HYTE (Hyeonggaeyeongyotang Extract), a polyherbal formula has been used as folk medicine, 28days repeat oral dose toxicity was tested in SD rats according to KFDA Guideline[2005-60]. Methods : In this study, mortality, clinical signs, body weight and gains, food and water consumption, ophthalmologic observation, urinalysis, hematology, serum biochemistry, gross findings, organ weight and histopathological observations were conducted during 28days of dosing periods. Results: 1. No HYTE treatment-related mortalities and clinical signs were detected in all dosing levels tested in male and female rats during the whole experimental periods. 2. No HYTE treatment-related changes on body weight, gains and food consumption were detected in all dosing levels tested in male and female rats during the whole experimental periods except for 2000mg/kg-dosing female groups in which significantly increase of body weight, gains, food and water consumption were detected compared to that of vehicle control in some points. 3. No HYTE treatment-related changes on ophthalmologic examination were detected in all dosing levels tested in male and female rats. 4. No HYTE treatment-related changes on urinalysis were detected in all dosing levels tested in male and female rats except for 2000mg/kg-dosing female groups in which, significantly increase of urine volume and related decrease on the urine specific gravity were detected as secondary effects of increase on the water consumptions not HYTE treatment-related toxicological signs. 5. No HYTE treatment-related changes on hematology were detected in all dosing levels tested in male and female rats except for increases in the total WBC count and lymphocytes of 2000mg/kg-dosing male and female groups with decrease of large unstained cells as pharmacological effects of immune enhancements not HYTE treatment-related toxicological signs. 6. No HYTE treatment-related changes on serum biochemistry were detected in all dosing levels tested in male and female rats. 7. No HYTE treatment-related changes on gross findings, organ weight and histopathology were detected in all dosing levels tested in male and female rats except for 2000mg/kg-dosing male and female groups in which, spleen and thymus organ weights, hypertrophy at gross observation and hyperpalsia of lymphoid cells and follicles at histopathological observation in spleen and thymus were detected as pharmacological effects of immune enhancements not HYTE treatment-related toxicological signs. Conclusions : Based on these results, the NOAEL and MTD of HYTE in SD rats were considered as over 2000mg/kg, respectively at 28days repeat oral dose toxicity test because most of these findings were considered as results of pharmacological effects of immune enhancements not HYTE treatment-related toxicological signs or secondary effects.
Objectives: This study aims to compare the physicochemical characteristics, toxicological data with Occupational Exposure Limits (OELs) of chemicals under the Occupational Safety and Health Act(OSHA) regulated by the Ministry of Employment and Labor of Korea. Methods: Information on chemicals which have OELs on physicochemical characteristics and toxicological data was collected using Material Safety Data Sheet(MSDS) from Korea Occupational Safety and Health Agency(KOSHA) and the Korea Information System for Chemical Safety Management(KISChem) in 2014. Statistical analyses including correlation and simple regression were performed to compare the OELs with chemical characteristics including molecular weight, boiling point, odor threshold, vapor pressure, vapor density, solubility and octanol-water partition coefficient(OWPC) and toxicological data such as median lethal dose($LD_{50}$) and median lethal concentration($LC_{50}$). Results: A total of 656 chemicals have OELs under OSHA in Korea. The numbers of chemicals which have eight-hour time weighted average(TWA) and short term exposure limits(STEL) are 618 and 190, respectively. TWA was significantly correlated with boiling point and STEL was only correlated with vapor pressure among physicochemical characteristics. Solubility and OWPC between "skin" and "no skin" substances which indicate skin penetration were not significantly different. Both $LD_{50}$ and $LC_{50}$ were correlated with TWA, while the $LC_{50}$ was not with STEL. As health indicators, health rating and Emergency Response Planning Guidelines(ERPG) rating as recommended by the National Fire Protection Association(NFPA) and American Industrial Hygiene Association(AIHA) were associated with OELs and reflect the chemical hazards. Conclusions: We found relationships between OEL and chemical information including physicochemical characteristics and toxicological data. The study has an important meaning for understanding present regulatory OELs.
Background: Korean Red Ginseng has been widely used in traditional oriental medicine for a prolonged period, and its pharmacological effects have been extensively investigated. In addition, Angelica gigas and deer antlers were also used as a tonic medicine with Korean Red Ginseng as the oriental herbal therapy. Methods: This study was conducted to evaluate the potential toxicological effect of KGC-HJ3, Korean Red Ginseng with angelica gigas and deer antlers, on reproductive and developmental functions including fertility, early embryonic development, maternal function, and embryo-fetal development. KGC-HJ3 was administered by oral gavage to Sprague-Dawley rats (22 animals per sex per group) at dose levels of 0 mg/kg (control), 500 mg/kg, 1000 mg/kg, and 2000 mg/kg to evaluate the potential toxicological effect on fertility and early embryonic development. In addition, KGC-HJ3 was also administered by oral gavage to mating-proven Sprague-Dawley rats (22 females per group) during the major organogenesis period at dose levels of 0 mg/kg (control), 500 mg/kg, 1000 mg/kg, and 2000 mg/kg to evaluate the potential toxicological effect on maternal function and embryo-fetal development. Results and conclusion: No test item-related changes in parameters for fertility, early embryonic development, maternal function, and embryo-fetal development were observed during the study period. On the basis of these results, it was concluded that KGC-HJ3 did not have toxicological potential on developmental and reproductive functions. Therefore, no observed adverse effect levels of KGC-HJ3 for fertility, early embryonic development, maternal function, and embryo-fetal development is considered to be at least 2000 mg/kg/day.
