Physicochemical and microbial characteristics of longissimus lumborum and biceps femoris muscles in Korean native black goat with wet-aging time

  • This study examined the effects of different wet-aging times on the physicochemical characteristics and microbial profile of longissimus lumborum (LL) and biceps femoris (BF) muscles from Korean native black goat (KNBG) meat. The water holding capacity (WHC), pH, cooking loss, shear force, meat color, free amino acid, total bacteria, and coliform count of KNGB meat were analyzed at 0, 5, 10, and 15 days of wet-aging at 4℃ under vacuum packaging. The results showed that different wet-aging times led to significant pH variations between the muscles throughout the aging period. The wet-aging time did not affect the WHC and cooking loss in meat from the LL muscle. In the BF muscle, however, meat wet-aged for five days had a significantly higher WHC and less cooking loss than meat aged for 0, 10, and 15 days. The meat from the LL muscle wet-aged for five days produced tenderer meat (low shear force value) than the unaged meat (p < 0.05). Moreover, the color was similar in the LL muscle regardless of the number of aging days. In the BF muscle, the redness (a*) was higher in the meat wet-aged for 15 days compared to that aged for 0, 5, and 10 days (p < 0.05). Regardless of the muscles, an increase in wet-aging time led to an increase in the total free amino acids contents in both muscles (p < 0.05). On the other hand, the tasty/bitter amino acid ratio was significantly higher for five days of wet-aged meat than 10 and 15 days of aging from the BF muscle. In addition, regardless of the muscles, the total bacteria and coliform counts were significantly lower for five days of wet-aged meat than 10 and 15 days of aging (p < 0.05). Therefore, chevon wet-aged for five days is an optimal aging period under vacuum packaging that fortifies meat quality with a minimal microbial negative defect.

Effects of Surfactant Tween 80 on Forage Degradability and Microbial Growth on the In vitro Rumen Mixed and Pure Cultures

  • Effect of a surfactant Tween 80 on the bacterial growth in the rumen was examined on the in vitro pure cultures of Streptococcus bovis, Selenomonas ruminantium, Butyrivibrio fibrisolvens, Prevotella ruminicola, Megasphaera elsidenni, Fibrobacta succinogenes, Ruminanococcus albus and Ruminococcus flavefaciens. Dry matter degradability (DMD), concentrations and compositions of volatile fatty acids (VFA), and the most probable number (MPN) of cellulolytic bacteria and total number of bacteria in the presence of Tween 80 were also examined on the in vitro rumen mixed culture either with barley grain or orchardgrass hay. The growth of S. bovis, S. ruminantium, B. fibrisolvens, P. ruminicola, M. elsidenni and F. succinogenes were significantly higher (p<0.05) at over 0.05% concentrations of Tween 80 than those of the control cultures, while was not changed with R. albus and R. flavefaciens. With rumen mixed culture the DMD of barley grain and orchardgrass hay was significantly higher (p<0.05) at a 0.2% concentration of Tween 80 than the control, being reflected in the significantly higher (p<0.05) VFA production (mmol $g^{-1}$DDM) with orchardgrass hay. The higher (p<0.05) ratio of propionate to acetate at a 0.2% concentration of Tween 80 was also observed with orchardgrass hay, showing a similar trend with barley grain. No changes in the total bacterial number and MPN of cellulolytic bacteria were observed.

도시에서 다양한 토양의 물리화학적 특성과 미생물 활성 (Physicochemical Characteristics and Microbial Activity in the Various Urban Soils)

  • 도시에서 토양을 보존하고 합리적으로 이용하기 위하여 도시 토양을 과학적으로 이해하는 것이 필요하지만 아직까지 이처럼 과도하게 관리되고 교란된 토양의 특성에 대하여 집중적으로 연구되지 않았다. 본 연구에서 조사된 인천의 산림, 잔디밭, 가로수, 나지 및 포장 밑 토양에서 답압에 의한 견밀화에 의하여 토양 가밀도가 높았다. 또한 다양한 도시 토양에서 수분함량은 전원지 산림 토양에 비하여 적었고 지온은 높았다. 화학적으로는 도시 토양에서 pH가 중성에 가깝고 유기물 함량이 적었다. 도시 토양의 총세균수는 전원지 산림 토양의 단지 5∼50% 수준이었다. 단계식 다중회귀분석의 결과, 토양 유기물함량이 총세균수에 영향을 미치는 주요한 요인으로서 선정되었다. 대부분의 도시 토양에서 탈수소효소 활성도는 전원지 산림 토양과 유의한 차이가 없었으나, 아스팔트 포장 밑 토양에서는 낮았다 그러므로 견밀한 도시 토양에서 낮은 유기물함량이 물질 순환에 관여하는 미생물 수에 악영향을 미친다고 판단되었다.

