  • Nine hundred and eighty fungal strains were isolated from lesions of Trifolium repens L. and various weeds. Among them, F40362, F40496, F40497 strains were selected by the screening of herbicidal activity on Trifolium repens L. and Zoysia japonica. Selected three strains showed selective activity between Trifolium repens L. and Zoysia japonica, and were identified as Penicillium sp.

Efficient Transformation of Trifolium repens L. Using Acetosyringone (Acetosyringone을 이용한 효율적인 White Clover의 형질전환)

  • Transformants of White Clover(Trifolium repens L.) were efficiently produced from immature seed derived callus cocultivated with Agrobacterium twnefaciens LBA4404 harboring plant binary vector. pBI121, using acetosyringone. The mean frequencies of transformants on the two kanamycin-containing media were 16 to 19% when the immature seed-derived calli were infected with bacteria cultured in the presence of 100$\mu$M acetosyringone compared with 7% in media without acetosyringone. Transgenic white clover was subject to molecular analysis for integration into plant nuclear genome and expression of $\beta$-glucuronidase(GUS) gene. PCR and Northern blot analyses demonstrated that GUS gene was integrated into white clover nuclear genome and expressed into its mRNA. The expression of GUS gene into its protein was confirmed by spectrophotometric assay of GUS activity.

Esterase Isozyme Variation in the Tissues of Glycine soja S. and Z. , Vigna vexillata var. tsusimensis Matsmura , and Trifolium repens L. in Korea (돌콩 , 돌동부 , 화이트 클로버의 조직별 Esterase Isozyme 변이에 관한 연구)

  • The esterase isozyme in tissue of wild legume plants were separated by horizontal starch gel electrophoresis. Extracts used in this study were prepared from fully expanded young leaf, cotyledon and radicle of seedling and root-nodule of Glycine sola, Vigna vexillata var. tsuscmensis and Trifoliwn repens. The results are as follows; 1. Each tissue examined had a characteristic banding pattcrn. Number of bands in each species, G. soja, V. vexillata, and T . repens, were 14, 8 and 1 1 bands, respectively. And difference in esterase isozyme bands were greater from tissue to tissue than difference between habitat. 2. Est-I, Est-2. Est-3 and Est-4 in G. soja, Est-I in V. vexillata and Est-l and Est-2 in T. repens showed strong cnzyme activity than other enzyme. 3. Esterase isozyme variation in G. soja and T . repens showed more variety than V. vexillata. This is resulted from many genotypic differences within species. 4. The main enLyme among thc esterase isozyme were Est-I. Est-2, Est-3 and Est-4.

Molybdate Alters Sulfate Assimilation and Induces Oxidative Stress in White Clover (Trifolium repens L.)

  • Molybdenum (Mo) in rhizosphere influences sulfate assimilation as well as a number of other physiological aspects. In this study, the activity of key enzymes in sulfate assimilatory pathways, such as ATP sulfurylase (ATPs), adenosine 5'-phosphosulphate reductase (APR), as well as the responses of reactive oxygen species (ROS), were analyzed to elucidate the metabolic and physiological effects of external Mo supply to detached leaves of Trifolium repens L. Mo supply with a range from 1 mM to 40 mM depressed the activity of ATPs throughout the entire time course. In the leaves exposed to 1 mM Mo, a continuous decrease in the activity of ATPs was confirmed by Native-PAGE. The APR activity was also declined by Mo treatment. The accumulation of $H_2O_2$ and ${O_2}^{{\cdot}-}$ were not significant up to 10 mM Mo, whereas a remarked accumulation was detected under 40 mM Mo supply. The data suggest that an external supply of Mo has an inhibitory effect on sulfate assimilation, and induces oxidative stress only at an extremely high concentration.

