Cinnamomum loureiroi 추출물의 항산화 활성 (The Activity of Anti-oxidation of Cinnamomum loureiroi Extract)

  • 본 연구는 Cinnamomum loureiroi 추출물(CLE)의 항산화 활성을 확인하기 위해 수행되었다. 실험 전에 세포독성을 확인하였으며, 본 연구수행에서 사용할 농도의 안전성을 확보하였다. 추출물의 폴리페놀과 플라보노이드 함량을 측정하고 ABTS 라디칼과 DPPH 라디칼 소거능을 측정하였으며, LPS에 유도된 Raw 264.7 세포에서 ROS의 생성 감소를 확인하였다. 연구 결과 총 폴리페놀의 함량은 397.7±8.3 mg/g, 총 플라보노이드 함량은 101.9±0.8 mg/g으로 BHA 검량선 기준으로 선행연구들과 비교한 결과 함량비가 높은 것으로 나타났다. ABTS 라디칼과 DPPH 라디칼 소거율은 농도 의존적으로 증가하는 것으로 나타나 농도에 따라 항산화 활성이 높아지는 것으로 확인되었다. ROS 생성은 농도 의존적으로 유의한 감소를 나타내어 라디칼 소거능과 부합되는 결과를 나타냈다. 결과적으로 CLE의 항산화 활성이 확인되었으며, 향 후 천연원료로서 기능성화장품 또는 심화 연구를 통한 천연화장품의 응용할 가능성이 있는 것으로 사료된다.

팔미지황탕(八味地黃湯)의 ROS 생성 및 p53 활성 조절을 통한 시스플라틴 신장독성 완화효과 (Palmijihwang-tang Alleviates Cisplatin-induced Nephrotoxicity through Inhibiting ROS Production and p53 Activation)

  • Palmijihwang-tang is an herbal formula frequently used to treat many symptoms, such as lumbago, pollakiuria, cold hands and feet, nephritis, sterilitas virilis, and prostatic disorders. The aim of this study was to investigate the effects of Palmijihwang-tang on cisplatin-induced nephrotoxicity in rat kidney proximal tubular NRK-52E cells. NRK-52E cells were treated with Palmijihwang-tang in absence or presence of 30 µM cisplatin for 12 or 24 h. Palmijihwang-tang at concentrations of 50-800 ㎍/ml did not change the cell viability in NRK-52E cells, and showed no significant toxicity. Palmijihwang-tang at concentrations of 400 and 800 ㎍/ml significantly increased the cell viability and reduced apoptotic cells in NRK-52E cells exposed to cisplatin. Also, Palmijihwang-tang markedly inhibited cisplatin-induced caspase-3 activation, PARP cleavage, ROS production and p53 activation in NRK-52E cells. Furthermore, Palmijihwang-tang did not interfere with the antitumor activity of cisplatin in AGS and A549 cancer cells. Particularly, Palmijihwang-tang enhanced antitumor activity of cisplatin in A549 cells. Taken together, these results suggest that Palmijihwang-tang ameliorated cisplatin-induced nephrotoxicity through reduction of ROS production and p53 activation, and did not interrupt antitumor efficacy of cisplatin against cancer cells.

Impact of imatinib administration on the mouse ovarian follicle count and levels of intra-ovarian proteins related to follicular quality

  • Objective: The impact of imatinib, a tyrosine kinase inhibitor, on ovarian follicles and several proteins related to follicular function and apoptosis was investigated in mice. Methods: Saline, cyclophosphamide (Cp; 50 or 75 mg/kg), or imatinib (7.5 or 15 mg/kg) was injected once intraperitoneally into female B6D2F1 mice (18 mice in each group). In multiple ovarian sections, the number of various types of follicles and the proportion of good-quality (G1) follicles were counted. The levels of six proteins (anti-Müllerian hormone [AMH], BCL-xL, BAX, acid sphingomyelinase [A-SMase], caspase-3, and α-smooth muscle actin [α-SMA]) within the whole ovaries were quantified using Western blots. Results: Compared to the saline group, a significant reduction of the primordial follicle count was observed in the group treated with imatinib 7.5 and 15 mg/kg, as well as in the group treated with Cp 75 mg/kg. Administration of Cp significantly decreased the proportion of G1 primordial follicles, but administration of imatinib did not. No differences in the AMH, anti-apoptotic BCLX-L, pro-apoptotic BAX, and A-SMase levels in the ovarian tissues were observed among the five groups. However, caspase-3 and α-SMA levels were significantly higher in the imatinib and Cp groups than in the saline group. Conclusion: The administration of imatinib to mice significantly reduced the primordial follicle count and increased the protein levels of caspase-3 and α-SMA. Our findings suggest that imatinib potentially exerts ovarian toxicity via apoptotic processes, similarly to Cp.

