현재까지 진행된 다양한 인공지능 교육 관련 연구 성과에서는 인공지능의 원리와 개념 교육에 관한 내용이 중심이었다. 따라서 본 연구에서는 인공지능을 타 학문과 탈 학문적 융합교육을 할 수 있도록 교육 프로그램을 개발하였다. 중심 교육 내용으로는 고등학교 화학에서 다루고 있는 분자 구조의 이해를 도울 수 있도록 머신러닝을 활용하는 것으로 하였고, 총 8차시 분량으로 설계하였다. 본 연구에서 개발한 프로그램은 전문가 검토를 통해 I-CVI(Item Content Validity Index) 값을 산출하였고, 그 결과 모든 문항이 .80 이상으로 기각되는 항목이 없었으나. 추가 세부 의견을 반영하여 수정 및 완성하였다. 본 연구의 프로그램은 화학 교과와 정보(인공지능) 교과의 내용적 요소를 탈 학문적으로 융합하였기 때문에 학습자로 하여금 융합인재소양이 높아질 수 있을 것으로 기대한다. 또한, 이 교육 프로그램을 위한 별도의 시수 확보가 요구되지 않아 교사에게는 수업 부담을 낮출 수 있을 것이다.
Myocarditis was previously attributed to an epidemic viral infection. Additional harmful reagents, in addition to viruses, play a role in its etiology. Coronavirus disease 2019 (COVID-19) vaccine-induced myocarditis has recently been described, drawing attention to vaccine-induced myocarditis in children and adolescents. Its pathology is based on a series of complex immune responses, including initial innate immune responses in response to viral entry, adaptive immune responses leading to the development of antigen-specific antibodies, and autoimmune responses to cellular injury caused by cardiomyocyte rupture that releases antigens. Chronic inflammation and fibrosis in the myocardium eventually result in cardiac failure. Recent advancements in molecular biology have remarkably increased our understanding of myocarditis. In particular, microRNAs (miRNAs) are a hot topic in terms of the role of new biomarkers and the pathophysiology of myocarditis. Myocarditis has been linked with microRNA-221/222 (miR-221/222), miR-155, miR-10a*, and miR-590. Despite the lack of clinical trials of miRNA intervention in myocarditis yet, multiple clinical trials of miRNAs in other cardiac diseases have been aggressively conducted to help pave the way for future research, which is bolstered by the success of recently U.S. Food and Drug Administration-approved small-RNA medications. This review presents basic information and recent research that focuses on myocarditis and related miRNAs as a potential novel biomarker and the therapeutics.

The cyclic Arg-Gly-Asp (cRGD) peptide is well-known as a binding molecule to the integrin αvβ3 receptor which is highly expressed on activated endothelial cells and new blood vessels in tumors. Although numerous results have been reported by the usage of cRGD peptide-based ligands for cancer diagnosis and therapy, the distinct mechanisms, and functions of cRGD-integrin binding to cancer cells are still being investigated. In this study, we evaluated the internalization efficacy of different types of cRGD peptides (monomer, dimer and tetramer form) in integrin αvβ3 overexpressing cancer cells. Western blot and flow cytometric analysis showed U87MG expresses highly integrin αvβ3, whereas CT-26 does not show integrin αvβ3 expression. Cytotoxicity assay indicated that all cRGD peptides (0-200 µM) had at least 70-80% of viability in U87MG cells. Fluorescence images showed cRGD dimer peptides have the highest cellular internalization compare to cRGD monomer and cRGD tetramer peptides. Additionally, transmission electron microscope results clearly visualized the endocytic internalization of integrin αvβ3 receptors and correlated with confocal microscopic results. These results support the rationale for the use of cRGD dimer peptides for imaging, diagnosis, or therapy of integrin αvβ3-rich glioblastoma.
