Upper gastrointestinal contrast studies were performed to compare the quality of images using two different commercialized barium products in 10 healthy beagle dogs of each group; 30% barium suspension in group 1, and 35% dilute barium suspension in group 2. Contrast media were administered through gastric tube at a dose rate of 10 ml/kg in all groups. All procedures were evaluated by three criteria: the quality of the images obtained, the transit time and the distensibility of the bowel. The imaging in group 1 showed good image quality, a rapid transit time, and good distensibility of bowel loop.
This study sought to investigate any additive or interactive effects that soy isoflavones may have on the ecosystem of the gut, which is influenced by fructooligosaccharide (FOS) in colon-cancer model rats. Male Sprague-Dawley rats treated with 1,2-dimethylhydrazine were given experimental diets containing 0, 3, 6, or 9% FOS with or without 0.1% soy isoflavone for 12 weeks. In addition to the effects of FOS dosage on the gut ecosystem, dietary supplementation with soy isoflavone reduced the number of colonic aberrant crypts (ACs). The fecal weight, fecal pH, and gut transit time significantly decreased in a dose-dependent manner in rats fed FOS and the fecal concentration of bifidobacteria was higher in rats fed FOS than in control rats. The fecal output of total short-chain fatty acids, acetate, and propionate was significantly increased by the presence of FOS and was negatively correlated with the number of ACs, whereas the fecal output of butyrate showed no significant correlation with FOS dosage. The addition of soy isoflavone to the diet did not result in any significant differences in gut ecosystem parameters. Therefore, we conclude that the suppressive effect of soy isoflavone on ACs was not associated with the intestinal ecosystem, which was significantly altered by the dosage of FOS.
Background: The purpose of this study was to assess the the efficacy of oral glutamine (GLN) in prevention of acute radiation-induced esophagitis in patients with lung cancer and determine the predictive role of clinical and dosimetric parameters. Materials and Methods: Thirty-two patients diagnosed with lung cancer were studied prospectively. Sixteen patients (50%) received prophylactic powdered GLN orally in doses of 10g/8h. Patients were treated 2 Gy per fraction daily, 5 days a week. We evaluated the grading of esophagitis daily at the end of each fraction of each treatment day until a cumulative dose of 50 Gy was reached. The primary end point was radiation-induced esophagitis. Results: All patients tolerated GLN well. Toxicity grade, weight loss, serum cytokine levels and esophageal transit times exhibited statistically significant improvement in the GLN receiving group. GLN suppressed the inflammation related to the disease and treatment and reduced toxicity with statistical significance. Conclusions: This study suggests a benefical role of oral GLN use in prevention and/or delay of radiation-induced esophagitis, in terms of esophageal transit time and serum immunological parameters, as well as weight loss.
This study investigated the effects of high amylose starch (HAS) consumption on gut functions in male Sprague-Dawley rats. Experimental animals were fed an diet containing HAS for 4 weeks (0, 125, 250, 500 g/kg diet). Stool weights, transit time, the pH of cecum, Bifidobacterium growth, short chain fatty acid production, and prostaglandin E$_2$production in colon mucus were measured. HAS intake did not affect body weight gain or food efficiency ratio during experimental period. There were no significant differences in kidney weight, epididymal fat pad weights or spleen weights, but the weights of the liver and thymus were significantly lower in the HAS100 group. The length of the large intestine, the weights of the cecum wall and cecum contents, and stool weights significantly increased through HAS intake. But transit time was not affected by the experimental diet. Although Bifidobacterium growth in the cecum increased through the HAS intake dose dependently, there were significant differences in the HAS50 and HAS100 groups. HAS intake increased the production of short chain fatty acid in the cecum contents. In particular, acetate and butyrate concentrations grew significantly. And the production of prostaglandin E$_2$in the colon mucus significantly decreased through HAS intake. These results demonstrate that high amylose starch intake significantly improves gut function.
The dried rhizomes and roots of Gentiana scabra (GS) have been frequently prescribed as a traditional folk medicine in East Asia (including China, Japan, and Korea) for the treatment of various pathophy-siological conditions, such as inflammatory skin diseases, anorexia, indigestion, gastric infections, and diabetes mellitus. In the present study, the effects of aqueous (GS-W) and ethanolic (GS-E) extracts of GS on gastrointestinal (GI) motor function were investigated by measuring the in vivo gastric emptying rate (GER) and the intestinal transit rate (ITR) in mice. The GER was significantly increased by GS-W at a dose of 1 g/kg. The ITR was significantly increased by GS-W (at doses of 0.1 and 1 g/kg) or GS-E (at a dose of 1 g/kg) in a dose-dependent manner. Furthermore, the ITR value of GS-W (at a dose of 1 g/kg) appeared to be higher than that of cisapride, which was the most prominent prokinetic agent in the 1900s but was removed from the market in 2000 due to its fatal side effects. The above results suggest that GS-W might be a potential prokinetic agent to replace cisapride.

