A magnetic resonance imaging study on changes in rat mandibular bone marrow and pulp tissue after high-dose irradiation

  • Purpose: This study was designed to evaluate whether magnetic resonance imaging (MRI) is appropriate for detecting early changes in the mandibular bone marrow and pulp tissue of rats after high-dose irradiation. Materials and Methods: The right mandibles of Sprague-Dawley rats were irradiated with 10 Gy (Group 1, n=5) and 20 Gy (Group 2, n=5). Five non-irradiated animals were used as controls. The MR images of rat mandibles were obtained before irradiation and once a week until week 4 after irradiation. From the MR images, the signal intensity (SI) of the mandibular bone marrow and pulp tissue of the incisor was interpreted. The MR images were compared with the histopathologic findings. Results: The SI of the mandibular bone marrow had decreased on T2-weighted MR images. There was little difference between Groups 1 and 2. The SI of the irradiated groups appeared to be lower than that of the control group. The histopathologic findings showed that the trabecular bone in the irradiated group had increased. The SI of the irradiated pulp tissue had decreased on T2-weighted MR images. However, the SI of the MR images in Group 2 was high in the atrophic pulp of the incisor apex at week 2 after irradiation. Conclusion: These patterns seen on MRI in rat bone marrow and pulp tissue were consistent with histopathologic findings. They may be useful to assess radiogenic sclerotic changes in rat mandibular bone marrow.

The relationship between 3D bone architectural parameters and elastic moduli of three orthogonal directions predicted from finite elements analysis (돼지 하악 과두의 해면골에서 유한요소분석법으로 예측한 방향에 따른 탄성율과 3차원 골 미세지표 간의 상관관계)

  • Purpose: To investigate the relationship between 3D bone architectural parameters and direction-related elastic moduli of cancellous bone of mandibular condyle. Materials and Methods: Two micro-pigs (Micro-$pig^R$, PWG Genetics Korea) were used. Each pig was about 12 months old and weighing around 44 kg. 31 cylindrical bone specimen were obtained from cancellous bone of condyles for 3D analysis and measured by micro-computed tomography. Six parameters were trabecular thickness (Tb. Th), bone specific surface (BS/BV), percent bone volume (BV/TV), structure model index (SMI), degree of anisotropy (DA) and 3-dimensional fractal dimension (3DFD). Elastic moduli of three orthogonal directions (superior-inferior (SI), medial-lateral (ML), andterior-posterior (AP) direction) were calculated through finite element analysis. Results: Elastic modulus of superior-inferior direction was higher than those of other directions. Elastic moduli of 3 orthogonal directions showed different correlation with 3D architectural parameters. Elastic moduli of SI and ML directions showed significant strong to moderate correlation with BV/TV, SMI and 3DFD. Conclusion: Elastic modulus of cancellous bone of pig mandibular condyle was highest in the SI direction and it was supposed that the change into plate-like structure of trabeculae was mainly affected by increase of trabeculae of SI and ML directions.

Morphological Changes of Bones and Joints with Rheumatoid Arthritis and Osteoarthritis

  • Arthritis is a common disease in aged people, and is clinically divided into rheumatoid arthritis (RA) and osteoarthritis (OA). Although common symptoms such as pain are present, the underlying pathological mechanisms are slightly different. Therefore, the objectives of the present study were to compare joint damage induced by RA and OA by analyzing the major morphological and molecular differences, and to propose a suitable therapeutic intervention based on the pathophysiological conditions of bones and joints. For the RA animal model, 8-week-old DBA1/J mice were immunized with bovine type II collagen emulsified in complete Freund's adjuvant (CFA). Normal C57BL/6 mice (over 2 years of age) were used for OA. The clinical arthritis score was calculated using a subjective scoring system, and paw thicknesses were measured using calipers. The serum TNF ${\alpha}$ level was analyzed using an ELISA kit. Micro-CT was used to identify pathological characteristics and morphological changes. In collagen-induced RA mice, there were increased ankle joint volumes and clinical scores (p<0.01). The concentration of TNF ${\alpha}$ was significantly increased from 3 to 7 weeks after immunization. Micro-CT images showed trabecular bone destruction, pannus formation, and subchondral region destruction in RA mice. OA among aged mice showed narrowed joint spaces and breakdown of articular cartilage. This study suggests that a careful therapeutic intervention between RA and OA is required, and it should be based on morphological alteration of bone and joint.

  • Glaucoma is an eye disease which is caused by abnormal high IOP (Intra Ocular Pressure). High IOP is caused by the aqueous humor which is produced consistently but not drained due to malfunction of the trabecular system which has a role of draining the aqueous humor into the venous system. Currently, there are three methods to treat glaucoma-using medicines, surgical operation, and using implant device. The first and second methods are not long acting, so the use of implants is increasing in these days in order to drain out the aqueous humor compulsory. However, though conventional implants have a capability of pressure regulation, they cannot maintain IOPs desired for different patients, and too much aqueous humor are usually drained, to cause hypotony. To solve these problems, it is needed to develop a new implant which is capable of controling the IOP actively and copes with personal difference of patients. An active glaucoma implant consists of the valve actuator, pressure sensor, controller, and power supply. In this paper, firstly, we make an analysis of the operation of a conventional implant using a bond graph and show defects and limitations of the conventional valve analytically. Secondly, we design and analyze a valve actuator considering actuation principles, resistance elements, control methods, and energy sources focused on power saving problem. Finally, using simulations the possibility of the proposed valve actuator is investigated.