To produce an intestinal immunomodulatory beverage containing Centella asiatica extract (CAE), three types of CAE-added beverage prototypes were prepared, and their immunomodulatory activities and marker compounds were analyzed. As a result of the cytotoxicity assessment, all the beverages did not show significant toxicity compared to the control group. Next, the immunomodulatory activities of the beverage prototype were evaluated using the inflammatory model of IL-1β-induced intestinal epithelial cell line. All the samples significantly reduced the production of IL-6, IL-8, and MCP-1 in a CAE concentration-dependent manner. In addition, CAE-added beverages inhibited NO, IL-6, and IL-12 production in LPS-induced RAW 264.7 cells. When the major triterpenoids, as marker compounds for the production of CAE-added beverages, were analyzed by HPLC-DAD, only asiaticoside was detected beyond the limit of quantification, while madecassoside, madecassic acid, and asiatic acid were not detected. The amounts of asiaticoside in CAE-added beverage prototypes were confirmed in No. 1 (19.39 ㎍/mL), 2 (19.25 ㎍/mL), and 3 (19.98 ㎍/mL). In conclusion, the results of this study suggested that CAE-added beverage prototypes induced immunomodulatory effects in the intestinal inflammatory cell line models and asiaticoside could be used as a marker compound for CAE-added beverage production.
To increase industrial applicability of Astragalus membranaceus (AM) as immunostimulating materials, hot-water extract (AME) was prepared from AM and fermented with Kimchi-lactic acid bacteria (Lactobacillus sakei & Leuconostoc mesenteroides) to prepare fermented AM-postbiotics (FAME). Although FAME prepared from AM-postbiotics did not show a significant enhancement in macrophage stimulating activity compared to non-fermented AME, crude polysaccharide (FAME-CP) fractionated by EtOH precipitation from FAME showed significantly higher macrophage stimulating activity than AME-CP. Compared to AME-CP, FAME-CP showed dramatic changes in component sugar and molecular weight distribution. FAME-CP was a polysaccharide with a major molecular weight distribution of 113.4 kDa containing Man (44.2%), Glc (19.3%), Gal (10.2%), GalA (10.2%), and Ara (7.4%) as sugar components. FAME-CP with enhanced macrophage stimulatory activity not only increased expression levels of mRNA genes encoding macrophage-activated factors (iNOS, TNF-α, MCP-1, IL-6, and COX-2), but also led the nuclear translocation of activated p65 and c-Jun. In conclusion, crude polysaccharide from AM-postbiotics fermented with lactic acid bacteria could increase industrial applicability as a functional material with enhanced immunostimulating activity than AME-CP.