Anti-inflammatory and antioxidant activities of Sargassum horneri extract in RAW264.7 macrophages

  • [Purpose] In this study, we investigated whether a 70% ethanolic (EtOH) extract of Sargassum horneri had antioxidant and anti-inflammatory effects in lipopolysaccharide (LPS)-stimulated macrophage-like RAW 264.7 cells. [Methods] The proximate composition, fatty acids, amino acids, and dietary fiber of S. horneri, various biologically active compounds, and antioxidant activity were analyzed. [Results] The DPPH and ABTS free radical scavenging activities, as well as the reduction power, of the S. horneri extract used here were significantly increased in a concentration-dependent manner. This indicates that S. horneri contains bioactive compounds, such as phenols and flavonoids, that have excellent antioxidant activity. The cellular viability and metabolic activity results confirmed that the extract had no discernible toxicity at concentrations up to 100 ㎍/mL. The levels of nitrites and cytokines (PGE2, TNF-α and IL-6), which mediate pro-inflammatory effect, were significantly inhibited by treatment with either 50 or 100 ㎍/mL S. horneri extract, whereas that of IL-1β was significantly inhibited by treatment with 100 ㎍/mL of the extract. Similarly, the expression of iNOS and COX-2 proteins also decreased according to 50 or 100 ㎍/mL extract concentrations. NF-κB binding to DNA was also significantly inhibited by treatment with 100 ㎍/mL of extract. [Conclusion] These results suggest that 70% EtOH extracts of S. horneri can relieve inflammation caused by disease or high intensity exercise.

Co-administration of alcohol and combination antiretroviral therapy (cART) in male Sprague Dawley rats: a study on testicular morphology, oxidative and cytokines perturbations

  • Alcohol consumption alongside combination antiretroviral therapy (cART) has attracted research interest, especially because of increasing male infertility. This study investigated the combined effects of alcohol and cART on testicular morphology, biomarkers of oxidative stress, inflammation, and apoptosis. Rats, weighing 330-370 g, were divided into four groups of six animals each; control, alcohol treated (A), cART, and alcohol plus cART treated (A+cART). Following 90 days treatment period, animals were euthanized, testis extracted, and routinely processed for histology and immunohistochemical analysis. Significantly decreased epithelial area fraction, increased luminal and connective tissue area fractions, and reduction of epithelial height and spermatocyte number, were recorded in the treated groups compared to control. Extensive seminiferous epithelial lesions including widened intercellular space, karyolysis, and sloughing of germinal epithelium were recorded in all the treated groups. Furthermore, upregulation of inducible nitric oxide synthase and 8-hydroxydeoxyguanosine, interleukin-6, and caspase 3 recorded in treated animals, was more significant in A+cART group. Also, the levels of interleukin-1β and tumor necrosis factor-α were more elevated in A and cART treated groups than in A+cART, while MDA was significantly elevated in cART and A+cART treated groups compared to control group. Altogether, the results indicate testicular toxicity of the treatments. It is concluded that consuming alcohol or cART induces oxidative stress, inflammation, and apoptosis in testis of rats, which lead to testicular structural and functional derangements, which are exacerbated when alcohol and cART are consumed concurrently. The result will invaluably assist clinicians in management of reproductive dysfunctions in male HIV/AIDS-alcoholic patients on cART.

