Change of Fatty Acid Compositions during Hepatic Triacylglycerol Accumulation in Dietary Orotic Acid-induced Fatty Liver (오로토산 유발 지방산의 간장 트리아실글리세롤 축적간 지방산 조성의 변화)

  • Dietary orotic acid is known to induce the fatty liver. Fatty acid profiles in the lipid fraction of the liver and the serum in rats fed with or with orotic acid diet were analyzed. In all the hepatic lipid fraction of rats fed on the supplemented orotic acid diet, there was a significant increased in linoleic acid. In addition, linoleic acid was also increased in the triacylglycerol fraction of hepatic endoplasmic reticulum and the triacylglycerol and diacylglycerol fractions of hepatic Golgi apparatus of the orotic acid-feeding rats. In the time course study of the fatty acid profile in the hepatic triacylglycerol and diacylgycerol fractions, an increase of linoleic acid was observed similarly in the initial stages of orotic acid intake in the both fractions. However, linoleic acid in the serum triacylglycerol fraction of orotic acid-feeding rats increased from day 1, but it began to decrease the increment from day 2, resulting in the lower level of linoleic acid in the serum triacylglycerol fraction of orotic acid-feeding rats than that of rat fed a orotic acid-free diet after 10 days. Oleic acid (18:1) was increased in the only cholesteryl ester fraction of helpatic. However, oleic acid level in other fractions was not changed. The compositions of 14:0, 16:0 and 18:0 was reduced in the hepatic triacylogylcerol, diacylglycerol and cholesteryl ester fractions by orotic acid-feeding. However, these saturated fatty acids were significantly increased in the serum triacylglycerol fractions. The orotic acid indcued changes in linoleic acid level in hepatic triacylglycerol may be explained by the impaired fatty acid metabolism and limited excretion of this fatty acid from liver to serum.

Effects of Taurine and Orotic Acid on Lipid Concentration of Hepatic and Serum in Mouse (마우스의 간장과 혈청 지질농도에 미치는 타우린 및 오로트산의 영향)

  • Male mice (ddY strain) were fed with the chow diet containing 10% sucrose supplemented with orotic acid at the 1% level or/and taurine at the 5% levels for 14 days. The concentrations of triacylglycerol and cholesterol in the liver were significantly lower in the orotic acid group than the control group. When taurine and orotic acid were administered simultaneously, the concentrations of triacylglycerol and cholesterol in the liver were higher and lower, respectively, compared to the orotic acid group. The concentration of triacylglycerol in the serum was higher in the taurine group than that of the control or the orotic acid groups, and the simultaneous supplementation of orotic acid and taurine further enhanced. There were no significantly difference in body weight gain, diary food intake, and the concentrations of serum cholesterol and hepatic phospholipid. These results suggest that dietary taurine stimulated the increasion of hepatic triacylglycerol by orotic acid in mouse.

Effects of citrus aglycone flavonoids, hesperetin and naringenin, on triacylglycerol metabolism in hamsters fed with a cholesterol diet

  • Effects of hesperetin and naringenin on the concentration of triacylglycerol in the serum and liver were studied in male golden hamster fed with the semipurified diet containing at 1% level of them for 3 weeks. The concentration of triacylglycerol in serum of the naringenin group decreased by 31%, whereas that in liver increased by 37% compared to the control group. The concentration of triacylglycerol in the serum and liver of the hesperetin group was slightly lower than the control group. The activity of microsomal phosphatidate phosphohydrolase in the liver, which is a key enzyme for biosynthesis of triacylglycerol, was significantly inhibited in the hesperetin group, whereas it was not affected in the naringenin group. The effect of hesperetin on phosphatidate phosphohydrolase was also measured in vitro. Hesperetin decreased the activity of phosphatidate phosphohydrolase with a dose-dependent manner. Both naringenin and hesperetin did not statistically affect the daily food consumption, body weight, liver weight, and total cholesterol in the serum. The observation accounts for the hypotriglyceridemic effect of hesperetin in the hyperlipidemic hamster.

