Streptomyces virginiae 유래의 virginiamycin 생합성 유도인자인 VB-C를 이용하여 다른 방선균에의 항생물질 생산 유도 기능을 검토하였다. Erythromycin 생산균인 Streptomyces erythraeus를 공시균주로 사용하였으며, VB-C 결합 receptor 유전자가 포함된 plasmid(pARS 701, 9 kb)를 공시균주에 도입하여 transformant로 사용하였다. Parent와 transformant 모두 유도인자인 VB류를 생산하였으며, VB-C의 signal 전달에 관여하는 결합 단백질의 존재가 확인되었다. Parent는 본배양 0, 20, 44시간째에 합성 VB-C 300 ng/ml를 첨가시 초기 항생물질 생산시기가 약 8시간 이상 단축되었다. Transformant는 본배양 44시간에 첨가시 6시간 이상 항생물질 생산시기가 단축되었고 항생물질 생산량도 증가되었으며, parent에 비해 B. subtilis에도 강한 항균력을 나타내었다. 또한 항균 spectrum은 parent보다는 transformant가 생산한 항생물질의 항균호과가 더 큰 것으로 나타났으며, VB-C를 첨가한 경우 미첨가 경우보다 넓은 항균 spectrum을 보였다. 이들 결과에서 공시균인 S. erythraeus에서 VB signal 전달에 따른 항생물질 생산유도가 입증되었으며, VB-C receptor gene의 도입에 따른 발현과 함께 VBs의 유도, 항생물질의 생산시기의 단축 및 생산량의 증가가 예상됨에 따라 다양한 Streptomyces sp. 균주를 대상으로 항생물질의 대량생산을 비롯한 새로운 2차 대사산물 생산에의 응용이 예상되었다.

It was found that Shinorhizobium meliloti hemoprotein (SM) was more effective than Vitreoscilla hemoglobin (Vhb) in promoting secondary metabolites production when overexpressed in Streptomyces lividans TK24. The transformant with sm (sm-transformant) produced 2.7-times and 3-times larger amounts of actinorhodin than the vhb-transformant in solid culture and flask culture, respectively. In both solid and flask cultures, a larger amount of undecylprodigiocin was produced by the sm-transformant. It is considered that the overexpression of SM especially has activated the pentose phosphate pathway through oxidative stress, as evidenced by an increased NADPH production observed, and that it has promoted secondary metabolites biosynthesis.
Bradyrhizobium japonicum ROKS 26 균주를 glycine및 Iysozyme을 처리하여 spheroplast를 형성시킨 후 Agrobacterium tumefaciens에서 분리한 Ti plasmid를 분리하여 형질전환 시키고 octopine 영양 요구성에 의하여 transformants를 분리하였다. 그 결과 Ti plasmid가 도입된 transformants의 형질전환율은 $1{\times}10^{-7}$ 정도 되었다. 얻어진 transformant의 plasmid를 조사한 결과 도입된 Ti plasmid 존재가 확인되었으며, 2차원 전기영동법으로 균체 단백질을 분석한 결과 수용세포인 Bredyrhizobium japonicum ROKS 26과 transformant간의 단백질 구성 차이도 화인되었다. 또한 transformant의 근류 형성력을 조사한 결과 본래의 근류의근류형성력을 가지고 있었으나, crown gall형성에 있어서는 Agrobacterium tumefaciens 15955에 의한 gall보다는 크기에 있어 차이가 있었다.
