파라자일렌(p-xylene)을 산화반응시켜 테레프탈산(terephthatic acid)을 제조하는 과정 중에 발생하는 주요 유기불순물을 효과적으로 분석하는 방법을 수립하였다. 이 제조공정 중에 발생하는 유기불순물은 매우 다양하나, 여기에서는 주로 benzoic acid, p-toluic acid, p-tolualdehyde, 4-carboxybenzaldehyde, phthalic acid, isophthalic acid, trimellitic acid와 4-hydroxymethyl benzoic acid 등의 유기불순물을 분석하는데 촛점을 맞추었다. 이 유기물들은 공정 중의 모액이나 고체상태의 테레프탈산 입자에 존재하는데, 이들을 동시에 분석하기 위해 99% bis(trimethylsilyl)trifluoroacetamide와 1% trimethylchlorosilane의 내부표준용액과 pyridine의 혼합용액내에서 trimethylsilylation으로 시료를 전처리하고 gas chromatography를 이용하여 분석한 결과 상기한 유기물이 모두 성공적으로 분리되어 50분 이내에 정량분석할 수 있었다.
본 연구는 [$H_2O$]/[TEOS]=1.7에서 산촉매를 첨가한 부분가수분해를 행하여 졸을 합성하였다. 최적의 방사성을 갖는 졸을 결정하기 위하여 부분가수분해에 의하여 합성된 졸을 trimethylsilylation하여 안정화시킨 후에 반응시간에 따르는 분자량과 점도의 변화를 관찰하였다. 최적조건하에서 제조된 졸용액에서 용매를 제거한 후 겔섬유를 제조하였고 이를 $1,000^{\circ}C$에서 열처리하여 실리카섬유를 제조하였다. 제조된 섬유는 단면적이 원형이며 인장강도는 $83{\pm}20kg/mm^2$이었고, 순도는 약 99.997%이었다.
A gas chromatographic method using nitrogen phosphorous selective detection was developed for simultaneous determination of haloperidol and its metabolite, reduced haloperidol, in human plasma. Combelen was used as internal standard, The method involved extraction and trimethylsilylation followed by the injection of $2-4\;{\mu}l$ of benzene layer, which was used to dissolve the trimethylsilylated derivatives of haloperidol and reduced haloperidol, onto SE-54 column [5% phenyl methyl silica fused capillary column, $16m{\times}0.22\;mm$$(I.D.){\times}0.33\;{\mu}m$ (coated thickness)]. The temperature of column oven was programmed from $200^{\circ}C\;to\;300^{\circ}C$ at the increase rate of $10^{\circ}C/min and also the temperatures of injector and detector were set at $300^{\circ}C$. Helium was used as carrier gas and its flow rate was maintained at 30 ml/min. The detection was conducted with nitrogen phosphorous selective detector. The retention times for combelen, reduced haloperidol and haloperidol were found to be 9.14, 9.75 and 9.99 min, respectively. The detection limits for haloperidol and reduced haloperidol in human plasma were both 0.2 ng/ml. The coefficients of variation of the intra-assay were generally low (below 9.8%). The mean absolute recoveries of added haloperidol and reduced haloperidol from plasma were 72% and 84%, respectively. No interferences from endogenous substances were found.
Analytical method for synephrine and octopamine in citrus fruits, drinks containing citrus fruit, and human urine was developed using gas chromatography / mass spectrometry(GC/MS), Silylation with MSTFA, acetylation with MBTFA, and trimethylsilylation with MSTFA followed by trifiuoroacetylation with MBTFA were compared. The selective derivatization of synephrine and octopamine was optimized with two derivatizing reagents ; MSTFA and MBTFA. The ion at m/z 267 was monitored to characterize the benzyl group of the both compounds. Synephrine was detected in the concentrations of 0.46∼1.88 ug/g for citrus fruits and 1.2∼8.1 ug/ml for drinks. The urinary excretion data of synephrine showed the highest concentration at the period of 8-20 hours after drinking orange juices and total amounts of its urinary excretion calculated as a parent compound was 11-14% of a dose during 48 hours. Octopamine was not detected in citrus fruits, drinks, and human urine.
The carpophores of three Korean mushrooms, Coriolus versicolor, Pleurotus ostreatus, and Lentinus edodes were respectively extracted with hot water and the extract were dialyzed through Visking tube. They were found to contain an antinumor activity against sarcoma 180 implanted in mice. The components of these aqueous extracts were found to be polysaccharide and protein by color reactions including anthrone and Lowry-Folin tests. The hydrolysis of the polysaccharide with 3% HCI-MeOH and trimethylsilylation yielded four monosaccharides : glucose, mannose, galactose and xylose which were identified by G. L. C. After hydrolysis of protein with 6N HCL, fourteen to seventeen amino acids including aspartic and glutamic acids were detected by an amino acid analyzer.
