돼지풀의 수용추출물이 수종 식물에 미치는 알레로파시 효과 (The Allelopathic Effects of Aqueous Chemicals of Ambrosia artemisiifolia on Selected Plants)

  • 돼지풀의 수용추출액에 함유된 화학물질중 benzoic acids와 phenolic compounds가 주를 이루었고 일부 non-acids 화학물질도 검출되었다. 또한 수용추출액에 함유된 화학물질은 부위에 따라 차이를 나타내었으며, 잎에서 60종류, 뿌리에서도 53종류의 물질이 검출되었다. 총 함량은 잎이 0.48 mg/g.f,w, 뿌리가 0.37 mg/g.f,w이었다. 수용추출액은 수용체식물 종자발아와 유식물 생장에 뚜렷한 억제효과를 나타내었고, 식물 종에 따라 큰 차이를 보였다. 추출액의 농도가 증가됨에 따라 억제효과도 증가하였다. 발아와 유식물 생장에 억제효과가 높은 식물은 고추, 상치, 쇠무릎 등이었고, 배추와 무는 비교적 낮은 억제 효과를 보였다. 종자발아 및 유식물 생장 실험 모두에서 잎 수용추출액의 억제효과가 뿌리보다 높게 조사되었다.

Phellinus linteus KACC 93057P, '한경상황버섯'인공재배 자실체 페놀추출물의 항산화 활성 및 화학적 동정 (Chemical identification and antioxidant activity of phenolic compounds extracted from the fruiting body of 'Hankyong Sanghwang', Phellinus linteus KACC 93057P)

  • 참나무 단목원목으로 재배한 P. linteus KACC93057P(PLHS)2년산 자실체 60%에탄올 추출물의 총 페놀함량은 $19.05{\pm}0.32mg\;GAE/g$로 다른 비교구 버섯 종에 비하여 4배에서 10배이상의 높게 나타났다. $125{\mu}g/mL$농도에서 DPPH와 ABTS 라디칼 소거능은 62%와 100%를 나타냈으며 FRAP 환원력은 $0.7{\mu}MFeSO_4/g$으로 비교 구 버섯종 보다 2에서 10배이상의 유효활성이 검출 되었다. PLHS 자실체 ethyl acetate분배 추출물의 HPLC분석에서 styrylpyrone 계열의 화합물 davallialactone, hispidin, hypholomine B와 phenylpropanoid 계열의 화합물인 caffeic acid가 동정 되었다.

곰취 메탄올 추출물의 생리활성 및 암세포 증식억제 효과 (Physiological Activity of Methanol Extracts from Ligularia fischeri and Their Hyperplasia Inhibition Activity of Cancer Cell)

  • 곰취(Ligularia fischeri)의 이용성을 높이데 필요한 기초자료를 확보하고자 메탄올을 용매로 하여 추출 온도 및 시간에 따른 생리활성 효과를 조사하였다. 곰취 메탄올추출물 $1,000mg{\cdot}L^{-1}$에 함유된 총 페놀함량, 총플라보노이드 함량은 각각 $75.8-297.7mg{\cdot}L^{-1}$ and $45.6-173.6mg{\cdot}L^{-1}$이었다. 추출온도와 시간은 총 페놀함량, 총 플라보노이드 함량, 전자공여 능 의 경우 $95^{\circ}C$에서 6시간 동안 추출했을 때 가장 좋았다. 그러나 아질산염 소거능은 $75^{\circ}C$에서 12시간 추출한 것에서 97.4%로 가장 높게 나타났다. 곰취 메탄을 추출물 $200mg{\cdot}L^{-1}$$400mg{\cdot}L^{-1}$ 농도는 각각 폐암과 위암세포 증식을 90% 이상 억제시켰다. 따라서 곰취는 높은 생리활성 기능을 나타내는 채소로 나타났으며, 곰취를 가공품으로 이용하기 위해 메탄올로 추출할 때는 $95^{\circ}C$에서 6시간 정도 하는 것이 좋을 것으로 생각되었다.

고온고압증기멸균이 우황청심원에 미치는 영향 (The Effects of High Temperature High Pressure Steam Sterilization on Woohwangchungsimwon)