Impact of peripheral blood mononuclear cells preconditioned by activated platelet supernatant in managing gastric mucosal damage induced by zinc oxide nanoparticles in rats

  • The world has witnessed tremendous advancements in nano-base applications. Zinc oxide nanoparticles (ZON) are widely used in food industry and medicine. Although their application is of important value, they may cause toxicity to body tissues. Peripheral blood mononuclear cells (PBMCs) proved its efficacy in tissue regeneration especially when it is preconditioned by activated platelet supernatant (APS). The aim of this study is to evaluate the effect of ZON on the gastric mucosa and the therapeutic role of the PBMCs preconditioned by APS in rats. Ten rats were donors and fifty rats were recipients. The recipients were divided into; control group, ZON group (10 mg/kg/day orally for five days) and preconditioned PBMCs group (1×107 once intravenously 24 hours after ZON). Gastric specimens were processed for histological, immunohistochemical, biochemical and quantitative real-time polymerase chain reaction studies. ZON group showed marked structural changes in the gastric mucosa. There was desquamation or deep ulceration of the epithelium. Cytoplasmic vacuoles and pyknotic nuclei were in glandular cells. Reduced proliferating cell nuclear antigen and increased tumor necrosis factor-α were in epithelial cells. There were significant elevation in malondialdahyde and reduction in glutathione, superoxide dismutase, and catalase. Enhancement in mRNA expression of nuclear factor kappa-B and cyclooxygenase-2 was detected. The preconditioned PBMCs group showed significant improvement of all parameters. So, ZON had cytotoxic effects on the gastric mucosa and the preconditioned PBMCs had a therapeutic effect on gastric mucosal damage after ZON.

중금속(Cd, Cu, Zn)이 해산규조류(Skeletonema costatum)의 개체군 성장률에 미치는 독성영향 (Toxic Effects of Heavy Metal (Cd, Cu, Zn) on Population Growth Rate of the Marine Diatom (Skeletonema costatum))

  • 해산규조류 Skeletonema cosatatum의 개체군성장률 (r)을 사용하여 중금속 3종(Cd, Cu, Zn)의 독성영향 평가를 수행하였다. S. costatum을 Cd (0, 0.63, 1.25, 2.50, 5.00, 10.00 ppm), Cu (0, 0.25, 0.50, 0.75, 1.00, 1.25, 1.50 ppm) 그리고 Zn (0, 0.31, 0.63, 1.25, 2.00, 2.50, 5.00 ppm)에 노출하고 96 h 이후에 r 값을 도출하였다. 대조구(Cd, Cu 그리고 Zn을 포함하지 않은)에서 r은 0.05보다 높았으며 중금속 농도가 증가할수록 r은 감소하였다. 중금속에 의해 농도의존적으로 r이 감소되었고 Cd 농도 1.25, Cu 농도 1.25 그리고 Zn 농도 2.50 ppm 이상의 농도에서 명확한 감소를 나타냈다. 중금속의 독성영향은 Cu>Zn>Cd 순서이며, 이때 $EC_{50}$ 값은 각각 1.11, 2.13 그리고 6.84 ppm으로 나타났다. 중금속 3종(Cd, Cu 그리고 Zn)에 노출된 r의 최소영향농도는 각각 1.25, 1.00, 2.00 ppm으로 나타났다. 이 결과를 바탕으로, 해양환경에서 S. costatum의 r은 Cd 농도가 1.25 ppm 이상, Cu 농도가 1.00 ppm 이상, Zn 농도가 2.00 ppm 이상인 경우에 독성영향을 받았다. 그러므로 S. costatum의 r은 해양환경에서 중금속 독성영향 평가를 위한 매우 유용한 방법으로 판단된다.

라이코펜이 사이토카인에 의해 유도된 베타세포 사멸에 미치는 효과 및 기전 연구 (Protective effect of lycopene against cytokine-induced β-cell apoptosis in INS-1 cells)

  • 본 연구는 베타세포에서 라이코펜의 항사멸 효과와 그 기전에 대해 조사하기 위해 실시하였다. 라이코펜에 의한 베타세포독성을 조사하기 위해 다양한 농도 (0.1, 1, 10 nM)로 처리하였을 경우, 저농도에서 세포독성이 나타나지 않음을 관찰하였다. 선택한 농도를 사이토카인 혼합물과 함께 처리하였을 경우, 세포 생존율이 증가하는 것을 관찰하였고, 세포사멸 유도 단백질인 Bax의 발현양은 감소하고, 세포사멸억제 단백질인 Bcl-2 발현양은 증가하는 것을 관찰하였다. 또한 사이토카인 혼합물에서 증가하였던 세포내 산화스트레스가 라이코펜과 함께 처리하였을 경우 감소되는 것을 관찰하였고 이러한 효과는 항산화 유전자인 GCLC, NQO1, HO-1의 발현양이 증가함으로서 일어난 현상임을 알 수 있었다. 라이코펜은 미토콘드리아의 생성 및 기능과 관련된 유전자의 발현을 증가시키고 사이토카인 혼합물에 의해 감소되었던 세포내 ATP 생성량을 증가시켰다. 이러한 결과는 라이코펜의 항산화효과와 미토콘드리아 기능 개선 효과가 사이토카인에 의한 베타세포 사멸을 억제하는 기전 중의 하나로 작용할 수 있음을 의미한다. 향후 라이코펜이 베타세포를 타겟으로 하는 제 2형 당뇨 치료의 기능성 소재로 개발될 가능성이 있음을 시사하는 바이다.