  • This study was designed to investigate the effects of antagonistic microorganism, Bacillus subtilis, on the growth of forage seedlings in repeated cultivation soils and unrepeated cultivation soils. The field experiment was wnducted in pots in a vinyl house using repeated and unrepeated cultivation soils. Forage types were 'Suwon 19' wrn(Zea mqs L.), 'Califbmia' white clover(Tr~oIium repens L.) and 'Fawn' tall fescue (Festuca arundianacea Schreb.). Samples of white clover and tall fescue were taken h m each pot at 36 days after seeding. Samples of wm were examined at 50 days after seeding. The most active antagonistic bacterium was isolated h m forage rhizosphere soil, and selected by reference to it's antagonistic ability on the growth of pathogenic fungi, Rhizoctonia solmi and Fusarium oxyspomm, and it was identified as Bacillus subtilis. This strain strongly suppressed the growth of fungal pathogens among isolated rhizobacteria. The dry weight of forage shoots and roots cultivated in unrepeated cultivation soils was higher than that cultivated in repeated cultivation soils. The dry weight of forage was positively affected by the inoculation of the antagonistic bacterium, Bacillus subtilis, in both repeated cultivation soils and unrepeated cultivation soils. In conclusion, the growth of forage was more affected by the inoculation of the antagonistic bacterium in unrepeated cultivation soils than that in repeated cultivation soils, and bacterization of forage with B. subtilis resulted in an inrreased yield.

  • The study was to introduce the revegetation measures for the improvement of the landscape in October, 2012 on the slope; two and four years after the introduction, the slope was examined to see the status of the revegetation. The result is as follows. The result from examining the soil on the slope, the soil was Loamy sand; which infers that the soil is in a poor condition for improving the scenery. Examining the characteristics of the vegetation-base materials, the soil acidity was pH 6.4 to 6.7; and the soil hardness was 21 to 24mm (hardness quotient of soil), this can be considered a favourable condition, which has no effect on the growth and development of plants. When it comes to the discovery of the species in the area, Lotus corniculatus var., Silene armeria L., Papaver rhoeas, Trifolium pratense L. developed and grew in spring after two years. And in the summer, Lotus corniculatus var., Dianthus chinensis L., Cosmos bipinnatus, Coreopsis drummondii L., Trifolium pratense were founded. And there were Lotus corniculatus var., Dianthus chinensis L., Aster yomena, Coreopsis tinctoria, Trifolium pratense L. in the autumn. According to the examination conducting after four years, Lotus corniculatus var., Dianthus chinensis L., Silene armeria, Cosmos bipinnatus, Coreopsis drummondii L., Papaver rhoeas, Trifolium pratense L. were founded in the spring, and Lotus corniculatus var., Dianthus chinensis L., Cosmos bipinnatus, Coreopsis drummondii L., Trifolium pratense grew in the summer. Lotus corniculatus var., Dianthus chinensis L., Astragalus sinicus, Aster yomena, Coreopsis tinctoria, Trifolium pratense L. were discovered in the autumn. Among the exotic species founded in the area, were Festuca arundinacea, Lolium perenne, Festuca rubra. When it comes to the protection of vegetation and dominance, Lotus corniculatus var., Metaplexis japonica, Coreopsis drummondii L. are the dominant species among the spring plants; and Lotus corniculatus var., Trifolium repens L., Erigeron annuus (L.) Pers. were the dominant plants in the summer. In the autumn, Lotus corniculatus var., Coreopsis drummondii L., Poa pratensis appear to be the dominant plants. The colonies of Pinus densiflora and Castanea crenata var. were formed around the South Sejong IC. Part of the area is occupied by the colony of Alnus hirsuta Turcz. ex Rupr. In terms of shrub, there were Indigofera pseudo-tinctoria, Robinia pseudoacacia, and Rosa polyantha var. genuina NAKA. As ground cover plants, Sasa borealis, Rubus crataegifolius Bunge were founded.

