Promoting effects of Transforming growth Factor-$\beta$2 on Chondrogenic Differentiation in vitro (Transforming Growth Factor-$\beta$2에 의한 연골세포 분화 촉진 효과)

  • 계배 limb bud 간충직 연골원성 세포로부터 연골세포로의 분화에 미치는 transforming growth factor-f2(TGF-$\beta$2)의 영향을 알아보기 위하여, Hamburger-Hamilton stages 23/24의 간충직 세포들을 미세배양법으로 배양하면서. TGF-$\beta$2의 농도 및 처리시간에 따른 연골세포의 분화에 미치는 영향을 조사하였다 그 결과 TGF-$\beta$2는 배양 첫 24시간 동안 1-2 ng/ml의 농도로 처리하였을 때 가장 효과적으로 연골세포의 분화를 촉진하였으며, 또한 TGF-$\beta$2의 처리군에서 배양 3일째에 tsss) sulfate의 glycosaminoglvcan으로의 유입량이 현저히 증가함을 보였다 한편. 배양 48시간내에TGF-$\beta$2를 처리한 경우 분화를 촉진 유도한 반면, 배양 48시간 이후에 처리하였을 때에는 분화 촉진 효과가 나타나지 않았다. 이상의 결과로부터 TGF-$\beta$2는 연골원 세포의 분화 초기단계에 세포외기질의 합성을 촉진시켜 세포응축을 유발하고. 세포-세포 및 세포-세포외기질의 상호작용을 증대시킴으로써 연골세포로의 분화를 촉진시킬 것으로 추정되었다.

Purification of TGF-$\beta$1 from Bovine Colostrum (젖소 초유로부터 TGF-$\beta$1의 정제)

  • Bovine colostrum contains various bio-functional Proteins. Especially, transforming: growth factor-${\beta}$1 (TGF-${\beta}$1) has a function in concerns with immune response. The purpose of this study was to establish the purification Processing of transforming growth factor-${\beta}$1(TGF-${\beta}$1). The highest concentration of TGF-${\beta}$1 was measured within 48 h after parturition in bovine colostrum using ELISA kit. Purification of TGF-${\beta}$1 from whey protein was carried out by the gel filtration, AF-heparin chromatography and AF-heparin rechromatography. After final purification step, TGF-${\beta}$1 with a molecular weight of 25 kD was obtained, and confirmed by silver staining and western blotting. Finally, TGF-${\beta}$1 was identified native form of 25 kD and reducing form of 12.5 kD by reducing agent.

Effects of Transforming Growth Factor $\beta$ on In-vitro Maturation of Porcine Oocytes (Transforming Growth Factor $\beta$가 돼지 난자의 체외성숙에 미치는 영향)

  • 돼지 수정란의 체외생산은 난자의 체외성숙과 체외수정에 관한 기술의 부족으로 아직까지 만족스럽지 못한 수준이다. 특히 돼지 수정란의 체외생산에는 복잡한 세포질의 성숙과정과 높은 다정자침입율 및 전핵형성의 억제등의 문제점이 있다. 본 연구에서는 돼지 난자의 체외성숙 체계를 개선하기 위하여 transforming growth factor$\beta$(TGF$\beta$)의 첨가가 난자 및 난구세포에 미치는 효과에 대하여 검토하였다. 체외성숙용 배지에 TGF$\beta$를 1~10ng/$m\ell$의 농도로 첨가하여 미성숙 난자를 배양한 결과 성숙율이 높아졌다. TGF$\beta$의 효과는 난구세포가 제거된 난자의 성숙에도 효과적이었다. TGF$\beta$(를 첨가하지 않은 배양액 내에서는 배양 24시간 까지 metaphase-II로 성숙된 난자가 관찰되지 않았으나 TGF$\beta$를 첨가한 배양액 내에서는 관찰되었다. 한편, 난구세포가 부착된 난자의 성숙배양시 TGF$\beta$의 첨가시기에 따른 차이는 없었으나, 난구세포를 제거한 난자의 경우에는 성숙배양 전반기(59%) 또는 후반기(57%) 24시간 동안에만 TGF$\beta$를 첨가하는 것이 48시간 동안 계속하여 첨가(27%)하는 경우 또는 비첨가(38%)에 비하여 유의적으로 높은 성숙율을 나타냈다(p<0.05). 이와 같은 결과는 난구 세포가 돼지 난자의 체외성숙에 필수적이지만 TGF$\beta$는 난구세포가 제거된 난자의 체외성숙에 어 느정도 유익한 효과를 발휘하는 것으로 추측된다.