Effects of Rosa rwgosa Radix on Lipid Metabolism in Rats (흰쥐의 지질대사에 미치는 해당화 뿌리 추출물의 효과)

  • The feeding effects of Rosa rugosa roots powder extracted with two different extracting solvents on lipid metabolism in rats were evaluated. Rats were fed with purified diets containing methanol or 70% aqueous acetone extracts at a 1% level for 4 weeks. In addition, to examine the effects of age, rats with different ages (4weeks old vs. 3 months old) were fed with purified diets containing methanol extracts at a 1% level for 4weeks. Feeding of methanol extracts decreased the concentration of triacylglycerol in serum and liver, whereas the aqueous acetone extracts lowered the concentration of liver triacylglycerol with concomitant elevation of the concentration of serum triacylglycerol. The methanol extracts ameliorated the degree of increase in the serum triacylglycerol level in relation to age and decreased especially the concentration of free fatty acid in serum of adult rats. The results suggested possibility that the regulatory mechanism responsible for triacylglycerol-lowering effects of Rosa rugosa roots extracts in the liver could be modulated by differences in the components of extracts due to the variation in extracting solvents, and that the methanol extracts might be effective in lowering an age-dependent increase in the concentration of serum and liver triacylglycerol in rats.

Effects of ${\alpha}-linolenic$, eicosapentaenoic and docosahexaenoic acids administration on lowering of triacylglycerol level in the hepatic and serum of rats (n-3계 지방산 투여가 성장기 흰쥐의 간장 및 혈청 Triacylglycerol 농도에 미치는 영향)

  • We studied the difference effects of dietary ${\alpha}-linolenic\;acid\;({\alpha}-LA,\;18:3\;n-3)$, eicosapentaenoic acid (EPA, 20:5 n-3) and docosahexaenoic acid (DHA, 22:6 n-3) on the lowering of triacylglycerol in the liver and serum on lipid metabolism in rats. Rats were fed semipurified diets containing 10% fat with constant polyunsaturated/monounsaturated/saturated fatty acids (1:1:1) and n-6/n-3 ratio (1:2). EPA (98%) and DHA (98%) were added in diets as the ethyl esters. The concentration of liver triacylglycerol was significantly lower in rats fed both EPA and DHA than in those fed ${\alpha}-LA$. The concentration of liver phospholipid was significantly higher in rats fed DHA than in those fed ${\alpha}-LA$ and EPA. Both EPA and DHA reduced serum triacylglycerol concentration compared with ${\alpha}-LA$, but this effect was more pronounced in the EPA diet. The activity of phophatidate phosphohydrolase in the liver microsome was significantly lower in rats fed both EPA and DHA than in those fed ${\alpha}-LA$. but, there was no significant difference on the activities of diacylglycerol acyltransferase among the three groups. The concentration of liver triacylglycerol were correlated with changes in the microsomal phosphatidate phosphohydrolase activity (r=0.84). Hepatic NADPH generating enzyme, glucose-6-phosphate dehydrogenase was more effective to reduce the activity in rats fed both EPA and DHA than in those fed ${\alpha}-LA$. In conclusion, EPA or DHA reduced the hepatic triacylglycerol concentration by inhibiting microsomal phosphatidate phosphohydrolase, thereby inhibiting synthesis of triacylglycerol in the liver.

