Background: A quantitative structure-activity relationship (QSAR) model was adopted in the Registration, Evaluation, Authorization, and Restriction of Chemicals (REACH, EU) regulations as well as the Act on Registration, Evaluation, etc. of Chemicals (AREC, Republic of Korea). It has been previously used in the registration of chemicals. Objectives: In this study, we investigated the correlation between the predicted data provided by three prediction programs using a QSAR model and actual experimental results (acute fish, daphnia magna toxicity). Through this approach, we aimed to effectively conjecture on the performance and determine the most applicable programs when designating toxic substances through the AREC. Methods: Chemicals that had been registered and evaluated in the Toxic Chemicals Control Act (TCCA, Republic of Korea) were selected for this study. Two prediction programs developed and operated by the U.S. EPA - the Ecological Structure-Activity Relationship (ECOSAR) and Toxicity Estimation Software Tool (T.E.S.T.) models - were utilized along with the TOPKAT (Toxicity Prediction by Komputer Assisted Technology) commercial program. The applicability of these three programs was evaluated according to three parameters: accuracy, sensitivity, and specificity. Results: The prediction analysis on fish and daphnia magna in the three programs showed that the TOPKAT program had better sensitivity than the others. Conclusions: Although the predictive performance of the TOPKAT program when using a single predictive program was found to perform well in toxic substance designation, using a single program involves many restrictions. It is necessary to validate the reliability of predictions by utilizing multiple methods when applying the prediction program to the regulation of chemicals.
본 연구에서는 건축물 내장 재료에 대한 연소독성평가와 독성지수연구로써 FT-IR을 이용한 연소가스 분석실험을 하였다. 독성지수를 산정하기 위한 실험의 화재모델로 콘 칼로리미터 화재모델(KS F ISO/TR 9122-4)을 사용하였으며 ISO 19702의 절차에 따라 FT-IR을 이용하여 건축 재료의 연소로부터 발생되는 가스의 분석을 수행하였다. 국제규격에서 제시하고 있는 몇 가지 독성지수 산정법 중 ISO 13344에서 규정하는 방법에 따라 FED 값을 산정하였으며, 30분간 시험동물에 노출 시 대상의 50 %가 사망하는 농도인 $LC_{50}$을 기준으로 하여 3가지 재료의 독성지수화를 통해 상대적인 독성 위험도를 평가하였다. 그 결과로 0.025~0.285의 FED가 산출되었으며 우레탄에서는 HCN이 검출되었다.
Background: Plastic particles less than 5 mm in diameter (microplastics) are well-known for causing various toxicities such as lung inflammation, oxidative stress, genotoxicity, and reproductive toxicity. As microplastics become smaller, they can move across cell membranes, the placenta, and the blood-brain barrier. Objectives: We evaluated the toxicities of polyethylene microplastics (PE-PMs) in dams and neonates through intragastric intubation of pregnant ICR mice. Methods: Low concentrations (0.01 mg/mouse/day) and high concentrations (0.1 mg/mouse/day) of polyethylene microplastics were administered from the ninth day of pregnancy to postnatal day seven. The control group was administered with distilled water. On the day of sacrifice, the weight of dams and neonates and the organ weight of neonates was measured. Further, acetylcholinesterase levels and glutathione peroxidase levels were evaluated by using a blood sample obtained on the sacrifice day. Results: No significant difference in the number of neonates was found, but the body weight gain of dams was seen to be lower in the low-dose group. On the other hand, we observed a consecutively declining trend in the weight gain and organ weight of neonates among the high-, control, and low-dose groups. Meanwhile, the serum acetylcholinesterase and glutathione peroxidase level were higher in the low-dose group compared to the control group. Further, the dose-dependent accumulation of microplastics in the organs of neonates revealed the transport of plastic particles from dams to their offspring. Conclusions: Although the exact mechanism of toxicity caused by microplastics could not be confirmed, it was validated that exposure to microplastics during pregnancy and lactation causes its migration between generations and accumulation throughout the body. Hence, it is necessary to evaluate the systemic toxicity of microplastics and assessment of co-morbidities such as second-generation toxicity, neurotoxicity, and depression following long-term exposure.
