Effect of Copper Source (Cupric Citrate vs Cupric Sulfate) and Level on Growth Performance and Copper Metabolism in Pigs

  • Two experiments were conducted to evaluate the efficacy of cupric citrate (Cu-citrate) relative to cupric sulfate $(CuSO_4)$ as a Cu source for weanling and grow-finish pigs. In addition, the use of liver and bile Cu concentrations as indices of the bioavailability of Cu sources was investigated. Experiment one consisted of a nursery phase (35 d; initial BW=6.4 kg, final BW=21.4 kg) followed by a grow-finish phase (103 d; initial BW=21.5 kg, final BW=111.7 kg). Experiment two only consisted of a nursery phase (35 d; initial BW=6.3 kg, final BW=18.6 kg). Dietary treatments were identical for both experiments and consisted of: control (10 ppm $CuSO_4$); control+66 or 225 ppm $CuSO_4$; control+33, 66, or 100 ppm Cu-citrate. An antibiotic was included in diets for Exp. 1 but not Exp. 2. In both experiments, growth performance variables were similar for pigs receiving Cu-citrate and $CuSO_4$; however, growth performance was not improved by high concentrations of $CuSO_4$. Liver and bile Cu were increased (p<0.05) by 225 ppm $CuSO_4$; however, lower dietary concentrations of Cu from either $CuSO_4$ or Cu-citrate did not affect the Cu concentration of liver or bile relative to that observed in the control pigs. Irrespective of Cu source, there was no linear (p>0.10) increase in plasma Cu with increasing Cu concentrations in the diet for both experiments. However, the plasma Cu concentrations were highest (p<0.10) in pigs receiving diets supplemented with 225 ppm $CuSO_4$. Sixteen randomly chosen pigs per treatment in Exp. 1 were continued through the grow-finish phase. Body weight gain and feed intake were improved (p<0.10) by 66 ppm $CuSO_4$, but other dietary Cu treatments did not alter pig performance compared to the control diet. Plasma Cu concentrations were increased (p<0.10) by 225 ppm $CuSO_4$ in the growing phase and by 225 ppm $CuSO_4$ and 100 ppm Cu-citrate in the finishing phase. These data reveal no consistent effect of $CuSO_4$ on performance; therefore, it is difficult to assess the efficacy of these two Cu sources. In addition, these studies demonstrate that liver and bile Cu are not good indicators of Cu bioavailability in pigs fed adequate to pharmacological concentrations of Cu.

Royal jelly enhances migration of human dermal fibroblasts and alters the levels of cholesterol and sphinganine in an in vitro wound healing model

  • Oral administration of royal jelly (RJ) promotes wound healing in diabetic mice. Concerns have arisen regarding the efficacy of RJ on the wound healing process of normal skin cells. In this study, a wound was created by scratching normal human dermal fibroblasts, one of the major cells involved in the wound healing process. The area was promptly treated with RJ at varying concentrations of 0.1, 1.0, or 5 mg/ml for up to 48 hrs and migration was analyzed by evaluating closure of the wound margins. Furthermore, altered levels of lipids, which were recently reported to participate in the wound healing process, were analyzed by HPTLC and HPLC. Migration of fibroblasts peaked at 24 hrs after wounding. RJ treatment significantly accelerated the migration of fibroblasts in a dose-dependent manner at 8 hrs. Although RJ also accelerated the migration of fibroblasts at both 20 hrs and 24 hrs after wounding, the efficacy was less potent than at 8 hrs. Among various lipid classes within fibroblasts, the level of cholesterol was significantly decreased at 8 hrs following administration of both 0.1 ug/ml and 5 mg/ml RJ. Despite a dose-dependent increase in sphinganines, the levels of sphingosines, ceramides, and glucosylceramides were not altered with any concentration of RJ. We demonstrated that RJ enhances the migration of fibroblasts and alters the levels of various lipids involved in the wound healing process.

Clinical Trial to Evaluate the Efficacy of Extract of Citri Pericarpium on Serum Lipid Profiles in Subjects: a Randomized, Double-blind (혈중지질에 대한 진피추출물의 유효성 평가를 위한 인체적용시험 : 무작위배정, 이중맹검)

  • Objectives : This study was performed to evaluate the efficacy of extract from Citri Pericarpium (CP) on lipid-lowering effect in semi-healthy subjects with increased serum levels of total cholesterol and triglyceride. Methods : A experimental group (n=45) and placebo group (n=46) were randomly established based on the plasma lipid profiles (triglyceride, total cholesterol etc.) in the semi-healthy subjects, then all subjects randomly took experimental or placebo capsules, which contain 1,200 mg of CP or the same dose of soluble starch per day with regular meals for 8 weeks by double-blind method. Results : One-hundred forty (140) subjects were screened, then 91 subjects enrolled in the study. No serious adverse events were reported for trial period. The CP treatment showed statistically significant decrease in serum levels of triglyceride both at 4-weeks (p<0.01) and at 8-weeks (p=0.0001). Futhermore, there was significant difference in the triglyceride levels between CP-treated and placebo group. The CP-treated but not placebo group, experienced decrease in serum levels of total cholesterol at 8-weeks. Then there was no significant difference in the total cholesterol levels between the two groups. In addition, statistically significant difference of AST, ALT, ${\gamma}$-GT serum levels wasn't shown before and after trial in two groups. Conclusion : These results demonstrate that CP can significantly suppress the elevated triglyceride but not total cholesterol level. Therefore, CP may play a role in the improvement of blood lipid levels, as an effective functional food.

