  • Organic solvent tolerant bacterium, Pseudomonas savastanoi BCNU 106 could utilize trichloroacetic acid, monochloroacetic acid, trichloroethylene, p-dichlorobenzene as a sole carbon source. But Pseudomonas savastanoi BCNU 106 didn't have tolerance about trichloroacetic acid, monochloroacetic acid, trichloroethylene, p-dichlorobenzene. Strain BCNU 106 could utilize to the extend of 30 mM trichloroacetic acid as a sole carbon source on mineral salt medium.

The Relationship Between Airborne Trichloroetnylene Concentrations and Total Trichloro-compounds and Trichloroacetic Acid in Urine (근로자의 트리클로로에틸렌 폭로 농도와 요중 총삼염화물 및 삼염화초산 농도와의 관계)

  • This study was designed to establish the relationship between airborne trichloroethylene concentrations and total trichloro-compounds and trichloroacetic acid in urine samples of the trichloroethylene exposed workers, to examine if the biological screening value for trichloroacetic acid in urine set by the Ministry of Labor is appropriate, and to suggest a suitable biological screening value for total trichloro-compounds in urine. Seventy male workers from the cleaning, the packing, and the inspcetion areas were selected as the study group and eighty male office workers were chosen as the control group. The results were as follows: 1. The mean values of total trichloro-compounds and trichloroacetic acid in the exposed group ($48.1{\pm}1.5mg/{\ell}$, $19.7{\pm}1.9mg/{\ell}$) were significantly higher than those in the control group($4.3{\pm}1.5mg/{\ell}$, $1.8{\pm}1.2mg/{\ell}$). 2. The airborne tichloroethylene concentrations were significantly related with the concentrations of total trichloro-compounds in urine(r=0.8212) and the concentrations of trichloroacetic acid in urine(r=0.7216). 3. The average trichloroethylene concentrations in the manual cleaning plants and that in the automatic cleaning plants were 40.1 ppm and 7.7 ppm, respectively. The difference between two groups was statistically significant. 4. The geometric mean of 49.6 ppm trichloroethylene concentration was resulted in the $185.4mg/{\ell}$ total trichloro-compounds in urine, and the 50 ppm trichloroethylene concentration was expected to produce $170.4{\pm}28.5mg/{\ell}$ total trichloro-compounds in urine. 5. With the geometric mean of 49.6 ppm trichloroethylene concentration, the corresponding geometric mean concentration of trichloroacetic acid in urine was $74.7mg/{\ell}$. In conclusion, the level of personal exposure to trichloroethylene concentration was significantly correlated with the concentrations of total trichloro-compounds and trichloroacetic acid in urine. Current biological screening value of $75mg/{\ell}$ for trichloroacetic acid in urine set by the Ministry of Labor was thought to be appropriate, and a biological screening value for total trichloro-compounds in urine should be set in the range of $170.4{\pm}28.5mg/{\ell}$ as a reference value for trichoroethylene exposure.

  • Dichloroacetic acid (DCAA) and trichloroacetic acid (TCAA) in fresh-cut salads available from the market were determined by gas chromatography-mass spectrometry (GC-MS). The target compounds in 3 g of acidified homogenates were extracted with 20 mL of methyl t-butyl ether (MTBE). The extract was concentrated to 1 mL and heated for 1 h at $55^{\circ}C$. The analytes were separated using a DB-1701 column and detected with a mass spectrometer. The method detection limit was approximately $6{\mu}g/kg$, and both analytical accuracy and precision were found to be satisfactory. The linearity of the calibration curves expressed as the coefficients of determination was >0.996. The analysis of seven samples using the established method showed that the four samples contained considerable amounts of analytes ($25.4-31.2{\mu}g/kg$ of DCAA and $18.8-46.1{\mu}g/kg$ of TCAA). These results raised a concern about the impact of fresh-cut salad consumption on human health.

  • Based on the turbidimetric response of protein with 50% trichloroacetic acid (TCA), this study aims to introduce an assay method for protein in solution. The standard procedure consists of mixing equal volume of sample solution (standard or unknown) with 50%-TCA solution and measuring the absorbance at 450 nm after 20 min. The absorbances of the solutions were almost stable over 120 min at room temperature. This assy method is simple, reproducible, and tolerant to many interfering substances. It can detect less amount than $10\mu$g/ml of bovin serum albumin. The assay method has low protein-to-protein variability over wide range of molecular weight.

  • TCA is widely used in cosmetic treatments such as chemical peels, tattoo removal, and also in gynecological procedures for managing cervical intraepithelial neoplasia. However, storage of high concentration of TCA in a gynecology office is a potential hazard, as it may accidentally cause severe chemical burns. We report a case of deep dermal TCA chemical burn on genital area, which occured accidentally and resulted from misuse of high concentration (90%) of TCA coated vaginal gauze packing dressing during gynecological procedures.