3-Monochloro-1,2-propanediol(3-MCPD) is currently being a matter of concern because of its toxicity. 3-MCPD produced during the acid hydrolysis of soybean products has been reported to be mutagenic, neurotoxic, nephrotoxic and spermatotoxic. Howerer, the carcinogenicity of 3-MCPD is a controversial issue over the past several decades. 3-MCPD characteristically showed a variety of toxicities in reproductive system such as, decrease in sperm number and sperm motility, infertility, loss of sperm function, and weight decrease in ovary. Due to the toxicity of 3-MCPD, exposure to 3-MCPD has been proposed to be reduced to as low a level as technologically feasible. 3-MCPD can be detected in soy sauce or non-soy sauce products. The legal limit for 3-MCPD this year has been suggested to be 20 ppb($\mu\textrm{g}$/kg)in the European Community. In Korea, the permissible level of 3-MCPD is expected to be 0.3 ppm. In this study, 3-MCPD was toxicologically evaluated in terms of risk assessment in humans.
As the NOAFL of copper based on liver toxicity in the human body is set and the TDI of copper is lower, it is necessary to strengthen the drinking water standard of copper according to toxic effects and the TDI of copper in humans. It is difficult to calculate the accurate drinking water standard because of the part of uncertainty for chronic effects of acute human with Wilson's disease and baby in the current studies. In order to improve the drinking water standard of copper considering of liver toxicity, it is desired to set the drinking water standard with concerning of the revising tendency in the foreign countries such as US, EC and WHO.
Earthworm (Eisenia fetida) acute toxicity test was carried out and ecological risk assessment in soil was performed with national monitoring data. 14 day - $LC_{50}$ of nonylphenol and bisphenol A were 288.1 mg/kg and 90.1 mg/kg, respectively. And NOECs of nonylphenol and bisphenol A were 250 mg/kg and 50 mg/kg, respectively. Significant weight decrement was appeared at 70 mg/kg of bisphenol A, however, nonylphenol at concentrations tested did not severe adverse effect on the weight decrement. The environmental monitoring has been carrying out by NIER since 1999. Exposure levels of nonylphenol in soil were ND$\sim$10.55 $\mu$g/kg and those of bisphenol A were ND$\sim$15.50$\mu$g/kg in National Monitoring data which had been performed from 2000 to 2004. The measured soil exposure level was applied to evaluate the environmental risk assessment. The values of PNEC for bisphenol A and nonylphenol were determined as 0.5 mg/kg and 2.5 mg/kg, respectively using the safety factors which were suggested in EU and OECD. The values of HQ (PEC/PNEC) were determined to be below I for bisphenol A and nonylphenol when the maximum exposure levels for bispheol A (15.50$\mu$g/kg) and nonylphenol (10.55$\mu$g/kg) were applied. Conclusively, the environmental risk assessment of bisphenol A and nonylphenol was not critical in soil.
Polychlorinated hydrocarbons are continuously released into the environment from various industrial processes. Trichloroethylene (TCE) and tetrachloroethylene (perchloroethylene, PCE) are of primary concern because of their large-scale production, wide industrial application, poor biodegradability, and tendency to circulate in the air and water. The common routes of human exposure to these compounds include inhalation, ingestion, and dermal adsorption. Additionally, they have been detected in various plant foods. Prolonged exposure to these contaminants is associated with certain risks. They are carcinogenic and have other toxic effects, including gastrointestinal, developmental, neurological, and hematological toxicity. To analyze these contaminants, they are generally extracted from various matrices, followed by instrumental analysis. Gas chromatography, often in combination with different detectors, is the most widely used analytical method. This review covers the toxicity, analytical methods, occurrence in foods, and risk assessment of these contaminants.