  • Osteoclast has been known as a primary responsible cell for the bone resorption. The activation of osteoclast, therefore, may be the key event in the regulation of bone growth and remodeling. Various factors were reported to have influence on the resorbing activity of osteoclast in organ culture. Among those factors, transforming growth factor-$\beta$ (TGF-$\beta$) has been known to have a profound effect on bone metabolism. Since a large amount of TGF-$\beta$ presents in bone tissue, it may be important for the understanding the regulatory mechanism of bone resorption to elucidate the effect of TGF-$\beta$ on the osteoclast. We have reported the dlsaggregated chick embryonic osteoclast culture as an useful assay method for determining the resorption activity of osteoclast. In this culture, we found that TGF-$\beta$ significantly enhaced the osteoclastic bone resorption activity. We also found that the timulatory effect seemed to be an indirect one that is mediated by other cells. As nordihydroguaiaretic acid significantly inhibited the TGF-$\beta$1-induced osteoclastic bone resorption, we suggest that the lipoxygenase derivative of arachidonic acid may participate in the action of TGF-$\beta$ as a paracrine or an autocrine mediator.

Study on the Purification of Transforming Growth Factor-$\beta$ in Canine Platelets (개 혈소판에서 변형성장인자 베타의 분리에 관한 연구)

  • To purify transforming growth factor type beta(TGF-$\beta$) in canine platelets, Sephadex G-75 gel filtration and semipreparative HPLC were carried out. The column of $2.0 {\times}120cm$ was used for gel filtration and one inch semipreparative column filled with SP-Toyopeal for HPLC. Electrophoresis and bioassay using African green monkey kidney cell were used for identification of TGF-$\beta$ Crude TGF-$\beta$ of 2.75mg was extracted from 5.2g of the platelets by the treatment of acid/ethanol. In gel filtration of crude TGF-$\beta$, 4 peaks were observed at the detection of spectrophotometer at 280nm. Electrophoresis and bioassay identified the 3rd peak TGF-$\beta$. Linear gradient elution from 0 to 3M NaCl in sornipreparative HPLC showed TGF-$\beta$ at 1.5M NaCl. Gel filtration was less expensive and useful method for the purification of TGF-$\beta$.

Changes of Interleukin-12 and Transforming Growth Factor Beta 1 before and after Antipsychotic Treatments in Schizophrenic Patients (정신분열병 환자에서 Interleukin-12와 Transforming Growth Factor Beta 1의 치료 전후의 변화)

  • Background:Several reports have suggested that cytokine alterations could be related to the pathophysiology of schizophrenia. In this study, we measured plasma level of interleukin-12(IL-12), a proinflammatory T helper 1(Th1) cytokine and transforming growth factor-${\beta}1$(TGF-${\beta}1$), an anti-inflammatory Th3 cytokine before and after antipsychotic treatment in schizophrenic patients. Methods:The plasma concentrations of IL-12 and TGF-${\beta}1$ were measured by using quantitative ELISA in 23 schizophrenic patients and 31 normal controls at admission and 8 weeks later. The psychopathology was measured by Brief Psychiatric Rating Scale(BPRS). Results:IL-12 and TGF-${\beta}1$ levels were significantly higher in schizophrenic patients than in controls before treatment. At the 8 week of treatment, the TGF-${\beta}1$ levels returned to control values, while IL-12 levels were not significantly changed. There were no significant correlations between the changes of BPRS scores and the changes of IL-12 or TGF-${\beta}1$ levels in schizophrenic patients. Conclusion:Cytokine abnormalities in schizophrenia might be involved in the pathophysiology of the illness. It is possible that TGF-${\beta}1$ plays an important role in the schizophrenia.

