인삼 모상근의 색소 생성 및 엽록체 발달에 미치는 광의 효과 (Effects of Light on the Pigment Production and Chloroplast Development of Ginseng Hairy Roots)

  • The effects of light on the pigment production and chloroplast development were examined on ginseng hairy roots cultured in 1/2MS liquid medium. The chlorophyll and carotenoid production were increased from 1,000 to 3,500 lux condition, but decreased drastically in 7,000 lux condition. The anthocyanin production was significantly increased with increment light intensity(1,000∼7,000 lux). The thylakoid membrane of chloroplast was proplastid in dark condition and it began to develop into thylakoid membrane in 1,000 lux condition and then intact thylakoid membrane was developed in 3,500 lux condition. However, the development of thylakoid membrane in 7,000 lux condition was inhibited comparing to 3,500 lux condition. The total chlorophyll production in blue light condition were high comparing to other wavelength and same as 40% of total chlorophyll on white light(3,500 lux) condition. The chlorophyll and carotenoid production by sucrose concentration were high in 3% sucrose condition and anthocyanin production was high in 4% condition. The production of chlorophyll and carotenoid by light periods was high when explants were cultured in dark condition for 1 week and then transferred to light condition for 4 weeks. Our results suggest that pigment production and chloroplast development could be accelerated by light Intensity of specific wavelength in cultures of ginseng hairy root.

고추와 가바쌀을 첨가한 기능성 마카롱 개발에 관한 연구 (Development of Macaronè with Korean Red Peppers (Capsicum annuum L.) and GABA rice and Evaluation of Physiological Characteristics)

  • The aims of this study were to develop $macaron{\grave{e}}$ with Korean red pepper (Capsicum annuum L.) and GABA (${\gamma}$-aminobutyric acid) rice powder and analyze their physico-chemical and physiological characteristics. Korean red pepper (Capsicum annuum L.) was added to $macaron{\grave{e}}$ at a weight percentage of 0, 2.5 and 4.5%. Color values (L-value, redness, and yellowness), total sugar content, total phenolics, total carotenoids, ABTS radical scavenging activity, textures, and sensory characteristics with varying various Korean red pepper were measured. As the ratio of the powders in the $macaron{\grave{e}}s$ increased, total phenolic content, total carotenoid content, and radical scavenging activity increased. There were significant differences in total phenolic and total carotenoid content of $macaron{\grave{e}}s$ (p<0.05). Sensory evaluation indicated significant differences (p<0.05) in color, sweetness, texture, and overall acceptability depending on the addition of Korean red pepper. KR30 $macaron{\grave{e}}$, showed higher overall acceptability, as compared to other $macaron{\grave{e}}$.

녹숙 고추의 추숙에 미치는 빛의 영향 (The Effect of Light on the Matured Hot Green Pepper Fruits during the After-Ripening Period)

  • This investigation was designed to determine the influence of light on the changes of capsaicin, carotenoid and sugar in hot green pepper fruits during the after-ripening period. The results were as follows; 1. In capsaicin content, the sample in light was increased about twice of that of the sample under the darkness. 2. In total carotenoid content, both samples were increased in the same trend. And so, it might be able to assume that carotenoid pigments were produced without the light. 3. In sugar content, the sample in light was more rapidly decreased than that of the sample in darkness.

고추의 건조방법(乾燥方法)에 따른 성분(成分) 변화(變化)에 관(關)한 연구(硏究) - 제 1보 Carotenoid, Capsaicin, Vitamin C의 변화(變化) - (A Study on the Influence of Drying Methods upon the Chemical Changes in Red Pepper)

  • This investigation was designed to determine the influence of drying methods on the chemical changes in red pepper. Three different drying methods were employed: 1) Sun-drying at $25{\sim}28^{\circ}C$ for 15 days, 2) Drying in oven at $60^{\circ}C$ for 49 hours, and 3) Drying in oven at $90^{\circ}C$ for 8 hours. Dried and ground peels were used for the analysis of carotenoid pigments, capsaicin, and vitamin C. The results were as follows; 1. The total carotenoid content of sun-dried sample was more significantly in creased about 4% than that of the fresh sample, but both samples dried in oven were decreased about 30%. Among carotenoid groups, diol-polyol group was chiefly increased, and both samples dried in oven were decreased in the same trend; $17{\sim}18%$ reduction was found in hydrocarbons and monols and $36{\sim}38%$ was in diol-polyols. 2. In Capsaicin content in red pepper, the sun-dried sample was increased about twice of that of the fresh sample and both samples dried in oven were also tended to increase. 3. High reduction of vitamin C content was found in each treatment; 76% for the sun-dried and 89% for the oven-dried samples, respectively.