Rumen Microbial Population in the In vitro Fermentation of Different Ratios of Forage and Concentrate in the Presence of Whole Lerak (Sapindus rarak) Fruit Extract

  • This experiment was designed to investigate the effect of lerak extract on the dynamic of rumen microbes in the in vitro fermentation of diet with different ratios of forage and concentrate. In vitro fermentation was conducted according to the method of Tilley and Terry (1963). The design of experiment was a factorial block design with 2 factors. The first factor was the ratio of forage and concentrate (90:10, 80:20, and 70:30 w/w) and the second factor was the level of lerak extract (0, 0.6, and 0.8 mg/ml). Total volatile fatty acid (VFA) concentration, proportional VFA and NH3 concentration were measured at 4 h incubation. Protozoal numbers in the buffered rumen fluid after 4 and 24 h of incubation were counted under a microscope. Bacterial DNAs of buffered rumen fluid were isolated from incubated samples after 24 h of incubation using a QiaAmp kit. Total bacteria, Fibrobacter succinogenes, Ruminococcus albus, and Prevotella ruminicola were quantified using real time polymerase chain reaction (PCR). Lerak extract markedly reduced protozoal numbers in buffered rumen fluid of all diets after 24 h of incubation. Total bacteria did not change with lerak extract addition. While no difference in F. succinogenes was found, there was a slight increase in R. albus number and a significant enhancement in P. ruminicola number by increasing the level of lerak extract in all diets. Propionate concentration significantly increased in the presence of lerak extract at level 0.8 mg/ml. It was concluded that the addition of lerak extract could modify rumen fermentation and had positive effects on rumen microbes.

바이오디젤의 미생물 안정성 조사 (Microbiological Stability Test of Biodisel)

  • 동ㆍ식물성 기름으로부터 산이나 알카리 촉매를 사용하여 제조한 친환경 에너지인 바이오디젤의 미생물학적 안정성을 테스트하기 위하여 온도와 저장기간에 따른 미생물에 의한 변질정도를 측정하였다. 측정방법으로 역상 현미경 관찰과, 3M petrifilm분석법으로 생균수를 측정하였고, 균체 단백질의 양을 측정하기 위하여 Bradford protein assay법을 사용하였다. 바이오디젤과 석유디젤, 그리고 각각의 시료를 혼합한 시료를 서로 다른 온도조건(25℃와 35℃)에서 혐기적인 조건으로 90일동안 장기간 저장했을 때 바이오디젤, 석유디젤 그리고 혼합한 시료에서 미생물의 생육은 관찰되지 않았다.

미생물 및 생화학적 질량역적분석에 의한 퇴비화단계별 부숙도 평가 (Assessment of Compost Maturity on Their Different Stages with Microbial and Biochemical Mass Dynamics)

  • 유기물의 퇴비화과정중 미생물과 이에 관련된 퇴비의 생화학적 질량의 변화는 퇴비화공정 최적화와 최종산물의 품질은 매우 중요하다. 본 연구에서는 퇴비화단계중 미생물과 관련 생화학적 변수의 질량변화가 퇴비부숙도의 기준으로서의 적합성을 평가하였다. 전국 5개 퇴비공장 (용인축협, 양평축협, 논산축협, 전주연초, 지리산낙협)에서 세 단계 (초기, 부숙, 후숙)의 퇴비시료를 채취하여, Total Aerobic Bacteria(TAB), Coliforms, Escherichia coli, Actinomycetes, Fungi 등의 군집농도를 분석하였다. 연구결과, 5개 퇴비공장의 시료에서 Coliforms과 E.coli는 부숙단계에서 급격히 감소되어 후숙단계에서는 완전 사멸되었으나 다른 미생물은 완전 사멸되지 않았다. 그러므로 Coliforms과 E.coli 군집을 부숙도의 기준으로 제시하였다. 미생물탄소질량/질소질량비 (MBC/MBN)는 부숙단계에서 약간 감소하였으며, 후숙 단계에서는 증가하였다. 이는 부숙단계에서 Coliforms, E. coli, Fungi 등의 군집감소 때문으로, 후숙단계에서는 Fungi 및 TAB 군집증가 때문으로 이해된다. 또한 중금속성분 농도는 방선균 군집과 매우 강한 음(陰)의 상관성을 나타내었다. 본 연구의 성과는 Coliforms과 E.coli 군집 농도를 퇴비부숙도 기준으로 제시하였으며, 중금속농도와 미생물군집농도 상관관계를 이용하여 퇴비품질의 평가기준을 제시한 데 있다.