  • Objectives To check marker content for appropriate quality control of Woohwangchungsimwon sterilized to ensure microbiological safety and to observe antioxidant activity for any changes in efficacy. Methods To measure any effects of sterilization on the effective compounds, 8 ingredients of Woohwangchungsimwon were screened for any changes in marker content using HPLC-DAD. Using the colorimetric method on the microplate reader any changes in total phenolic compound and flavonoid levels were observed. Antioxidant activity was measured using the DPPH, ABTS, and FRAP. Results Of the ingredients of Woohwangchungsimwon, 8 were subject to quantitative analysis before and after sterilization. 21.6 mg and 1.93 mg of Glycyrrhizin was found in Glycyrrhiza uralensis Fischer pre and post sterilization, respectively. Decursin found in Angelica gigas Nakai increased from 0.16 mg to 0.29 mg after sterilization. Bilirubin found in Gallstone of Bostaurusvar. domesticus increased from 0.24 mg to 0.33 mg. Cinnamic acid found in Cinnamomum cassia Blume increased from 0.02 mg to 0.05 mg. Ginsenoside Rb1 found in Panax ginseng C. A. Meyer decreased from 0.02 mg to 0.14 mg. Paeoniflorin found in Paeonia lactiflora Pallas increased from 1.05 mg to 1.13 mg. Amygdalin found in Armeniacae Amarum Semen increased from 2.68 mg to 2.83 mg. L-muscone found in Musk increased from 0.63 mg to 0.76 mg. As for total phenolic compound and total flavonoid content, there was a 1.22 and 4.15-fold increase. DPPH and ABTS increased by 20.45% and 20.69%, respectively. FRAP activity was 2.78 times more active post stabilization. Conclusions This study confirmed that high temperature high pressure steam sterilization, a method used to ensure microbiological safety of Woohwangchungsimwon, does not affect marker content; in other words, does not affect quality of the Woohwangchungsimwon. It could also be seen that total phenolic compound and flavonoid content increased after sterilization. An antioxidant activity test showed that there was significantly increased activity of antioxidants.

Phenolic Composition and Antioxidant Activities of Different Solvent Extracts from Pine Needles in Pinus Species

  • The purpose of this study was to investigate the phenolic acid, proanthocyanidin (PAs), and flavonol glycoside contents, as well as the antioxidant activities of pine needle extracts from six species of young pine trees. The extracts were prepared from Section Pinus (Diploxylon): P. densiflora, P. sylvestris, P. pinaster and P. pinea, and Section Strobus (Haploxylon): P. koraiensis and P. strobus. Phenolics were extracted from pine needles with 80% acetone to obtain the soluble free fraction, and insoluble residues were digested with 4 M NaOH to obtain bound ethyl acetate and bound water fractions. Phenolics were analyzed by HPLC, and the hydrophilic antioxidant activity was measured using oxygen radical absorbance capacity (ORAC). Total phenolic and flavonoid contents of the soluble free fraction were higher than those of the bound ethyl acetate and bound water fractions. The main phenolics were monomers and polymers of PAs in the soluble free fraction, and phenolic acids and flavonol glycosides in bound ethyl acetate fraction. Flavonol glycosides found in different species of pine needles were qualitatively similar within fractions, but composition varied among Pinus sections. High levels of kaempferol arabinoside and an unknown compound were present in all Strobus species. The soluble free fraction had the highest antioxidant activity, followed by bound ethyl acetate and bound water fractions.

국내 재배 상추로부터 UPLC-DAD-QToF/MS를 이용한 페놀화합물 성분 비교 연구 (Study on Phenolic Compounds in Lettuce Samples Cultivated from Korea Using UPLC-DAD-QToF/MS)

  • The chemical informs about 70 individual phenolic compounds were constructed from various lettuce samples based on literature sources and analytical data. A total of 30 phenolic compounds including quercetin 3-O-glucuronide, quercetin 3-O-(6''-O- malonyl) glucoside, cyanidin 3-O-(6''-O-malonyl)glucoside, chlorogenic acid and chicoric acid as major components were identified in 6 lettuce samples from Korea using UPLC-DAD-QToF/MS on the basis of constructed library. Among these, quercetin 3,7-di-O-glucoside(m/z 627 [M+H]+), quercetin 3-O-(2''-O-malonyl)glucoside(morkotin C, m/z 551 [M+H]+), quercetin 3-O-(6''- O-malonyl)glucoside methyl ester(m/z 565 [M+H]+), 5-O-cis-p-coumaroylquinic acid(m/z 339 [M+H]+) and 5-O-caffeoylquinic acid methyl ester(m/z 369 [M+H]+) were newly confirmed from the lettuce samples. In total content of phenolic compounds, 4 red lettuce samples(2,947.7~7,535.6 mg/100 g, dry weight) showed higher than green lettuce(2,687.3 mg) and head lettuce(320.1 mg).

다양한 열처리 조건에 따른 우엉뿌리의 항산화 활성 (Antioxidant Activities of Burdock Root (Arctium lappa L.) with Various Heat Treatment Conditions)

  • This study examined the changes in antioxidant activity and contents of phenolic compounds inblanched, steamed, and autoclaved burdock root (BR). The total polyphenolic and flavonoids contents of raw and cooked BR were determined spectrophotometrically. The antioxidant activity of BR was measured using 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), and oxygen radical absorbance capacity (ORAC) assays. The main phenolic compounds in BR were quantified by HPLC (high performance liquid chromatography). Both blanching and steaming treatments significantly increased the antioxidant activities of BR in all groups (5 min, 15 min, and 30 min), whereas in autoclaving treatment, the 30 min treatment only showed an increase in the antioxidant activities of BR. The 30 min blanched BR exhibited the strongest DPPH and ABTS radical scavenging activities and possessed the highest total polyphenol and flavonoid phenolic contents. The 15 min-steamed BR showed the highest ORAC value. The main phenolic compound of the 15 min-steamed BR was CGA (chlorogenic acid). These results suggest that heat cooking methods, such as blanching and steaming, improve the antioxidant activity of BR by increasing the concentration of phenolic compounds.

