Ecotoxicity Test Using E. agilis Biomonitoring System (Euglena 운동성 측정장치를 이용한 생태독성평가)

  • The toxic responses of flagellate Euglena agilis Carter to 8 heavy metals (Ag, Cd, Cr (VI), Cu, Hg, Ni, Pb, Zn) were measured using E. agilis system (E-Tox), an automated biomonitoring system. The E-Tox measures cell movement parameters, such as velocity, motility, and forms of the cells, as biological endpoints. $EC_{50}$ values from the E. agilis biomonitoring test were compared with the literature data from the tests with Daphnia magna, Vibrio fischeri and Euglena gracilis. Measurement of the E. agilis movement behavior and D. magna acute toxicity test were also conducted for the wastewater samples. E. agilis is less sensitive than D. magna but is comparable to or more sensitive than V. fischeri and E. gracilis for the heavy metals tested in this study. E. agilis shows prompt changes of these parameters for the toxic metal plating wastewater. Major advantages of the E-tox are automatic, easy to handle and fast ecotoxicity monitoring system compared to other biological test systems. These results imply that E. agilis biomonitoring test using E-Tox can be a putative ecotoxicity test as a good early warning tool for the monitoring of toxic wastewater.

Synthesis of I-125 Labelled Compound of Taxol Analogue for Radioimmunoassay (Taxol의 방사면역측정을 위한 I-125 표지화합물 합성)

  • Taxol, an anticancer drug that has diterpenoid conformation, has been used as an effective chemotherapeutical agent in the treatment of breast and ovarian cancers. Because of its toxicity like other anticancer drugs, monitoring the taxol level in serum is important procedure during cancer therapy. The various monitoring methods using HPLC, ELISA, and RIA have been adopted, and RIA technique is known to be superior than other methods in trems of sensitivity and convenience. In this study, in order to develope taxol RIA system using $^{125}$I labelled antigen, first of all we synthesized taxol derivatives. 2'-hemisuccinyltaxol was obtained with about 80% yield by esterification of taxol at C-2' hydroxyl group on C-13 carbon with succinic anhydride. [$^{125}$I]iodotyramine was prepared with 58% labelling yield by radioiodination of tyramine and purified by gel chromatography. 2'-[$^{125}$I]iodotyramine-hemisuninyltaxol, $^{125}$I labelled antigen for taxol RIA, was synthesized with 96% yield from conjugation of 2'-hemisuccinyltaxol and [$^{125}$I]iodotyramine. Anti-taxol serum was produced from the rabbit immunized with 2'-hemisuccinyltaxol-BSA synthesized by 2'-hemisuccinyltaxol and BSA. The antiserum titer was determined by RIA using 2'-[$^{125}$I]iodotyramine-hemisuccinyltaxol. The titer of 1:20 was obtained with about 40% of B/T. The results suggest that taxol RIA using $^{125}$I labelled antigen can be applied to monitor the taxol level in serum.

Effects of Vinyl Chloride to Human Health and Aquatic Ecosystems (염화비닐(Vinyl Chloride)의 인체 및 수생태계 영향 연구)

  • Vinyl chloride (VC) is a known human carcinogen, and it is released to multi-environmental media via several exposure routes. VC was potentially evaluated as a water quality pollutant based on human health risk assessment in Korean water environments. In this study, we investigated physicochemical and toxicological properties of VC, human health and ecological risk assessment, and the regulation of VC as a water quality pollutant in developed countries. Currently there are no standard or guideline values of VC in Korean legal system for the protection of human health and aquatic ecosystem, except that it is designated as a specific toxic substance and a water quality pollutant. Human health risk assessment of VC was previously performed based on the limited water quality monitoring data. The monitoring level of VC in Korean water system is more higher than other countries'. VC was assessed as potential hazardous chemical based on the US EPA's cancer risk assessment. There were a few ecotoxicity data of VC available using very limited kinds of aquatic organisms, and the toxicity results obtained seem to be overestimated without considering the losses of VC in open exposure system. Therefore it is needed to monitor the VC in various areas and to carry out the ecotoxicity research using multi-level organisms. We expect that these results can be useful information for implementing VC as a water quality pollutant in legal system for the protection of human health and aquatic ecosystem in near future.

The Study on the Analysis Method of Tetrodotoxin in Puffer Fish (복어 중 테트로도톡신 분석법에 관한 연구)