출토복식유물에 대한 훈증소독의 영향에 관한 연구 (A Study on the Effects of Fumigation on Excavated Costumes)

  • This study examines the effects and influences of fumigation on the antimicrobial treatment of excavated dresses in two different museum settings using chemical composite of Methyl Bromide and Ethylene Oxide. The microbial test indicated that the prewashed samples contained Micrococcus, Bacillus, Staphylococcus, and some unidentified fungi and that the total number of microbes decreased after washing. While it was observed that most germs were removed immediately after fumigation, Bacillus was still found and its number even increased. After 5 months, the number of microbes found in Museum A was smaller than Museum B, which shows that Museum A equipped with better environment control facility was superior to Museum. B in terms of exhibition and preservation of dresses against organic damages. In addition, the result indicates that fumigation is an effective measure against the growth and reproduction of microorganisms, considering that the number of microorganisms decreased after fumigation. The color of test sample showed immediate change after fumigation but color difference became stabilized and decreased with the passage of time. The test samples from both museums showed similar patterns.

Monitoring of petroleum hydrocarbon degradative potential of indigenous microorganisms in ozonated soil

  • Diesel-contaminated soils were ozonated for different times (0 - 900 min) and incubated for 9 wk to monitor petroleum hydrocarbons (PH)-degradative potential of indigenous microorganisms in the soils. Increased ozonation time decreased not only concentration of PH but also number of microorganisms in the soils. Microorganisms in the ozonated soils increased during 9-wk incubation as monitored by culture- and nonculture-based methods. Higher (1-2 orders of magnitude) cell number was observed by quantitative analysis of soil DNA using probes detecting genes encoding 165 rRNA(rrn), naphthalene dioxygenase (nahA), toluene dioxygenase (todC), and alkane hydroxylase (alkB) than microbial abundance estimated by culture-based methods. Such PH-degraders were relatively a few or under detection limit in 900-min ozonated soil. Further PH-removal observed during the incubation period supported the presence of PH-degraders in ozonated soils. Highest reduction (25.4%) of total PH (TPH) was observed in 180-min ozonated soil white negligible reduction was shown in 900-min ozonated soil during the period, resulting in lowest TPH-concentration in 180-min ozonated soil among the ozonated soils. Microbial community composition in 9-wk incubated soils revealed slight difference between 900-min ozonated and unozonated soils as analyzed by whole cell hybridization using group-specific rRNA-targeted oligonucleotides. Results of this study suggest that appropriate ozonation and subsequent biodegradation by indigenous microorganisms may be a cost-effective and successful remediation strategy for PH-contaminated soils.

Aqueous Chlorine Dioxide Treatment Decreases Microbial Contamination and Preserves Sensory Properties of Mackerel During Storage

  • Effect of aqueous chlorine dioxide ($ClO_{2}$) treatment on quality change of mackerel during storage was examined. Mackerel treated with 0, 5, 10, and 50 ppm of $ClO_{2}$ solution, respectively was stored at $4^{\circ}C$. $ClO_{2}$ treatment decreased populations of aerobic bacteria in mackerel during storage. The number of total aerobic bacteria of mackerel treated with 50 ppm $ClO_{2}$ increased from 2.45 to 3.44 log CFU/g after 9 days of storage, while that of the control increased from 3.47 to 4.72 log CFU/g. The pH values of mackerel increased during storage, with no significant changes among treatments. Volatile basic nitrogen values of mackerel were decreased by $ClO_{2}$ treatment. Quality of mackerel treated with $ClO_{2}$ was better than that of the control during storage based on sensory evaluation. These results indicate that aqueous $ClO_{2}$ treatment could be useful for improving the microbial safety and qualities of mackerel.

Metagenomic and Proteomic Analyses of a Mangrove Microbial Community Following Green Macroalgae Enteromorpha prolifera Degradation

  • A mangrove microbial community was analyzed at the gene and protein levels using metagenomic and proteomic methods with the green macroalgae Enteromorpha prolifera as the substrate. Total DNA was sequenced on the Illumina HiSeq 2000 PE-100 platform. Two-dimensional gel electrophoresis in combination with liquid chromatography tandem mass spectrometry was used for proteomic analysis. The metagenomic data revealed that the orders Pseudomonadales, Rhizobiales, and Sphingomonadales were the most prevalent in the mangrove microbial community. By monitoring changes at the functional level, proteomic analyses detected ATP synthase and transporter proteins, which were expressed mainly by members of the phyla Proteobacteria and Bacteroidetes. Members of the phylum Proteobacteria expressed a high number of sugar transporters and demonstrated specialized and efficient digestion of various glycans. A few glycoside hydrolases were detected in members of the phylum Firmicutes, which appeared to be the main cellulose-degrading bacteria. This is the first report of multiple "omics" analysis of E. prolifera degradation. These results support the fact that key enzymes of glycoside hydrolase family were expressed in large quantities, indicating the high metabolic activity of the community.
