운향과 식물의 과실 유래 한약재들이 마우스의 위 배출 속도 및 장 이송률에 미치는 영향 비교 (Comparison of Effects of Oriental Medicines from Rutaceae Fruits on Gastric Emptying and Intestinal Transit in Mice)

  • 운향과(Rutaceae) 식물의 과실을 소재로 하는 일군의 한약재들은 위장관 운동 기능 항진을 목적으로 우리나라와 중국 등지에서 오래 전부터 널리 사용되어 왔으며, 대표적인 약재로 각각 탱자나무 및 광귤나무의 미성숙 과실인 지실 및 지각, 각각 광귤나무 및 귤나무의 성숙 과피인 등피 및 진피 등이 있다. 본 연구에서는, 이러한 운향과 과실 유래 한약재들이 위장관 운동 기능에 미치는 영향을 검정하고 이 약재들 간의 효능을 비교하기 위해, 이들 한약재의 열수 및 에탄올 추출물을 제조하여 마우스에 경구 투여한 후 위 배출 속도 및 장 이송률을 측정하는 실험을 수행하였다. 본 연구는, 공통 성분을 다수 함유하는 운향과 과실 유래 한약재들 간의 위장관 운동 기능개선 효과를 in vivo 동물 실험을 통해 직접 비교한 최초의 연구 자료로서 의미를 가진다. 특히, 등피 에탄올 추출물 투여 시 위 배출 속도에는 유의적인 변화 없이 장 이송률의 유의적 및 용량 의존적 증가 양상을 보였으며, 이전 연구들을 통해 장 이송률 증가 효능이 보고된 바 있는 지실, 지각, 진피 열수 추출물 투여군들과 비교했을 때에도 수치 상으로 등피 에탄올 추출물의 장 이송률이 가장 높았다.

  • This work was on the reduction of exposure dose and contrast improvement by Use of Heavy Elements Filter From the result of experimental evaluation, it was found that the beam harding of X-ray was not showed in Ho and Gd, heavy elements filters, contrast to Cu and Al filters In which the harding showed. And the ratio of transit dose to surface dose and the load of X-ray tube increase in order of Al, Cu, Gd and Ho, respectively. The contrast of X-ray images using the intensifying screen and the input phosphor showed the higher value in order of Cu, Al, Gd and Ho. Therefore, in the case of using contrast media and phosphor in region of diagnostic radiology, X-ray image quality depends primarily on kVp and heavy elements filters.

  • 본 연구의 목적은, X선조영촬영을 이용한 콘크리트강도의 평가 및 콘크리트열화의 수치화를 수행하는 것이다. 콘크리트강도를 추정하기 위하여, 물-시멘트비가 서로 다른 콘크리트를 제작하여, X선투과선량과 콘크리트강도의 관계를 구하였다. 이 실험에는 콘크리트 외에 몰탈, Non-AE 콘크리트를 이용한 실험도 수행하였다. 또한, 동결융해작용을 가해 상대동탄성계수를 변화시킨 공시체에 대하여, 열화의 수치화를 수행하였다. 실험의 결과, X선조영촬영법을 사용하면, 투과선변화량이 구해지므로, 콘크리트강도의 추정과 열화의 수치화가 가능하다는 것을 알았다.

식이섬유의 섭취가 만성 기능성 변비에 미치는 영향 (Effects of Fiber Supplements on Functional Constipation)

  • This study aimed to evaluate the effect of dietary fiber supplementation with snack type on functional constipation. We conducted a double blind case-control study with 3 groups; placebo, low dose group (LD: insoluble fiber 7 g/d + soluble fiber 1.2 g/d) and high dose group (HD: insoluble fiber 14 g/d + soluble fiber 2.4 g/d) , which were randomly assigned out of subjects with functional constipation, defined by modified Rome II criteria. We measured anthropometric parameters and evaluated the bowel movement frequency, stool type, straining, sense of complete evacuation, abdominal discomfort/pain, abdominal inflation, relief and colon transit time before, during the supplementation. Both LD and HD groups had significant improvement in straining, sense of complete evacuation and satisfactory relief compared with placebo group (p < 0.05). When subdivide by baseline colon transit time (less than 24 hrs/ 24 hrs or more and less than 72 hrs / 72 hrs or more), only HD group had significantly improvement; colon transit time recovered near by 24 hrs in subjects of 'less than 24 hrs' (from $7.57{\pm}1.40$ hrs to $25.2{\pm}3.91$ hrs, p < 0.01) and reduced in those of '24hr or more and less than 72 hr' (from $47.0{\pm}3.36$ hrs to $31.3{\pm}4.31$ hrs, p < 0.01) and '72 hr or more' (from $106.7{\pm}10.7$ hrs to $85.0{\pm}13.1$ hrs, P < 0.05) subjects. Particularly, positive effect of fiber supplementation on straining and sense of complete evacuation in test groups seemed to be greater in subjects of '24 hrs or less' and '24 hrs or more and less' than 72 hrs' than those of '72 hrs or more'. In conclusion, subjects with functional constipation, particularly those having colon transit time less than 72 hrs can significantly improve, at least in part, symptom related to constipation by fiber supplementation of snack type without serious side effects.

