Effect of Dry Powders, Ethanol Extracts and Juices of Radish and Onion on Lipid Metabolism and Antioxidative Capacity in Rats (무, 양파의 시료제조 방법에 따른 흰쥐의 지방대사와 항산화능에 관한 연구)

  • This study was performed to investigate the effects of dry powders, ethanol extracts and juices of radish and onion on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in rats. Forty-nine male Sprague-Dawley rats weighing 157$\pm$6g were blocked into seven groups according to body weight and raised for four weeks with diets containing 5%(w/w) dry powders of two different vegetables consumed frequently by Korean-radish(Raphanus sativus L.) and onion(Allium cepa L.), ethanol extracts and juices from equal amount of each dry powder. All the powders, ethanol extracts and juices of radish and onion decreased total lipids, triglycerides and total cholesterol concentrations in plasma and liver. Above all, onion ethanol extract decreased them most remarkably. It was thought that organosulfur compounds and flavonoids extracted from onion by ethanol inhibited biosynthesis and absorption of lipid and promoted degradation of lipid. Radish powder also decreased them by increasing fecal excretions of total lipids, triglycerides and total cholesterol most effectively. Catalase and glutathine peroxidase(GSH-px) activities in red blood cell(RBC) were most remarkably increased by radish powder and onion powder respectively. Superoxide dismutase(SOD), catalase and GSH-px activities in liver were most remarkably increased by onion ethanol extract, radish powder and onion ethanol extract respectively. Xanthine oxidase(XOD) activities in liver were most effectively decreased by ethanol extracts of radish and onion. Thiobarbituric acid reactive substance (TBARS) levels in plasma and liver of experimental groups were significantly lower than those of controls. Above all, onion powder decreased them most effectively. It was thought that vitamin E and high flavonoids in onion powder inhibited lipid peroxidation, promoting liver and RBC SOD, catalase and GSH-px activities and inhibiting XOD activities effectively. Flavonoids in onion ethanol extract inhibited lipid peroxidation, promoting three antioxident enzyme activities and inhibiting XOD activities most remarkably. Also flavonoids and high vitamin C in radish powder inhibited lipid peroxidation, promoting liver and RBC catalase most remarkably and inhibiting XOD activities. In conclusion, radish and onion were effective in lowering lipid levels and inhibiting of lipid peroxidation in animal tissue. From these data, radish and onion can be recommended in the treatment and prevention of diseases such as cardiovascular disease and cancer and in delaying aging. As ethanol from onion were most effective in lowering lipid level and promoting three antioxident enzymes, and inhibited lipid peroxidation as did we should try to utilize onion skin which is discarded though reported to have abundant flavonoids. Optimization of Extraction Process for Antioxidant from Persimmon Leaf and Thistle Using Response Surface Methodology (반응표면분석법을 이용한 감잎과 엉겅퀴로부터 항산화성분의 추출공정 최적화)

  • In this study, we extracted an antioxidant from natural products which are known to have a high antioxidant content and also optimized the extraction process by applying a response surface methodology (RSM). In addition, by measuring the total flavonoids and total polyphenols of the active ingredient extracted persimmon leaf and thistle, the functionality of the active ingredient was evaluated. Both ultrapure water and alcohol were used as extraction solvents and the ratio of ultrapure and alcohol, amount of sample, extraction time, extraction temperature were set as independence variables. Also, the yield, total polyphenols, and flavonoids was set as the response. Optimal extraction conditions were as follows; for persimmon leaf, the extraction time = 3.1 h, ratio of alcohol/ultrapure = 63.4 vol%, and temperature = $54.6^{\circ}C$ while for thistle the extraction time = 2.9 h, ratio of alcohol/ultrapure = 40.7 vol%, and temperature = $68.4^{\circ}C$. Also, the response were as follows; for persimmon leaf, the yield = 27.7%, total polyphenols = 33.2 mg GAE/g, and total flavonoids = 47.8 mg QE/mg dw, whereas for thistle the yield = 27.0%, total polyphenols = 17.9 mg GAE/g, and total flavonoids = 28.8 mg QE/mg dw at the optimal conditions. The overall satisfaction level was 71.7%.