  • The current standard for testing tetrodotoxin (TTX) in foodstuffs is the mouse bioassay (MBA) in Korea as in many other countries. However, this test suffers from potential ethical concerns over the use of live animals. In addition, the mouse bioassay does not test for a specific toxin thus a sample resulting in mouse incapacitation would need further confirmatory testing to determine the exact source toxin (e.g., TTX, STX, brevotoxin, etc.). Furthermore, though the time of death is proportional to toxicity in this assay, the dynamic range for this proportional relationship is small thus many samples must be diluted and new mice be injected to yield a result that falls within the quantitative dynamic range. Therefore, in recent years, there have been many efforts in this field to develop alternative assays. High performance liquid chromatography (HPLC) coupled with mass spectrometry (MS) has been emerged as one of the most promising options. A LC-MS-MS method involves solid-phase extraction (SPE) and followed by analysis using an electrospray in the positive ionization mode and multiple reactions monitoring (MRM). To adopt LC-MS-MS method as alternative standard for testing TTX, we performed a validation study for the quantification of TTX in puffer fish. This LC-MS-MS method showed good sensitivity as limits of detection (LOD) of $0.03{\sim}0.08{\mu}g/g$ and limits of quantification (LOQ) of $0.10{\sim}0.25{\mu}g/g$. The linearity ($r^2$) of tetrodotoxin were 0.9986~0.9997, the recovery were 80.9~103.0% and the relative standard deviations (RSD) were 4.3~13.0%. The correlation coefficient between the mouse bioassay and LC/MS/MS method was higher than 0.95.

Study on the Distributions of VOCs, Aldehydes, PAHs Concentration in Seoul Metropolitan Area (수도권 지역에서 환경대기 중 유해대기오염물질 (VOCs, Aldehydes, PAHs) 농도분포 특성 연구)

  • Although concentrations of hazardous air pollutants(HAPs) are very low in the atmosphere, a growing attention has been paid on such compounds due to their high toxicity and bioaccumulation potentials into human body. In order to control and manage the amount of these materials in ambient air, it is necessary to construct monitoring system of them and to know the current concentration level of HAPs above all. In this work, a wide range of HAPs has been measured in metropolitan area to recognize the present state of HAPs in this area. The measured concentration of VOCs was higher in order of Jeonnongdong, Jeongdong, and Yangsuri. The regional difference of VOCs concentration was also highest in spring. Its total VOCs was ranged from $15.17{\sim}41.45$ ppb. Benzene $0.43{\sim}2.32$ ppb showed similar concentration level with the result of previous researches in Seoul. This value is a little higher than the average concentration 0.92 ppb for national ambient air quality standards in Japan. The concentration of aldehydes in this study was lower than those of other researches. Previous works in Seoul metropolitan area showed that the concentration of formaldehyde and acetaldehyde were higher than 5 ppb. The concentration of gaseous and particulate PAHs was high in order of winter, spring, and summer More than 90% of PAHs with low molecular weight such as 2-rings and 3-rings PAHs existed in gas phase. On the other hands, PAHs with high molecular weight more than 5-rings PAHs almost existed in particulate. In spring, the concentration of gaseous PAHs was 24.38 $ng/m^3$ in Jeongdong. Among the particulate PAHs, the concentrations of Naphthalene, Benzo(b)fluoranthene, and Benzo(g, h, i)perylene were higher than others. Especially, the concentration of Benzo(a)pyrene, a important carcinogenic pollutant, was highest in winter 0.5 $ng/m^3$ and ranged from 0.03 to 0.3 $ng/m^3$ in spring and summer, which is lower than the monitoring result in 90's. These components were mainly originated from the vehicle exhaust or heating equipment use.

Biosensor System for the Detection of Agrichemicals and Its Applications (농약 검출을 위한 바이오센서 시스템 연구 및 그 응용)

  • In the recent years, some organic toxic chemicals were used for obtaining high-yield productivity in agriculture. The undegraded pesticides may remain in the agricultural foods through atmosphere, water, and soil and cause public health problems to environmental resources and human beings even at very low concentrations. Small amounts of pesticides can affect a central nervous system, resulting in immunogenic diseases, infertility problems, respiratory diseases and born marrow diseases, which can lead even to death. Monitoring of the environmental pesticide is one of the important issues for the human well-being. Several kinds of biosensors have been successfully applied to the detection of agrichemical toxicity. Also, few platforms for biocide detection have been definitely developed for the degradation and reaction of pesticides. Biochip and electrochemistry experiments involve immobilizing a receptor molecule on a solid substrate surface, and monitoring its interaction with an analyze in a sample solution. Furthermore, nanotechnology can be applied to make high-throughput analyses that are smaller, faster and sensitive than conventional assays. Some nanomaterials or nanofabricated surfaces can be coupled to biomolecules and used in antibody-based assays and enzymatic methods for pesticide residues. The operation procedure has become more convenient as it does not require labeling procedure. In this paper, we review the recent advances in agrichemical defection research and also describe the label-free biosensor for pesticides using various useful detection methods.

Detection and Monitoring of Benzylpenicillin Residues in Livestock and Marine Products (유통 축·수산물 중 잔류벤질페니실린의 검출 및 모니터링)

  • Penicillin and its salts, including the benzatine, procaine, and sodium salts, have been widely used in human and veterinary medicine. Owing to their low toxicity, they currently form the most important group of antibiotics. However, overdose and abuse of these antibiotics may lead to potential risk in human health. Therefore, this study was conducted to validate the analysis method established by the Korea Food Code in 2012 and to monitor the levels of benzylpenicillin residues in products with reference to the maximum residue level (MRL). Of the 232 product samples tested, benzylpenicillin was detected in 11 livestock products and 2 marine products. Benzylpenicillin concentrations were found to be lower than the MRL in 12 products; however, the concentration of benzylpenicillin was found to be greater than the MRL in 1 pork product. The limit of quantification (LOQ) for benzylpenicillin was found to be 0.001-0.002 mg/kg, with an average recovery of 90.4-115.3%. Calibrations showed good linearity of 0.995 over a range of 0.002-0.05 mg/kg.