Efficacy of Low Dose Barbiturate Coma Therapy for the Patients with Intractable Intracranial Hypertension Using the $Bispectral^{TM}$ Index Monitoring

  • Objective : Barbiturate coma therapy (BCT) is a useful method to control increased intracranial pressure (IICP) patients. However, the complications such as hypotension and hypokalemia have caused conditions that stopped BCT early. The complications of low dose BCT with $Bispectral^{TM}$ index (BIS) monitoring and those of high dose BCT without BIS monitoring have been compared to evaluate the efficacy of low dose BCT with BIS monitoring. Methods : We analyzed 39 patients with high dose BCT group (21 patients) and low dose BCT group (18 patients). Because BIS value of 40-60 is general anesthesia score, we have adjusted the target dose of thiopental to maintain the BIS score of 40-60. Therefore, dose of thiopental was kept 1.3 to 2.6 mg/kg/hour during low dose BCT. However, high dose BCT consisted of 5 mg/kg/hour without BIS monitoing. Results : The protocol of BCT was successful in 72.2% and 38.1% of low dose and high dose BCT groups, respectively. The complications such as QT prolongation, hypotension and cardiac arrest have caused conditions that stopped BCT early. Hypokalemia showed the highest incidence rate in complications of both BCT. The descent in potassium level were $0.63{\pm}0.26$ in low dose group, and $1.31{\pm}0.48$ in high dose group. The treatment durations were $4.89{\pm}1.68$ days and $3.38{\pm}1.24$ days in low dose BCT and high dose BCT, respectively. Conclusion : It was proved that low dose BCT showed less severe complications than high dose BCT. Low dose BCT with BIS monitoring provided enough duration of BCT possible to control ICP.

The efficacy and safety of Montelukast sodium in the prevention of bronchopulmonary dysplasia

  • Purpose: The purpose of this study was to evaluate the efficacy and safety of Montelukast sodium in the prevention of bronchopulmonarydysplasia (BPD). Methods: The Interventional study was designed as a multicenter, prospective, and randomized trial, with open labeled and parallel-experimental groups, 66 infants were enrolled and allocated to either the case group (n=30) or the control group (n=36) based on gestational age (GA). Infants in the case group were given Montelukast sodium (Singulair) based on their body weight (BW). Zero week was defined as the start time of the study. Results: The incidence of moderate to severe BPD was not different between the groups (case group: 13 of 30 [43.3%] vs. control group: 19 of 36 [52.8%], P=0.912). Additionally, secondary outcomes such as ventilation index, mean airway pressure and resort to systemic steroids were not significantly different. There were no serious adverse drug reactions in either group, and furthermore the rate of occurrence of mild drug related-events were not significantly different (case group: 10 of 42 [23.8%] vs. control group: 6 of 48 (15.8%), P=0.414). Conclusion: Montelukast was not effective in reducing moderate or severe BPD. There were no significant adverse drug events associated with Montelukast treatment.

Insecticidal activity of mixed formulation with buprofezin and single formulation without buprofezin against citrus mealbug, Planococcus citri Risso (Hemiptera: Pseudococcidae) (귤가루깍지벌레에 대한 단제 및 buprofezin 혼합제의 살충 활성)

  • Nineteen registered insecticides including 8 mixed formulations with buprofezin and 11 single formulations which is not including buprofezin were evaluated by spray application for their toxicity against adult and nymph of citrus mealybug, Planococcus citri. Five mixed formulations with buprofezin such as buprofezin 15EC+acetamiprid 4EC, buprofezin 10SC+clothianidin 3SC, buprofezin 20WP+dinotefuran 15WP, buprofezin 20SC+thiacloprid 5SC, and buprofezin 20SC+thiamethoxam 3.3SC showed high insecticidal activity (>93%) against nymph and adult of P. citri. Insecticidal activities of EPN 45EC, fenitrothion 50EC and methidathion 40EC in organophosphorous group showed 90 to 93% against nymph only. In addition to, insecticidal activities of acetamiprid 8WP, clothianidin 8SC, dinotefuran 20WG and thiamethoxam 10WG in neonicotinoids group showed above 90% against nymph only. In systemic and residual effect, five mixed formulations that was already proved to have high insecticidal activity showed low toxicity with below 60% against $3^{rd}$ instar nymph of P. citri in tomato and rose under greenhouse. Control efficacy of five mixed formulations with buprofezin was above 90%, 80% and 70% at 5 days after treatment (DAT), 10 DAT and 15 DAT, respectively.

