달걀.메추리알.오리알의 Retinol Equivalent 측정과 Carotenoids 분석 (Determination of Retinol Equivalent and Identification of Carotenoids in Hen, Quail and Duck Eggs)

  • 시중에서 판매되는 달걀, 메추리알, 오리알의 총 carotenoid, provitamin A carotenoids, retinol함량을 측정하여 Retinol Equivalent를 산출하고, 이들 난황의 carotenoid 조성을 비교분석하였다. 각 난황에서 단위 g당 총 carotenoid함량은 달걀>메추리알>오리알의 순서로, retinol은 메추리알>오리알>달걀의 순서로, Retinol, Equivalent는 메추리알>오리알>달걀의 순서로 높았다. 간 난황의 carotenoid 분석결과, carotenoid 조성은 거의 비슷했으나, 달걀과 메추리알의 경우 lutein과 zeaxanthin이 총 carotenoid의 75% 정도였고, 오리알의 경우는 60% 정도였다. 거의 대부분을 차지하고 있는 carotenoids임을 보였다.

메기과에 속하는 메기와 미유기의 Carotenoid 색소 성분의 비교 (Comparision of Carotenoid Pigments in Catfish, Parasilurus asotus and Slender catfish, Parasilurus microdorsalis in the Family Siluridae)

  • 미이용자원인 어류 carotenoid에 관한 비교 생화학적 연구의 일환으로서, 메기과의 자연산 및 양식산 메기와 한국 특산의 담수어류인 미유기의 표피 carotenoid를 TLC, column chromatography 및 HPLC로 분리하고 NaBH$_4$에 의한 환원반응, I$_2$에 의한 이성화반응 및 UV-visible spectrophotometer로 비교하고 IR 및 Mass spectrum을 측정하여 carotenoid를 동정한 결과는 다음과 같다. 자연산 메기 표피의 총 carotenoid 함량은 0.27 mg($\pm$0.7)%였으며 carotenoid의 조성은 zeaxanthin 25.8%, diatoxanin 9.7% 및 cynthiaxanthin 9.1%로서 주성분을 이루고 있었으며 그 외 isocryptoxanthin 6.5%, $\beta$-carotene 5.1%, $\beta$-cryptoxanthin 4.3%, lutein 3.4% 및 $\alpha$-cryptoxantin 2.4%의 순으로 함유하고 있었다. 양식산 메기 표피의 총 carotenoid 함량은 0.07 mg($\pm$0.03)%로서 자연산 메기에 비해 다소 낮았으며, carotenoid의 조성은 lutein 48.5%, zeaxanin 13.9% 및 isocryptoxanin 13.3%로서 주성분을 이루고, 그 외 $\beta$-carotene 9.5%, $\beta$-cryptoxantin 1.4% 및 $\alpha$-cryptoxanthin 4.6%의 순으로 함유하고 있었다. 한편 미유기 표피의 총 carotenoid 함량은 0.75 mg($\pm$0.25)%로서 메기과의 다른 어종에 비해 함량이 높았으며, carotenoid 조성은 zeaxanthin 24.5%, 7'8'-dihydro-$\beta$ -carotene-4-ol 24.1%, 7'8'-dihydro-$\beta$-carotene 17.9% 및 7'8'-dihydro-$\beta$-carotene-3-ol 10.8%로서 주성분을 이루고 있었으며 그 외 diatoxanthin 8.7%, cynthiaxanthin 6,7% 및 lutein 5.0%의 순으로 함유하고 있었다. 이러한 결과는 parasiloxanthin과 7',8'-dihyo-parasiloxanthin을 주성분으로 함유하는 biwa호수 메기, P. asotus 표피 carotenoid 조성과 비교하여, 미유기에서는 7'8'-dihydro-$\beta$-carotene의 구조를 가진 carotenoid가 존재하였으나 자연산 및 양식산 메기에서는 발견되지 않아 특이하였다.