Study of bony trabecular characteristics using bone morphometry and micro-CT (골형태분석법과 micro-CT를 이용한 골소주 특성에 대한 연구)

  • Purpose : The research was done to investigate the effectiveness of 2D bony morphometry and microstructure of micro-computed tomography (micro-CT) on the osteoporotic bony change. We performed the bone morphometric analysis of proximal femur in ovariectomized rabbits with BMD and micro-CT examination. Materials and Methods : Twenty-one female (Newzealand, about 16 weeks old, 2.9-3.4kg) rabbits were used. Three rabbits were sacrificed on the day when experiment began (Baseline). The remaining 18 rabbits were divided into two groups. One group was ovariectomized bilaterally (OVX) and the other animals were subjected to sham operation (Sham). Bone specimens were obtained from the right and left femur of sacrificed rabbits. At intervals of 1, 2, 3, 5, 6 months respectively, BMD tests were performed on the proximal femur by using PIXlmus 2 (GE Lunar Co. USA), 2-dimensional bone morphometric analysis by custome computer program and 2D/3D bone structure analysis by micro-CT (Skyscan 1072, Antwerpen, Belgium). Statistical analysis was carried out for the correlation between bone morphometry, micro-CT and BMD Result : BV/TV, Tb.Th, Tb.N of micro-CT parameters showed higher values in sham group than OVX group. N.Nd/Ar.RI, N.NdNd, N.NdTm, N. TmTm, PmB/Ar.RI, 3-D BoxSlope of 2D morphometric parameters showed higher values in Sham group than OVX group. The micro-CT parameters of Tb.Sp, Tb.N were statistically significant correlated with BMD respectively. Several 2D morphometric parameters were statistically significant correlated with BMD respectively. Conclusion : Several parameters of 2D bony morphometry and micro-CT showed effective aspects on the osteoporotic bony change.

Analysis on correlation between bone strength by FEA, micro-CT parameters and bone mineral density (유한요소법에 의한 골강도와 micro-CT 지표 및 골밀도간의 상관관계)

  • Purpose : Bone mineral density (BMD) and bone microarchitecture are important determinants for bone strength. Recently micro-CT have provided possibilities for measuring a variety of structural indices to characterize bone microarchitecture. The objective of this study was to compare the BMD and micro-CT parameters with Young's modulus calculated by finite element analysis (FEA) for the evaluation of bone strength. Materials and Methods Bone specimens were obtained from the 18 female rabbits aged 16 weeks. Of those, 36 samples (right and left femur) were selected for 3D micro-CT analysis $(ANT^{TM},\;SKYSCAN,\;Belgium)$ and BMD by PIXlmus 2 (GE Lunar Co. USA). Five microstructural parameters of micro-CT, such as trabecular thickness (Tb.Th), bone specific surface (BS/BV), percent bone volume (BV/TV), structure model index (SMI) and degree of anisotropy (DOA) were studied. Young's modulus was obtained by software program (ANSYS 9.0, ANSYS Inc, Canonsburg, PA) based on micro-CT three dimensional images. Results : Young's modulus assessed by FEA correlated significantly with Tb.Th, BV/TV, BS/BV and SMI respectively. Young's modulus showed higher correlation with these rnicrostructural parameters of micro-CT than BMD. Microstructural parameters except DOA showed significant correlations within the examined group. Conclusion The microarchitectural parameters o( micro-CT and BMD represented some informations in the evaluation of bone strength assessed by FEA.

A prospective study on the effectiveness of newly developed autogenous tooth bone graft material for sinus bone graft procedure

  • PURPOSE. The purpose of this prospective study was to evaluate the effectiveness of newly developed autogenous tooth bone graft material (AutoBT)application for sinus bone graft procedure. MATERIALS AND METHODS. The patients with less than 5.0 mm of residual bone height in maxillary posterior area were enrolled. For the sinus bone graft procedure, Bio-Oss was grafted in control group and AutoBT powder was grafted in experimental group. Clinical and radiographic examination were done for the comparison of grafted materials in sinus cavity between groups. At 4 months after sinus bone graft procedure, biopsy specimens were analyzed by microcomputed tomography and histomorphometric examination for the evaluation of healing state of bone graft site. RESULTS. In CT evaluation, there was no difference in bone density, bone height and sinus membrane thickness between groups. In microCT analysis, there was no difference in total bone volume, new bone volume, bone mineral density of new bone between groups. There was significant difference trabecular thickness ($0.07{\mu}m$ in Bio-Oss group Vs. $0.08{\mu}m$ in AutoBT group) (P=.006). In histomorphometric analysis, there was no difference in new bone formation, residual graft material, bone marrow space between groups. There was significant difference osteoid thickness ($8.35{\mu}m$ in Bio-Oss group Vs. $13.12{\mu}m$ in AutoBT group) (P=.025). CONCLUSION. AutoBT could be considered a viable alternative to the autogenous bone or other bone graft materials in sinus bone graft procedure.