Effect of Trichloroacetic Acid on the Solubility of Caseinomacropeptide

  • Crude caseinomacropeptide (CMP) was prepared from Na-caseinate using a commercial renneting enzyme. Most of the crude CMP was released from the Na-caseinate by hydrolyzing with the enzyme for 40 min. The hydrolysis of the k-casein with carbohydrate was slower than that of the k-casein without carbohydrate, as shown by the analyses of the sialic acid content and the tricine-SDS-polyacrylamide gel electrophoresis. The yield of crude CMP from Na-caseinate was 3.7%. Cation exchange chromatography showed that the crude CMP consisted of 40.5% CMP and 59.5% caseinogylcomacropetide (CGP). The effect of the TCA concentration on the solubility of CMP and CGP was determined by using crude CMP. The amounts of crude CMP and sialic acid decreased in the proportion to the increase of trichloroacetic acid (TCA) concentration from 2 to 12%, suggesting that the CGP containing carbohydrate, as well as the CMP having no carbohydrate, was precipitated in a range of 4 to 12%, depending on the TCA concentration. This result supports the hypothesis that the different non-glycosylated and glycosylated forms of CMP have different sensitivities to TCA precipitation.

  • The pyriform sinus fistula can cause a recurrent abscess in the neck and the current treatment of choice involves complete excision of the sinus tract. But, because of excisional difficult, chemical cautery has been intermittently used as a successful substitute. Recently we experienced a case of pyriform sinus fistula of 9 year-old female who was successfully treated with chemocauterization with trichloroacetic acid(TCA) and ligation of the internal opening of the fistula tract on suspension laryngoscopy. So we report this rate case with review of literatures.

  • Lupoid leishmaniasis is a unique form of cutaneous leishmaniasis characterized by unusual clinical features and a chronic relapsing course, mostly caused by infection with Leishmania tropica. In this clinical form, 1-2 yr after healing of the acute lesion, new papules and nodules appear at the margin of the remaining scar. Herein, we describe a case of this clinical form that was resistant to 2 courses of treatments: systemic glucantime and then a combination therapy with allopurinol and systemic glucantime. However, marked improvement was seen after a combination therapy with topical trichloroacetic acid solution (50%) and systemic glucantime, and there were no signs of recurrence after 1 yr of follow-up.

Extraction of Glycosaminoglycans from Styela clava Tunic (미더덕 껍질로부터 Glycosaminoglycans의 추출)

  • Glycosaminoglycans (GAGS was extracted from sea squirt, Styela clava with sodium phosphate at 105$^{\circ}C$ for 2 hr and deproteinized with trichloroacetic acid or hydrochloride. The GAGs obtained from tunic consist 41.7% crude carbohydrates, 31.8% crude protein, and 31.2% sulfate. It was mainly constituted of galactose, glucosamine, glucose, mannose, and glacrosamine. The prominent amino acid were phenylalanine, threonine, glutamic acid, and aspartic acid. Mineral contents was mainly constituted 3.0 mg% sodium, 1.6 mg% potassium, and 1.2 mg% phosphorus. Trichloroacetic acid, hydrochloride and 5-sulfosalicylic acid were used for deprotein of the GAGs. Effective volume for deprotein of crude GAGs were 5.0% trichloroacetic acid (w/v) and 10.0% HCI (v/v) treatment. The deproteinized GAGs contained 35.1%, 35.4% of protein and 22.0%, 18.5% of sulfate, respectively.

Analysis of the effect of trichloroacetic acid and epidermal growth factor release on cytoskeleton gene expression using the nano-controlled releasing system (나노방출제어시스템을 이용한 trichloroacetic acid와 epidermal growth factor 방출이 세포골격형성 유전자 발현에 미치는 영향 분석)

  • Purpose: Here, we verified that the actin cytoskeletal gene expression of human gingival fibroblasts was altered by the administration of trichloroacetic acid (TCA) and epidermal growth factor (EGF) using the nano-controlled releasing system. Materials and methods: The control and experimental groups were divided into 3 groups: the group with the TCA-only nano-controlled releasing system (EXP1), the group with the TCA- and EGF nano-controlled releasing system (EXP2), and the control group (CON) with 48-h incubation. Expression of 26 genes involved in the regulation of actin cytoskeleton were analyzed by real-time PCR followed by the determination of correlations and influential factors using the Pearson correlation analysis and multiple regression analysis. Results: Among 23 genes upregulated in EXP1 and EXP2, expression of 14 genes were significantly increased in EXP2 compared to EXP1. On the other hand, LPAR1 was downregulated only in EXP1, GNA13 was upregulated only in EXP2, and F2R was downregulated only in EXP2. Three Rac1-related genes and CDC42 were identified as the influential factors of the actin gene upregulation. Conclusion: The actin cytoskeleton genes in human gingival fibroblast were upregulated by the administration of TCA and EGF using HGC-based nano-controlled releasing system.