Daucus carota 현탁 배양에서 배지 조성 및 세포 응집 크기가 Carotenoid 생합성에 미치는 영향 (Effects of Nutrients and Cell Aggregate Size on the Biosynthesis of Carotenoid in Daucus carota Suspension Culture)

  • The effects of nutrients and cell aggregate size on the cell growth of D. carota and the biosynthesi of carotenoid were investigated. Highest carotenoid content was obtained with sucrose as a carbon source and the equal ratio of ammonium to nitrate. High phosphate concentration stimulated the carotenoid biosynthesis in D. corota. 2,4-D inhibited the cell growth but stimulated the specific carotenoid content at high concentration. By modifying the medium composition based on these findings, three times higher specific carotenoid content and 2.5 times higher total carotenoid content were obtained as compared with the results obtained with basic MS media. Biosynthesis of carotenoid was found to be affected by cell aggregate size; high carotenoid production was obtained from the large aggregated cells resulted from high sucrose concentration.

저온처리가 케일(Brassica oleracea)잎 내 Carotenoid에 미치는 영향 (Effect of Cold Stress on Carotenoids in Kale Leaves (Brassica oleracea))

  • BACKGROUND: Kale (Brassica oleracea) biosynthesizes various phytochemicals including glucosinolates, flavonoids, and carotenoids. Phytochemicals of plants are influenced by light, temperature, carbon dioxide, and growing conditions. Specifically, carotenoids are affected by temperature, light, and oxygen. The aim of this study was to investigate the effect of cold stress (day/night: $25^{\circ}C/20^{\circ}C$, $20^{\circ}C/15^{\circ}C$, $15^{\circ}C/10^{\circ}C$) on carotenoids in kale leaves. METHODS AND RESULTS: Kale was grown in pots for up to 50 days after sowing (DAS) in a greenhouse. For cold acclimation experiments, kale grown in growth chambers for 3 days and was subjected to low temperature for 4 days. The conditions maintained in the growth chambers were as follows: photoperiod, 12/12 h (day/night); light, fluorescent; and relative humidity, 60%. Carotenoid (lutein, ${\alpha}-carotene$, zeaxanthin, ${\beta}-carotene$) contents were analyzed by high-performance liquid chromatography (HPLC). The total carotenoid content gradually increased during cold acclimation for 3 days. When kale was subjected to cold stress, the total carotenoid content was high at $25^{\circ}C/20^{\circ}C$ treatment, but low at $15^{\circ}C/10^{\circ}C$ treatment. The total carotenoid content of kale leaves continuously grown in greenhouse decreased from 50 to 57 DAS (1,418 and 1,160 mgkg-1 dry wt., respectively). The lutein, ${\alpha}-carotene$, and ${\beta}-carotene$ contents were very low and the zeaxanthin contents were very high at $15^{\circ}C/10^{\circ}C$ treatment. When kale was subjected to cold stress, the ratio of individual to the total carotenoid contents of kale leaves was 4553% for -carotene and 210% for zeaxanthin. CONCLUSION: The ${\beta}-carotene$ and zeaxanthin contents in kale leaves indicate their sensitiveness toward cold stress.

  • 신미종(辛味種)고추의 추숙(追熟)에 따른 carotenoid 색소(色素)와 chlorophyll의 변화(變化)를 측정(測定) 고찰(考察)한 결과(結果)는 다음과 같다. 1. 신미종(辛味種) 고추의 carotenoid 색소(色素)를 column chromatography로 6 fraction으로 분리(分離)한 다음 각(各) fraction을 다시 TLC에 의하여 54종(種)을 분리(分離)하고 그 가운데서 30종(種)을 동정(同定)하였다. 2. carotenoid색소(色素) 총량(總量)은 cli. stage에서 급증(急增)하였고 group별(別)로 보니 diol가 가장 함량(含量)이 높고 추숙(追熟)에 따른 함량증가율(含量增加率)이 가장 높았다. 3. carotenoid 색소(色素) 개별당(個別當) 함량(含量) 변화(變化)에서 주목(注目)되는 것은 phytoene이 post cli. stage.에서 감소(減少)하고 고추의 중요(重要)한 carotenoid 색소(色素)인 ${\beta}$-carotene, capsanthin, capsorubin, violaxanthin의 함량(含量)이 크게 증가(增加) 하였다. 4. chlorophyll은 a, b, total 함량(含量)이 다같이 추숙(追熟)에 따라 감소(減少)하고 post-cli. stage에서 소실(消失)하였다.

