우리나라에 보급된 상업용 저온저장고는 총 1,529개동에 239,732평이고 전국평균 가동율이 21.1% 밖에 되지 않아 규모상으로는 저온저장고의 공급이 과도한 상태에 있다는 지적도 있다. 이러한 양적인 문제에 대한 논란은 접어두더라도 저온저장된 농산물의 손실이 10-30%에 이르고 있어 저장환경의 조절에 문제가 있는 것으로 지적되고 있다. (중략)
Leaf mesophyll protoplasts of Dianthus superbus were cultured in MSP1 liquid medium supplemented with 0.5 mg/L BAP, 2.0 mg/L NAA and 9% mannitol. Protoplast-derived colonies were formed after 3 to 4 weeks of culture in the dark at 27$^{\circ}C$. These colonies were kept under continuous illumination (21.5 $\mu$E. m-2 sec-1) for 2 weeks and finally most of the colonies became green microcalli, about 3 mm in diameter. When green microcalli were transferred to MS solidified medium with 2.0 mg/L 2,4-D, they formed embryogenic calli after 4 week of culture. These calli were then transferred onto $N_{6}$ medium containing 0.1mg/L 2,4-D, 0.1 mg/L NAA, 2.0 mg/L kinetin and 2.0 g/L casein hydrolysate and cultured under illumination. After 5 weeks of culture the calli gave rise to multiple shoots of 10 to 15 per callus. Upon transfer onto MS medium containing 2.0 mg/L NAA, they were noted. The regenerates were successfully transplanted into potting soil.
Levodopa or L-3,4-dihydroxyphenylalanine (L-DOPA) is the precursor of the neurotransmitter dopamine. L-DOPA is a famous treatment for Parkinson's disease symptoms. In this study, electroenzymatic synthesis of L-DOPA was performed in a three-electrode cell, comprising a Ag/AgCl reference electrode, a platinum wire auxiliary electrode, and a glassy carbon working electrode. L-DOPA had an oxidation peak at 376 mV and a reduction peak at -550 mV. The optimum conditions of pH, temperature, and amount of free tyrosinase enzyme were pH 7, $30^{\circ}C$, and 250 IU, respectively. The kinetic constant of the free tyrosinase enzyme was found for both cresolase and catacholase activity to be 0.25 and 0.4 mM, respectively. A cyclic voltammogram was used to investigate the electron transfer rate constant. The mean heterogeneous electron transfer rate ($k_e$) was $5.8{\times}10^{-4}$ cm/s. The results suggest that the electroenzymatic method could be an alternative way to produce L-DOPA without the use of a reducing agent such as ascorbic acid.

Lipid transfer proteins (LTPs) are widely distributed in the plant kingdom, but their functions remain elusive. The proteins AlLTP2-4 were isolated from three related Allium plants: garlic (A. sativum L.), Welsh onion (A. fistulosum L.), and Nanking shallot (A. ascalonicum L.). These novel proteins comprise a new class of LTPs associated with the Ace-AMP1 from onion (A. cepa L.). The AlLTP genes encode proteins harboring 132 common amino acids and also share a high level of sequence identity. Protein characteristics and phylogenetic analysis suggest that LTPs could be classified into five distinct groups. The AlLTPs were clustered into the most distantly related plant LTP subfamily and appeared to be restricted to the Allium species. In particular, the number of amino acids existing between the fourth and fifth Cys residue was suggested as a conserved motif facilitating the categorization of all the LTP-related proteins in the family. Unlike other LTPs, AlLTPs harboring both the putative C-terminal propeptide and N-terminal signal peptide were predicted to be localized to cytoplasmic vacuoles. When a chimeric GFP protein fused with both N-terminal and C-terminal AlLTP2 signal peptides was expressed in rice cells, the fluorescence signal was detected in the endomembrane compartments, thereby confirming that AlLTPs are an unprecedented intracellular type of LTP. Collectively, our present data demonstrate that AlLTPs are a novel type of LTP associated with the Allium species.
This study was carried out to investigate the effect of activation timing, cell cycle and passage on the development of embryos produced by cumulus cell nuclear transfer in Hanwoo (Korean cattle). Nuclear donor cumulus cells were cultured in Dulbecco's modified Eagle medium supplemented with 10% fetal bovine serum at $38.5^{\circ}C$ in a humidified atmosphere of 5% $CO_2$ in air. The 1~6 passages of serum deprived or actively dividing cumulus cells were isolated and used as donor cells. The in vitro matured oocytes were enucleated and then the isolated donor cells were introduced. One pulse of 180 volts for $15{\mu}s$ was applied to induce the fusion between karyoplast and cytoplast. The activation was done before or after the fusion. To activate, oocytes were treated with $10{\mu}M$ calcium ionophore for 5 min immediately followed by 2 mM 6-dimethylaminopurine for 3 h. The nuclear transfer embryos were cultured in $500{\mu}l$ of modified CRlaa supplemented with 3 mg/ml BSA in four well dish covered with mineral oil. After 3 days culture, culture medium was changed into modified CRlaa medium containing 1.5 mg/ml BSA and 5% FBS for 4 days. The incubation environment was 5% $CO_2$, 5% $O_2$, 90% $N_2$ at $38.5^{\circ}C$. There was no blastocyst formation when the nuclear transfer embryos were activated before the fusion, whereas, 29.9% of blastocyst formation was shown when the nuclear transfer embryos were activated after the fusion. When serum deprived and actively dividing cumulus cells were used as nuclear donor cells, the developmental rates to blastocyst were 38.5% and 40.6%, respectively. There was no significant difference between serum deprived and actively dividing cells in the developmental rates. The developmental rates to blastocyst according to 1~6 passages were 37.5~44.4%. However, there were no significant differences among passages. These results indicate that 1~6 passage cumulus cell irrespective of cell cycle could support development of nuclear transfer embryos activated after the fusion.