The objective of this work is to determine the sugar profiles in commercial fruit juices, and to obtain chemometric characteristics. Sugar compositions of fruit juices were determined by HPLC-RID and GC-FID via methoxymation and trimethylsilylation with BSTFA. The appearance of multiple peaks in GC analysis for carbohydrates was disadvantageous as described in earlier literatures. Fructose, glucose, and sucrose were major carbohydrates in most fruit juices. Glucose/fructose ratios obtained by GC were lower than those by HPLC. Orange juices are similar to pineapple juices in the sugar profiles. However, grape juices are characterized by its lower or no detectable sucrose content. In addition, it was also found that unsweeten juices contained considerable level of sucrose. Chemometric technique such as principal components analysis was applied to provide an overview of the distinguishability of fruit juices based on HPLC or GC data. Principal components plot showed that different fruit juices grouped into distinct cluster. Principal components analysis was very useful in fruit juices industry for many aspects such as pattern recognition, detection of adulterants, and quality evaluation.
케톤체는 생체 에너지 생산과정이 탄수화물보다는 지방산의 의존도가 높을 때 생성되며, 과도한 분비는 당뇨병성 케토시스나 선천성 유전성 대사이상질환을 의심할 수 있는 근거가 된다. 따라서 이의 신속 정확한 분석법의 개발이 필요하다. 본 연구에서는 혈장을 제단백한 후 hydroxylamine을 가하여 60 ℃에서 30 분간 반응시켜 oximation 후 BSTFA를 가한 후 trimethylsilylation 유도체화하여 혈장 중에서 케톤체를 신속하게 정량할 수 있는 GC-MS/SIM 분석법을 개발하였다. 케톤체의 직선성의 범위는 0.001-250 μg/mL 이었고, 혈장에서의 검출한계는 0.1 pg 이었다. 직선성을 가지는 범위에서의 상관계수(R2)는 0.998-0.999이었고, 회수율은 1 μg/mL의 표준품을 첨가하였을 때 88.2-92.3 %, 10 μg/mL의 농도를 첨가하였을 때 89.5-94.8 % 였으며 RSD는 6.3-9.4 %였다. 이 분석법을 정상인과 케토시스 환자의 검체에 적용하여 벨리데이션 하였으며, 본 분석방법은 어린이나 성인의 당뇨성 케토시스나 여러 유전성대사질환 환자 중 케토시스를 보이는 환자의 혈장 중 β-hydroxybutyric acid/acetoacetic acid의 비를 계산하여 케톤체를 신속하고 효율적으로 임상검체 분석에 응용할 수 있음을 보여주었다.
본 연구는 건강한 성인남자로부터 ibuprofen의 인체 내 대사물질의 규명 및 배설에 대한 연구를 gas chromatography-mass spectrometry (GC/MS) 법으로 수행하였다. Ibuprofen 복용 후 3시간 단위로 15시간까지 소변을 채취하여 실험하였다. 이 중 glucuronide가 포합된 대사체로부터 포합체를 떼어내기 위해 6 M HCl을 통해 $100^{\circ}C$에서 30분 동안 가열하여 산 가수분해 시켰다. 가수분해 된 뇨 중 ibuprofen과 그 대사체를 추출하기 위하여 액체-액체 추출법을 적용하였다. 모약물과 대사체의 추출 최적조건을 찾기 위하여 pH 3, 5 및 8에서 수행하였으며 그 중 pH 3에서 가장 좋은 회수율을 보여 주었다. 또한 미량의 대사체 검출을 위하여 trimethylsilylation (TMS) 유도체 반응을 적용시킨 후 GC-MS를 이용하여 분석하였다. 본 연구에서는 모약물을 포함한 총 5개의 대사체를 검출하였으며, 이들 대사물질들은 주로 산화과정에 의해 약물 모핵에 hydroxylation 또는 carboxylation된 화합물이었다. 또한 시간대별로 채취한 시료에서 각각의 대사체의 배설율의 변화를 관찰함으로써 시간대별 배설율 및 축적률을 구할 수 있었다.
본 연구는 건강한 성인남자로부터 비스테로이드성 항염증제인 diclofenac약물을 복용 후 26시간 동안 배설된 소변을 채취하여 이 약물의 대사과정과 배설에 대하여 연구를 수행하였다. 소변 중에 diclofenac의 포합대사체를 검출하기 위하여 산가수분해 과정을 수행하였고 이 과정에서 diclofenac과 대사체들은 탈수반응에 의해 락탐환이 형성되어 이들의 극성은 산성에서 염기성으로 전환된다. 그러나 변환된 락탐환은 염기조건에서 수산화이온에 의해 쉽게 분해가 일어나므로 오히려 염기성 조건에서의 추출율이 낮아지는 경향이 있어 적정 추출 조건에 대하여 연구하였다. 미량의 대사체 검출을 위하여 trimethylsilylation (TMS) 유도체 반응을 시킨 후 gas chromatograph-mass spectrometer (GC-MS)를 이용하여 분석하였다. 본 연구를 통해 4개의 대사체를 검출할 수 있었으며, 이들은 모두 산화과정에 의해 약물모핵에 hydroxylation된 화합물이었다. 각 대사체는 질량스펙트럼의 해석과 이전 연구결과의 비교를 통하여 구조가 규명되었으며, 이를 바탕으로 시간에 따른 모 약물과 대사체의 배설율을 조사하였다. 본 실험 결과로부터 diclofenac의 체내 대사 경로를 제안하였다.