  • A whole body physiologically based pharmacokinetic (PBPK) model was applied to investigate absorption, distribution, and physiologic variations on pharmacokinetics of imatinib in human body. Previously published pharmacokinetic data of the drug after intravenous (i.v.) infusion and oral administration were simulated by the PBPK model. Oral dose absorption kinetics were analyzed by adopting a compartmental absorption and transit model in gut section. Tissue/plasma partition coefficients of drug after i.v. infusion were also used for oral administration. Sensitivity analysis of the PBPK model was carried out by taking parameters that were commonly subject to variation in human. Drug concentration in adipose tissue was found to be higher than those in other tissues, suggesting that adipose tissue plays a role as a storage tissue for the drug. Variations of metabolism in liver, body weight, and blood/plasma partition coefficient were found to be important factors affecting the plasma concentration profile of drug in human body.

『상한론(傷寒論)』 처방에서 감초(甘草) 용량의 안전성 및 임상 사용에 대한 문헌 고찰 (A Study for Safe Dose and Clinical Use of Glycyrrhizae Radix in Shanghanlun)

  • Objectives : The purpose of this paper is to find out the safe dose and clinical use for administration of Glycyrrhizae Radix in Shanghanlun(傷寒論). Methods : Web-databases(OASIS, NDSL, Pubmed, Google) were searched with keywords including 'Licorice', 'Pseudoaldosteronism', 'Glycyrrhizin', 'Testosterone' on 14/10/2016. The searched about 40 papers and books were reveiwed. Results : Glycyrrhizin(GL) and 3-monoglucuronyl glycyrrhetinic acid(3MGA) in Glycyrrhizae Radix are found to be the main compounds vulnerable for inducing pseudoaldosteronism. The dose range of Glycyrrhizae Radix in Shanghanlun prescriptions is from 0.25 g to 12 g as a daily administration, and this dose satisfies the guidelines of WHO, European Union, ABC etc. And risk factors contributing for personal sensitivities are old age(>60), female sex, liver dysfunction, hypokalemia, prolonged gastrointestinal transit time, anorexia nervosa, decreased 11-${\ss}$-hydroxysteroid dehydrogenase-2 activity and hypertension. Conclusions : As a result, dose of Glycyrrhizae Radix in Shanghanlun(傷寒論) is safe. However, the personal sensitivity and unexpected drug interactions are independent from doses of GL, so doctors should monitor those risk factors and symptoms of pseudoaldosteronism when administering Glycyrrhizae Radix.

Dose-response assessment of the anti-cancer efficacy of soy isoflavones in dimethylhydrazine-treated rats fed 6% fructooligosaccharide

  • We investigated the combinatorial effects of different doses of dietary soy isoflavones (SI) and fructooligosaccharide (FOS) in a rat model of colon cancer. We hypothesized that increased bioavailability of SI metabolites due to dietary FOS may increase production of bioactive equol and affect colon carcinogenesis in a dose-dependent manner. Sprague-Dawley male rats were injected with 12-dimethylhydrazine (DMH) and were providec experimental diets that contained 0, 10, 50, 150, or 500 mg SI per kg of diet and 6% FOS for 12 weeks. The number of aberrant crypt foci (ACF) and the expression of cyclooxygenase-2 (COX-2) in colonic tissues were significantly decreased in the 6% FOS-fed groups compared to the control group. Gut transit time and fecal pH were significantly lower, and fecal concentrations of bifidobacteria were increased with 6% FOS. However, dietary SI supplementation in combination with 6% dietary FOS did not affect ACF formation or COX-2 expression. Plasma equol concentrations were dose-dependently increased by supplementation of SI up to 500 mg/kg of diet. In conclusion, SI supplementation up to 500 mg/kg of diet appeared to have no additive beneficial effects in rats with chemically-induced colon cancer that were fed 6% FOS, although plasma equol was dose-dependently increased.