Recently, entomopathogenic nematodes have received a considerable attention as biologicalcontrol agents. For in vitro cultivation, storage and transportation of nematodes, oxygen supply isextremely impotant due to its limited solubility and mass transfer problem. The oxygen uptake rates(OURs) of four different Steinernema species were measured in a 5L bioreactor at varying temperatures.The OURs of the Steinernema spp. were below 0.5 x mmolO'||'&'||' . min in the range of 13-17$^{\circ}$C. TheOURs (mmo102/L - min) of S. glaseri Dongrae and S. carpocapsae Pocheon strains were 0.4 x lo-', 0.75x lo-\ulcorner at 21$^{\circ}$C, 1.5 x lo-\ulcorner, 3.2 x 10-2 at 25"C, and 2.8 x lo-', 5.8 x lo-\ulcorner at 29"C, respectively. However,the OURs were not significantly altered by the agitation speed of 50-150 rpm. The specific oxygenuptake rates (qol) of S. glaseri NC, S. glaseri Dongrae, S. glaseri Mungyeong and S. carpocapsaePocheon strains were 0.3 x 0.5 x 0.3 x and 0.2 x mmolO~/cell min at 25"C,respectively. As the nematode size and temperature were increased, the qo, was also increased.the qo, was also increased.
Effects of substituents in the nucleophile(X), the substrate(Y) and the leaving group(Z) on the structure of $S_N2$ transition states have been analyzed by considering effects of four components, electrostatic($E_{es}$), exchange repulsion ($E_{ex}$), polarization($E)_{pl}$) and charge transfer($E_{ct}$) terms, of interaction between the reactants on the degree of bond making and bond breaking. Prediction of net effects of all substituents(X, Y and Z) on the degree of bond making were found to be clearcut whereas the effect of an electron withdrawing group on the substrate (Y = EWG) on the degree of bond breaking was complex; the substituent(Y = EWG) is normally carbon-leaving group($C^{\ast}$-L) bond tightening($E_{pl}$ dominance) but becomes $C^{\ast}$-L bond loosening when the bond is strongly antibonding ($E_{ct}$ dominance). Our model calculations on the reaction of $CH_2XNH_2$ with $YCH_2COOCH_2Z$ using energy decomposition scheme have confirmed that predictions based on our analysis are correct.
An experimental study of the absorption process of water vapor into a lithium bromide solution was performed. For the purpose of development of high performance absorption chiller/hater utilizing lithium bromide solution as working fluid, it is the most effective to improve the performance of absorber with the largest heat transfer area of the four heat exchangers. The experimental apparatus was composed of a plate type absorber which can increase the heat exchange area per unit volume to investigate more detail characteristics instead of the conventional type, horizontal tube bundle type. The size of plate absorbers were made for $0.4m{\times}0.6m$ and the design object of a refrigeration capacity was lRT. In this experiment, three kind plate absorbers which were flat plate, dimple plate and groove plate were used. The results were less than the design object values, that is, the refrigeration capacity was about $0.3{\sim}0.4RT$ and the overall heat transfer coefficient was $500{\sim}600kcal/m^2h^{\circ}C$ at the standard conditions.
In order to predict the temperature distribution of stainless steel strip in Bright Annealing (BA) furnace, we performed the analysis of heat transfer and fluid flow using STAR-CCM+. The analysis model included unsteady fluid flow, heat transfer with radiation and moving grid. Two kinds of radiative properties, emissivity and reflectivity, were applied to the stainless steel strip, one is constant and the other is variable with time. As we call, the BA furnaces of stainless steel strip have two different types, muffle and no-muffle. The using of muffle type has been faced with some problems such as rising in material price and shortening of life cycle, etc. So the development of no-muffle type BA furnace is very important in order to save energy cost, lower environmental load and increase the productivity. The designed (or expected) temperature of stainless steel strip coming out of BA furnace was about $1065^{\circ}C$ while the environment temperature maintains around $1100^{\circ}C$. The result of our calculation was very close (or similar) to design temperature, and the application of radiative properties variable with time produced more accurate result than applying constant ones.

Using twodimensional heat transfer model, first. the importance of several parameters will be identified by a parametric analysis. Then, the tcmpcmture distribution of thc cylindrical concrete specimen in the laboratory was mensuwti and compared with that yielded by thc theoretical considel.ations.