잉어아과에 속하는 묵납자루와 각시붕어의 Carotenoid 색소성분의 비교 (Comparison of Carotenoid Pigments in Korean Bittering, Cheilognathus signifer and Bride Bittering, Rhodeus ukekii in the Subfamily Cyprinidae)

  • Differences in carotenoid composition in the integument of Korean bittering, Cheilognathus signifer and bride bittering, Rhodeus ukekii which are Korean native fresh water fish were compared. Total ca rotenoid contents in the integument of wild Korean bittering was 2.11mg% and composed of 42.6% zeaxanthin, 12.1% diatoxanthin and 12.1% lutein epoxide which are predominant carotenoids and 10.3% cynthiaxanthin, 8.3% zeaxanthin epoxide, 6.4% lutein and 1.5% cryptoxanthin which are minor carotenoids. Total ca rotenoid contents in the integument of wild bride bittering was 4.99mg% during a spawning period but after the spawning period it was decreased to 4.17mg% and carotenoid composition of bride bittering during the spawning period was 46.7% zeaxanthin, 26.5% diatoxanthin and 12.3% lutein which are predominant carotenoids, and 6.2% zeaxanthin epoxide, 3.1% cynthiaxanthin, 2.9% cryptoxanthin and 0.7% canthax anthin which are minor carotenoids. These results indicated that the carotenoid composition of bride bittering during spawning period was very similar to that of Korean bittering and carotenoid composition of bride bittering after the spawning period was 30.5% diatoxanthin, 21.5% cynthiaxanthin and 16.8% zeaxanthin which are predominant carotenoids and 14.0% cryptoxanthin, 11.3% lutein and 3.4% can thaxanthin which are minor carotenoids, indicating that after the spawning period, the content of zeaxanthin was decreased while that of cryptoxanthin and cynthiaxanthin was increased as compared to that of the spawning period. Total carotenoid contents in Korean bittering and bride bittering was relatively higher than that in other species of cyprinidae whereas composition of the carotenoid was similar.

한국 여자 노인의 Carotenoid Bioavailability에 미치는 요인 조사 (Factors Effecting the Bioavailability of Carotenoid in Elderly Korean Women)

  • Carotenoid-rich foods focus one's attention on the prevention age-related diseases. This study was conducted to investigate the carotenoid status and look into the factors that affect the bioavailability of carotenoid in 121 elderly nonsmoking Korean women. Carotenoids and lipids in plasma, and nutrient intakes including carotenoid were studied. The mean plasma total-cholesterol, HDL-cholesterol, LDL-cholesterol and triacylglycerol concentrations were 220.0 mg/dl 49.5mg/dl , 139.2mg/dl and 157.4mg/dl , respectively. Significantly positive correlations were found between the plasma lutein + zeaxanthin, lycopene and $\beta$-carotene concentrations and the intake of fruits (r=0.17, r=0.20, r=0.19). However, significantly negative correlations were found between the plasma zeaxanthin, and $\beta$-carotene concentrations that adjusted for carotenoid intakes and intakes of vegetables (r=-0.21, r=-0.19), and between plasma lutein+zeaxanthin, lycopene and $\beta$-carotene concentrations that adjusted for carotenoid intakes and intakes of fruits (r= -0.21, r=-0.18, r=-0.24). After the adjustment for plasma lipids, there was no correlation between the plasma carotenoid concentrations and the carotenoid-rich foods. However, after adjustment for fiber intake, significantly strong positive correlations were found between the plasma carotenoid concentrations and carotenoid-rich foods. The plasma levels of carotenoid biomarkers (plasma carotenoid concentrations adjusted for dietary fiber intakes) decreased with age, and the plasma levels of lycopene biomarkers (plasma lycopene concentrations adjusted for dietary fiber intakes) increased with regular exercise. However alcohol drinking had no impact. These results suggested that age, physical activity, and dietary fiber intake affected the bioavailability of carotenoid. Therefore, when the elderly have carotenoid-rich foods, they should consider ways of increasing the bioavailability of carotenoid through cooking methods and Physical activity.