Lethal dose 50% ($LD_{50}$) has been commonly used as a parameter for the estimation of acute toxicity not only in animal experiment, but also in human study. Several methods to estimate $LD_{50}$ had been introduced, but Spearman-Karber and Berens-Karber method have been widely used due to their relative convenience and accuracy. However, $LD_{50}$ values estimated from the two methods showed inconsistency and variation depending on the characteristics of mortality data. In this study, the two methods were comparatively investigated in terms of accuracy and stability for the estimation of $LD_{50}$.
Diabetes is a major cause of morbidity and mortality, and associated with a high risk of atherosclerosis, and liver, kidney, nerve and tissue damage. Defective insulin secretion in pancreas and/or insulin resistance in peripheral tissues is a central component of diabetes. It is well established that, regardless of the degree of muscle insulin resistance, glucose levels in diabetic and non-diabetic individuals are determined by the rate of hepatic glucose production. Moreover recently studies using liver-specific insulin receptor knockout mice show the paramount role of the liver in insulin resistance and diabetes. Insulin exerts a multifaceted and highly integrated series of actions via its intracellular signaling systems. The first major section of this review defines the major insulin-mediated signaling pathways including phosphatidylinositol 3-kinase and mitogen activated protein kinases. The second major section of the review presents a summary and evaluation of methods for determination of the role and function of signaling pathways, including methods for determination of kinase phosphorylation, the use of pharmacological inhibitors of kinase and dominant-negative kinase constructs, and the application of new RNA interference methods.
Scientists have long used conventional toxicological methods to establish 'safe levels of exposure' for chemicals presumed to have threshold health effects or doses below which significant effects are unlikely to occur. These same methods cannot be used to establish safe levels of exposure for non-threshold pollutants. such as carcinogens. Therefore. Federal regulatory agencies in the United States are using risk assessment methods to provide information for public health policy decisions concerning increases in risk associated with increases in exposure to carcinogenic and other non-threshold pollutants. Acceptable exposure/risk levels are decided by policymakers who consider descriptions and estimates of risks together with social and economic benefits from the uses of the chemical. 1bis paper focuses on the development of quantitative risk assessment approaches by Federal regulatory agencies in the United States, and identifies the mathematical models currently being used for risk extrapolation. including their inherent uncertainties. The uncertainties and limitations of these methods have led some scientists to question the utility of quantitative risk extrapolation. The experience of the; U.S. Environmental Protection Agency (EPA). as summarized in this paper. can provide a realistic basis for evaluating the pros and cons. Finally. shortcomings in current risk assessment methods and their use in policy decisions are explored. and areas for possible improvement. given current scientific knowledge. are identified.
Cannabidiol derivatives (1, 2 and 3), 5-fluorouracil (4, 5-FU) and adriamycin (5, AM) were tested for their growth inhibitory effects against human tumor cell lines using two different 3-{4,5-dimeth-ylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay and sulforhodamine B protein (SRB) assay. The light microscopic study showed morphological changes of the treated cells. Disruptions in cell organelles were determined by colorimetric methods; MTT assay and STB assay. These results suggest that cannabidiol (1, CBD) retains the most growth-inhibitory activity against human tumor cell lines.
This study is an overview of toxicities and measurement techniques of ultrafine particles (UFPs), and their exposure controls. UFPs are ubiquitous in many working situations. Exposure to UFPs is possibly causing adverse health symptoms including cardio-respiratory disease to humans. In order to measure exposure levels of airborne UFPs, there are current available measurement guidelines, instruments and other techniques (i.e. contour mapping, control banding). However, these risk assessment techniques including measurement techniques, controls and guidelines are dependent on background levels, metrics (e.g. size, mass, number, surface area, composition), environmental conditions and controls. There are no standardized measurement methods available and no generic and specific occupational exposure standards for UFPs. It is thought that there needs to be more effort to develop Regulations and Exposure Standards for generic UFPs should be based on more exposure data, health surveys, toxicological data and epidemiological data. A carefully considered hierarchy of controls can also reduce the maximum amount of airborne UFPs being emitted from diverse sources in industries.

Carcinogens studied for biological human monitoring include benzo(a)pyrene, aflatoxin B1, UV light, ethylene oxide, 8-methoxypsoralen, 4-aminobiphenyl, vinyl choride, N-nitrosamine, cisplatin and other chemotherapeutic agents. Relevance of human monitoring for cancer research, progress in this field, methods to detect carcinogen-adducts are reviewed here. It is hoped that these approaches will be used for the risk assessment of carcinogen exposure, cancer etiology study and cancer prevention in humans.