  • This study was devised to investigate the effects of flavonoids on carbon tetrachloride toxicity of rats. Through the experiments, the contents of total cholesterol, transaminase(GOT, GPT) and alkaline phosphatase(Alk. P) activities in the liver and serum were determined. The results obtained are summarized as follows; (1) The liver cholesterol concentration of flavonoid injected groups were remarkably increased up to 45% at 2 days after $CCl_4$ injection and then the values were almost recovered to the level of control group at 4 days after $CCl_4$ injection. The increase of serum cholesterol content was inhibited by the flavonoids after $CCl_4$ injection, but the result was not significant. (2) Flavonoids injection was found to have the ability to decrease the elevated serum GOT, Alk. P activities resulting from injection of $CCl_4$ and induce rapid recovery from such an elevated level and the extent of such a decreasing action was greatest in hesperidin injection group. The liver microsomal Alk. P and GOT activities were not affected by the treatment of flavonoids. (3) The elevated serum and liver microsomal GPT activities induced by $CCl_4$ injection were inhibited by the injection of flavonoids.

Antioxidant Activity and Cytotoxicity of Different Taraxacum Species in Korea (국내 자생 민들레의 종류별 항산화성 및 세포독성 연구)

  • Contents of phenolics and flavonoids, antioxidant activity and cytotoxicity were investigated in the methanol extracts of three different $Taraxacum$ species, $Taraxacum$ $coreanum$, $Taraxacum$ $mongolicum$, and $Taraxacum$ $officinale$. Total phenolics content at $1000mg\;kg^{-1}$ was more present in shoot parts than in roots, and was highest in $T.$ $mongolicum$ shoot and root extracts (76.8 and $40.0mg\;kg^{-1}$, respectively), followed by $T.$ $coreanum$ and $T.$ $officinale$ ($p$ < 0.05). Total flavonoid level had same tendency to total phenolics among $Taraxacum$ species, showing lower amounts ($6.5{\sim}36.4mg\;kg^{-1}$) than total phenolics. The antioxidant activity of the methanol extracts from all the species dose-dependently increased. DPPH free radical scavenging activity at $1,000mg\;kg^{-1}$ was highest in shoot and root extracts from $T.$ $mongolicum$ by 89.6 and 83.4%, respectively. According to MTT assay, cell viability of Calu-6 (human pulmonary carcinoma) was lowest in the $T.$ $mongolicum$ shoot and root extracts ($IC_{50}$ values=83.4 and $66.4mg\;kg^{-1}$, respectively), and followed by $T.$ $coreanum$ and $T.$ $officinale$ (lowest). Calu-6 was more sensitive to the extracts than SNU-601 (human gastric carcinoma). Antioxidative and anticancer activities in three different $Taraxacum$ species was more correlated with total phenolics content ($r^2$=0.0097 to 0.6213) than with total flavonoids level ($r^2$=0.0027 to 0.4627). The results showed total phenolics content and total flavonoids level were highly correlated with anticancer activity and antioxidant activity, and their content and activities were different depending on species.

Phenolics Level and Antioxidant Activity of Methanol Extracts from Different Plant Parts in Youngia sonchifolia (고들빼기 부위별 메탄올 추출물의 폴리페놀 함량 및 항산화성 연구)

  • Proximate composition, total phenolics and total flavonoids level, DPPH radical scavenging activity, and cytotoxicity were determined in the methanol extracts of different plant parts of Youngia sonchifolia at reproductive growth stage. Crude protein and crude fat were present as the highest amount in flowers, and crude fiber in the stems and roots. The highest content of phenolics [mg ferulic acid equivalents (FAE) $kg^{-1}$ dry weight (DW)] was found in flowers (highest) and followed by leaves, stems and roots (lowest). Flavonoids [mg rutin equivalents $kg^{-1}$ DW] level, however, showed the highest in leaf extracts and lowest in root extracts. The antioxidant potential of the methanol extracts from the plants dose-dependently increased DPPH (1,1-diphenyl-2-picrylhydrazyl) free radical scavenging activity (%). DPPH radical scavenging activity were highest in root extracts ($IC_{50}=1,135.6\;mg\;kg^{-1}$) and followed by leaf, stem and flower extracts. By way of MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay, methanol extracts of roots showed the highest anticancer activity on human cancer cell line Calu-6 for human pulmonary carcinoma ($IC_{50}=196.3\;mg\;kg^{-1}$) and HCT-116 for human colon carcinoma ($IC_{50}=623.6\;mg\;kg^{-1}$).