제주산 감귤 품종별 carotenoid, 색도, UV 스펙트럼, 유기산 및 유리당 함량 (Carotenoid, Color value, UV Spectrum, Organic Acid and Free Sugar Contents of Citrus Varieties Produced in Cheju)

  • Total carotenoid contents, color values and spectrum characterization in the range of UV of fruit juice and organic acid and free sugar contents determined by HPLC were investigated on 10 varieties of Cheju citrus fruits. Carotenoid contents of juice were 0.47-9.20$\mu\textrm{g}$/ml which showed higher with Dangyooja while showed lowest content with Meiwa Kumquat. Lightness (L) of juice was highest in Meiwa Kumquat, and in the order of Sankyool, Navel orange and Hungjin. Redness(a) and yellowness(b) were in the order of Dangyooja, Natsudaidai, Sambokam and Kinkoji. The maximum absorption wavelengths if citrus Juices were variable at 269.5-285.5nm according to varieties and it was observed that were changed by mixing with Juices of other varieties. Major organic acids of citrus juice were citric and malic acid, and citric acid was 64.4-95.1% of total organic acids. Ascorbic acid was high in Navel orange, Sudachi and Dangyooja, and low in Meiwa Kumquat and Hungjin. Major free sugars were sucrose, glucose and fructose. Sucrose was 49.2-75.2% of total free sugars, and high in Sankyool and Meiwa Kumquat. Glucose and fructose were high in Meiwa Kumquat and Navel Orange.

구굴무치과에 속하는 동사리와 얼룩동사리의 Carotenoid 색소성분의 비교 (Comparison of Carotenoid Pigments on Korean Dark Sleeper, Odontobutis platycephala and Dark Sleeper, Odontobutis odontobutis interrupta in the Family Eleotridae)

  • This study was performed as a part of comparative biochemical studies of carotenoid pigment for the fresh water fish. Carotenoids in integument of Korean dark sleeper, Odontobutis platycephala, and dark sleeper, Odontobutis odontobutis interrupta, which are all the Korean native fresh water fish, were separated by thin layer chromatography, column chromatography and HPLC. The separated carotenoid were then reduced and isomerized by NaBH4 and I2 respectively to investigate UV-Vis spectrophotometeric patterns and chracterized by IR, 1H-NMR and Mass spectrum. The content of total carotenoids in the integument of Korean dark sleeper was 3.01mg% in April, but it was increased to 3.74mg% in September at the near of spawning period. The carotenoid isolated in April consisted of $\beta$-carotene(25.6%), lutein(18.5%) and zeaxanthin(12.0%) as major carotenoids and also contained isocryptoxanthin, diatoxanthin, tunaxanthin, cynthiaxanthin, canthaxanthin and $\alpha$-cryptoxanthin as minor carotenoids. Similarly, in September the carotenoid consisted of $\beta$-carotene(16.5%), zeaxanthin(13.7%) and cynthiaxanthin(13.6%) as major carotenoids and also contained lutein, isocryptoxanthin, tunaxanthin, $\alpha$-cryptoxanthin, diatoxanthin and canthaxanthin as minor carotenoids. At the near of spawning period, the content of cynthiaxanthin and $\alpha$-cryptoxanthin were increased. The content of total carotenoids in the integument of spawning period. T도 carotenoid isolated in April and September consisted of $\beta$-carotene(24.9%, 27.5%), zeaxanthin(14.4%, 20.9%) and lutein(12.6%, 11.4%) as major carotenoids and also contained cynthiaxanthin, tunaxanthin, diatoxanthin, isocryptoxanthin, $\alpha$-cryp-toxanthin and canthaxanthin as minor carotenoids. At the near of spawning period, the content of zeaxanthin was increased, indicating that the carotenoid composition were dependent upon their living conditions and their integument colors. Both Korean dark sleeper and dark sleeper contained high amount of cynthiaxanthin and diatoxanthin which are found as rare carotenoids in the other of fresh water fish. It is interes that they also contained tunaxanthin which is a specific carotenoid in marine fishes.

