IP 컨택센터에서 통화 녹음을 위한 VoIP 녹취 시스템 (A VoIP Transcript System for Call Recording in IP Contact Center)

  • 본 논문에서는 VoIP를 통화 수단으로 사용하는 IP 컨택센터에서 상담원과 고객간의 대화 내용을 기록할 수 있는 VoIP 녹취 시스템을 설계하고 구현하였다. 본 논문에서 구현한 녹취 시스템은 패킷 스니퍼링 기법을 이용하여 네트워크 부담 없이 실행될 수 있으며 호 제어 프로토콜인 H.323과 SIP에 구분 없이 녹취가 가능하다. 구현된 녹취 시스템은 CTI 시스템과 연동하여 고객과 상담원과의 통화 내용을 효율적으로 저장하고 관리한다. 녹취 시스템을 시스템에 독립적으로 설계하였으며 이를 바탕으로 Windows 및 Linux 환경에 모두 구현하였다.

U-937 세포에서 방사선 및 $H_2$O_2$에 의한 ceruloplasmin의 mRNA 유전자 발현 (Ceruloplasmin Gene Expression in U-937 Cells exposed to ${\gamma}$-Irradiation and $H_2$O_2)

  • In human U-937 cell exposed to ${\gamma}$-irradiation and $H_2O$$_2$, the level of mRNA efrpression in ceruloplasmin gene was measured by using comparative RT.PCR (reverse transcriptase-polymerase chain reaction). At the normal growth condition, the level of ceruloplasmin transcript was estimated as 8.2% and 0.0068% of hprt (hypoxantine phosphoribosyl transferase) transcript and of $\beta$-actin transcript, respectively. In U-937 cells exposed to a dose of 100 rad ${\gamma}$-irradiation, the level of ceruloplasmin transcript was increased about 2.7 and 1.6 fold compared to un-treated cell by using compensation with the levels of hprt and $\beta$-actin transcript. By contrast, the expression of ceruloplasmin gene in U-937 cells exposed to $H_2O$$_2$(50 $\mu$M, 24 h), was shown no significant difference compared to un-treated cell. These results indicated that the expression system of ceruloplasmin gene may react only some specific oxygen species, such as reactive oxygen species induced by ${\gamma}$-irradiation.

Cloning and Characterization of the Lactococcus lactis subsp. lactis ATCC 7962 pts HI Operon

  • The ptsH and ptsI genes of Lactococus lactis subsp. lactis ATCC 7962 (L. lactis 7962), encoding the general proteins of phosphotransferase system (PTS) components, HPr and enzyme I, respectively, were cloned and characterized. A 1.3 kb PCR product was obtained using a primer set that was hybridized to the internal region of the L. lactis 7962 pts HI genes and then subcloned into a low-copy number vector, pACYC184. The 5' upstream and 3' downstream region from the 1.3 kb fragment were subsequently clone using the chromosome walking method. The complete ptsHI operon was constructed and the nucleotide sequences determined. Two ORFs corresponding to HPr (88 amino acids) and enzyme I (575 amino acids) were located. The ptsHI genes of L. lactis 7962 showed a very high homology (84-90%) with those genes from other Gram-positive bacteria. A primer extension analysis showed that the transcription started at either one of two adjacent bases upstream of the start codon. Using a Northern analysis, two transcripts were detected; the first, a 0.3 kb transcript corresponding to ptsH and the second, a 2 kb transcript corresponding to ptsH and ptsI. The transcription level of ptsH was higher than that of ptsI. The concentration of the ptsH transcript in cells grown on glucose was similar to that in cells grown on lactose, yet higher than that in cells grown on galactose. The ptsI transcript was scarcely detected in cell grown on lactose or galactose. The ptsI transcript was scarcely detected in cells grown on lactose or galactose. The results of a sequence analysis and Northern blot confirmed that the ptsH and ptsI genes of L. lactis 7962 were arranged in an operon like other known ptsHI genes and the expression of the ptsHI genes was regulated at the transcriptional level in response to the carbon source.

