Dryfilm method by using 3M Petrifilm$^{TM}$ has been examined to replace conventional agar method for isolation of microorganisms from foods. The objectives of the present study were to evaluate suitability of dryfilm method as a microbial isolation method and to determine the effect of antimicrobial agent on dryfilm for isolation of microorganisms from foods. Five different foods, milk, ground beef, fishery surimi, Takju and wheat flour were used to isolate the natural microflora in foods and the inoculated Escheri chia coli. Standard method agar (SMA, Difco) and Petrifilm$^{TM}$ aerobic count (PAC, 3M) were used to isolate total microorganisms from foods. Violet red bile agar (VRBA), brilliant green lactose bile (BGLB) broth and Petrifilm$^{TM}$ coliform count (PCC, 3M) were used to isolate coliforms from foods. E. coli broth (EC broth) and Petrifilm$^{TM}$ E. coli count (PEC, 3M) were used to isolate E. coli from foods. Acidified potato dextrose agar (APDA) and Petrifilm$^{TM}$ yeast & mold count (PYMC, 3M) were used to isolate yeasts and molds from foods. Total aerobic plate counts isolated from five different foods by SMA and PAC (3M) were riot significantly different each other at P<0.05 level and were highly correlated each other ($\geq$0.96). Mugwort extract as an antimicrobial agent did not affect microbial enumeratiion of Dryfilm. Significantly higher number of coliform colonies were formed on VRBA than PCC (3M) from ground beef, but they were not significantly different in coliform colonies from milk samples. PCC (3M) and BGLB were not significantly different for enumeration of coliforms in milk and beef samples. Significantly higher number of E. coli were isolated by EC broth than PEC from ground beef, but these were not significontly different for enumeration of E. coli from milk. Yeast and mold counts isolated from Takju and wheat flour by APDA and PYMC (3M) were not significantly different at P<0.05 level. These data indicate that dryfilm method by using 3M Petrifilm$^{TM}$ can be successively used as an alternative to conventional agar method for enumeration of microorganisms in various foods.
The present study evaluated the variations in soil microbial population of controlled horticultural land used for lettuce (Lactuca sativa) cultivation by their fatty acid methyl ester and chemical properties. We utilized four treatment groups, no treatment (NT), culture medium (CM), Bacillus subtilis S37-2 (KACC 91281P) ${\times}10^6CFU\;mL^{-1}$ (BS1), and Bacillus subtilis $S37-2{\times}10^7CFU\;mL^{-1}$ (BS2) and analyzed these variations throughout the before treatment and harvesting stage. The chemical properties such as pH, organic matter, available phosphate, and electrical conductivity in soils before treatment and harvesting stage showed no significant difference among the treatments. Total numbers of bacteria and microbial biomass C in soil treated with BS1 were larger than those of NT, CM, and BS2, whereas total number of fungi at the harvesting stage was significantly lower in the BS1 soil than in the NT and CM soils (P < 0.05). On basis of leaf length, leaf width, leaf number and leaf weight, the growth characteristics lettuce on the soil treated with BS1 and BS2 was faster than those of NT and CM soils. Yield of lettuce with treated BS1 and BS2 were 35% and 29% more than that of NT, respectively.
The purpose of this study was that to estimate the microbial and sensory quality by the method and time of holding in the step of holding of Cow's Knee Broth, Stir-Fry Pork and Cooked Cuttlefish that were provide at the industry foodservice establishment, then to suggest safer feeding by the reasonable holding method. The results are as follows: Cow's Knee Broth: holding at room temperature was shown that the number of mi- crobiology by passage of the holding time was higer than the case of holding at steam table, but it was lower after reheating it. As a result of sensory test, it was exellent to holding at steam table and to reheat after holding room temperature within four hours and six hours each. Stir-Fry Pork: as a result of microbial analysis, in the case of holding at the steam table microbes increased safer two hours, but after four and six hours they little reduced. In the case of holding at room temperature the number of microbes increased according to the passage of time. As a result of sensory analysis, its taste of room temperature holding was fine until four and six hours each. Cooked Cuttlefish: as a microbial analysis, it was shown that the amount of total plate counts of room temperature holding ten times as high as the latter after six hours. As the result of sensory analysis, the cold holding was better until six hours and the room temperature holding was fine just until two hours. As a result of food poisoning bacteria, it was negative in every test in sample against Salmonella, Staphylococcus aureus, Vibrio parahaemolyticus.