Effects of Diet of Korean Safflower(Carthamus tinctorious L.) Seed Powder on Bone Tissue in Rats during the Recovery of Rib Fracture (토종홍화씨의 급여가 실험동물의 늑골골절 회복중 골조직에 미치는 영향)

  • This study was carried out to investigate the effects of diet of Korean safflower(Carthamus tinctorious L.) seed powder on bone tissue during the recovery of rib-fracture in rats. Male Sprague-Dawley rats of 10 weeks old, weighing 370$\pm$5g, were divided into two groups including the control group(C group, AIN-76 semipurified diet) and safflower seed group(S group, AIN-76 semipurified diet+10% safflower seed powder) and were fed experimental diets for 12 days after adaptation period. After this period, the 9th right rib was fractured surgically and sham-operation was also performed. Rats were fed with experimental diets for up to 30 more days after rib-fracture. The degree of bone repair was evaluated during the recovery period at the 8th, 11th, 16th, 21st, 30th days after the surgical operation by microscopic observation of the fractured rib tissue. In callus formation, the portion of hyaline cartilage was noticably higher in S group than C group. The intracatilagenous ossification was observed at the 8th day in S group, but at 11th day in C group. The intramembranous ossification in callus was widely found over the 8th day to the 11th day in S group, but it was shown over the 11th day to the 16th day in C group. Bone resorption was also occured more rapidly in S group as indicated by large numbers of osteoclasts observed. At the 30th day, most of trabecular bones were disappeared in S group, whereas still shwon in C group over wide ranges of fractured ribs. These results imply that the supplementation of Korean safflower seed powder influences in the recovery of bone fracture by accelerating the process of bone repair.

Consideration of Lateral Cortical Bone Thickness and IAN Canal Location During Mandibular Ramus Bone Grafting for Implant Placement

  • Purpose: This study aimed at examining the thickness of lateral cortical bone in the mandibular posterior body and the location of the inferior alveolar nerve canal as well as investigating the clinically viable bone grafting site(s) and proper thickness of the bone grafts. Subjects and Methods: The study enrolled a total of 49 patients who visited the Department of Oral and Maxillofacial Surgery at Kyung Hee University Dental Hospital to have their lower third molar extracted and received cone beam computed tomography (CBCT) examinations. Their CBCT data were used for the study. The thickness of lateral cortical bone and the location of inferior alveolar nerve canal were each measured from the buccal midpoint of the patients' lower first molar to the mandibular ramus area in the occlusal plane of the molar area. Results: Except in the external oblique ridge and alveolar ridge, all measured areas exhibited the greatest cortical bone thickness near the lower second molar area and the smallest cortical bone thickness in the retromolar area. The inferior alveolar nerve canal was found to be located in the innermost site near the lower second molar area compared to other areas. In addition, the greatest thickness of the trabecular bone was found between the inferior alveolar nerve canal and the lateral cortical bone. Conclusions: In actual clinical settings involving bone harvesting in the posterior mandibular body, clinicians are advised to avoid locating the osteotomy line in the retromolar area to help protect the inferior alveolar nerve canal from damage. Harvesting the bone near the lower second molar area is judged to be the proper way of securing cortical bone with the greatest thickness.

The effect of combined Rehmannia glutinosa Libosch and Eleutherococcus senticosus Max (OPB) extracts on bone mineral density in ovariectomized rats.

  • This study was conducted to investigate the preventing effects of OPB (Rehmannia glutinosa Libosch and Eleutherococcus senticosus Max extracts) and combined OPB/Calcium therapy on bone loss in ovariectomized rats. Sixty Sprague Dawley rats of 12-week-old were divided into eight groups: OVX (ovariectomized), OPBL (OPB 50 mg/kg), OPBM (OPB100 mg/kg), OPBH (OPB 200 mg/kg), OPBL/CAL(OPBL+CAL), OPBM/CAL (OPBM+CAL), OPBH/CAL (OPBH+CAL) and CAL (Calcium citrate 88.33 mg/kg+1$\alpha$, 25-dihydroxy-vitamin $D_3$ 33.33 IU/kg). Bone mineral density (BMD), bone mineral content (BMC), bone strength indices and cortical thickness were analyzed by peripheral quantitative computerized tomography (pQCT). pQCT scanning showed that OVX induced a significant decrease in trabecular bone mineral density and bone mineral content in the proximal tibia $(-36.4\pm2.4%,\;-21.8\pm12.7%)$. These decreases were significantly prevented by the administration of OPBM and OPBM/CAL. Cortical BMD and BMC of tibia were slightly enhanced by OPB and OPB/CAL. However there was no significant difference between OVX and OPB, OPB/CAL treated group. Bone strength indices and cortical thickness were not significantly different. Our results suggest that OPB and combined OPB/Calcium therapy are effective in preventing the development of bone loss induced by ovariectomy in rats.