본 연구에서는 $PRESAGE^{REU}$ 겔을 이용하여 방사선 치료계획 시 3차원 흡수선량 분포 검증을 위한 정도 관리 소프트웨어를 개발하여 겔을 이용한 3차원 선량분석 방법을 제시하고자 한다. 우선 치료계획상의 3차원 흡수선량 데이터와 측정한 겔 광학밀도 데이터의 입출력 기능을 구현하였고, 변환 테이블을 이용하여 광학밀도를 흡수선량으로 변환하는 기능을 구현하였다. 겔에서 측정된 흡수선량과 치료계획상의 흡수선량 분포간의 기하학적 매칭을 위하여 3D 볼륨 데이터의 x, y, z 방향 및 회전 변환을 구현하였다. 매칭이 완료된 두 선량 분포간에 일치도를 검증하기 위하여 3차원 감마 인덱스알고리듬을 구현하였고, 감마 통과 지도(gamma passing map) 기반의 일치도 확인 기능을 구현하였다. 광학밀도와 흡수선량간의 관계를 분석하기 위하여 원기둥 형태의 $PRESAGE^{REU}$ 겔을 대상으로 X-선 전산화 단층촬영기를 이용하여 CT 영상을 획득하였고, 방사선 치료계획 시스템(Eclipse, Varian, Palo Alto)을 이용하여 원반 형태의 6개의 가상 표적을 생성하여, 각각에 1 Gy에서 6 Gy까지 선량이 전달되도록 입체조형 방사선 치료계획을 수립하였다. 다음으로 광학 CT 스캐너($Vista^{TM}$, Modus Medical Devices Inc, Canada)를 이용하여 기준 투영 영상들을 획득하였고, 치료계획과 동일하게 겔에 방사선을 조사하였다. 조사2시간 후 매 2시간 간격으로 광학 CT 스캐너로 투영 영상 셋을 획득 후 3차원 광학밀도 데이터로 재구성하였다. 실린더 중심축을 따라 치료계획상의 흡수선량 프로파일과 광학밀도 프로파일을 추출하여 광학선량 대비 흡수선량 대응 테이블을 정의하였다. 이후 본 연구에서 개발한 소프트웨어를 이용하여 3차원상의 선량 분포의 일치도를 평가하였다. 광학밀도와 흡수선량간에는 supra-linear 관계가 나타났으며, 광학밀도는 그 크기에 따라 24시간당 60% 전후로 감쇄하였다. 측정된 흡수선량은 중심축 부근에서는 치료계획 선량과 잘 일치하였으나, 주변부로 갈수록 크게 낮아짐을 확인할 수 있었으며, 이로 인하여 3D 감마 통과율은 선량 차이율과 DtoA 각각 3%/3 mm의 조건하에 70.36%로 낮게 나타났다. 이러한 결과는 광학 CT 스캐너 내부의 오일과 $PRESAGE^{REU}$ 겔간의 굴절률이 정확하게 매칭되지 않아서 광학 스캔 시 빔이 굴절되어 부정확한 데이터를 만들어 내는 것으로 분석되었다. 본 연구에서 개발한 정도 관리소프트웨어는 3차원 겔 선량을 비교 분석하기에 유효한 것으로 평가되었으나, $PRESAGE^{REU}$ 겔로부터 정확한 흡수선량데이터를 획득하기 위해 겔 선량측정 과정의 많은 개선이 요구된다.
목적: 본 연구는 초음파 조영제를 직접 합성하고 그 크기 및 소멸 시간 등의 특성을 분석하며, 기존의 초음파 조영제와의 영상을 비교하여 앞으로 초음파 조영제를 매개로 한 영상 유도 하 약물 및 유전자 치료제의 전달 시스템 구축의 기본을 갖추고자 한다. 대상 및 방법: 초음파 조영제는 21마이크로 몰의 DPPC(1, 2-Dihexadecanoyl-sn-glycero-3-phosphocholine, $C_{40}H_{80}NO_8P$), 9 마이크로 몰의 콜레스테롤, 1.9 마이크로 몰의 DCP(Dihexadecylphosphate, $[CH_3(CH_2)_{15}O]_2P(O)OH$) 및 클로로포름 및 SF6 기체를 이용하여 합성하였다. 약 12-24시간의 동결건조 후에 초음파 조영제는 SF6 기체와 초음파 처리(sonication) 법을 이용하여 합성하였고 그 초음파 조영제의 크기는 압출기를 이용하여 일정한 크기로 조정하였다. 합성된 초음파 조영제는 동적 광산란 측정법(dynamic light scattering measurement)을 이용하여 그 크기를 분석하였다. 한편 합성한 초음파 조영제의 소멸 양상을 평가하기 위하여 광학 현미경을 이용하여 그 숫자를 합성 직후, 12시간, 24시간, 36시간, 48시간, 60시간, 72시간 및 84시간 후에 평가하였다. 초음파 조영제로서의 효과를 평가하기 위하여 모형을 만들어 현재 임상적으로 사용되고 있는 초음파 조영제와 그 에코를 비교 분석하였다. 결과: 초음파 조영제는 동결건조-초음파 처리 방법을 통하여 성공적으로 합성 할 수 있었다. 합성된 초음파 조영제는 154.2 nm의 정점 (peak)에서 가장 많이 분포하였으며 합성 후 24시간부터는 입자의 숫자들이 감소하는 양상을 보였다. 소노뷰와 합성된 초음파 조영제의 에코밝기는 유의한 차이를 보이지 않았으나 소노뷰와 생리식염수 및 합성된 초음파 조영제와 생리식염수의 에코 밝기는 통계적으로 유의한 차이를 보였다 (p < 0.01). 결론: 본 연구를 통하여 초음파 조영제를 직접 합성할 수 있는 방법을 습득할 수 있었고 그 크기 및 특성을 분석할 수 있었다.