모바일 오피스 환경을 위한 스마트폰 레코딩 시스템 (SmartPhone Recording System for Mobile Office Environment)

  • 최근 스마트폰의 발전에도 불구하고 통화 녹취 기술은 대부분 사내 유선망을 이용하여 통화내용을 서버에 저장, 검색 및 청취할 수 있도록 되어 있다. 따라서 본 논문은 스마트폰의 장점인 이동성을 활용하여 모바일간의 통화 및 모바일과 유선상의 통화내용을 녹취할 수 있는 안드로이드 기반의 스마트폰 레코딩 시스템을 소개한다. 스마트폰 레코딩 시스템은 어플리케이션이 설치된 스마트폰을 이용하여 통화가 이루어지면 단말기에 통화내용을 저장하고 사용자 권한에 의해 암호화되어 웹 서버에 업로드 할 수 있다. 업로드된 녹취 데이터는 웹을 통하여 관리할 수 있다. 이 시스템을 통하여 이동 업무에 적용가능하고 증거자료 확보 및 보이스 피싱과 같은 범죄를 예방하는데 활용될 수 있을 것이다.

Development of a Transformation System for the Medicinal Fungus Sanghuangporus baumii and Acquisition of High-Value Strain

  • To further explore the molecular mechanism of triterpenoid biosynthesis and acquire high-value strain of Sanghuangporus baumii, the Agrobacterium tumefaciens-mediated transformation (ATMT) system was studied. The key triterpenoid biosynthesis-associated gene isopentenyl diphosphate isomerase (IDI) was transformed into S. baumii by ATMT system. Then, the qRT-PCR technique was used to analyze gene transcript level, and the widely targeted metabolomics was used to investigate individual triterpenoid content. Total triterpenoid content and anti-oxidant activity were determined by spectrophotometer. In this study, we for the first time established an efficient ATMT system and transferred the IDI gene into S. baumii. Relative to the wild-type (WT) strain, the IDI-transformant (IT) strain showed significantly higher transcript levels of IDI and total triterpenoid content. We then investigated individual triterpenoids in S. baumii, which led to the identification of 10 distinct triterpenoids. The contents of individual triterpenoids produced by the IT2 strain were 1.76-10.03 times higher than those produced by the WT strain. The triterpenoid production showed a significant positive correlation with the IDI gene expression. Besides, IT2 strain showed better anti-oxidant activity. The findings provide valuable information about the biosynthetic pathway of triterpenoids and provide a strategy for cultivating high-value S. baumii strains.

Cloning and characterization of the psbA Gene from Panax ginseng(Characterization of the psbA Gene from P. ginseng)

  • The psbA gene of photo system II was cloned and characterized from the P. ginseng chloroplast. The psbA gene is composed of 1,062 nucleotides. The overall amino acid sequence shows 99% and 98% identities to dicots and monocots of higher plants, respectively. Southern blot analysis revealed that a single copy of the psbA gene existed in the chloroplast genome. Northern blot analysis of the in vivo accumulation of the psbA transcript, after being grown under the different intensities (5%, 10%, 20%, and 100%) of daylight, indicated that the steady-state level of the psbA transcript was not significantly affected by light intensity.

Text-driven Speech Animation with Emotion Control

  • In this paper, we present a new approach to creating speech animation with emotional expressions using a small set of example models. To generate realistic facial animation, two example models called key visemes and expressions are used for lip-synchronization and facial expressions, respectively. The key visemes represent lip shapes of phonemes such as vowels and consonants while the key expressions represent basic emotions of a face. Our approach utilizes a text-to-speech (TTS) system to create a phonetic transcript for the speech animation. Based on a phonetic transcript, a sequence of speech animation is synthesized by interpolating the corresponding sequence of key visemes. Using an input parameter vector, the key expressions are blended by a method of scattered data interpolation. During the synthesizing process, an importance-based scheme is introduced to combine both lip-synchronization and facial expressions into one animation sequence in real time (over 120Hz). The proposed approach can be applied to diverse types of digital content and applications that use facial animation with high accuracy (over 90%) in speech recognition.