Development on Metallic Nanoparticles-enhanced Ultrasensitive Sensors for Alkaline Fuel Concentrations (금속 나노입자 도입형의 초고감도 센서 개발 및 알칼라인 연료 측정에 적용 연구)

  • Alkaline fuel cells using liquid fuels such as hydrazine and ammonia are gaining great attention as a clean and renewable energy solution possibly owing to advantages such as excellent energy density, simple structure, compact size in fuel container, and ease of storage and transportation. However, common shortcomings including cathode flooding, fuel crossover, side yield reactions, and fuel security and toxicity are still challenging issues. Real time monitoring of fuel concentrations integrated into a fuel cell device can help improving fuel cell performance via predicting any loss of fuels used at a cathode for efficient energy production. There have been extensive research efforts made on developing real-time sensing platforms for hydrazine and ammonia. Among these, recent advancements in electrochemical sensors offering high sensitivity and selectivity, easy fabrication, and fast monitoring capability for analysis of hydrazine and ammonia concentrations will be introduced. In particular, research trend on the integration of metallic and metal oxide nanoparticles and also their hybrids with carbon-based nanomaterials into electrochemical sensing platforms for improvement in sensitivity and selectivity will be highlighted.

A Study on the Behavior Change of Zebrafish For Toxicity Evaluation of Residual Psychoactive Medication in Wastewater Treatment Plant Effluent (하수처리시설 방류수 내 잔류 향정신성 의약품의 독성평가를 위한 zebrafish의 행동성 변화 연구)

  • As interest in health issues increases, it is imperative to ensure good water quality from wastewater treatment plant effluent to preserve environmental health. In particular, currently there is a lack of water ecosystem risk assessment on pharmaceutical substances remaining in effluent. In this study, antidepressant escitalopram (ESC), antiepileptic carbamazepine (CBZ) and lead, which impact the behavior of aquatic organisms, were used to test their impact on the potential behavior of zebrafish. Zebrafish have been widely used in toxicological assessment studies due to the ease of handlinggenerically and genetically. It was possible to observe changes in the growth of organisms through monitoring the embryos' cognitive and behavior assessment. In this study, the embryo lethal dose test showed that the lethal concentration of ESC and CBZ was at 10 ppb, which is below the water quality criterion (100 ppb), increased by 32.5 % and 40 %, respectively. In the cognitive test, it was found that the cognitive ability function decreased by 22 % and 17% for ESC(500 ppb) and CBZ(1,000 ppb) respectively relative to control. Based on these results, it is necessary to initiate efforts to remove these trace pollutants from sewage treatment facilities to protect the health of aquatic organisms.

Thyroid Function after Postoperative Radiation Therapy in Patients with Breast Cancer

  • Objective: The aim of this study was to assess thyroid function in breast cancer patients exposed to therapeutic external beam radiation. The focus was on possible progressive changes and any relationships between the incidence of primary hypothyroidism, the time required to become hypothyroid, and factors such as chemotherapy, hormonotherapy and immunotherapy. Materials and Methods: Seventy females undergoing 3D conformal and IMRT radiation therapy for breast cancers were enrolled in a non-randomized prospective study. The patients was divided into two groups: those after mastectomy or breast conserving surgery (BCS) were irradiated to a scar of the chest wall/breast and the ipsilateral supraclavicular and the axillary areas (supraclavicular radiotherapy group - SC-RT group - 32 patients) and the control group receiving adjuvant chest wall/breast RT only (BCT group - 38 patients).The total doses were 50.0 to 70 Gy in 5 to 7 weeks. The median follow-up term was 24 months (range, 1-40 months). Thyroid function was evaluated by measuring thyroid stimulating hormone (TSH), free thyroxine (fT4), and free triiodothyronine (fT3) levels. The minimum, maximum and mean thyroid gland doses for 20 Gy (V20) were calculated for all patients. Results: Statistically significant results were obtained for the SC-RT group. Two yearsa fter the end of RT the chance of an event was increased in 6% of the population (p=0.009) in the SC-RT group. In the BCT group no significance was noted. No statistically significant differences were found for V20, chemio-, immunotherapy and hormonotherapy or Ki67 values (p=0.12). No significant results were obtained for development of hypothyroidism and clinical factors (age, thyroid volume, treatment modalities). Conclusion: Radiotherapy is associated with a higher incidence of thyroid toxicity in breast cancer patients. Routine thyroid function monitoring should be recommended in such cases.