Further Study on the Anti-hypercholesterolemic Effect of Ixeris sonchifolia (고들빼기의 고콜레스테롤혈증 개선효과)

  • Effect of various fractions prepared from the methanol extract of Ixeris sonchifolia and flavonoids isolated from the extract on hypercholesterolemic mice were examined. When various fractions $(CHCl_3,\;Interphase,\;EtOAc,\;n-BuOH\;and\;H_2O)$were administered to mice, the Interphase material produced a significant activity. Total cholesterol level in the Interphase material-treated group given doses of 25 and 50mg was decreased significantly by as much as 16% and 30%, respectively. Among the isolated flavonoids, cynaroside caused a significant hypocholesterolemic activity.

Effect of Dried Powders of Chamomile, Sage, and Green Tea on Antioxidative Capacity in 15-Month-Old Rats (캐모마일, 세이지, 녹차 건분의 섭취가 노령흰쥐의 항산화능에 미치는 영향)

  • This study was performed to investigate the effect of chamomile, sage, and green tea intakes on anti oxidative capacity in 15-month-old rats. Dried powders of three plants were analyzed to determine the amount of total flavonoids, $\beta$-carotene, vitamin C, vitamin E, and dietary fibers. In order to examine the change of antioxidative capacity in old rats, forty-eight Sprague-Dawley male rats weighing 621.2$\pm$9.5 g were divided into four groups according to body weight and fed for four weeks with each experimental diet of three dried powders and control diet. Plasma and liver thiobarbituric acid reactive substances (TBARS) levels and xanthine oxidase (XO) activities, erythrocyte and liver superoxide dismutase (SOD) activities and plasma vitamin A, C, E, and total carotenoids levels were measured. The total flavonoids and vitamin C contents were the highest in green tea powder. Beta-carotene and vitamin E contents were not significantly different among all three dried powders. Total dietary fiber contents also were not different among all three dried powders, but soluble dietary fiber contents of chamomile was higher than other two dried powders. Plasma TBARS level was found to be significantly lower in all the experimental groups as compared to control group. Chamomile powder group, especially, showed the lowest level among all experimental groups. Liver TBARS levels of experimental groups were also lower than that of control group and significant differences were observed in chamomile and green tea groups compared to control. Plasma XO activity of green tea group was significantly lower than control group. Liver XO activities of green tea and chamomile groups were significantly lower than control group. Erythrocyte SOD activity was not significantly different among all the groups. However, liver SOD activities of sage and green tea groups showed a significant increase as compared to control group. Plasma vitamin A level was not significantly different among all the groups. Plasma total carotenoids levels were found to be significantly higher in experimental groups as compared to control group. Plasma vitamin E level of chamomile group and vitamin C level of green tea group showed the highest level among all the groups. In conclusion, chamomile, sage, and green tea intakes had an effect on improving antioxidative capacity in 15-month-old rats. Effects of green tea and chamomile powders, especially, were higher than sage powder.

Estimated flavonoid intakes according to socioeconomic status of Korean adults based on the Korea National Health and Nutrition Examination Survey 2007~2012 (우리나라 성인의 사회경제적 수준에 따른 플라보노이드 섭취현황 : 2007~2012년 국민건강영양조사 자료를 이용하여)

