Effects of several herbs and Bogol-Tang on the experimental osteoporosis of rat (수종(數種)의 한약재(韓藥材)와 보골탕(補骨湯)이 흰쥐의 실험적(實驗的) 골다공증(骨多孔症)에 미치는 영향(影響))

  • Objective : The purpose of this studies was to determine the effect of Carthami semen, Bogol-Tang and Rhynchosia volubilis Lour. on changes of trabecular area and physiological metabolites in the ovariectomized osteoporotic Rats. Methods : In order to induce estrogen deficient osteoporosis, ovariectomy was done on rats. Then the Carthami semen, Bogol-Tang and Rhynchosia volubilis Lour. were orally administerd: Such indexes were measured as the changes of body weight, bone mineral density, trabecular area in tibia, and levels of osteocalcin, bone alkaline phosphatase, calcium, phosphorus, cholesterol, triglyceride, aspartate aminotransferase and alanine aminotransferase in serum. Results : 1. The change of bone mineral density in Bogol-Tang group and Rhynchosia volubilis lour. group. was significantly increased compare to control group. 2. The change of trabecular area % in epiphysis of tibia in Carthami semen group was significantly increased compare to control group. 3. The change of trabecular area % in diaphysis of tibia in Bogol-Tang group was significantly increased compare to control group. 4. The change of serum osteocalcin in Bogol-Tang group was significantly decreased compare to control group. 5. The change of serum bone alkaline phosphatase in Bogol-Tang group was significantly decreased compare to control group. 5. The change of serum bone alkaline phosphatase in Bogol-Tang group was significantly decreased compare to control group. 6. The change of1 serum calcium in Rhynchosia volubilis lour. group was significantly decreased compare to control group. 7. Evaluation of the Efficacy of Zoledronic Acid and Amifostine on Radiation-induced Bone Loss in Mice

  • Background: This study investigated the effects of zoledronic acid (ZA) on radiation-induced bone loss in C3H/HeN mice. Materials and Methods: C3H/HeN mice were divided into sham control and three irradiated groups (3 Gy, gamma ray). The irradiated mice were treated for 12 weeks with vehicle, amifostine (intraperitoneal injection), or ZA (subcutaneous injection). Grip strength, uterus weight, and serum alkaline phosphatase (ALP), and tartrate-resistant acid phosphatase (TRAP) levels were measured. Tibiae were analyzed using micro-computed tomography. Results and Discussion: Treatment of ZA ($100{\mu}g{\cdot}kg^{-1}{\cdot}week^{-1}$) significantly preserved trabecular bone volume, trabecular thickness, trabecular number, trabecular separation, bone mineral density of proximal tibia metaphysic, and cortical bone volume, but did not alter the uterus weight of the mice. The administration of ZA for 12 weeks lowered serum ALP and TRAP levels in irradiated mice, suggesting that ZA can reduce the bone turnover rate in mice. No differences were apparent between the amifostine-treated group and the irradiation control group. Conclusion: The results indicate that ZA can prevent radiation-induced bone loss in mice.

Correlations between Acoustic Properties and Bone Mineral Density in Bovine Femoral Trabecular Bone In Vitro (생체 외 조건의 소 대퇴골 해면질골에서 음향특성과 골밀도 사이의 상관관계)

  • The purpose of the present study is to investigate the correlations between acoustic properties, such as speed of sound and normalized broadband ultrasound attenuation, and bone mineral density in femur with high fracture risk. The speed of sound and the normalized broadband ultrasound attenuation in 15 bovine femoral trabecular bone samples in vitro were measured by using a through-transmission method with two matched pairs of ultrasonic transducers with center frequencies of 0.5 and 1.0 MHz. The volumetric bone mineral density of the trabecular bone samples was measured by using micro-computed tomography. The bone mineral density exhibited strong correlations with both the speed of sound and the normalized broadband ultrasound attenuation measured by using the 0.5 and the 1.0 MHz transducers. The highest correlation was found between the bone mineral density and the normalized broadband ultrasound attenuation measured by using the 0.5 MHz ultrasonic transducers. The results suggest that the acoustic properties measured in the femur in vitro can be used as indices for the prediction of femoral bone mineral density.