염장미역의 감마선 조사에 따른 이화확적 특성 변화 (Changes in Physico-Chemical Properties of Salted Sea Mustard (Undaria Pinnatifida) by Gamma Irradiation)

  • 염장미역 제품의 염농도 감소와 저장 조건 개선을 위한 연구의 일환으로 감마선 조사가 염장미역의 이화학적 특성에 미치는 영향을 조사한 결과는 다음과 같다. 미역의 chlorophyll과 carotenoid 색소는 감선 조사에 의해 부분적으로 감소되었고 $10^{\circ}C$ 내외의 저장조건에서 6개원 저장시 비조사 시료와 2kGy 조사시료의 색소 잔존률은 서리 유의적인 차이가 없이 chlorophyll이 47~77%, carotenoid가 54~56%였다. 미역의 관능적 품질과 관련된 유기산과 휘발산의 함량은 염농도가 낮고 저장온도가 높을수록 증가하였고, 감마선 조사에 의한 미역의 조직 연화는 알긴산 추출과 관련이 있는 것으로 나타났다.

토마토 종류에 따른 카로티노이드 함량 비교와 다중분석법 개발 (Method Development and Analysis of Carotenoid Compositions in Various Tomatoes)

  • BACKGROUND: Purpose of this research is HPLC analysis method development of lycopene in tomato. And then, three components of carotenoid in four kinds of tomatoes (general tomato, cherry tomato, red and orange date tomato) were compared with each other. METHODS AND RESULTS: Lycopene in tomato was extracted with hexane likes other carotenoid components using 500 mg of dried powder sample. HPLC analysis conditions were column temperature ($40^{\circ}C$), detection wavelength (454 nm), flow rate (1.0 mL/min) and injection volume ($20.0{\mu}L$). Lycopene was analyzed by the gradient elution ($60{\rightarrow}100%$) of the mobile phase solvents A[water: methanol=25: 75 (v/v)] and B[ethyl acetate]. CONCLUSION: Three components of carotenoids (lutein, ${\beta}$-carotene, lycopene) were observed in tomatoes. The total carotenoid contents was the highest in red date tomato (662.0 mg/kg dry wt.) and the lowest in orange date tomato (111.3 mg/kg dry wt.). Lycopene contents in tomatoes was the highest percentage (93%) among all the carotenoids.

고광도와 질소 결핍이 Haematococcus pluvialis의 색소 생합성에 미치는 영향 (Influence of High Light and Nitrate Deprivation on the Carotenoid Biosynthesis in Haematococcus pluvialis)

  • H. pluviails는 고광도와 질소 결핍 배지 조건에서 ketocarotenoid의 일종인 astaxanthin을 다량 축적하는 녹조류이다. 스트레스가 없는 조건에서 키운 green cell과 astaxanthin이 합성된 red cell을 HPLC를 통해 비교해 본 결과 각 색소의 양이 변화하는 것을 볼 수 있었다. 여러 ester 형태의 astaxanthin이 생합성 되고, zeaxanthin이 늘어난 반면, lutein과 ${\beta}$-carotene은 감소하였다. 또한 total chlorophyll 양이 줄어드는 대신 total carotenoid의 양이 늘어남을 보였다. H. pluvilalis에서 찾아낸 astaxanthin 생합성 경로에 있는 carotenoid hydroxylase, phytoene desaturase, isopentenyl pyrophosphate isomerase, ${\beta}$-carotene ketolase 유전자는 음성대조군인 chloroplast chlorophyll a-b binding protein와는 달리cell이 성장하기 좋은 조건의 상태보다 astaxanthin을 생합성하기 위해 고광도의 스트레스를 받았을 때 더 높은 발현양상을 보이는 것을 확인할 수 있었다.

Temperature determines post-harvest quality of spinach leaves after gamma irradiation

  • The relative importance of radiation dose, storage time, and temperature in radiation processing of spinach (Spinacia oleracea L.), was evaluated in terms of the postharvest quality through a model study using leaf disks. Physiological activity and chlorophyll and carotenoid contents were measured to represent the postharvest quality (in terms of external appearance) of spinach, a leafy green vegetable. At $22^{\circ}C$ and $30^{\circ}C$, the higher gamma-radiation dose caused a greater decrease in the physiological activity, depending on the storage time of 4 d. However, this decrease was not significant at $4^{\circ}C$ and $15^{\circ}C$. Total chlorophyll and carotenoid contents were substantially decreased by 3 kGy at $15^{\circ}C$, and dose-dependently by 0.5-3 kGy at $22^{\circ}C$. In contrast, the proportion of lutein in total carotenoid was significantly increased in the 2-3-kGy samples only at $22^{\circ}C$, while that of ${\beta}$-carotene was slightly decreased, indicating progression of leaf aging. These data suggest that the unfavorable effect of gamma irradiation on the postharvest quality of spinach could be avoided or controlled by the storage time or temperature rather than the radiation dose. The current study could be available to improve radiation processing of other leafy green vegetables.