  • Purpose: The purpose of this study was to estimate the dietary flavonoid intakes of Korean adults according to socioeconomic status. Methods: Using data from the 2007~2012 Korea National Health and Nutrition Examination Survey, a total of 31,112 subjects aged over 19 years were included in this study. We estimated individuals' daily intakes of total flavonoids and seven flavonoid subclasses, including flavonols, flavones, flavanones, flavan-3-ols, anthocyanins, proanthocyanidin, and isoflavones,by linking food consumption data with the flavonoids database for commonly consumed Korean foods. We compared intakes of flavonoids according to the levels of household income and education. Results: Average dietary flavonoid intakes of the study subjects were 321.8 mg/d in men and 308.3 mg/d in women. Daily flavonoid intakes were positively associated with household income level (p < 0.0001) and education level (p < 0.0001). The subjects in the highest household income and highest education level group (OR 0.37, 95% CI 0.30~0.45, p < 0.0001 in men, OR 0.50, 95% CI 0.41~0.60, p < 0.0001 in women) had a lower likelihood of having low total flavonoid intake (less than 25 percentile) compared to the lowest household income and lowest education level group. The food group that contributed to total flavonoid intake with the biggest difference between the lowest and highest groups for both household income level and education level was beverages. Conclusion: This study shows that socioeconomic status was positively associated with flavonoid intake in a representative Korean population. Further research is needed to analyze the association of flavonoid intake with health outcomes according to socioeconomic status such as household income and education level.

Total Phenolics Contents, Total Flavonoids Contents and Antioxidant Capacities of Commercially Available Korean Domestic and Foreign Intermediate Food Materials (국내외 시판 농산물 중간소재의 총페놀, 총플라보노이드, 총안토시아닌 함량 및 항산화 활성)

  • Commercial Korean domestic and foreign intermediate food materials (IFMs) of blueberry, Aronia, oat, sweet pumpkin, and ginger were purchased to compare their functional properties, including total phenolic content (TPC), total flavonoid content (TFC), total anthocyanin content (TAC), and antioxidant capacity. Each IFM was extracted using 80% (v/v) methanol for the analyses. The TPC (mg gallic acid equivalent/100 g dried sample) and TFC (mg catechin equivalent/100 g dried sample) levels in domestic blueberry (TPC, 2,916 ± 200; TFC, 1,327 ± 31) and sweet pumpkin (TPC, 476 ± 20; TFC, 23 ± 32) IFMs were significantly higher than those in the foreign IFMs. In the case of TAC (mg cyanidin-3-glucoside/100 g dried sample), the level in domestic blueberry IFM (949 ± 57) was significantly higher than that in the foreign product. Among the domestic IFMs, the antioxidant capacities (mg vitamin C equivalent/100 g dried sample) of blueberry, sweet pumpkin, and ginger were 7,057 ± 321, 311 ± 8, and 3,321 ± 109, respectively, being significantly higher than those of their foreign counterparts, based on the ABTS radical scavenging assay. In the DPPH radical scavenging assay, foreign Aronia (12,667 ± 437) and ginger (2,067 ± 99) IFMs showed significantly higher levels of free radical scavenging activity than did the domestic IFMs. These results provide basic information regarding the functional properties of Korean domestic IFMs, compared with their foreign counterparts.

Total Phenolics Levels and Antioxidant Properties in Methanol Extracts from Several Vietnamese Wild Plants

  • The aim of this study was to determine total phenolics (TP) content, total flavonoids (TF) level, and antioxidant activity of methanol extracts from leaf parts of 15 Vietnamese wild plants. TP content showed the highest amount in methanol extracts from Altenanthera sessilis ($409.7{\pm}1.4$ ppm), and followed by Eclipta prostrata ($183.6{\pm}3.8$ ppm) and Cassia tora ($159.3{\pm}5.7$ ppm). The highest TF level also was found in A. sessilis ($156.3{\pm}1.7$ ppm), followed by E. prostrata, and C. tora, showing similar tendency to TP. Methanol extracts of A. sessilis, E. prostrate and C. tora at 1000 ppm exhibited the highest DPPH radical scavenging activity by 94.5, 92.6 and 92.7%, respectively. The methanol extracts from A. sessilis showed the highest ABTS and nitrite scavenging activities by 97% and 92%, respectively. The highest correlation coefficient ($r^2$) was 0.9522 between TP and TF, and followed by 0.8919 between DPPH and ABTS activities. However, polyphenols and antioxidant activities showed low correlation coefficients, ranging from $r^2=0.4114$ to 0.4826. It was concluded that Vietnamese wild plants contain polyphenol compounds with antioxidant activities depending on plant species.