Starch로부터 직접적으로 ethanol을 발효 생산할 수 있는 새로운 효모균주의 개발을 목적으로 S. diastaticus의 glucoamylase gene을 cloning vector를 사용하지 않고 S. cerevisiae에 transformation시켜, soluble starch를 직접 발효 할 수 있는 transformants를 얻는데 성공하였으며 이들 중 glucoamylase 생성능이 가장 우수한 균주인 TSD-14를 전보에서 선별하였다. Transformant TSD-14의 glucoamylase 생성조건과 ethanol productivity를 parent strain과 비교 검토한 결과 이들 성질에 있어서 TSD-14는 donor인 S. diastaticus와 거의 유사하였다. 즉, 탄소원으로는 soluble starch가 균의 생육 및 효소생성에 가장 효과적이었으며 glucose, maltose 등은 균의 생육에는 효과적이었으나 효소생성은 저해하였다. 질소원으로 무기태 질소원에 비하여 유기태 질소원이 좋은 효과를 나타냈으며 특히, peptone과 yeast extract가 효과적이었다. 또한 금속염으로는 FeSO$_4$, MgSO$_4$, MnCl$_2$, NiSO$_4$등이 효과적이었으며 CoCl$_2$, HgCl$_2$, PbCl$_2$등은 오히려 균의 생육뿐만 아니라 효소생성을 크게 저해하였다. 한편, TSD-14의 ethanol productivity를 조사한 결과, 15% sucrose, soluble starch, liquefied potato starch 등에서 8.3% (v/v) , 4.8%(v/v), 7.5%(v/v)의 ethanol을 생성하여 각각 총당에 대해 84%, 45%, 70%의 발효율을 나타냈다.
Genetic transformation was affected by material of explant, age of callus, and medium of regeneration. Two rice seed cultivars (Ilpum and Baekjinju) and mediums were investigated in this study for enhancing regeneration of transgenic rice expressed AtBI-1 gene encoding the Arabidopsis thaliana Bax inhibitor. Regeneration rate of Ilpum rice transformant in gelrite of 5 and 8 g were 27.4% and 18.0%, respectively. In Baekjinju, regeneration rate of transformant was 5.4% and 4.3% in 5 and 8 g gelrite, respectively. The highest number of transformant plant in this study was regenerated from Ilpum cultivar on MS medium (30.4%) and was applied for the subsequent experiment. The callus regeneration rate of transformant were 40.7% in callus infection of up-side, it was higher regeneration then in the down-side (3.9%). The regeneration rate of callus of 25 days and 35 days were 14.7% and 38.6%, respectively. The most important application of this work is in genetic transformation of rice, particularly for improvement transgenic plant tissue culture protocol with high frequency of plant regeneration.
A gene(lsd1) encoding dextranase from Lipomyces starkeyi KSM22 has been previously cloned, sequenced, and expressed in Saccharomyces cerevisiae. The gene consisting of 1,824 base pairs and encoding a protein of 608 amino acids was then cloned into and secretively expressed in Pichia pastoris under the control of the AOX1 promoter. The dextranase productivity of the P. pastoris transformant(pPIC9K-LSD1, 134,000 U/I) was approximately 4.2-fold higher than that of the S. cerevisiae transformant(pYLSD1, 32,000 U/I) cultured in an 8-1 fermentor. Over 0.63 g/l of active dextranase was secreted into the medium after methanol induction. The dextranase of the P. pastoris transformant, as analyzed by SDS-PAGE and Western blotting, showed only one homogeneous band. This dextranase of the P. pastoris transformant showed a broad band near 73 kDa. Rabbit monoclonal antibodies against a synthetic LSD1 peptide mix also recognized approximately 73 kDa.
Starch로부터 ethanol을 직접적으로 발효 생산할 수 있는 새로운 효모 균주를 개발하고자 glucoamylase 생성균으로 알려진 Saceharomyces diastaticus의 glucoamylase gene을 cloning vector를 사용하지 않고 S, cerevisiae에 transformation시켰다. Li$_2$SO$_4$, 처리로써 competent화 한 S, cerevisiae의 Intact cells을 recipient로 하여 BamHI으로 partial digestion한 S, diastaticus의 chromosomal DNA를 transformation시키고 starch를 유일한 탄소원으로 함유한 최소 배지상에서 starch 자화능을 marker로 하여 transformant를 선별한 결과, 8.5$\times$$10^{-7}$ 빈도로 transformant를 얻었다. Transformant의 특성을 recipient 및 donor와 비교하기 위해 copper resistance와 당 발효능을 조사한 결과, donor인 S, diastaticus와 동일한 성질로서 표현된 maltose와 starch 발효능을 제외하고는 800ppm 농도까지 생육 가능한 copper resistance와 galactose 발효능 등에 있어서는 recipent와 동일하게 나타났다. 또한 transformant가 생성하는 glucoamylase의 그 작용에 있어서의 최적온도와 최적pH를 조사하여 본 바 각각 pH5.0, 50C로서 donor의 glucoamylase와 동일함을 알 수 있었다.