Ultrasound Attenuation in the Assessment of Bone Mineral Density and Elastic Modulus of Human Trabecular Bone

  • The objective of this study was to re-evaluate ultrasound attenuation as an indicator of bone properties. Ultrasound attenuation(BUA), were measured in the three orthogonal directions of trabecular bone cubes, Measurements of bone mineral density(BMD) were made using quantitative computed tomography and apparent density by weighing bone specimens and measuring their volume. Ultrasonic modulus was calculated from the standard equation with apparent density and ultrasound velocity. Ultrasound attenuation at a frequency of 0.5 MHz and BUA were correlated with BMD and ultrasonic modulus in the anterior/posterior, medial/lateral, and superior/inferior directions. Analysis of correlations demonstrated that attenuation at 0.5 MHz was superior to BUA in describing both BMD and elastic modulus of trabecular bone. This result may be used to improve current ultrasound diagnostic techniques for assessing bone status.

St. John's Wort (Hypericum perforatum) stimulates human osteoblastic MG-63 cell proliferation and attenuates trabecular bone loss induced by ovariectomy

  • BACKGROUND/OBJECFTIVES: The effect of St. John's Wort extract (SJW) on MG-63 cell proliferation and trabecular bone loss induced by ovariectomy was examined. MATERIALS/METHODS: Proliferation, expression of estrogen receptor (ER) ${\alpha}$ and ER ${\beta}$, and gene expressions of osteoprotegerin (OPG), osteocalcin (OC) and alkaline phosphatase (ALP) were examined in MG-63 cells treated with or without SJW. Ovariectomized rats were treated with SJW at the dose of 100 or 200 mg/kg/day, ${\beta}$-estradiol-3-benzoate (E2), or vehicle only (OVX-C), and sham operated rats were treated with vehicle only (Sham-C). Serum ALP and C-telopeptide (CTX), and femoral trabecular bone loss were examined. RESULTS: SJW increased MG-63 cell proliferation and expression of ER ${\alpha}$ and ER ${\beta}$, and positive effect was shown on gene expressions of ALP, OC and OPG. SJW also showed estrogen like effect on bone associated with slowing down in trabecular bone loss. Histopathology by H&E showed rats treated with SJW displayed denser structure in metaphyseal region of distal femur compared with rats in OVX-C. SJW was shown to reduce serum CTX in OVX rats. CONCLUSION: The present study provides new insight in preventing estrogen deficiency induced bone loss of SJW and possibility for its application in bone health supplement.

Bone Microarchitecture at the Femoral Attachment of the Posterior Cruciate Ligament (PCL) by Texture Analysis of Magnetic Resonance Imaging (MRI) in Patients with PCL Injury: an Indirect Reflection of Ligament Integrity

  • Purpose: (1) To evaluate the trabecular pattern at the femoral attachment of the posterior cruciate ligament (PCL) in patients with a PCL injury; (2) to analyze bone microarchitecture by applying gray level co-occurrence matrix (GLCM)-based texture analysis; and (3) to determine if there is a significant relationship between bone microarchitecture and posterior instability. Materials and Methods: The study included 96 patients with PCL tears. Trabecular patterns were evaluated on T2-weighted MRI qualitatively, and were evaluated by GLCM texture analysis quantitatively. The grades of posterior drawer test (PDT) and the degrees of posterior displacement on stress radiographs were recorded. The 96 patients were classified into two groups: acute and chronic injury. And 27 patients with no PCL injury were enrolled for control. Pearson's correlation coefficient and one-way ANOVA with Bonferroni test were conducted for statistical analyses. This protocol was approved by the Institutional Review Board. Results: A thick and anisotropic trabecular bone pattern was apparent in normal or acute injury (n = 57/61;93.4%), but was not prominent in chronic injury and posterior instability (n = 31/35;88.6%). Grades of PDT and degrees of posterior displacement on stress radiograph were not correlated with texture parameters. However, the texture analysis parameters of chronic injury were significantly different from those of acute injury and control groups (P < 0.05). Conclusion: The trabecular pattern and texture analysis parameters are useful in predicting posterior instability in patients with PCL injury. Evaluation of the bone microarchitecture resulting from altered biomechanics could advance the understanding of PCL function and improve the detection of PCL injury.

