol in Triton WR-1339-induced hyperlipidemic mice. However, Rhei Rhizoma did not show the hypolipidemic activity in high cholesterol diet-induced hyperlipidemic mice. When in vitro pancreatic lipase-inhibitory and in vivo antihyperlipidemic activities of Whangryunhaedoktang (WT) and Chunghyuldan (CD), which is consisted of ingredients of WT and Rhei Rhizoma, were measured, CD exhibited more potent inhibitory activities than WT. Therefore these results suggest that antihyperlipidemic activity of Rhei Rhizoma and CD may be more or less originated from the inhibition of pancreatic lipase.

Effect of Brassica rapa L. extracts and ${\beta}-sitosterol$ on hyperlipidemic rats (순무와 ${\beta}-sitosterol$의 고지혈증 억제에 대한 연구)

  • To evaluate the anti-hyperlipidemic effect of Brassica rapa L. and its major compound, $\beta$-sitosterol, the present study was undertaken, hypercholesterolemia was induced in rats with poloxamer-407, Triton WR-1339, 30% corn oil high cholesterol diet. The ethanol extract of Brassica rapa L. significantly decreased total cholesterol (TC), phospholipid, triglyceride at doses of 200 mg/kg, and $\beta$-sitosterol significantly exerted anti-hyperlipidemic activity at a dose of 15 mg/kg in rats with hyperlipidemia. Taken together, Brassica rapa L. and $\beta$-sitosterol can be useful agents for the prevention or treatment of hyperlipidemia.

Hypolipidemic and Antioxidative Effects of Bangpoongtongsungsankamibang on Diet-Induced Obesity Rats (비만유도 흰쥐에 대한 방풍통성산가미방 (防風通聖散加味方)의 항고지혈 효과 및 항산화 효과)

  • Obesity is associated with a number of pathological disorders such as non-insulin-dependent diabetes, hypertension, hyperlipidemia, and cardiovascular diseases. Bangpoongtongsungsankamibang (BTSK) has been widely used in the oriental medicine for the treatment of several diseases associated with inflammatory abnormalities in cardiovascular and nervous system. The BTSK is the modified prescription of Bangpoongtongsungsankamibang in which sea tangle (Laminaria japonica) were added. This study was carried out to detemine the anti-obestic effects of BTSK. Pretreatment with the BTSK at daily dose of 100 or 200 mg/kg (p.o.) far 4 weeks reduced serum triglyceride, total cholesterol contents in rat induced by Poloxamer-407 or Triton WR-1339, respectively. Furthermore, post-treatment with BTSK far four weeks also inhibited body weight gain, adipose tissue mass and hyperlipidemia induced by the high fat diet for six weeks. The BTSK shifted serum total-, HDL- and LDL-cholesterol levels toward the values of normal group, suggesting that BTSK has hypolipldemlc effects. The rats fed BTSK reduced lipid peroxide and hydroxy radical in the rat blood and increased superoxide dismutase (SOD) activity compared to the control group. Taken together, these results superoxide that BTSK improve hyperlidemia and obesity via the upregulation of anti-oxidative mechanism.

Anti-hyperlipidemic Effect of Insamsansa-eum in Mice

ISE and its ingredients, RG and CF, inhibited pancreatic lipase and HMG-CoA reductase activities. Of its ingredients, RG reduced the blood TC level and HMG-CoA reductase activity more than CF. CF reduced blood TG level and pancreatic lipase activity more than RG. These finding suggest that the antihyperlipidemic effect of ISE may be due to synergistic inhibition of HMG-CoA reductase and pancreatic lipase by RG and CF, and that ISE may be effective hypolipidemic agents.

The Effects of Sojadodamgangkitang on the injured Tracheal tissue induced SO2 in Rats (소자도담강기탕(蘇子導痰降氣湯)이 SO2에 의(依)한 흰쥐의 호흡기(呼吸器) 조직손상(組織損傷)에 미치는 영향(影響))

  • This experiment was performed in order to study the effects of Sojadodamgangkitang on the injured tracheal tissue induced $SO_2$ in rats. Healthy adult male rats weighting about 250g were divided into 4 groups-the Normal group, the Control group, the group of Sojadodamgangkitang administration for 5 days after $SO_2$ gas exposure (Sample I), and the group of Sojadodamgangkitang administration for 10 days before and for 5 days after $SO_2$ gas exposure (Sample II). The results were obtained as follows ; 1. In the trachea Control group. the lesion of the ciliated epithelium was severe and the mucus secretion of the respiratory tract was increased iginificantly. 2. In the trachea of Sample I group, the lesion of the ciliated epithelium and the mucus secretion of the respiratory tract were decreased compared with Control group. 3. In the trachea of Sample II group, the lesion of the ciliated epithelium and the mucus secretion of the respiratory tract were decreased compared with Control and Sample I group. According to the above results, Sojadodamgangkitang has significant effect on the injuried tracheal tissue caused by $SO_2$ in rats.