이 연구의 목적은 사회정의 주제를 중심으로 수학과 도덕을 통합한 통합교수모델을 개발하고 통합교수모델을 적용한 수업이 학생들에게 주는 효과를 알아보는 것이다. 통합교수모델을 개발하기 위해 2015 개정 수학 교육과과 3~4학년군, 5~6학년군 수학 교과서, 2015 개정 도덕 교육과정에 대한 문헌 연구를 실시하여 수학과 도덕 교과에 사회정의 소재가 얼마나, 어떻게 반영되어 있는지를 알아보고자 하였다. 앞선 교과서 분석 결과를 바탕으로 수학과 도덕, 사회정의를 3개의 축으로 하여 융합모형 틀과 사회정의를 위한 수학과 도덕의 통합교수모델을 구안하였다. 통합교수모델의 효과를 확인하기 위하여 수학과 도덕의 통합수업이 학생들의 수학에 대한 정의적 측면과 사회적 의식, 창의적 사고력 및 비판적 사고력에 어떤 영향을 주는지 확인하였다.
본 연구는 4차 산업혁명 시대의 공감능력 배양을 위한 국내 대학의 융합 디자인 교육과정의 나아가야 할 방향을 제안하는 데 그 목적이 있다. 연구방법은 문헌 연구를 통해 초연결 시대에 공감능력이 중요하게 된 배경 및 디자인 재정립에 관한 이론적 배경을 고찰하였고, 국내 외 대학 융합 디자인 교육 과정의 현황에 관한 사례 분석 연구를 진행하였다. 이를 통해 학부와 대학원 과정이 연계된 초학제적 융합 디자인 교육 과정을 제안하며, 이는 기본적으로 공감을 기반으로 한 디자인과정이 주도적으로 타 학문 간 연결의 핵심으로 작용하는 교육 과정이다. 디자인사고와 과정의 본질적 측면을 바탕으로 공감능력을 향상시키는 융합교육과정은 급변하는 환경에서 대체될 수 없는 경쟁력 향상에 기여할 것이라 고려된다. 앞으로 국내 대학 융합 디자인 교육의 활성화 참고자료로 사용될 것을 기대하며 후속 연구에 도움이 되길 기대한다.
To investigate the industrial availability of liquid fermentation (PL-ferment) by Phellinus linteus mycelium as a postbiotics for the inhibition of inflammation, PL-ferment was fractionated into culture supernatant (CS), hot-water extract (HW) from PL-ferment, EtOH-precipitate (CP) fractionated from HW, and the dialysate (DCP) of CP. Compared to the other fractions, DCP which is expected to contain exopolysaccharide (EPS) as the major component, significantly decreased the production of NO, IL-6, and MCP-1 in LPS-induced RAW 264.7 cells, and IL-6 and IL-8 in TNF-α and IFN-γ-induced HaCaT cells. The general component analysis results showed that no significant difference in components was observed between the fractions, whereas sugar composition analysis revealed that DCP had decreased glucose and increased mannose contents compared to the other fractions. This suggests that mannose played an important role in the anti-inflammatory activity of the active fraction, DCP. Molecular weight distribution analysis revealed that DCP was mainly composed of low-molecular-weight material-removed high-molecular-weight polysaccharides of 18-638 kDa, suggesting that EPS originated from P. linteus EPS. In conclusion, our results suggest that the DCP of P. linteus mycelium fermentation using the anti-inflammatory activity could be used industrially as postbiotic material.