This study was carried out to investigate the effects of soil microbial fertilizers on yields of Chinese cabbage (Brassica campestris L.). Five microbial fertilizers, MPK+Husk+Palma, Husk+Palma, MPK+Compost, BLCS(Bio livestock cattle system) cattle dropping, and Tomi, were used. All of microbial fertilizers significantly increased yields of Chinese cabbage, except BLCS cattle dropping. MPK+Husk+Palma was the most effective than any other treatments. Chinese cabbage treated with Tomi showed higher concentrations of K, Ca, Mg, Fe, Mn, and Zn than Compost treatment. MPK+ Husk+Palma was high in concentrations of Mg and Mn. MPK+Compost was high in concentrations of K, Mg, and Na. In a chemical components of soil, concentrations of K and P was increased with Tomi treatment, however, the other concentrations of plant and soil chemical components were not different. In a microbial properties of soil, Tomi, Husk+Palma, and MPK+Husk+Palma treatments increased in the number of total bacteria and bacilli. Tomi treatment increased in the number of actinomycetes and fungi. The other microbial properties of soil showed no significant differences. It would be assumed that yield increase in Chinese cabbage might be due to the beneficial microbial properties, therefore, those would increase yields of Chinese cabbage.
An in vitro experiment was conducted to evaluate the effects of Aspergillus oryzae culture (AOC) and 2-hydroxy-4-(methylthio)-butanoic acid (HMB) on rumen fermentation and microbial populations between different roughage sources. Two roughage sources (Chinese wild rye [CWR] vs corn silage [CS]) were assigned in a $2{\times}3$ factorial arrangement with HMB (0 or 15 mg) and AOC (0, 3, or 6 mg). Gas production (GP), microbial protein (MCP) and total volatile fatty acid (VFA) were increased in response to addition of HMB and AOC (p<0.01) for the two roughages. The HMB and AOC showed inconsistent effects on ammonia-N with different substrates. For CWR, neither HMB nor AOC had significant effect on molar proportion of individual VFA. For CS, acetate was increased (p = 0.02) and butyrate was decreased (p<0.01) by adding HMB and AOC. Increase of propionate was only occurred with AOC (p<0.01). Populations of protozoa ($p{\leq}0.03$) and fungi ($p{\leq}0.02$) of CWR were differently influenced by HMB and AOC. Percentages of F. succinogenes, R. albus, and R. flavefaciens (p<0.01) increased when AOC was added to CWR. For CS, HMB decreased the protozoa population (p = 0.01) and increased the populations of F. succinogenes and R. albus ($p{\leq}0.03$). Populations of fungi, F. succinogenes (p = 0.02) and R. flavefacien (p = 0.03) were increased by adding AOC. The HMB${\times}$AOC interactions were noted in MCP, fungi and R. flavefacien for CWR and GP, ammonia-N, MCP, total VFA, propionate, acetate/propionate (A/P) and R. albus for CS. It is inferred that addition of HMB and AOC could influence rumen fermentation of forages by increasing the number of rumen microbes.

Distribution changes in microalgae communities were studied in the Nakdonggang River at two sampling stations (St.1 Gyeongcheongyo Bridge (GB) and St.2 Daedong Wharf (DW)) at monthly intervals from January 2021 to November 2021. A total of 83 taxa included 82 species, 1 forma, belonging to 49 genera, 32 families, 21 orders, and 8 classes. The most important groups were Bacillariophyta and Chlorophyta. The number of species ranged from 5 to 24 in GB, and from 9 to 21 taxa in DW. The contribution of Bacillariophyta to the total species richness was the highest during all survey periods, and Chlorophyta yielded the next highest value in the study area. The dominant taxa were Aulacoseira ambigua, A. ambigua f. japonica, and Ulnaria acus in this study. Cluster analysis and non-metric multidimensional scaling (nMDS) analysis based on Bray- Curtis similarity identified 4 major groups, which corresponded to microalgae assemblages and their characteristic species. Correlation was analyzed through the CCA analysis. It was found that there was a correlation between the microalgae and environmental factors. It was revealed that the divided groups were distinguished because of the differences by the survey period. Therefore, seasonal change was judged as a major factor affecting the distribution of microalgae communities.

In this study, the TPH(Total Petroleum Hydrocarbon) contamination and microbial ecological characteristics in petroleum-contaminated site were investigated through the correlation among the vertical TPH contamination distribution of the site, the geochemical characteristics, and the indigenous microbial ecology. The high TPH concentration showed in the vicinity of 3~4 m or less which is thought to be affected by vertical movement due to the impervious clay layer. In addition, the TPH concentration was found to have a positive correlation with Fe2+, TOC concentration, and the number of petroleum-degrading bacteria, and a negative correlation with the microbial community diversity. The microbial community according to the vertical distribution of TPH showed that Proteobacteria and Firmicutes at the phylum level were dominant in this study area as a whole, and they competed with each other. In particular, it was confirmed that the difference in the microbial community was different due to the difference in the degree of vertical TPH contamination. In addition, the genera Acidovorax, Leptolinea, Rugoshibacter, and Smithella appeared dominant in the samples in which TPH was detected, which is considered to be the microorganisms involved in the degradation of TPH in this study area. It is expected that this study can be used as an important data to understand the contamination characteristics and biogeochemical and microbial characteristics of these TPH-contaminated sites.