  • Transforming growth factor (TGF)-$\beta$, a hormonally active polypeptide found in normal and transformed tissue, is a potent regulator of cell growth and differentiation. In this study, we examined the effect of TGF-$\beta$ on invasion and motility of MCF10A human breast epithelial cells. TGF-$\beta$-induced migration and invasive phenotype of the parental MCF10A cells in a dose-dependent manner. Activity of MMP-2 promoter was increased by TGF-b, suggesting that the TGF-$\beta$-induced invasive phenotype may possibly be mediated by MMP-2 rather than MMP-9. (omitted)

  • Background : Many investigators have found transforming growth factor-${\beta}1$ (TGF-${\beta}1$) to be elevated in tumors. Changes in responsiveness to TGF-${\beta}1$ have been linked to malignant transformation, tumor progression and tumor regression. Many malignant cell lines of epithelial or hematopoietic origin are refractory to the antiproliferative effects of TGF-${\beta}1$. However, a little is known about the association of TGF-${\beta}1$ with progression of malignant tumor. Methods : In this study, we measured the plasma level of TGF-${\beta}1$ in various cancer patients and evaluated the utility of plasma TGF-${\beta}1$ as a possible tumor marker. Plasma TGF-${\beta}1$ levels were measured using enzyme-linked immunosorbent assay in cancer patients and normal controls. Carcinoembryonic antigen (CEA) and alpha-fetoprotein (AFP) as tumor marker were compared with TGF-${\beta}1$ in the aspects of sensitivity and specificity. Results : The mean of plasma TGF-${\beta}1$ levels was $1.219{\pm}0.834ng/ml$ in normal controls, $5.491{\pm}3.598ng/ml$ in breast cancer, $12.670{\pm}10.386ng/ml$ in lung cancer, $5.747{\pm}3.228ng/ml$ in hepatocellular carcinoma and $10.854{\pm}7.996ng/ml$ in cervical cancer. In comparison with CEA and AFP, TGF-${\beta}1$ is more sensitive. Conclusion : We conclude that the high levels of TGF-${\beta}1$ are common in the plasma of cancer patients. These results suggest that the plasma TGF-${\beta}1$ level can be a potent tumor marker in various cancer patients.

  • Background: In our previous study, we demonstrated that transforming growth factor-beta 1 receptor II(TGF-${\beta}1RII$) may have a role in the formation of bullae. In this study, we investigated if expression of transforming growth factor-beta 1 (TGF-${\beta}1$) ligand was altered in a bullous lung tissue by immunohistochemical staining of bullous tissues from patients with primary spontaneous pneumothorax. Material and Method: Bullous lung tissues were obtained from 36 patients with primary spontaneous pneumothorax, including 34 males and 2 females aged 14 to 38 years old. Result: Of the 36 patients, 19 were TGF-${\beta}1$ positive and 24 were transforming growth factor-beta 1 receptor II(TGF-${\beta}1RII$) positive. Among the 19 TGF-${\beta}1$ positives, 15 were also TGF-${\beta}1RII$ positive, observation at high magnification showed that strong immunohistochemical stain was detected in the boundary region between the bullous and normal lung tissues. Conclusion: These results suggest that overexpression of TGF-${\beta}1$ may be involved in the formation of a bulla as well as the alteration of TGF-${\beta}1RII$ expression. Further molecular studies are needed to elucidate the more detailed molecular mechanisms of the bulla formation.

  • Transforming growth factor-$\beta$ is a pleiotropic growth factor that has enthralled many investigators for approximately two decades. In addition to many reports that have clarified the basic mechanism of transforming growth factor-$\beta$ signal transduction, numerous laboratories have published on the clinical implication/application of transforming growth factor-$\beta$. To name a few, dysregulation of transforming growth factor-$\beta$ signaling plays a role in carcinogenesis, autoimmunity, angiogenesis, and wound healing. In this report, we will review these clinical implications of transforming growth factor-$\beta$.