The Plant Succession of Improved Pasture Around Mt. Halla in Cheju Island (한라산 인공초지의 식물천이)

  • This study was conducted to examine the plant succession on the improved pasture of the mountain districts around Mt. Halla in Cheju Island. For this study, the researcher had investigated for ten years the improved pasture which had been used for grazing from 1976 to 1985. The pasture was reclaimed from native grassland. The mixed seeds of 17kgs' Dactylis glomerata, 7kgs' Festuca arundinacea, 2kgs' Lolium multiflorum and 2kgs' Trifolium repens were sowed per ha. The results of this study are the followings; The annual changes in the number of plant species were observed. 37 weed species were found in 1976 and increased year after year to 151 species in 1985. The changes in the distribution of annual and perennial plant, and one species of arbor were found in 1976 and increased respectively year by year to 56 species (annual plant), 95 species (perennial plant) and 9 species (arbor) in 1985. The average plant height of introduced grasses by year increased from 38.05cm (1976) to 47.30cm (1978) and decreased from 40.50cm (1979) to 10.36cm in 1985 (y =-0.501x$^2$+1.609x + 41.946). While the average plant height of invading weeds increased from 26.61cm to 42.84cm (y=-0.80$\chi$$^2$+2.540$\chi$+27.570) between 1976 and 1985. The density of introduced grasses was 70.90% in 1976 and was reduced to 0.49% in 1985 (y =-0.501$\chi$$^2$+1.609$\chi$+41.946); while that of introduing weeds was 29.10% in 1976 and was increased to 99.51% in 1985 (y=-0.080$\chi$$^2$+2.540$\chi$+27.570). The coverage of introduced grasses by year increased gradually from 72.8% (1976) to 74.86% (1978) and decreased from 43.01% (1979) to 1.21% in 1986 while that of intruding weeds developed a tendency to increase every year. Their coverage in 1976 was 22.09% and increased to 98.78% in 1985. The weight of introduced grasses by year increased from 2,808kg (1976) to 3,535kg (1978) per l0a and after 1979 decreased gradually from 2,326kg (1978) to 35kg per l0a in 1985. That of intruding weeds increased yearly from 308kg in 1976 to 3,178kg in 1985. The type of annual vegetation were changed as follows; Year Type 1976-1978 Dactylis glomerata / Trifolium repens type 1979 Trifolium repens / Imperata cylindrica type 1980-1982 Imperata cylindrica / Zoysia Japonica type 1983 Imperata cylindrica / Pteridium aquilinum type 1984-1985 Imperata cylindrica / Miscanthus sinensis type The plants whose plant height, coverage, density, and weight increased year after year were Imperate cylind-rica, Zoysia japonica, Pteridium equilinum, Miscanthus sinensis, Cirsium japonicum, Erigeron canadensis, Artemisia japonica, Lespedeza cuneata, Spondiopogon cotuUfer. Cymbopogon tortilis, Plantago asiatica, Rumex acetosella, etc. The vegetation of Digitaria sanguinalis, Hydrocotyl japonica, Artemisia asiatica, etc. was com-paratively remarkable in the beginning.

Studies on White Clover Yield Increase by Antagonistic Bacteria (길항미상물에 의한 White Clover ( Trifolium repens L. ) 생산성 향상에 관한 연구)

  • This study was conducted to investigate the effect of antagonistic bacteria and pathogenic fungi on growth and yields of white clover(Trifo1iurn repens L.) in continuous cropping soil(CCS) and non-continuous cropping soil(NCCS). The gowth experiment of white clover was conducted at pots in a vinyl house. White clover was established by seeding into pots of 12 cm in diameter and 9 cm in depth containing 1:l mixture of soil and vermiculite with antagonistic bacteria and pathogenic fungi. In dark culture experiment, white clover lived longer in treatment of antagonistic bacteria than in treatment of control, but lived shorter in CCS than in NCCS. Dry weight of white clover was increased by the inoculation of the antagonistic bacteria(p< 0.05), but decreased by the inoculation of the pathogenic fungi(p< 0.05) both CCS and NCCS. In conclusion, bacterization of white clover with antagonistic bacteria enhances the growth and yield of white clover.