Disease Control Efficacy of the Extract of Magnolia officinalis against Perilla and Zoysiagrass Rusts (후박 추출물의 들깨 녹병과 잔디 녹병에 대한 방제 효과)

  • Rusts are plant diseases caused by pathogenic fungi of the order Pucciniales. Rusts can affect a variety of plants. Development of new effective and environmentally benign agents against rusts is of great interest. In this study, we investigated the disease control efficacy of the emulsion concentrate (EC10) and wettable powder (WP20) type formulations of the extract of Magnolia officinalis (Magjia90; containing honokiol and magnolol at 90%) against rust diseases of perilla and zoysiagrass in fields. The treatment of EC10 and WP20 of Magjia90 showed control values of 47.9% to 69.6% and Magjia90-WP20 reduced more effectively the development of rust symptoms on perilla plants than Magjia90-EC10. Magjia90-WP20 also highly suppressed zoysiagrass rust with control values of 65.7% to 80.5%. On the other hand, no harmful effect of Magjia90-EC10 and Magjia90-WP20 was observed on the perilla and zoysiagrass plants tested. The results strongly indicate that the extract of M. officinalis (Magjia90) can be used as a natural fungicide for the control of rust diseases.

Analysis of Medicinal Ingredients that Compose Liu Hejian's Formulae (류하간(劉河間)의 처방 구성 약재에 대한 연구)

  • Liu Hejian maintained that the six types of energy can become fire and focused on the treatment of fire and heat symptoms along with the prescriptions of cryotherapy, believing that a vast majority of disease are due to fire. In this study, we investigated the characteristics of Hejian's formulae by classifying medicinal ingredients in consideration of medicinal properties and efficacy in order to examine how many cold and hot medicinal ingredients are included in the formulae of Hejian. We analysis 848 formulae among 1,055 formulae in total, excluding 207 redundant ones, which includes 366 Sunmyunglon formulae, 223 Bomyeongjip formulae, 8 Samsolon formulae, 61 Sanghanjigkyeok formulae, 68 Sanghanpyobon formulae, 22 Sanghansimyo formulae, 103 Hwalbeopgiyo formulae, 186 Bodongbiyo formulae, and 18 Gagamyoungbisippalbang formulae. In the analysis of the frequency of using each medicinal herb after classifying medicinal ingredients of Hejian's formulae by medicinal properties, we found that cold medicinal ingredients accounted for 40%, the biggest share in the formulae, and hot medicinal ingredients accounted for around 45%, which indicates that Hejian's formulae consist of both elimination and reinforcement and cold and heat, rather than cold medicinal ingredients only. As a comparison of the frequency of using each medicinal ingredients for the formulae by efficacy, we found that clear heat medicinal ingredients, totify qi and replenish blood medicinal herbs, and activate blood medicinal ingredients were most frequently used.

Ecological Characteristics of Bacteriophages Infecting Xanthomonas oryzae pv. oryzae and Their Use as Biocontrol Agents (벼 흰잎마름병균 파지의 생태학적 특성 및 이를 이용한 생물방제)

  • Bacterial leaf blight (BLB) caused by Xanthomonas oryzae pv. oryzae (Xoo) is a very serious disease in rice growing regions of the world. There are no effective ways of protecting rice from the disease. In this study, the bacteriophage (phage) mixtures infecting Xoo were investigated as biological control agent on BLB. The effects of pH, heat and ultraviolet on the stability of phages were investigated to check and increase the possibility of practical use in the field. Phages were rather stable between pH 5 and pH 10. The infectivity dropped sharply when the phages were incubated at $50^{\circ}C$ and more than 90% of the phages were inactivated after two minutes of ultraviolet treatment. The phages were stable for 7 days at the rice plant leaves, and the phages survived 10 times more than other treatments when mixed with skim milk. Although the skim milk increased the stability of the phages, the control efficacy was not effective. However, the phage mixtures reduced the occurrence of BLB when they were treated with Tecloftalam WP or Acibenzolar-S-methyl simultaneously. The results indicated that the Xoo phages could be used as an alternative control method to increase the control efficacy and reduce the use of agrochemicals.

The Efficacy of Lowering Blood Glucose Levels Using the Extracts of Fermented Bitter Melon in the Diabetic Mice (당뇨 마우스에서 여주발효추출물의 혈당 강하 효능)

  • Momordica charantia, commonly known as bitter melon, has interesting pharmacological activities such as anticancer, antiviral, antibacterial, anti-inflammatory, analgesic, and antioxidant. As supported by recent scientific reports on the beneficial effects of M. charantia, it is one of the most promising functional plants for diabetes today. In this study, we fermented the bitter melon with lactic acid bacteria and investigated the capability of controlling diabetic conditions by decreasing the blood glucose levels. After extracting the fermented bitter melon with hot water or ethanol, we tested several biological activities using mouse models. When we tested the efficacy of the glycemic control, the extracts of fermented bitter melon significantly lowered the blood glucose levels of the alloxan-induced diabetic mice. We also found that the lactic acid bacteria-fermented bitter melon protected liver damages from the treatment of alloxan monohydrates and maintained low levels of triglycerides and high levels of HDL cholesterol in these mouse models. These results suggest that our approach on fermenting bitter melon and the extracts of fermented bitter melon could lead to the possibility of the development of functional foods that contain the effectiveness of controlling blood glucose and lipid levels as well as preventing liver damages.