회재(晦齋) 『중용구경연의(中庸九經衍義)』의 경세론적 이해 (A Confucius Political Economics Based Understanding of the Chungyongkugyongyonui(中庸九經衍義))

  • 이 논문은 회재(晦齋) 이언적(李彦迪, 1491~1553)의 "중용구경연의"를 고찰한 것이다. 주지하다시피 회재는 16세기를 대표하는 사상가 가운데 한 사람으로, 일찍이 조한보(曺漢輔, ?~?) 사이에 벌어졌던 태극논쟁(太極論爭)은 주자성리학(朱子性理學)을 조선에 정착시키는데 큰 역할을 하였으며, 아울러 지치주의(至治主義)의 이론적 체계를 확립한 학자이기도하다. "대학"과 "중용"에 담긴 지치주의 이념을 군주(君主)에게 진헌하고자 저술된 책이 바로 회재의 "대학장구보유"와 "중용구경연의"이다. 회재는 수기(修己)의 도(道)에 속하는 조목에 있어서 "대학"이 "중용"보다 더 체계적이고 자세한 반면에 치인(治人)의 도(道)에 해당하는 조목은 "중용"이 "대학" 보다 더 구체적이라고 하여 '표리(表裏)의 관계'로 파악하고, 이 둘을 함께 보아야만 제왕위치(帝王爲治)의 규모가 갖추어 진다고 하였다. 특히 "중용구경연의"는 송나라 진덕수(眞德秀)의 "대학연의(大學衍義)"와 명나라 구준(丘濬)의 "대학연의보(大學衍義補)"를 모방하여 당시의 군주였던 명종(明宗)으로 하여금 이제 삼왕(二帝三王)의 지치(至治)를 실현시킬 목적으로 저술된 책으로, 이는 퇴계의 "성학십도"와 율곡의 "성학집요"에 많은 영향을 끼쳤다.

High Copy Rme1p Suppresses Iron-Induced Cell Growth Defect of Saccharomyces cerevisiae

  • In the yeast Saccharomyces cerevisiae, iron can be toxic. Because of this phenomenon, its metabolism of iron is strictly regulated. We have constructed a model system in which cell growth is defected during periods of iron over-load. When $Aft1-1^{up}$ protein was overexpressed with Ga110 promoter, a galactose inducible promoter, cell growth was defected and levels of CLN2 transcript decreased. However transcript levels of AFT1 and FET3 genes increased over time in a consistent manner throughout the course of $AFT1-1^{up}$ overexpression. We have screened to find genes to suppress cell growth defect by iron overload with YEp-derived high copy yeast genomic DNA library and found that high copy of Rmelp suppressed cell growth defects. Rme1p has been known as an activator protein of CLN2 gene expression. Taking these results together, we suggest that the yeast cell cycle is arrested at the $G_1$, phase by iron overload via Cln2p.

Structural Analysis and Transcriptional Regulation of the Chloroplast psbC Gene from Panax ginseng

  • The psbC gene, encoding the intrinsic chlorophyll-binding protein of CP43, one of the PS core complex polypeptides, was cloned from the Panax ginseng chloroplast, which is composed of 1,422 nucleotides and the overall nucleotide sequence shows more than 84% identity to those of eukaryotic photosynthetic organisms. The predicted topology of CP43, based on hydropathy analysis, includes six membrane-spanning ${\alpha}-helices$ resulting in three lumenal and four stromal loops. The putative translation start codon for the psbC gene is located at 48 nucleotides upstream from the stop codon of the psbD gene whose product is also a component of the PSII reaction center, implying that the promoter of the psbC gene is possibly located in the middle of the structural gene of the psbD gene. Northern blot analysis of the in vivo accumulation of the psbC transcript from the plants grown under the various growth light intensities (5%, 10%, 20%, and 100%) of daylight indicated that the steady-state level of the psbC transcript was not significantly affected by light intensity.