The purpose of this study was to investigate the effects of ionized calcium treatment on total bacterial cross-contamination of chicken carcass surface in the slaughtering process. The growth of Escherichia coli was strongly inhibited in a medium prepared by using a 0.5% ionized calcium solution. The total bacterial cross-contamination of chicken carcass surface and the scalding water was significantly increased as the number of scalding was increased (p<0.05). The total bacterial cross-contamination of chicken carcass surface reached a plateau without a further increase as scalding was performed consecutively for 10 or more times. The total bacterial cross-contamination of the scalding water was significantly increased as the number of scalding was increased (p<0.05). The total bacterial cross-contamination of chicken carcass surface of the chickens raised on a floor type farm was significantly higher than that of the chickens raised in a battery cages (p<0.05). The total bacterial cross-contamination of chicken carcass surface of the chickens raised on a floor type farm was significantly lower in the 0.5% ionized calcium solution treatment group than in the control group (p<0.05).
This study was carried out to obtain some useful data for increasing an effective ginseng production. There was a direct relationship (r=0.2645) between spore germination of Fusarium solani and soil pH, and (r=0.315) between Cylindrocarpon destructans and soil pH. On the other hand, there was a direct relationship (r=0.19) between relative hyphal growth of Rhizoctonia solani and soil pH. There was a direct relationship (r=0.21) between number of total bacteria and F. solani, (r=0.37) between actinomycetes and F. solani and (r=0.20) between celluloytic bacteria and F. solani. However, there was an inverse relationship (r=-0.20) between number of total fungi and F. solani. There was a direct relationship (r=0.24) between number of actinomycetes and R. solani. Each ginseng pathogen-suppressive soil screened was 40 in F. solani, 20 in C. destructans and 9 soil samples in R. solani among 146 soil samples, respectively. The mean contents of K, Ca and Mg were fairly lower in each ginseng pathogen-suppressive soil than conducive soil, whereas Na were somewhat lower. The mean contents of organic matter were over 2 times higher in each ginseng pathogen-suppressive soil than conducive soil. The mean contents of phosphate were fairly lower in F. solani and R. solani-suppressive soil than conducive soil and, on the other hand, were somewhat higher in C. destructans-suppressive soil than conducive soil. The mean soil pH was somewhat lower in each ginseng pathogen-suppressive soil than conducive soil. The mean contents of sand were about 2 times higher in each ginseng pathogensuppressive soil than conducive soil, whereas silt and clay were somewhat lower. The microbial numbers of total bacteria, total fungi and celluloytic fungi were higher in F. solani-suppressive soil than conducive soil, whereas actinomycetes and celluloytic bacteria were lower. Each microbial number of total bacteria or total fungi indicated a significant difference (p=0.05) between F. solanisuppressive and conducive soil, and the microbial number of actinomycetes was a highly significant difference (p=0.01) between F. solani-suppressive and conducive soil. The microbial numbers of total bacteria, total fungi, actinomycetes and celluloytic fungi were higher in C. destructans-suppressive soil than conducive soil, whereas celluloytic bacteria were about 2 times lower. On the other hand, the microbial numbers of total fungi were higher in R. solani-suppressive soil than conducive soil, whereas total bacteria, actinomycetes, celluloytic bacteria and celluloytic fungi were lower. Fourteen of 16 F. solani-suppressive soils tested were suppressive to ginseng root rot, whereas fifteen of 16 C. destructans-suppressive soils were suppressive. Ginseng root rots of ginseng disease-suppressive soils were in the range of 1.0-17.4% in F. solani-suppressive soil and 0.2-20.4% in C. destructans-suppressive soil, respectively.

The microbial response after treatment of antibiotics was studied for investigating the resistance pattern under simulated aquaculture environment. A marine microcosm was developed for marine fish farm environment using artificial seawater and sediment. Oxolinic acid, which has been commonly used in aquaculture, was employed for the experiment. Resistance patterns and the changes of microbial community were monitored before. during and after use of oxolinic acid. Vibrionaceae was the dominant bacterial species throughout the experiment, consisting 65-75% of total bacterial number in fish farm environment. However, some gram-positive bacteria, Micrococcos sp. and Bacillus sp. strains in marine farm environment were increased in proportion to their number during the treatment. ETS activity of the bacterial communities in aquaculture environment was reduced to 42-67% during the treatment of oxolinic acid. But recovering trends of bacterial number were also detected immediately after cease of oxolinic acid treatment. Frequent treatment of oxolinic acid under the simulated fish farm environment showed bacterial resistance to increase sharply.