본 연구에서는 안정성이 보장된 효모를 숙주로 하여 이미 lysozyme 생산능이 확인된 저 copy수의 YCp type인 pHK101의 생산능을 높이기 위해 고발현 벡터인 다 copy 수의 YEp type인 pHK501을 구축하였다. pHK501과 pHK101형질전환체의 M. luteus를 기질로 한 lysoplate assay 비교에서 확실한 생산량의 증가를 생육저지환을 통해 확인하였다. 또한 E. coli에서 peptidoglycan만을 추출하여 기질로 사용한 lysoplate assay에서도 pHK501형질전환체의 배양액 중에는 정상적 인 HLY의 생산을 직접 확인할 수 있었다. 플라스크 배양에서 배양시간에 따른 HLY의 최대 생산량은 81시간만에 pHK501형질전환체가 55 units/$m\ell$에 도달됨으로써 pHK101(7 units/$m\ell$)에 비해 약 8배 증가됐다. 발효조규모에서의 HLY 생산은 24시간만에 26.8 units/$m\ell$(1.12 units/$m\ell$/h)에 도달하였고 전체 생산성은 플라스크배양(0.625 units/$m\ell$/h)에 비해 약 1.8배 정도 증가되었다.
To illustrate whether a hemolysin in $\delta$-endotoxins of Bacillus thuringiensis strain 73E10-2 and subsp. israelensis had immunological identity, a cyt gene of the strain 73E10-2 which encodes a hemolysin was cloned to B. subtilis (transformant 2753). The transformant 2753 containing cyt gene produced the hemolysin which lysed sheep erythrocytes after treatment of proteinase K. The hemolysin was proved also to be toxic against mosquito larvae (Aedes aegypti). The molecular weight of the hemolysin produced from the transformant 2753 was determined to be about 25 kDa by SDS-PAGE and immunoblot. The hemolysin in $\delta$-endotoxin of subsp. israelensis and subsp. kyushensis did not react on immunoblot using polyclonal anti-$\delta$-endotoxin of the strain 73E10-2, but 70-140 kDa mosquitocidal toxins in $\delta$-endotoxin of subsp. kyushuensis reacted.
NADPH has been known as a regulating factor the biosynthesis of polyhydroxyalkanote(PHA), and the flux of NADPH for PHA biosynthesis could be enforced by the amplification of zwf gene encoding glucose 6-phosphate dehydrogenase. The recombinant plasmid pCZWF harboring PHA synthase, phbC from R. eutropha and zwf from E. coli were constructed, and were transformed to R. eutropha by electroporation. The biosynthesis of P(3HB-3HV) copolymer were carried out in transformant R. eutropha through the two-stage cultivation method using valerate as a precursor. The biosynthesis rate and PHA content of transformant R. eutropha harboring pCZWF were increased compared with transformant R. eutropha harboring only phbC. Especially, the molar fraction of 3HV was increased from 68% to 74% due to amplification of zwf gene. And the biosynthesis P(3HB-3HV) and P(3HB-4HB) carried out using propionate and ${\gamma}-butyrolactone$ as a precursor, respectively. But the rate, content, and molar fraction of biosynthesis copolymers were not influenced appreciably. This may be due to the reduced availability of NADPH.