자궁주위 방사선 근접치료시 MIRD 팬텀을 이용한 주변장기의 피폭환경평가 (Assessment of Dose Distribution using the MIRD Phantom at Uterine Cervix and Surrounding Organs in High Doserate Brachytheraphy)

  • Manchester system 타입의 장착기중 상, 하부에 차폐체가 장착되어 있는 Henschke 장착기를 이용하여 자궁암 근접치료시 자궁 및 주변장기의 선량분포를 평가하기 위하여 치료계획수립에 사용되는 실용프로그램 결과와 몬테칼로 모의계산 결과를 비교하였다. 또한 자궁 및 주변 정상조직이 받은 선량을 계산하기 위해 ORNL(Oak Ridge National Laboratory)에서 수립한 여성의 MIRD (Medical Internal Radiation Dose)형 모의피폭체를 이용 하여 주변장기가 받는 선량을 MCNP로 계산하였다. 몬테칼로 모사에는 MCNP 4B코드를 사용하였으며, 실용계산프로그램에는 GAMMADOT를 이용하였다 MCNP계산에는 $^{192}Ir$ 선원과 장착기의 기하학적 모양을 정밀하게 모사하여 계산 오차를 줄이도록 하였으며, 치료계획용 실용계산프로그램의 계산 조건과 동일하게 치료선원의 강내 체류시간과 체류위치를 적용하여 선량을 계산하였다. 주요 선량 비교 평가점은 Manchester system에서 사용되는 4곳과 ICRU 38에서 Manchester system을 보완하기 위해 제시한 방광표면 및 직장이였다. 실용계산 결과는 MCNP모의계산의 결과와 비교했을 때 대부분 위치에서 상대오차 4% 이내의 결과를 보였고, 난형체의 차폐체 장착효과로 인한 방광과 직장에서의 선량감쇠효과는 각각 19%, 20%였다.

Effects of Cassia obtusifolia L. Extract on Loperamide-Induced Constipation in Rats

  • Cassia obtusifolia L. is commonly used as medicinal foods to treat gastrointestinal disease in many countries. This study evaluated the anti-constipation effects of ethanolic extract of Cassia obtusifolia L. in rats with loperamide-induced constipation. Diffferent concentrations of ethanolic extract of Cassia obtusifolia L. (0, 10, 250, and 500 mg/kg) was administered to rat orally once daily for 3 weeks following loperamide treatment. Six-week-old Sprague-Dawley rats (N=25) were randomly divided into five groups: normal control and experimental groups and injected loperamide intraperitoneally to induce constipation in the latter four experimental groups. Following the induction of loperamide-induced constipation, a marked decrease was observed in the fecal weight and water content discharged for over 1 week and intestinal transit time while an increase was observed in the number of fecal pellets remaining in the colonic lumen as compared with the normal control group. These conditions were significantly alleviated following the administration of the two doses (250 and 500 mg/kg) of ethanolic extract of Cassia obtusifolia L. when compared to the loperamide-treated (constipation control) group. More specifically, the number of fecal pellets in the colon was 3, 5, 4, 2.8, and 1.2 in the five groups, respectively and the corresponding GIT ratio was 70.77, 56.59, 58.67, 66.81, and 72.10%, respectively. The fecal water content, transit distance, and GIT ratio in the high-dose treatment group were significantly higher than that in the medium-dose treatment group, but a significant decrease in the number of fecal pellets in the colon. In conclusion, the Cassia obtusifolia L. extract is suggested to have beneficial effects as a therapeutic and preventive strategy to alleviate constipation and to relieve the symptoms of constipation such as pain, flatulence, distention, bloating, and unpleasant taste.

생맥산 및 평위산 추출물의 위장관 운동 조절 효능에 관한 연구 (Effects of Traditional Chinese Herbal Medicine Shengmai-San and Pyungwi-San on Gastrointestinal Motility in Mice)

  • Objectives: The purpose of this study was to investigate the effects of Shengmai-san and Pyungwi-san, a herbal product used in traditional Chinese medicine, on gastrointestinal (GI) motility in mice. Methods: The in vivo effects of Shengmai-san and Pyungwi-san on GI motility were investigated by measuring the intestinal transit rates (ITRs) using Evans blue in normal mice and in mice with experimentally induced GI motility dysfunction (GMD). GMD was induced by injecting acetic acid or streptozotocin intraperitoneally. Results: In normal Institute of Cancer Research mice, ITRs were significantly and dose-dependently increased by Shengmaisan (0.01~1 g/kg) and Pyungwi-san (0.01~1 g/kg). The ITRs of acetic acid induced peritoneal irritation model and streptozotocin-induced diabetic model mice were significantly reduced compared to normal mice, and these reductions were significantly and dose-dependently inhibited by Shengmai-san (0.01~1 g/kg) and Pyungwi-san (0.01~1 g/kg). Conclusions: These results suggest that both Shengmai-san and Pyungwi-san are a good candidate for the development of a prokinetic agent that may prevent or alleviate GMD.