신미종(辛未種) 고추의 추숙(追熟)에 관(關)한 생리화학적(生理化學的) 연구(硏究) -제7보 주요성분(主要成分)에 미치는 Ethephon의 효과(?果)- (Physio-Chemical Studies on the After-Ripeng of Hot Pepper Fruit -Part VII. Effects of Ethephon on the Major Compoments-)

  • 재래신미종(在來辛味種) 녹숙(綠熟) 고추의 추숙(追熟)에 따른 2-chloro ethyl phosphonic acid capsaicin생합성(生合成)의 sub strate인 phenylalanine의 처리효과(處理效果)를 검토(檢討)코저 color score, 고추 공포부(空胞部)의 $O_2$$CO_2$, 과피부(果皮部)의 total carotenoid, ${\beta}-carotene$, 전당(全糖), 유리환원당(遊離還元糖) 및 capsaicin의 함량변화(含量變化)를 측정(測定)한 결과(結果)는 다음과 같다. 1. ethephon 500ppm의 처리(處理)는 control구(區)에 비(比)하여 현저한 착색촉진(着色促進)의 효과(效果)를 볼 수 있었으며 특히 약(約) 20%의 적변불가(赤變不可)의 고추를 적변가능(赤變可能)케 하였다. 2. 추숙기간중(追熟期間中) 고추 공포부(空胞部)의 gas조성(組成)은 control 구(區)에서는 $O_2$의 함량(含量)이 증가(增加)하는 시기(時期)에 color score가 증가(增加)하였으며, 반대(反對)로 ethephon 처리구(處理區)에서는 color score가 급증하는 시기(時期)에 $O_2$의 함량(含量)이 저하(低下)하였다. 3. 추숙(追熟)에 따른 전당(全糖) 및 환원당(還元糖)의 변화(變化)는 처리구(處理區) 다같이 현저히 감소(減少)하였고, total carotenoid 함량(含量)은 ethephon 처리(處理)로서 약(約) 50%가 증가(增加)하였으며, ${\beta}-carotene$은 약(約) 2배(培)의 증가를 보였다. 4. 처리별(處理別) capsaicin의 함량변화(含量變化)는 ethephon처리(處理)로서 약(約) 20%의 증가(增加)를 볼 수 있었고, substrate인 phenylalanine의 단독 처리(處理)는 그 효과(效果)가 나타나지 않았으나, ethephon과 동시(同時) 처리(處理)함으로써 다소(多少)의 효과(效果)를 인정(認定)할 수 있었다.

사료내 paprika 및 spirulina 첨가가 쉬리(Coreoleuciscus splendidus) 표피 색택에 미치는 영향 (Effect of Dietary Supplementation of Paprika and Spirulina on Pigmentation of Swiri Coreoleuciscus splendidus)

  • 본 연구는 쉬리(Coreoleuciscus splendidus)를 대상으로 사료내 색소원으로서 paprika 및 spirulina 첨가와 원료 각각의 첨가량을 달리하여 쉬리의 생존율과 색택개선에 미치는 영향을 조사하였다. 실험어는 평균체중 약 4.4 g인 쉬리를 사용하였으며, 총 8개(Control, P1, P2, P3, S1, S2, S3 및 P2+S2)의 실험 사료를 설계하였다. 사료 P1, P2 및 P3는 paprika를 각각 2%, 5% 및 10%를 첨가하였으며, 사료 S1, S2 및 S3는 spirulina를 각각 5%, 10% 및 20%를 첨가하여 제조하였다. 또한 P2+S2 사료는 paprika 5%와 spirulina 10%를 혼합하여 제조하였다. 실험종료 후, 쉬리의 생존율은 spirulina가 첨가된 모든 실험구인 S1, S2, S3 및 P2+S2에서 100%로 대조구(80%) 및 paprika 첨가구($73{\sim}93%$)보다 높게 나타났다. 쉬리의 표피 및 지느러미의 총 carotenoids 함량은 색소원이 첨가되지 않은 대조구에서 1.58 mg/100 g으로 가장 낮았으며, spirulina 10% 첨가구인 S2 실험구에서 2.23 mg/100 g으로 가장 높았다. Paprika와 spirulina가 혼합되어 첨가된 실험구인 P2+S2는 1.61 mg/100 g으로 paprika와 spirulina가 각각 첨가된 P2 (2.14 mg/100 g) 및 S2 (2.23 mg/100 g)보다 낮은 총 carotenoids 함량을 보였다. 쉬리의 표피를 육안으로 관찰한 결과 색소원이 첨가된 모든 실험구에서 대조구에 비하여 체색이 더욱 선명화 됨을 관찰 할 수 있었다. 이상의 결과를 고려할 때, 쉬리의 사료내 paprika 첨가시 5%, spirulina 첨가시 10%가 쉬리의 색택개선에 좋을 것으로 판단된다.