Background/Aims Functional dyspepsia (FD) and irritable bowel syndrome (IBS) are common gastrointestinal (GI) disorders and these patients frequently overlap. Trimebutine has been known to be effective in controlling FD co-existing diarrhea-dominant IBS, however its effect on overlap syndrome (OS) patients has not been reported. Therefore, we investigated the effect of trimebutine on the model of OS in guinea pigs. Methods Male guinea pigs were used to evaluate the effects of trimebutine in corticotropin-releasing factor (CRF) induced OS model. Different doses (3, 10, and 30 mg/kg) of trimebutine were administered orally and incubated for 1 hour. The next treatment of $10{\mu}g/kg$ of CRF was intraperitoneally injected and stabilized for 30 minutes. Subsequently, intragastric 3 mL charcoal mix was administered, incubated for 10 minutes and the upper GI transit analyzed. Colonic transits were assessed after the same order and concentrations of trimebutine and CRF treatment by fecal pellet output assay. Results Different concentrations (1, 3, and $10{\mu}g/kg$) of rat/human CRF peptides was tested to establish the OS model in guinea pigs. CRF $10{\mu}g/kg$ was the most effective dose in the experimental OS model of guinea pigs. Trimebutine (3, 10, and 30 mg/kg) treatment significantly reversed the upper and lower GI transit of CRF induced OS model. Trimebutine significantly increased upper GI transit while it reduced fecal pellet output in the CRF induced OS model. Conclusions Trimebutine has been demonstrated to be effective on both upper and lower GI motor function in peripheral CRF induced OS model. Therefore, trimebutine might be an effective drug for the treatment of OS between FD and IBS patients.
The aim of this study is to evaluate a efficient w/v% barium suspension and w/v% sodium carboxy methyl cellulose (SCMC) for small bowel examination. Between november 2001 and june 2002, 370 patients were examined small bowel examination with oral administration of barium suspension mixed with SCMC. we classified into six groups including A(fine type 40% $BaSO_4$ mixed 0.5% SCMC and 600 ml 0.5% SCMC administration), B(fine type 30% $BaSO_4$ mixed 0.5% SCMC), C(fine type 20% $BaSO_4$ mixed 0.5% SCMC), D(fine type 25% $BaSO_4$ mixed 0.5% SCMC), E(D mixed coarse type 25% $BaSO_4$ mixed 0.5% SCMC), F(D mixed coarse type 25% $BaSO_4$ mixed 0.75% SCMC). We measured transparency rate of contrast media transit time, administration dose, viscosity and particle size. The transparency rate was higher then 80 kV and 90 kV in 100 kV in the same 20 mAs condition. Transit time was the fastest in A group(mean trans time 56 minutes) and the slowest in E group(mean 100 minutes), Administration dose was the smallest in A group(mean dose 541 ml and the most in E group(mean 1,100 ml), viscosity was the lowest in E(125.1 mpa/s) and the highest in A(375.5 mpa/s), and particle size was $1.0\;{\mu}m$ in A, B, C, and D group, $0.6\;{\mu}m,\;1.0\;{\mu}m,\;10.0\;{\mu}m$ in E, F group. In conclusion, we propose that the efficient condition for small examination is high voltage technique, high density $BaSO_4$, and 0.625% w/v SCMC.

Although major advances have been achieved in our understanding and treatment of diseases in many areas of medicine, relatively few improvements have been made in the area of gastrointestinal (GI) motor function. The dried root of Codonopsis pilosula (Franch.) Nannf. (CP) has been used as a traditional folk medicine for improving poor GI function in East Asia, including China and Korea. In the present study, neither aqueous (CP-W) nor ethanolic (CP-E) extracts of CP showed significant toxicity, even at an oral dose of 5 g/kg to mice. The effects of CP-W and CP-E on GI motor function were investigated by measuring in vivo the gastric emptying rate (GER) and intestinal transit rate (ITR) in mice. In normal mice, the ITR was significantly increased by CP-W in a dose-dependent manner, whereas the GER was not significantly affected by any CP extracts. The ITR was significantly retarded in the mice with experimental GI motility dysfunction (i.e., peritoneal irritation by acetic acid) compared with that in normal mice. However, the retardation was significantly recovered by the pre-treatment of CP-W in a dose-dependent manner. The above results suggest that CP-W might be a potential prokinetic agent preventing or alleviating GI motility dysfunctions in human patients.
Dried Citrus unshiu peels (Aurantii Nobilis Pericarpium; ANP) are used as a traditional folk medicine for the treatment of gastrointestinal (GI) motility disorders in East Asia, including Korea. In the present study, an ethanolic extract of ANP (ANP-E) exhibited no significant toxicity in mice, even at an oral dose of 5 g/kg. The effects of ANP-E on GI motor function were investigated by measuring the intestinal transit rate (ITR) of Evans blue in normal mice and mice with experimental GI motility dysfunction (i.e., peritoneal irritation by acetic acid; PIA). In normal mice, ANP-E significantly increased the ITR in a dose-dependent manner. The ITR in the PIA mice was significantly retarded compared to that in the normal mice. However, ANP-E significantly inhibited this retardation in a dose-dependent manner. Furthermore, in all the models, the potency of ANP-E appeared to be same or higher than that of cisapride, which was used predominantly for the treatment of various GI motility disorders in humans in the 1900s but was removed from the market in 2000 due to fatal side effects. The results suggest that ANP-E has potential as a new prokinetic agent that could be used as a substitute for cisapride.
Objectives: The purpose of this study was to investigate the effects of Socheongryong-tang, a traditional herbal medicine, on gastrointestinal (GI) motility disorders (inflammatory and diabetic models) in mice. Methods: The in vivo effects of Socheongryong-tang on GI motility were investigated by calculating percent intestinal transit rates (ITR) and gastric emptying (GE) values using Evans Blue and phenol red, respectively, in normal mice and in mice with experimentally induced GI motility dysfunction (GMD). GMD models were induced by acetic acid and streptozotocin. Results: In normal ICR mice, ITR and GE values were significantly and dose-dependently increased by the intragastric administration of Socheongryong-tang (0.0025~0.25 g/kg). The ITR values (%) of GMD mice were significantly lower than those of normal mice, and these reductions were significantly and dose-dependently inhibited by Socheongryong-tang. Conclusions: These results suggest that Socheongryong-tang is a good candidate for the development of a prokinetic agent that may prevent or alleviate GMD.