화학물질로부터 근로자의 건강보호를 위해 사용량이 많으나 작업환경 및 유해성 평가 자료가 부족한 gasoline에 대해 유해성(hazards)을 조사하고 국내 취급실태 및 일부 취급 사업장의 작업환경 측정을 통해 위해성(risk assessment)을 평가하였다. 연구결과 gasoline은 생식독성 추정 및 생식세포 변이원성 물질인 1B, 그리고 IARC Group 2B, ACGIH A3 물질이었다. 작업환경 측정결과는 TLV-TWA 기준 $900mg/m^3$미만이었으며, 유해성 평가결과 발암 $RfC_{(Worker)}$는 $0.3mg/m^3$, 만성흡입독성 $RfC_{(Worker)}$는 $2.7mg/m^3$, 발달독성 $RfC_{(Worker)}$는 $2.7mg/m^3$이었다. 그리고 위험도 평가에서 발암성은 459, 만성흡입독성은 51, 발달독성은 51이었으며, 이를 토대로 한 gasoline의 위험도는 1을 초과하는 물질로 평가되었다.
해산로티퍼 (Brachionus plicatilis)의 생존율 및 개체군 성장률을 사용하여 중금속 As, Cr 및 Pb에 대한 독성평가를 수행하였다. 중금속에 24시간 노출한 생존율은 As와 Cr의 30과 $150mg\;L^{-1}$ 농도에서 급격한 감소가 시작되어 농도의존적으로 감소하였으나, Pb에서는 영향이 나타나지 않았다. 중금 속에 72시간 노출한 개체군 성장률은 As, Cr 및 Pb의 5, 25 및 $50mg\;L^{-1}$ 농도에서 급격한 감소가 시작되어, 3종 중금속에서 모두 농도의존적으로 감소하는 경향을 보였다. As, Cr 및 Pb에 노출된 개체군 성장률의 $EC_{50}$ 값은 각각 12.98, 82.34 및 $110.14mg\;L^{-1}$이고 독성의 세기는 As>Cr>Pb로 생존율과 동일하였다. 또한 As, Cr 및 Pb에 대한 개체군 성장률의 NOEC는 각각 1, 12.5 및 $50mg\;L^{-1}$, LOEC는 각각 5, 25 및 $50mg\;L^{-1}$로 나타났다. 해양환경에서 각각의 LOEC 이상의 농도는 B. plicatilis에게 독성영향을 미칠 수 있는 농도로 판단되며, NOEC와 $EC_{50}$ 값은 WET test를 위한 혼합독성과 해양생태계 내 중금속독성에 대한 가이드라인으로 적절하게 활용될 수 있을 것이다.
Objectives: An adverse outcome pathway is a biological pathway that disturbs homeostasis and causes toxicity. It is a conceptual framework for organizing existing biological knowledge and consists of the molecular initiating event, key event, and adverse output. The AOP concept provides intuitive risk identification that can be helpful in evaluating the carcinogenicity of chemicals and in the prevention of cancer through the assessment of chemical carcinogenicity predictions. Methods: We reviewed various papers and books related to the application of AOPs for the prevention of occupational cancer. We mainly used the internet to search for the necessary research data and information, such as via Google scholar(http://scholar.google.com), ScienceDirect(www.sciencedirect.com), Scopus(www.scopus. com), NDSL(http: //www.ndsl.kr/index.do) and PubMed(http://www.ncbi.nlm.nih.gov/pubmed). The key terms searched were "adverse outcome pathway," "toxicology," "risk assessment," "human exposure," "worker," "nanoparticle," "applications," and "occupational safety and health," among others. Results: Since it focused on the current state of AOP for the prediction of toxicity from chemical exposure at work and prospects for industrial health in the context of the AOP concept, respiratory and nanomaterial hazard assessments. AOP provides an intuitive understanding of the toxicity of chemicals as a conceptual means, and it works toward accurately predicting chemical toxicity. The AOP technique has emerged as a future-oriented alternative to the existing paradigm of chemical hazard and risk assessment. AOP can be applied to the assessment of chemical carcinogenicity along with efforts to understand the effects of chronic toxic chemicals in workplaces. Based on these predictive tools, it could be possible to bring about a breakthrough in the prevention of occupational and environmental cancer. Conclusions: The AOP tool has emerged as a future-oriented alternative to the existing paradigm of chemical hazard and risk assessment and has been widely used in the field of chemical risk assessment and the evaluation of carcinogenicity at work. It will be a useful tool for prediction, and it is possible that it can help bring about a breakthrough in the prevention of occupational and environmental cancer.