해산로티퍼(Brachionus plicatilis)의 생존 및 개체군 성장률을 이용한 신방오도료(Zinc undecylenate)의 독성평가 (Toxicity Assessment of Antifouling Agent using the Survival and Population Growth Rate of Marine Rotifer, Brachionus plicatilis)

  • 해산로티퍼(Brachionus plicatilis)의 생존 및 개체군 성장률(r)을 사용하여 Zinc undecylenate (ZU)에 대한 독성평가를 실시하였다. 24 h 동안 ZU에 노출된 B. plicatilis의 생존율은 실험 최고농도 $100mg\;L^{-1}$에서도 영향이 나타나지 않았으나, ZU에 72 h 노출된 개체군 성장률(r)은 농도 의존적으로 감소하는 경향을 나타내, $12.5mg\;L^{-1}$에서 유의적인 감소를 나타냈고 최고농도 $50.0mg\;L^{-1}$에서 개체군 성장률이 90% 이상 감소되었다. ZU에 노출된 B. plicatilis의 개체군 성장률의 반수영향농도($EC_{50}$)값은 $26.4mg\;L^{-1}$, 무영향농도(NOEC)는 $6.3mg\;L^{-1}$, 최소영향농도(LOEC)는 $12.5mg\;L^{-1}$로 나타났다. 자연생태계 내에서 ZU 물질이 해수 중에서 $12.5mg\;L^{-1}$ 이상을 초과하여 나타낼 때 B. plicatilis와 같은 동물성플랑크톤의 개체군 성장률이 영향을 받을 것으로 판단되며, 이러한 연구결과는 신방오도료물질의 생태안정성 평가를 위한 기준치 설정 및 다른 방오도료물질과의 독성치를 비교할 수 있는 유용한 자료로 활용될 것으로 판단된다.

중금속(As, Cr, Pb)이 해산 규조류(Skeletonema costatum)의 개체군 성장률에 미치는 영향 (Effect of Heavy Metals (As, Cr, Pb) on the Population Growth Rates of Marine Diatom, Skeletonema castatum)

  • 본 연구에서는 해산 규조류(Skeletonema cosatatum)의 개체군성장률(r)을 사용하여 중금속 3종 (As, Cr, Pb)의 독성영향 평가를 실시하였다. S. costatum을 As (0, 0.32, 0.63, 1.25, 2.50 및 5.00 mg L-1), Cr (0, 0.16, 0.32, 0.63, 1.25 및 2.50 mg L-1) 및 Pb (0, 1.25, 2.50, 5.00, 10.00 및 20.00 mg L-1)에 96시간 노출한 이후에 r 값을 측정하였다. 대조구에서 r은 0.04보다 높았으며 As 농도 0.63, Cr 농도 0.63 및 Pb 농도 10.00 mg L-1 이상에서 r이 대조구 대비 유의하게 감소하였고 중금속 농도가 증가할수록 농도 의존적으로 감소되는 경향을 나타냈다. EC50(반수영향농도)을 이용한 중금속의 독성세기는 Cr>As>Pb 순서를 나타냈으며, EC50 각각 0.80, 2.57 및 13.88 mgL-1으로 나타났으며, 최소영향농도(LOEC)는 각각 0.63, 0.63 및 10.00 mg L-1으로 나타났다. LOEC를 기준으로 해양환경에서 Cr, As, Pb 농도가 0.63, 0.63 및 10.00 mg L-1 이상이 되었을 때, S. costatum에게 독성 영향이 나타날 수 있을 것으로 판단된다. 또한, 본 연구를 통해 S. costatum의 r은 해양생태계 내에서 생산자 역할을 하는 생물에 대한 중금속의 독성평가를 위하여 유용하게 사용될 수 있을 것으로 판단된다.