Macrophoma sp.에 의한 사과 부패의 생화학적특성 II. 감염과일의 페놀함량 (Biochemical Characteristics of Apple Rot Caused by Macrophoma sp. II. Phenolic Compound Content in Infected Fruits)

  • 사과 부패병균(Macrophoma sp.)에 감염된 과일에서 총페놀, flavonol, anthocyanin 함량의 병화를 사과의 여러 발육시기에서 조사하였다. 사과가 성숙됨에 따라 과육, 과피의 총페놀함량은 급격히 감소하였다. 부패병균에 감염된 과육에서 건전한 과육보다 다소 총페놀함량이 낮았으며 품종밀러보다 더 감수성인 후지의 감염된 과육에서 이의 감소가 뚜렷하였다. 건전한 과피에서 보다 감염된 과피에서 높은 페놀함량을 보였으며 밀러에서 이의 증가가 현저하였다. 부패된 사과에서 flavonol 함량이 건전 사과보다 높았으며 7월 10일에 Macrophoma에 완전 저항성이었던 사과의 감염된 과피에 flavonol이 크게 축적되었다. anthocyanin 생성도 감염된 과피에 상당히 증가되었으며 품종 밀러에서 뚜렷했다. 이들 결과에서 미루어 사과의 페놀대사변동은 사과 부패병진전과 관계가 있을지 모른다.

국내외 수집종 구기자 잎과 줄기의 페놀화합물 함량 (Determination of Total Content of Phenolic Compounds in Chinese Matrimony Vine's Accessions)

  • 본 연구는 우리나라에서 보존하고 있는 구기자(Lycium chinesis Miller) 유전자원 131점의 잎과 줄기를 에탄올로 추출한 후 페놀화합물의 함량을 측정하여 비교 분석하기 위하여 수행되었다. 구기자 잎의 페놀 함량은 $8.8{\sim}14.9mg\;g^{-1}$을 보였으며 $11.6{\sim}13.5mg\;g^{-1}$ 사이에는 약 60%의 유전자원이 분포되어 있었다. CB03286-89 유전자원은 총 페놀함량이 $14.9{\pm}0.3mg\;g^{-1}$으로 가장 높은 반면 CBP03310-250은 $8.8{\pm}0.2mg\;g^{-1}$으로 가장 낮은 함량을 보였다. 줄기에 대한 총 페놀함량은 $6.8{\sim}12.4mg\;g^{-1}$의 함량 범위를 보이고 있으며 잎보다는 상대적으로 다소 낮은 것으로 나타났다. 구기자 잎의 페놀화합물 중 33가지 물질을 정량 분석한 결과 China collection No.1에서 가장 높게 나타났으며 99797 수집종에서 가장 낮게 나타났다. (+)catechin 함량은 CB03286-89에서 가장 많았다. Myricetin은 Geumsan jaerae, Japan No.1, China collection No.1, CL32-13, CB04329-13, China collection No.12 그리고 CB03286-89 등에서 분석되었지만 나머지 구기자 유전자원의 잎에서는 검출되지 않았다. 줄기에 대한 페놀화합물은 잎에 비하여 상대적으로 함량이 적었으며 99797 수집종이 가장 많은 함량을 보였으나 CB04329-13에서는 가장 적은 함량을 보였다. (+)catechin 함량은 Japan No.1이 $231.0{\pm}0.5{\mu}g\;g^{-1}$으로 가장 높았으며, myricetin은 Japan No.1, China collection No.1, China collection No.12, CB03286-89 및 99797에서 분석되었지만 나머지 구기자 유전자원의 줄기에서는 검출되지 않았다.

탈지들깨박 Ethanol 추출물의 항산화 효과 (Studies on Antioxidant Activity of Ethanol Extracts from Defatted Perilla Flour)

  • The antioxidant activity of ethanol extracts from defatted perilla flour was investigated by measuring peroxide value of perilla oil during storage at $45^{\circ}C$. The antioxidant activity of ethanol extracts was also compared with BHA, BHT and tocopherol. Anti-oxidant activity of ethanol extracts was also examined in corn oil and lard. The ethanol extracts contents of defatted perilla flour and the original perilla seed were 7.69 and 4.56% respectively. The antioxidant activity of ethanol extracts was superior to that of 0.02% BHT, BHA and tocopherol in the perilla oil substrate, merely in concentration of one-twentieth as much as that contained in original perilla oil seeds. The fractions of non-polar solvent (hexane and chloroform) obtained from silicic acid column chromatography are less effective than that of polar solvent as an antioxidant. Antioxidant activity of partially purified ethanol fraction is slightly inferior to that of original crude ethanol extracts. Ethanol extracts were also effective in corn oil and lard almost same as in perilla oil. The total phenolic compound contents of crude ethanol extracts and partially purified ethanol fraction were 9.3, 6.4%, respectively.