Anti-Menopausal Effect of Heat-Killed Bifidobacterium breve HDB7040 via Estrogen Receptor-Selective Modulation in MCF-7 Cells and Ovariectomized Rats

  • Menopause is induced by spontaneous ovarian failure and leads to life quality deterioration with various irritating symptoms. Hormonal treatment can alleviate these symptoms, but long-term treatment is closely associated with breast and uterine cancer, and stroke. Therefore, developing alternative therapies with novel anti-menopausal substances and improved safety is needed. In our study, heat-killed Bifidobacterium breve HDB7040 significantly promoted MCF-7 cell proliferation in a dose-dependent manner under estrogen-free conditions, similar to 17β-estradiol. This strain also triggered ESR2 expression, but not ESR1, in MCF-7 cells. Moreover, administrating HDB7040 to ovariectomized (OVX) Sprague-Dawley (SD) female rats reduced estrogen deficiency-induced weight gain, fat mass, blood triglyceride, and total cholesterol levels. It also recovered collapsed trabecular microstructure by improving trabecular morphometric parameters (bone mineral density, bone volume per tissue volume, trabecular number, and trabecular separation) and decreasing blood alkaline phosphatase levels with no significant changes in uterine size and blood estradiol. HDB7040 also significantly regulated the expression of Tff1, Pgr, and Esr2, but not Esr1 in uteri of OVX rats. Heat-killed B. breve HDB7040 exerts an anti-menopausal effect via the specific regulation of ERβ in vitro and in vivo, suggesting its potential as a novel substance for improving and treating menopausal syndrome.

Changes in the fractal dimension of peri-implant trabecular bone after loading: a retrospective study

  • Purpose: To assess bony trabecular changes potentially caused by loading stress around dental implants using fractal dimension analysis. Methods: Fractal dimensions were measured in 48 subjects by comparing radiographs taken immediately after prosthesis delivery with those taken 1 year after functional loading. Regions of interest were isolated, and fractal analysis was performed using the box-counting method with Image J 1.42 software. Wilcoxon signed-rank test was used to analyze the difference in fractal dimension before and after implant loading. Results: The mean fractal dimension before loading ($1.4213{\pm}0.0525$) increased significantly to $1.4329{\pm}0.0479$ at 12 months after loading (P<0.05). Conclusions: Fractal dimension analysis might be helpful in detecting changes in peri-implant alveolar trabecular bone patterns in clinical situations.

A Study of Osteoporosis Prediction using Morphological Measuring of Proximal Femoral Part and Trabecular Characteristics Based on Femoral Radiographic Image (대퇴부 방사선영상에서 대퇴골 근위부의 형태학적 측정과 골소주의 특성을 이용한 골다공증 예측에 관한 연구)

  • This study was designed to examine the morphological measurement and characteristics of trabecullae based on femoral radiographic image for prediction of osteoporosis. Study subjects were 34 females (average age of 62.1 years) and 6 males (average age of 60.1 years), they were categorized into normal group and osteoporosis group in accordance with the T-score value. Measurement of the bone density of femoral bone was measured with DEXA(Dual Energy X-ray absorptiometry). ROI(Region of interests) was selected on femoral neck and trochanter. Characteristics of trabecullae was analyzed by using the skeletonization analysis of trabecular image. Morphological measurement was analyzed through femoral radiographic image in order to examine the correlation with osteoporosis. The result demonstrated statistically significant correlation between neck cortical thickness, shaft width, shaft cortical thickness, periphery, mean gray level and trabeculae area with BMD average (T-score) of femoral part. The results show that morphological measurement and characteristics of trabecullae based on femoral radiographic images for osteoporosis prediction could be effective.

Relationship between the morphologic features of alveolar trabecular bone and systemic osteoporosis (치조 골소주 모폴로지 특성과 골다공증의 연관관계에 대한 연구)

  • Purpose : The purpose of this study was to investigate the preliminary use of morphologic operation (MO) in analyzing trabecular pattern of alveolar bone for the predicting systemic osteoporosis. Materials and Methods : Study subjects consisted of 35 females (average age 48.5 years) and 25 males (average age 25.8 years). Bone mineral density BMD $(grams/cm^2)$ of lumbar spine and proximal femur of these subjects were measured by a dual energy X-ray absorptiometry (DEXA). Regions of interest (ROIs) were selected from the digitized periapical radiographs of subjects' posterior jaw. A custom computer program processed morphology operations of ROIs. We compared mean values of 11 MO variables according to the osteoporotic group divided by the T-scores of DEXA. We also studied correlation between radiographic density and these MO variables. Results : The mean radiographic densities insignificantly correlated with MO variables. There were statistically significant differences among the values of 9 MO variables according to the osteoporotic group. Conclusion Morphologic operation can be effective in analyzing trabecular pattern of alveolar bone for the predicting osteoporosis.