Expression Profiles of Triacylglycerol Biosynthesis Genes on Fattening Stages in Hanwoo (한우 비육기간 중 중성지방 생합성 관련 유전자의 발현양상)

  • Muscle fat content including intramuscular fat content(IMF) is the most important parameter in meat quality of cattle. Triacylglycerol is the major component of fat and is found in adipocyte and skeletal muscle. In present study, we carried out the determined of triacylglycerol contents and mRNA expression patterns of genes related with triacylglycerol biosynthesis such as ACSS2, GPAT, MGAT and DGAT in Hanwoo longissimus muscle using the real-time PCR. The triacylglycerol contents were continuously increased during the fattening stages. Interestingly, the contents of triacylglycerol were 7.4 fold higher(0.34 vs. 2.51, P<0.01) in 27 months old group than 12 months old group. The mRNA levels of ACSS2, GPAT and DGAT genes were also increased during fattening stages, whereas that of MGAT gene did not show difference among the stages. Thus, these results suggested that increasing of the triacylglycerol contents in longissimus muscle during fattening stages may be related with increased expressions of triacylglycerol biosynthesis genes(ACSS2, GPAT and DGAT). These results will be helpful to understand the mechanism of muscle fat deposition in skeletal muscle.

Effect of Schizandra chinensis Extract in Hyperlipidemic Rats (오미자 추출물이 고지혈증 흰쥐에 미치는 영향)

  • The purpose of this study was to investigate the effect of Schizandra chinensis extract on the serum and hepatic lipids inhyperlipidemic rats fed with high-fat diet. The Schizandra chinensis extract decreased significantly in total serum cholesterol, serum triacylglycerol and heptic triacylglycerol but not in the total hepatic cholesterol. The Schizandra chinensis extract also decreased in very low density lipoprotein increased by the highfat diet without affecting high density lipoprotein cholesterol. These results suggest that Schizandra chinensis extract may be benificial for the regulation of hyperlipidemia.

A Convergent Synthesis of Bis-2-oxo Amide Triacylglycerol Analogues as Potent Lipase Inhibitors Using Acyl Cyanophosphorane Methodology

  • A number of bis-2-oxo amide triacylglycerol analogues, a recently reported potent human gastric lipase inhibitor and its new analogues, have been prepared starting from 1,3-dibromo-2-propanol utilizing acyl cyanophosphorane methodology as a key step in a convergent manner. The key coupling reaction has been accomplished at -$78^{\circ}C$ between 1,3-diamino-2-propanol derivative and the labile diketo nitriles, derived from acyl cyanotriphenylphosphoranes upon oxidizing with $O_3$, under mild condition in moderate yields.

Effects of Hesperetin and Naringenin on Lipid Concentration in Orotic Acid Treated Mice (오르트산 투여 마우스의 지질농도에 미치는 Hesperetin 및 Naringenin의 영향)

  • Male mice (ddY strain) were fed a laboratory chow diet containing 10% sucrose supplemented with orotic acid, hesperetin or naringenin at the 1% level for 14 days. the concentrations of liver triacylglycerol and cholesterol were significantly lower in the OA group than in the control group. When both flavonoids and orotic acid were administered simultaneously, the orotic acid-dependent decrease in liver triacylglycerol and cholesterol were attenuated slightly. The concentration of serum cholesterol in the orotic acid group or the control group was lower than in the orotic acid groups supplemented with hesperetin or naringenin. There were no significantly difference in body weight gain, diary food intake, and the serum concentrations of triacylglycerol and high-density-lipoprotein cholestrol. It was concluded that the inducement of fatty liver in mice failed to feeding a laboratory chow diet containing 10% sucrose supplemented with 1% orotic acid for 14 days.

  • The effects of the prevention of micelle formation and bile acid reabsorption, by using cholestyramine (CHOLN), a bile acid binding polymer, on the plasma lipid of Single Comb White Leghorn male chicks given diets containing medium chain triacylglycerol (MCT) and long chain triacylglycerol (LCT) were investigated. Corn oil and glyceryl tricaprylate were used as LCT and MCT sources, respectively. Plasma HDL cholesterol was reduced by CHOLN in all treatments. Plasma LDL cholesterol was reduced by CHOLN in chicks given LCT diet but not in MCT diet which could be accounted to the reduced plasma total cholesterol in LCT diet with CHOLN. It is concluded that bile acid binding does not alter the cholesteremic effect of MCT in the plasma of chicks.