개불의 carotenoid 색소성분 (Carotenoid Pigments in Echiurid, Urechis unicinctus)

  • 어류 이외의 해양생물의 carotenoid 조성에 관한 비교 생화학적 연구의 일환으로 환형동물문의 개불의 carotenoid 색소성분을 Thin layer chromatography, Column chromatography 및 HPLC로 분리한 후, UV spectrometer, IR spectrometer 그리고 NMR 및 Mass spectrometer를 이용하여 분석, 비교, 검토한 결과는 다음과 같다. 개불의 총 carotenoid 함량이 3월에 1.19mg/100g 4월에 0.98mg/100g 그리고 5월에는 0.84mg/100g로서 서식 수온이 상승할수록 총 carotenoid의 함량이 감소했다. 채취시기에 따른 개불의 carotenoid 조성의 함량비의 변화를 보면, 3월에는 diatoxanthin monoester $16.3\%$, $\beta$-carotene $14.8\%$ 및 cynthiaxanthin monoester $12.6\%$가 주성분을 이루고 그 외 anthiaxanthin diester $8.4\%$, zeaxanthin monoester $8.2\%$%, diatoxanthin diester $7.3\%$, astaxanthin $4.2\%$, diatoxanthin $2.9\%$, triol $2.4\%$, anthiaxanthin $2.3\%$, isocryptoxanthin $1.7\%$, zeaxanthin diester $1.5\%$, zeaxanthin $0.8\%$ 및 lutein $0.5\%$의 순으로 함유하였으며, 4월에는 diatoxanthin monoester $21.9\%$, anthiaxanthin monoester $17.2\%$$\beta$-carotene $16.6\%$가 주성분을 이루고 그 외 zeaxanthin monoester $10.9\%$, cynthiaxanthin diester $5.6\%$, diatoxanthin diester $4.9\%$, astaxanthin $3.1\%$, triol $2.4\%$, diatoxanthin $2.3\%$, isocryptoxanthin $1.7\%$, lutein $1.5\%$, zeaxanthin $1.1\%$, cynthiaxanthin $1.0\%$ 및 zeaxanthin diester $1.0\%$의 순으로 함유하였고 그리고 5월에는 diaoxanthin monoester $25.3\%$, anthiaxanthin monoester $19.7\%$, $\beta$-carotene $13.0\%$ 및 zeaxanthin monoester $12.6\%$가 주성분을 이루고 그 외 cynthiaxanthin diester $5.8\%$, diatoxanthin $5.1\%$, astaxanthin $3.0\%$, triol $2.4\%$, diatoxanthin $2.2\%$, isocryptoxanthin $1.3\%$, zeaxanthin $1.2\%$, zeaxanthin diester $1.1\%$, lutein $1.0\%$ 및 cynthiaxanthin $0.9\%$의 순으로 함유하여 개불의 총 carotenoid에는 monoester형 carotenoid의 함량비가 $37.1\~57.6\%$ 및 diester형 carotenoids의 함량비가 $11.5\~17.2\%$로서 주성분을 이룬다. 한편, 개불에서 총 cantenoid 함량이 감소함에 따라 또한 해수의 수온이 상승함에 따라 zeaxanthin monoester, diatoxanthin monoester 및 cynthiaxanthin monoester의 함량비는 증가하는 반면, zeaxanthin diester, diatoxanthin diester 및 anthiaxanthin diester의 함량 비는 감소하여 특이하였다.
